The electrical measures of the soil have been used as variables that correlate with its characteristics. This study aimed at developing an electrical capacitance sensor of low cost, to evaluate its performance on the field and verify the correlation between the measurements of electrical capacitance with physical properties (sand, silt and clay) and chemical properties of soil (pH, MO, P resin, H + Al, K, Ca, Mg, SB, CTC and V%) and the moisture content. The data sampling was performed at the farm named "Capão da Onça" which belongs to the State University of Ponta Grossa. The samples collection was conducted in an area of approximately 13 hectares, totalizing 81 samples. In each sampling the electrical capacitance of the soil was measured. After the sensor withdrawal, soil samples were collected and sent to be analysed in the laboratory of the College of Agronomics Science of the Paulista State University. The measuring instrument used to collect data on electric capacitance of the soil a digital multimeter was used. The data were submitted to the analysis of correlation and regression. The developed system presented a low cost and it was capable to measuring variation of the electrical capacitance of the soil. The obtained measures satisfactorily correlated with the levels of clay and sand, and weakly with the moisture content. This had demonstrated the possibility to use a sensor to verify the soil texture in not homogeneous areas. The measures of the electrical capacitance of the soil obtained by the sensor had significantly correlated with the soil attributes: calcium, magnesium, pH, SB and CTC. These results had demonstrated the possibility to use a sensor for soil fertility control.

Abstract. Based on prior field observations, we hypothesized that individual and interacting effects of plant size, density, insect herbivory, and especially fungal disease, influenced seedling and juvenile plant growth in native Platte thistle populations (Cirsium canescens Nutt.). We worked at Arapaho Prairie in the Nebraska Sandhills (May - August 2007), monitoring plant growth, insect damage, and fungal infection within different density thistle patches. In the main experiment, we sprayed half of test plants in different density patches with fungicide (Fungonil© Bonide, containing chlorothalonil) and half with a water control. Fungal infection rates were very low, so we found no difference in fungal attack between these treatments. However, plants that received the fungicide treatment had significantly faster growth over the season than did the control plants. At the same time, plants in the fungicide treatment had significantly reduced insect herbivory. These results strongly suggest that the fungicide had insecticidal effects and that insect herbivory significantly decreases juvenile Platte thistle growth. Further, damage by insect herbivores tended to be higher for larger plants, and herbivory was variable among different patches. However, plant density did not appear to have a large effect on the amount of insect herbivory that individual juvenile Platte thistle plants received. In the second experiment, we examined germination and survival success in relationship to seed density, and found that germination success was higher in areas of lower seed density. In the third experiment, we tested germination for filled seeds categorized primarily by color variation and size, and found no difference in germination related to either color or seed weight. We conclude that seed density, but not seed quality as estimated by color or size, affects germination success. Further, although herbivory was not significantly affected by plant density at any of the scales examined, insect herbivory significantly reduces the growth and success of juveniles of this characteristic native sand prairie plant.

We compared the microbial community composition in soils from the Brazilian Amazon with two contrasting histories; anthrosols and their adjacent non-anthrosol soils of the same mineralogy. The anthrosols, also known as the Amazonian Dark Earths or terra preta, were managed by the indigenous pre-Colombian Indians between 500 and 8,700 years before present and are characterized by unusually high cation exchange capacity, phosphorus (P), and calcium (Ca) contents, and soil carbon pools that contain a high proportion of incompletely combusted biomass as biochar or black carbon (BC). We sampled paired anthrosol and unmodified soils from four locations in the Manaus, Brazil, region that differed in their current land use and soil type. Community DNA was extracted from sampled soils and characterized by use of denaturing gradient gel electrophoresis (DGGE) and terminal restriction fragment length polymorphism. DNA bands of interest from Bacteria and Archaea DGGE gels were cloned and sequenced. In cluster analyses of the DNA fingerprints, microbial communities from the anthrosols grouped together regardless of current land use or soil type and were distinct from those in their respective, paired adjacent soils. For the Archaea, the anthrosol communities diverged from the adjacent soils by over 90%. A greater overall richness was observed for Bacteria sequences as compared with those of the Archaea. Most of the sequences obtained were novel and matched those in databases at less than 98% similarity. Several sequences obtained only from the anthrosols grouped at 93% similarity with the Verrucomicrobia, a genus commonly found in rice paddies in the tropics. Sequences closely related to Proteobacteria and Cyanobacteria sp. were recovered only from adjacent soil samples. Sequences related to Pseudomonas, Acidobacteria, and Flexibacter sp. were recovered from both anthrosols and adjacent soils. The strong similarities among the microbial communities present in the anthrosols for both the Bacteria and Archaea suggests that the microbial community composition in these soils is controlled more strongly by their historical soil management than by soil type or current land use. The anthrosols had consistently higher concentrations of incompletely combusted organic black carbon material (BC), higher soil pH, and higher concentrations of P and Ca compared to their respective adjacent soils. Such characteristics may help to explain the longevity and distinctiveness of the anthrosols in the Amazonian landscape and guide us in recreating soils with sustained high fertility in otherwise nutrient-poor soils in modern times.

The macrophages are the first host cells that interact with the fungus Paracoccidioides brasiliensis, but the main mechanisms that regulate this interaction are not well understood. Because the role played by P. brasiliensis lipids in macrophage activation was not previously investigated, we aimed to assess the influence of diverse lipid fractions from P. brasiliensis yeasts in this process. The possible participation of TLR2 and TLR4 signaling was also evaluated using TLR2- and TLR4-defective macrophages. Four lipid-rich fractions were studied as follows: F1, composed by membrane phospholipids and neutral lipids, F2 by glycolipids of short chain, F3a by membrane glycoproteins anchored by glycosylphosphatidylinositol (GPI) groups, and F3b by glycolipids of long chain. All assayed lipid fractions were able to activate peritoneal macrophages and induce nitric oxide (NO) production. Importantly, the F1 and F3a fractions exerted opposite effects in the control of P. brasiliensis uptake and killing, but both fractions inhibited cytokines production. Furthermore, the increased NO production and expression of costimulatory molecules induced by F3a was shown to be TLR2 dependent although F1 used Toll-independent mechanisms. In conclusion, our work suggests that lipid components may play a role in the innate immunity against P. brasiliensis infection using Toll-dependent and independent mechanisms to control macrophage activation.

Extremophiles are organisms adapted to grow at extreme ranges of environmental variables, such as high or low temperatures, acid or alkaline medium, high salt concentration, high pressures and so forth. Most extremophiles are micro-organisms that belong to the Archaea and Bacteria domains, and are widely spread across the world, which include the polar regions, volcanoes, deserts, deep oceanic sediments, hydrothermal vents, hypersaline lakes, acid and alkaline water bodies, and other extreme environments considered hostile to human life. Despite the tropical climate, Brazil has a wide range of ecosystems which include some permanent or seasonally extreme environments. For example, the Cerrado is a biome with very low soil pH with high Al+3 concentration, the mangroves in the Brazilian coast are anaerobic and saline, Pantanal has thousands of alkaline-saline lakes, the Caatinga arid and hot soils and the deep sea sediments in the Brazilian ocean shelf. These environments harbour extremophilic organisms that, coupled with the high natural biodiversity in Brazil, could be explored for different purposes. However, only a few projects in Brazil intended to study the extremophiles. In the frame of astrobiology, for example, these organisms could provide important models for defining the limits of life and hypothesize about life outside Earth. Brazilian microbiologists have, however, studied the extremophilic micro-organisms inhabiting non-Brazilian environments, such as the Antarctic continent. The experience and previous results obtained from the Brazilian Antarctic Program (PROANTAR) provide important results that are directly related to astrobiology. This article is a brief synopsis of the Brazilian experience in researching extremophiles, indicating the most important results related to astrobiology and some future perspectives in this area. The rainforest of Mexico has been degraded and severely fragmented, and urgently require restoration. However, the practice of restoration has been limited by the lack of species-specific data on survival and growth responses to local environmental variation. This study explores the differential performance of 14 wet tropical early-, mid- or late-successional tree species that were grown in two abandoned pastures with contrasting land-use histories. After 18 months, seedling survival and growth of at least 7 of the 14 tree species studied were significantly higher in the site with a much longer history of land use (site 2). Saplings of the three early-successional species showed exceptional growth rates. However, differences in performance were noted in relation to the differential soil properties between the experimental sites. Mid-successional species generally showed slow growth rates but high seedling survival, whereas late-successional species exhibited poor seedling survival at both the study sites. Stepwise linear regressions revealed that the species integrated response index combining survivorship and growth measurements, was influenced mostly by differences in soil pH between the two abandoned pastures. Our results suggest that local environmental variation among abandoned pastures of contrasting land-use histories influences sapling survival and growth. Furthermore, the similarity of responses among species with the same successional status allowed us to make some preliminary site and species-specific silvicultural recommendations. Future field experiments should extend the number of species and the range of environmental conditions to identify site generalists or more narrowly adapted species, that we would call sensitive.

Occurrence of Zoophthora radicans infecting nymphs and adults of Thaumastocoris peregrinus Carpintero and Dellape, 2006 is reported in Brazil. This is a new record of host for this fungal species and the first fungal pathogen associated with this pest worldwide. Infection of Z. radicans on T. peregrinus populations on commercial Eucalyptus plantation (Eucalyptus spp.) reached up to 100%, and low insect densities were associated with high levels of fungal infection in three out of seven plots. This pathogen seems to be virulent against T. peregrinus and may play an important role in population regulations of this invasive pest through naturally induced epizootics. (c) 2012 Elsevier Inc. Among the soils in the Mato Grosso do Sul, stand out in the Pantanal biome, the Spodosols. Despite being recorded in considerable extensions, few studies aiming to characterize and classify these soils were performed. The purpose of this study was to characterize and classify soils in three areas of two physiographic types in the Taquari river basin: bay and flooded fields. Two trenches were opened in the bay area (P1 and P2) and two in the flooded field (P3 and P4). The third area (saline) with high sodium levels was sampled for further studies. In the soils in both areas the sand fraction was predominant and the texture from sand to sandy loam, with the main constituent quartz. In the bay area, the soil organic carbon in the surface layer (P1) was (OC) > 80 g kg(-1), being diagnosed as Histic epipedon. In the other profiles the surface horizons had low OC levels which, associated with other properties, classified them as Ochric epipedons. In the soils of the bay area (P1 and P2), the pH ranged from 5.0 to 7.5, associated with dominance of Ca2+ and Mg2+, with base saturation above 50 % in some horizons. In the flooded fields (P3 and P4) the soil pH ranged from 4.9 to 5.9, H+ contents were high in the surface horizons (0.8-10.5 cmol(c) kg(-1)), Ca2+ and Mg-2 contents ranged from 0.4 to 0.8 cmol(c) kg(-1) and base saturation was < 50 %. In the soils of the bay area (P1 and P2) iron was accumulated (extracted by dithionite - Fed) and OC in the spodic horizon; in the P3 and P4 soils only Fed was accumulated (in the subsurface layers). According to the criteria adopted by the Brazilian System of Soil Classification (SiBCS) at the subgroup level, the soils were classified as: P1: Organic Hydromorphic Ferrohumiluvic Spodosol. P2: Typical Orthic Ferrohumiluvic Spodosol. P3: Typical Hydromorphic Ferroluvic Spodosol. P4: Arenic Orthic Ferroluvic Spodosol.

The availability and uptake of Cd by lettuce (Lactuca sativa L.) in two common tropical soils (before and after liming) were studied in order to derive human health-based risk soil concentration. Cadmium concentrations ranging from 1 to 12 mg kg(-1) were added to samples from a clayey Oxisol and a sandy-loam Ultisol under glasshouse conditions. After incubation, a soil sample was taken from each pot, the concentration of Cd in the soil was determined, lettuce was grown during 36 d, and the edible parts were harvested and analyzed for Cd. A positive linear correlation was observed between total soil Cd and the Cd concentration in lettuce. The amount of Cd absorbed by lettuce grown in the Ultisol was about twice the amount absorbed in the Oxisol. Liming increased the soil pH and slightly reduced Cd availability and uptake. CaCl2 extraction was better than DTPA to reflect differences in binding strength of Cd between limed and unlimed soils. Risk Cd concentrations in the Ultisol were lower than in the Oxisol, reflecting the greater degree of uptake from the Ultisol. The derived risk Cd values were dependent on soil type and the exposure scenario.

Leaching of nitrate (NO3-) can increase the groundwater concentration of this anion and reduce the agronomical effectiveness of nitrogen fertilizers. The main soil property inversely related to NO3- leaching is the anion exchange capacity (AEC), whose determination is however too time-consuming for being carried out in soil testing laboratories. For this reason, this study evaluated if more easily measurable soil properties could be used to estimate the resistance of subsoils to NO3- leaching. Samples from the subsurface layer (20-40 cm) of 24 representative soils of São Paulo State were characterized for particle-size distribution and for chemical and electrochemical properties. The subsoil content of adsorbed NO3- was calculated from the difference between the NO3- contents extracted with 1 mol L-1 KCl and with water; furthermore, NO3- leaching was studied in miscible displacement experiments. The results of both adsorption and leaching experiments were consistent with the well-known role exerted by AEC on the nitrate behavior in weathered soils. Multiple regression analysis indicated that in subsoils with (i) low values of remaining phosphorus (Prem), (ii) low soil pH values measured in water (pH H2O), and (iii) high pH values measured in 1 moL L-1 KCl (pH KCl), the amounts of surface positive charges tend to be greater. For this reason, NO3- leaching tends to be slower in these subsoils, even under saturated flow condition.

South American blastomycosis, paracoccidioidomycosis (Pb mycosis) or Lutz disease is an endemically fungal infection in Latin America. It is caused by the dimorphic fungus Paracoccidioides brasiliensis and may cause oral mucosal lesions. The incidence of Pb mycosis oral lesions was evaluated in patients assisted at a Brazilian Dental School's Specialized Oral Diagnosis Service with special focus on the different clinical forms of these lesions, its location, patients' occupation, deleterious habits, and diagnosis methodology. Students' and professionals' initial diagnoses were compared with the definitive diagnosis. Lesions were detected 31 cases (18 patients). The results show that 88.8% of the patients were male with a mean age of 50 years and 39% work(ed) with activities related to agriculture. As much as 88.9% were smokers and 72.2% were alcohol users. Exfoliative cytology was performed in 66.6% of the patients. Oral mucosa (30%), gingiva (16.6%) and lips 16.6% were the most common sites of Pb mycosis oral lesions. Comparing the initial with the definitive diagnosis made by the professionals their accuracy was 33% (6 out of 18 patients). Students' diagnosis was more accurate demonstrating 72.5% of initial correct diagnosis (13 out of 18). Statistical analysis by ANOVA (α=0.05, SPSS WIN) demonstrated a significant difference between the diagnosis of Pb mycosis made by students and professionals when considering initial diagnosis and final diagnosis (after histopathological analysis) (p=0.25). Incisional biopsy and exfoliate cytology are efficient for an early diagnosis of this disease in mouth. Students' training in diagnosis of oral pathologies to recognize lesions is urgent to improve public health.

In der vorliegenden Arbeit wurden Miniemulsionen als räumliche Begrenzungen für die Synthese von unterschiedlichen funktionellen Materialien mit neuartigen Eigenschaften verwendet. Das erste Themengebiet umfasst die Herstellung von Polymer/Calciumphosphat-Hybridpartikeln und –Hybridkapseln über die templatgesteuerte Mineralisation von Calciumphosphat. Die funktionalisierte Oberfläche von Polymernanopartikeln, welche über die Miniemulsionspolymerisation hergestellt wurden, diente als Templat für die Kristallisation von Calciumphosphat auf den Partikeln. Der Einfluss der funktionellen Carboxylat- und Phosphonat-Oberflächengruppen auf die Komplexierung von Calcium-Ionen sowie die Mineralisation von Calciumphosphat auf der Oberfläche der Nanopartikel wurde mit mehreren Methoden (ionenselektive Elektroden, REM, TEM und XRD) detailliert analysiert. Es wurde herausgefunden, dass die Mineralisation bei verschiedenen pH-Werten zu vollkommen unterschiedlichen Kristallmorphologien (nadel- und plättchenförmige Kristalle) auf der Oberfläche der Partikel führt. Untersuchungen der Mineralisationskinetik zeigten, dass die Morphologie der Hydroxylapatit-Kristalle auf der Partikeloberfläche mit der Änderung der Kristallisationsgeschwindigkeit durch eine sorgfältige Wahl des pH-Wertes gezielt kontrolliert werden kann. Sowohl die Eigenschaften der als Templat verwendeten Polymernanopartikel (z. B. Größe, Form und Funktionalisierung), als auch die Oberflächentopografie der entstandenen Polymer/Calciumphosphat-Hybridpartikel wurden gezielt verändert, um die Eigenschaften der erhaltenen Kompositmaterialien zu steuern. rnEine ähnliche bio-inspirierte Methode wurde zur in situ-Herstellung von organisch/anorganischen Nanokapseln entwickelt. Hierbei wurde die flexible Grenzfläche von flüssigen Miniemulsionströpfchen zur Mineralisation von Calciumphosphat an der Grenzfläche eingesetzt, um Gelatine/Calciumphosphat-Hybridkapseln mit flüssigem Kern herzustellen. Der flüssige Kern der Nanokapseln ermöglicht dabei die Verkapselung unterschiedlicher hydrophiler Substanzen, was in dieser Arbeit durch die erfolgreiche Verkapselung sehr kleiner Hydroxylapatit-Kristalle sowie eines Fluoreszenzfarbstoffes (Rhodamin 6G) demonstriert wurde. Aufgrund der intrinsischen Eigenschaften der Gelatine/Calciumphosphat-Kapseln konnten abhängig vom pH-Wert der Umgebung unterschiedliche Mengen des verkapselten Fluoreszenzfarbstoffes aus den Kapseln freigesetzt werden. Eine mögliche Anwendung der Polymer/Calciumphosphat-Partikel und –Kapseln ist die Implantatbeschichtung, wobei diese als Bindeglied zwischen künstlichem Implantat und natürlichem Knochengewebe dienen. rnIm zweiten Themengebiet dieser Arbeit wurde die Grenzfläche von Nanometer-großen Miniemulsionströpfchen eingesetzt, um einzelne in der dispersen Phase gelöste Polymerketten zu separieren. Nach der Verdampfung des in den Tröpfchen vorhandenen Lösungsmittels wurden stabile Dispersionen sehr kleiner Polymer-Nanopartikel (<10 nm Durchmesser) erhalten, die aus nur wenigen oder einer einzigen Polymerkette bestehen. Die kolloidale Stabilität der Partikel nach der Synthese, gewährleistet durch die Anwesenheit von SDS in der wässrigen Phase der Dispersionen, ist vorteilhaft für die anschließende Charakterisierung der Polymer-Nanopartikel. Die Partikelgröße der Nanopartikel wurde mittels DLS und TEM bestimmt und mit Hilfe der Dichte und des Molekulargewichts der verwendeten Polymere die Anzahl an Polymerketten pro Partikel bestimmt. Wie es für Partikel, die aus nur einer Polymerkette bestehen, erwartet wird, stieg die mittels DLS bestimmte Partikelgröße mit steigendem Molekulargewicht des in der Synthese der Partikel eingesetzten Polymers deutlich an. Die Quantifizierung der Kettenzahl pro Partikel mit Hilfe von Fluoreszenzanisotropie-Messungen ergab, dass Polymer-Einzelkettenpartikel hoher Einheitlichkeit hergestellt wurden. Durch die Verwendung eines Hochdruckhomogenisators zur Herstellung der Einzelkettendispersionen war es möglich, größere Mengen der Einzelkettenpartikel herzustellen, deren Materialeigenschaften zurzeit näher untersucht werden.rn
