Glioblastoma (GBM) is the most common, malignant adult primary tumor with dismal patient survival, yet the molecular determinants of patient survival are poorly characterized. Global methylation profile of GBM samples (our cohort; n = 44) using high-resolution methylation microarrays was carried out. Cox regression analysis identified a 9-gene methylation signature that predicted survival in GBM patients. A risk-score derived from methylation signature predicted survival in univariate analysis in our and The Cancer Genome Atlas (TCGA) cohort. Multivariate analysis identified methylation risk score as an independent survival predictor in TCGA cohort. Methylation risk score stratified the patients into low-risk and high-risk groups with significant survival difference. Network analysis revealed an activated NF-kappa B pathway association with high-risk group. NF-kappa B inhibition reversed glioma chemoresistance, and RNA interference studies identified interleukin-6 and intercellular adhesion molecule-1 as key NF-kappa B targets in imparting chemoresistance. Promoter hypermethylation of neuronal pentraxin II (NPTX2), a risky methylated gene, was confirmed by bisulfite sequencing in GBMs. GBMs and glioma cell lines had low levels of NPTX2 transcripts, which could be reversed upon methylation inhibitor treatment. NPTX2 overexpression induced apoptosis, inhibited proliferation and anchorage-independent growth, and rendered glioma cells chemosensitive. Furthermore, NPTX2 repressed NF-kappa B activity by inhibiting AKT through a p53-PTEN-dependent pathway, thus explaining the hypermethylation and downregulation of NPTX2 in NF-kappa B-activated high-risk GBMs. Taken together, a 9-gene methylation signature was identified as an independent GBM prognosticator and could be used for GBM risk stratification. Prosurvival NF-kappa B pathway activation characterized high-risk patients with poor prognosis, indicating it to be a therapeutic target. (C) 2013 AACR.

Bent-core mesogens are an important class of thermotropic liquid crystals as they exhibit unusual properties as well as morphologies distinctly different from rodlike mesogens. Two bent-core mesogens with differing center rings namely benzene and thiophene are considered and investigated using high-resolution oriented solid state C-13 NMR method in their liquid crystalline phases. The mesogens exhibit different phase sequences with the benzene-based mesogen showing a B-1 phase, while the one based on thiophene showing nematic and smectic C phases. The 2-dimensional separated local field (2D-SLF) NMR method was used to obtain the C-13-H-1 dipolar couplings of carbons in the center ring as well as in the side-wing phenyl rings. Couplings, characteristic of the type of the center ring, that also provide orientational information on the molecule in the magnetic field were observed. Together with the dipolar couplings of the side-wing phenyl ring carbons from which the local order parameters of the different subunits of the core could be extracted, the bent angle of the mesogenic molecule could be obtained. Accordingly, for the benzene mesogen in its B-1 phase at 145 degrees C, the center ring methine C-13-H-1 dipolar couplings were found to be significantly larger (9.5-10.2 kHz) compared to those of the side-wing rings (1.6-2.1 kHz). From the local order parameter values of the center (0.68) as well as the side-wing rings (0.50), a bent-angle of 130.3 degrees for this mesogen was obtained. Interestingly, for the thiophene mesogen in its smectic C phase at 210 degrees C, the C-13-H-1 dipolar coupling of the center ring methine carbon (2.11 kHz) is smaller than those of the side-wing phenyl ring carbons (2.75-3.00 kHz) which is a consequence of the different structures of the thiophene and the benzene rings. These values correspond to local order parameters of 0.85 for the center thiophene ring and 0.76 for the first side-wing phenyl ring and a bent-angle of 149.2 degrees. Thus, the significant differences in the dipolar couplings and the order parameter values between different parts in the rigid core of the mesogens are a direct consequence of the nature of the center ring and the bent structure of the molecule. The present investigation thus highlights the ability of the C-13 2D-SLF technique to provide the geometry of the bent-core mesogens in a straightforward manner through the measurement of the C-13-H-1 dipolar couplings.

Molecules in their liquid crystalline phase undergo rotational motion about the long axis of the molecule and the shape adopted by the rotating molecule plays an important role in influencing the mesophase morphology. In this context, obtaining the topology and the relative orientation of the different sub-units are important steps. For studying the liquid crystalline phase, C-13 NMR spectroscopy is a convenient method and for certain specifically designed nematogens, 2-dimensional separated local field (2D-SLF) NMR spectroscopy provides a particularly simple and straightforward means of arriving at the molecular topology. We demonstrate this approach on two three ring based nematogens designed with a phenyl or a thiophene ring at one of the termini. From the C-13-H-1 dipolar couplings of the terminal carbon obtained using the 2D-SLF NMR technique, the order parameter of the local symmetry axis of the terminal phenyl ring as well as of the long molecular axis could be easily estimated. For the thiophene nematogen, the lack of symmetry of the thiophene moiety necessitates some additional computational steps. The results indicate that the thiophene unit has its local ordering axis oriented away from the long molecular axis by a small angle, consistent with a bent structure expected in view of the thiophene geometry. The experiment also demonstrates the ability of 2D-SLF NMR to provide high resolution spectra by separation of several overlapped resonances in terms of their C-13-H-1 dipolar couplings. The results are consistent with a rod-like topology of the core of the investigated mesogens. The investigation demonstrates the potential of 2D-SLF NMR C-13 spectroscopy for obtaining atomistic level information and its utility for topological studies of different mesogens.

Se evaluaron cinco dosis de fertilizantes y tres densidades de siembra para producir semilla de tomate, en vista de suprimir mayor volumen de alimento y obtener semilla de calidad. El ensayo se estableció en el Valle de Sébaco, periodo comprendido de noviembre 2003 a marzo 2004, el Valle se situa en una planicie con precipitaciones anuales de 800 mm y temperaturas entre 22°C y 35°C, los suelos son arcillosos, drenados con un contenido alto de M.O. el diseño utilizado fue un (B.C.A) evaluando variables de crecimiento, rendimiento agronómico y de la semilla calidad I, la germinación de la semilla producida y se realizo un análisis económico para determinar la rentabilidad de los tratamientos. Los resultados evidencian que la dosis 517.2 kg ha-1 de NPK obtiene las mayores alturas con 69.91 cm y con un diámetro de 14.40 mm. La mejor densidad fue 33,333 plantas ha-1 con 67.58 cm de altura de planta. En el número de frutos la mejor combinación fue 387.9 kgha-1 * 33,333 plantas ha-1 con 350,209 frutos ha-1. En el peso kg ha-1 la dosis 387.9 kg ha-1 obtiene 36,948 kg ha-1 y la mejor densidad 33,333 plantas ha-1 con 37,809 kg ha1. El rendimiento de la semilla la mejor combinación fue 387.9 kg ha-1 * 33,333 plantas ha-1 con 151.9 kg ha -1 y el mejor porcentaje de germinación se obtuvo con 258.6 kg ha-1 con 88.6% y con densidad de 33,333 plantas ha-1 con 88.1%. La mayor relación valor costo lo obtiene la combinación 387.9 kg ha-1 * 33,333 plantas ha-1 con $ 75.23 por cada dólar invertido.

An analytical fluid model for JxB heating during the normal incidence by a short ultraintense linearly polarized laser on a solid-density plasma is proposed. The steepening of an originally smooth electron density profile as the electrons are pushed inward by the laser is included self-consistently. It is shown that the JxB heating includes two distinct coupling processes depending on the initial laser and plasma conditions: for a moderate intensity (a <= 1), the ponderomotive force of the laser light can drive a large plasma wave at the point n(e)=4 gamma(0)n(c) resonantly. When this plasma wave is damped, the energy is transferred to the plasma. At higher intensity, the electron density is steepened to a high level by the time-independent ponderomotive force, n(e)> 4 gamma(0)n(c), so that no 2 omega resonance will occur, but the longitudinal component of the oscillating ponderomotive field can lead to an absorption mechanism similar to "vacuum heating." The ritterazine and cephalostatin natural products have biological activities and structures that are interesting to synthetic organic chemists. These products have been found to exhibit significant cytotoxicity against P388 murine leukemia cells, and therefore have the potential to be used as anticancer drugs. The ritterazines and cephalostatins are steroidal dimers joined by a central pyrazine ring. Given that the steroid halves are unsymmetrical and highly oxygenated, there are several challenges in synthesizing these compounds in an organic laboratory.
Ritterazine B is the most potent derivative in the ritterazine family. Its biological activity is comparable to drugs that are being used to treat cancer today. For this reason, and the fact that there are no reported syntheses of ritterazine B to date, our lab set out to synthesize this natural product.
Herein, efforts toward the synthesis of the western fragment of ritterazine B are described. Two different routes are explored to access a common intermediate. An alkyne conjugate addition reaction was initially investigated due to the success of this key reaction in the synthesis of the eastern fragment. However, it has been found that a propargylation reaction has greater reactivity and yields, and has the potential to reduce the step count of the synthesis of the western fragment of ritterazine B.

Tryptophan and unnatural tryptophan derivatives are important building blocks for the total synthesis of natural products, as well as the development of new drugs, biological probes, and chiral small molecule catalysts. This thesis describes various catalytic methods for the preparation of tryptophan derivatives as well as their functionalization and use in natural product total synthesis.
Herein, the tandem Friedel–Crafts conjugate addition/asymmetric protonation reaction between 2-substituted indoles and methyl 2-acetamidoacrylate to provide enantioenriched trytophans is reported. This method inspired further work in the area of transition metal catalyzed arylation reactions. We report the development of the coppercatalyzed arylation of tryptamine and tryptophan derivatives. The utility of these transformations is highlighted in the five-step syntheses of the natural products (+)-naseseazine A and B. Further work on the development of a mild and general Larock indolization protocol to access unnatural tryptophans is also discussed.

O Orthognathic Quality of Life Questionnaire (OQLQ) foi desenvolvido em 2000 e validado em 2002, com o objetivo de analisar os impactos e benefícios do tratamento orto-cirúrgico na qualidade de vida dos pacientes. O objetivo deste trabalho foi avaliar a validade de construto e confiabilidade da versão brasileira deste instrumento, B-OQLQ, para verificar se, no processo de adaptação transcultural, as propriedades psicométricas do questionário original foram mantidas. Para tal, foi realizado um estudo seccional com uma amostra composta por cento e um pacientes com necessidade de tratamento ortodôntico-cirúrgico no Hospital Universitário Pedro Ernesto (HUPE) e na Faculdade de Odontologia da Universidade do Estado do Rio de Janeiro (UERJ), utilizando o B-OQLQ na forma de autopreenchimento. A média de idade dos pacientes foi de 26,51 (dp= 9,25), sendo a maioria do sexo feminino (58,42%; n=59) e maior que 21 anos (n=68, 67,33%). Quanto ao tipo de cirurgia, 42,57% (n=43) da amostra necessitavam fazer procedimento cirúrgico que envolveria as duas bases ósseas, 16,83% (n=17) necessitavam operar apenas uma das bases ósseas e para 40,59% (n=41) dos pacientes ainda não era possível definir qual o tipo de cirurgia seria necessária. Uma ortodontista treinada examinou os dentes dos pacientes para registrar o Índice de Dentes Cariados, Perdidos e Obturados (CPOD). A validade de construto foi acessada através do teste de correlação de Spearman entre as pontuações do B-OQLQ e do questionário Oral Health Impact Profile (OHIP-14) e das pontuações do B-OQLQ com as pontuações obtidas através de indicadores subjetivos e objetivos de saúde. A confiabilidade foi acessada em termos de consistência interna e estabilidade (teste-reteste), utilizando-se o alfa de Cronbach e o Coeficiente de Correlação Intraclasse (CCI), respectivamente. Os escores do B-OQLQ se correlacionaram significativamente com: o escore total do OHIP-14 (rs=0,70, p<0,001), a saúde bucal percebida (rs=-0,24, p=0,02), a qualidade de vida medida por um item único de avaliação (rs=-0,29, p=0,03), a satisfação com a aparência física (rs=-0,40, p<0,001) e a satisfação com a aparência facial (rs=-0,39, p=0,0001). Foi encontrada uma associação entre a faixa etária e o escore total do B-OQLQ (p=0,0012), sendo que pacientes maiores de 21 anos obtiveram escores mais elevados que pacientes mais novos. A associação entre o escore total do B-OQLQ e o tipo de cirurgia não foi estatisticamente significativa. O alfa de Cronbach e o CCI foram 0,95 e 0,90, respectivamente. Os domínios do B-OQLQ que mais causaram impacto na qualidade de vida foram aspectos sociais da deformidade (13,0; dp= 10,54) e estética facial (11,81; dp= 6,23). A versão brasileira do questionário OQLQ se mostrou um instrumento válido e confiável, com boas propriedades psicométricas podendo, portanto, ser considerada um instrumento apropriado para acessar o impacto da deformidade dentofacial na qualidade de vida de pacientes portadores desta condição.

A series of zinc tellurite glasses of 75TeO(2)-20ZnO-(5-x)La2O3-xEr(2)O(3) (x=0.02, 0.05, and 0.1 mol%) with the different hydroxl groups were prepared by the conventional melt-quenching method. Infrared spectra were measured in order to estimate the exact content of OH- groups in samples. The observed increase of the fluorescence lifetime with the oxygen bubbling time has been related to the reduction in the OH- content concentration as evidenced by IR transmission spectra. Various nonradiative decay rates from I-4(13/2) of Er3+ with. the change of OH content were determined from the fluorescence lifetime and radiative decay rates were calculated on the basis of Judd-Ofelt theory. Photoluminescence of undoped and B-doped ZnO in silicate glasses was investigated by varying the concentration of ZnO (3550 mol%) and B dopant (0-10 mol%) in the glass matrices. The broad and intense near band edge emissions were observed while the visible light emission was very weak. UV luminescence in all samples was red-shifted relative to the exciton transition in bulk ZnO and enhanced by decreased ZnO concentration due to higher degree of structural integrity and the lower aggregation degree of ZnO. Donor B dopant played the double roles of filling conduction bands to broaden band gap when its concentration was lower than 5 mol%, and emerging with conduction bands to narrow the gap when B dopant exceeded this value.
