The Hsp70 family is one of the most important and conserved molecular chaperone families. It is well documented that Hsp70 family members assist many cellular processes involving protein quality control, as follows: protein folding, transport through membranes, protein degradation, escape from aggregation, intracellular signaling, among several others. The Hsp70 proteins act as a cellular pivot capable of receiving and distributing substrates among the other molecular chaperone families. Despite the high identity of the Hsp70 proteins, there are several homologue Hsp70 members that do not have the same role in the cell, which allow them to develop and participate in such large number of activities. The Hsp70 proteins are composed of two main domains: one that binds ATP and hydrolyses it to ADP and another which directly interacts with substrates. These domains present bidirectional heterotrophic allosteric regulation allowing a fine regulated cycle of substrate binding and release. The general mechanism of the Hsp70s cycle is under the control of ATP hydrolysis that modulates the low (ATP-bound state) and high (ADP-bound state) affinity states of Hsp70 for substrates. An important feature of the Hsp70s cycle is that they have several co-chaperones that modulate their cycle and that can also interact and select substrates. Here, we review some known details of the bidirectional heterotrophic allosteric mechanism and other important features for Hsp70s regulating cycle and function.

Purpose – This research focuses on finding the reasons, why members from different sectors join a cross-sector/multi-stakeholder CSR network and what motivates them to share (or not to share) their knowledge of CSR and their best practices. Design/methodology/approach – Semi-structured interviews were conducted with members of the largest cross-sector CSR network in Sweden. The sample base of 15 people was chosen to be able to represent a wider variety of members from each participating sectors. As well as the CEO of the intermediary organization was interviewed. The interviews were conducted via email and telephone. Findings – The findings include several reasons linked to the business case of CSR such as stakeholder pressure, competitive advantage, legitimacy and reputation as well as new reasons like the importance of CSR, and the access of further knowledge in the field. Further reasons are in line with members wanting to join a network, such as access to contact or having personal contacts. As to why members are sharing their CSR knowledge, the findings indicate to inspire others, to show CSR commitment, to be visible, it leads to business opportunity and the access of others knowledge, and because it was requested. Reasons for not sharing their knowledge would be the lack of opportunity, lack of time and the lack of experience to do so. Originality/value – The research contributes to existing studies, which focused on Corporate Social Responsibility and cross-sector networking as well as to inter-organizational knowledge sharing in the field of CSR.

The Levine family held an extensive reunion during the Summer of 2009 during which 29 DVDs of raw material were recorded for use in the creation of a Levine family mini-documentary. Many of these DVDs contain oral history interviews conducted by Wendy Miller, one of the organizers of the reunion. Although these interviews were not designed for historical research, they contain valuable historical information. O objetivo deste trabalho foi avaliar a influência da imersão em ácido peracético sobre a resistência flexural e rugosidade das cerâmicas do sistema Procera AllCeram. Para cada ensaio foram analisados, o grupo controle e experimental cuja variável foi a imersão em ácido peracético (0,2%), por 50 minutos. A resistência flexural biaxial foi avaliada de acordo com a especificação 6872, da International Organization for Standardization (ISO), que requer um valor mínimo de 50MPa para a cerâmica de cobertura (AllCeram) e 100MPa para a cerâmica de subestrutura (Procera). Para todos os ensaios foram confeccionados 10 corpos de prova, de cada material para compor os grupos experimental (imerso em desinfetante) e controle, com dimensões de (16±0,2)mm de diâmetro por (1,6±0,1)mm de espessura para o AllCeram e, (12±0,2)mm de diâmetro e (1,2±0,2)mm de espessura para o Procera, totalizando 60 corpos de prova. A rugosidade foi avaliada com um rugosímetro utilizando o parâmetro Ra. Os valores médios de resistência flexural biaxial e o desvio padrão dos grupos controle e experimental do Procera e AllCeram foram respectivamente, (440,7±114,77)MPa, (411,6±45,02)MPa, (76,5±3,59)MPa e (86,5±15,41)MPa. Os valores médios da rugosidade e o desvio padrão dos grupos controle e experimental do Procera e AllCeram foram respectivamente: (0,090±0,014)µm, (0,086±0,009) µm, (0,022±0,004)µm e (0,0210±0,006) µm. Quando comparados com a ISO 6872, todos os corpos de prova foram aprovados quanto à resistência flexural. Quando comparados entre si, através do teste “t” de Student não houve diferença significativa entre os grupos experimental e controle de cada um dos ensaios, mostrando que o ácido peracético não interferiu significativamente nas propriedades de resistência flexural e rugosidade dos materiais. Sendo assim, pode-se prever que tal procedimento não trará prejuízos ao desempenho clínico das próteses confeccionadas com as cerâmicas do sistema Procera AllCeram quando consideradas as propriedades analisadas, podendo ser um procedimento clinicamente recomendável. A madeira roliça possui grande emprego nas construções civis, desempenhando a função de vigas, colunas, fundações, postes para distribuição de energia elétrica, entre outras, apresentando a vantagem de não ser processada, como é o caso da madeira serrada. O projeto envolvendo elementos roliços requer, além de outras variáveis estruturais, o conhecimento do módulo de elasticidade. No Brasil, os documentos normativos que tratam da determinação das propriedades de rigidez e resistência para peças roliças de madeira estão em vigência há mais de vinte anos sem revisão técnica. A madeira roliça, por geralmente possuir eixo com curvatura não nula, pode apresentar, segundo a posição da peça no ensaio de flexão, valores diferentes do módulo de elasticidade. Este trabalho tem como objetivo analisara influência da posição de peças roliças de madeira de Eucalyptus grandis na determinação do módulo de elasticidade na flexão. O ensaio de flexão utilizado é o de três pontos, sendo cada peça avaliada em duas posições distintas, definidas mediante o giro da seção transversal em torno do eixo. Os resultados encontrados indicam a necessidade do ensaio de flexão em, pelo menos, duas posições distintas da peça.

Usually masonry structures has low tension strength, hence the design to flexural efforts can results in high reinforcement ratio, specification of high unit and prism strength, structural members with larger section dimensions and modification in structural arrangement to be possible to use masonry members. The main objective of this study is to evaluate the stiffness, the efforts distribution and the effect of horizontal elements (girders) and vertical elements (counterforts) distribution on the behavior of masonry blocks retaining walls. For this purpose, numerical modeling was performed on typical retaining wall arrangements by varying the amount and placement of horizontal and vertical elements, beyond includes elements simulating the reactions of the soil supporting the foundation of the wall. The numerical modeling also include the macro modeling strategy in which the units, mortar and grout are discretized by a standard volume that represents the masonry elastic behavior. Also, numerical model results were compared with those ones of simplified models usually adopted in bending design of masonry elements. The results show horizontal displacements, principal and shear stresses distribution, and bending moments diagrams. From the analysis it was concluded that quantity and manner of distribution of the girders are both important factors to the panel flexural behavior, the inclusion of the foundation changed significantly the behavior of the wall, especially the horizontal displacements, and has been proposed a new way of considering the flanges section of the counterforts

The paper presents a methodology to model three-dimensional reinforced concrete members by means of embedded discontinuity elements based on the Continuum Strong Discontinuous Approach (CSDA). Mixture theory concepts are used to model reinforced concrete as a 31) composite material constituted of concrete with long fibers (rebars) bundles oriented in different directions embedded in it. The effects of the rebars are modeled by phenomenological constitutive models devised to reproduce the axial non-linear behavior, as well as the bond-slip and dowel action. The paper presents the constitutive models assumed for the components and the compatibility conditions chosen to constitute the composite. Numerical analyses of existing experimental reinforced concrete members are presented, illustrating the applicability of the proposed methodology.

In socio-environmental scenario increased the nature resources concern beyond products and subproducts reuse. Recycling is the approach for a material or energy reintroducing in productive system. This method allows the reduction of garbage volume dumped in environment, saving energy and decreasing the requirement of natural resources use. In general, the ending of expanded polystyrene is deposited sanitary landfills or garbage dumps without control that take large volume and spreads easily by aeolian action, with consequently environmental pollution, however, the recycling avoids their misuse and the obtainment from petroleum is reduced. This work recycled expanded polystyrene via merger and/or dissolution by solvents for the production of integrated circuits boards. The obtained material was characterized in flexural mode according to ASTM D 790 and results were compared with phenolite, traditionally used. Specimens fractures were observed by electronic microscopy scanning in order to establish patterns. Expanded Polyestirene recycled as well as phenolite were also thermo analyzed by TGA and DSC. The method using dissolution produced very brittle materials. The method using merger showed no voids formation nor increased the brittleness of the material. A major difficulty to achieve maximum weight savings in the manufacture of composite structural components, is the tendency of these materials have the formation of voids and cracks in the interior and surface components. In aeronautical applications, controlling the volume fraction of fibers, resins and empty the components of composite is very hard. In this work, composites of epoxy matrix RTM6 reinforced with NCF (non crimp fabric carbon) processed by resin transfer molding (RTM) were characterized for porosity (P-ap) and density (rho(ad)). We used a method based on Archimedes' principle (ASTM C830) and the technique of helium pycnometer. The porosity values were compared with those determined by acid digestion (ASTM D3171). The mechanical properties of processed composites was evaluated by testing on the performing flexural and the results were correlated with the porosity value. All techniques tested to determine void content are satisfactory. The differents results can be justified for heterogeneous void distribution on laminate and differences among techniques characteristics. (C) 2011 Published by Elsevier Ltd. Este estudo se reporta às funções de células natural killer (NK), como adesão, lise e citotoxicidade e de subpopulações de células T em uma família com alta prevalência de pacientes com câncer e que apresentaram: glioblastoma, leucemia mielóide crônica, osteoblastoma, melanoma e carcinomas gástrico, pancreático e cólon retal. Quinze membros dessa família foram estudados, sendo 13 sadios, acompanhados por 5 anos e dois com câncer: glioblastoma e leucemia mielóide crônica. Duas pessoas sadias, no momento da avaliação, desenvolveram posteriormente osteoblastoma mandibular ou melanoma maligno. Como controle, foram avaliados 19 indivíduos saudáveis de faixa etária equivalente. A determinação de linfócitos T CD3+ e de suas subpopulações CD4+ e CD8+ foi realizada empregando-se anticorpos monoclonais e a atividade citotóxica de células NK, avaliada pelo teste de single-cell contra células alvo da linhagem eritroleucêmica K562. Os resultados mostraram que as percentagens de células T totais (CD3+), da subpopulação CD4+ e da relação CD4/CD8 foram significativamente menores nos indivíduos da família estudada em comparação aos valores observados no grupo controle. em todos os membros dessa família a percentagem de formação de conjugados entre células NK-células alvo foi inferior ao valor mínimo observado nos controles. Essa alteração poderia estar relacionada a defeito na expressão de moléculas de adesão, presentes na membrana de células NK, como provável causa das alterações funcionais dessas células. A herança dos mecanismos determinantes desta deficiência pode ser um fator de risco, com valor prognóstico para o desenvolvimento de cancer.

INTRODUÇÃO: Piedra branca é micose superficial causada por fungos do gênero Trichosporon e caracterizado por nódulos firmemente aderidos à haste do pêlo. MÉTODOS: Os autores relatam casos familiares encaminhados como pediculose. Foram utilizados cultura em ágar Mycosel® e identificação molecular. RESULTADOS: Exame micológico revelou a infecção por Trichosporon spp. A identificação molecular demonstrou se tratar do Trichosporon inkin. CONCLUSÕES: Piedra branca e infecção pelo T. inkin são raramente relatados na região sudeste do Brasil. A identificação molecular é essencial para correta determinação de espécies no gênero Trichosporon.

Despite the large number of studies addressing the effect of microwave polymerization on the properties of acrylic resin, this method has received limited clinical acceptance. This study evaluated the influence of microwave polymerization on the flexural strength of a denture base resin. A conventional heat-polymerized (Classico), a microwave-polymerized (Onda-Cryl) and a autopolymerizing acrylic (Jet) resins were used. Five groups were established, according to polymerization cycles: A, B and C (Onda-Cryl, short cycle - 500W/3 min, long - 90W/13 min + 500W/90 see, and manufacturing microwave cycle - 320W/3 min + OW/3 min + 720W/3 min); T(Classico, water bath cycle - 74 degrees C/9h) and Q (Jet, press chamber cycle - 50 degrees C/15 min at 2 bar). Ten specimens (65 x 10 x 3.3 mm) were prepared for each cycle. The flexural strength of the five groups was measured using a three-point bending test at a cross-head speed of 5 mm/min. Flexural strength values were analyzed by one-way ANOVA and the Tukey's test was performed to identify the groups that were significantly different at 5% level. The microwave-polymerized groups showed the highest means (p<0.05) for flexural strength (MPa) (A = 106.97 +/- 5.31; B = 107.57 +/- 3.99; C = 109.63 +/- 5.19), and there were no significant differences among them. The heat-polymerized group (T) showed the lowest flexural strength means (84.40 +/- 1.68), and differ significantly from all groups. The specimens of a microwavable denture base resin could be polymerized by different microwave cycles without risk of decreasing the flexural strength.
