The purpose of this study was to determine whether or not blind children perseverate during a modified Piagetian A-not-B reaching task, with conditions that employ luminous AB targets and acoustic AB targets. Ten congenitally blind children, ages 1-4 years, with residual vision for light, took part in this study. Behavioral and kinematic data were computed for participants' reaches, performed in six A trials and in two B trials, in both stimulus conditions. All of the children perseverated in the luminous condition, and none of them perseverated in the condition using acoustic targets. The children tilted their heads in the direction of the target as they reached towards it. However, this coupling action (head-reaching) occurred predominantly in the A trials in the acoustic condition. In the luminous condition, in contrast to the acoustic condition, the children took longer times to initiate the reaching movement. Also, in the luminous condition, the children explored the target surroundings, unlike the acoustic condition, in which they reached straight ahead. For these blind children, sound was more relevant to reaching than was the luminous stimulus. The luminous input caused perseveration in congenitally blind children in a similar way that has been reported in the literature for typically-developing, sighted infants, ages 8-12 months, performing A-not-B tasks with visual inputs. O experimento foi realizado no Departamento de Produção Vegetal da Escola Superior de Agricultura Luiz de Queiroz, ESALQ/USP, Piracicaba, São Paulo, Brasil, utilizando-se as culturas de guaranazeiro (Paullinia cupana Kunth), cafeeiro (Coffea arabica L.), cajueiro (Anacardium occidentale L.), goiabeira (Psidium guajava L.) e seringueira (Hevea brasiliensis Muell. - Arg.). No período de seca (setembro/94) e de chuvas (novembro/94), realizaram-se determinações de resistência estomática (RE) (s cm-1) e transpiração (T) (µg cm-1 s-1) nas diferentes espécies. O delineamento experimental foi em blocos casualizados com cinco repetições. A partir das análises dos dados pode-se concluir: 1. diferenças significativas entre espécies, em termos das variáveis avaliadas no período de deficiência hídrica, com valores decrescentes de resistência estomática e crescente de transpiração na seguinte ordem: guaranazeiro > cafeeiro > cajueiro > goiabeira > seringueira; 2. Nas águas as diferenças entre espécies, para ambas as variáveis, foram menos evidentes, continuando a cultura da seringueira a apresentar menor resistência estomática e maior transpiração dentre as espécies; 3. As culturas de guaraná e café apresentaram maior resistência à perda de água, em relação às demais culturas.

Genome sequencing efforts are providing us with complete genetic blueprints for hundreds of organisms. We are now faced with assigning, understanding, and modifying the functions of proteins encoded by these genomes. DBMODELING is a relational database of annotated comparative protein structure models and their metabolic pathway characterization, when identified. This procedure was applied to complete genomes such as Mycobacteritum tuberculosis and Xylella fastidiosa. The main interest in the study of metabolic pathways is that some of these pathways are not present in humans, which makes them selective targets for drug design, decreasing the impact of drugs in humans. In the database, there are currently 1116 proteins from two genomes. It can be accessed by any researcher at http://www.biocristalografia.df.ibilce.unesp.br/tools/. This project confirms that homology modeling is a useful tool in structural bioinformatics and that it can be very valuable in annotating genome sequence information, contributing to structural and functional genomics, and analyzing protein-ligand docking.

P>BackgroundThe nonclassical human leucocyte antigen (HLA)-G molecule has been well recognized as a tolerogenic molecule and few studies have evaluated the role of the molecule in inflammatory cutaneous autoimmune diseases.ObjectivesTo evaluate the expression of HLA-G in skin specimens of patients with psoriasis and to analyse its correlation with epidemiological and clinical variables.MethodsThirty untreated patients with psoriasis and 32 healthy individuals were enrolled. Immunohistochemistry was applied to identify HLA-G expression in formalin-fixed paraffin-embedded cutaneous skin biopsies.ResultsSoluble and membrane-bound HLA-G expression was detected in 30 (90%) of the skin specimens from patients presenting clinical and histopathological features of psoriasis. Although infiltrating lymphomononuclear cells of the dermis exhibited HLA-G expression, the epidermis was primarily targeted. HLA-G expression was also observed in 27% (three of 11) of the specimens that exhibited no clinical and histopathological features of psoriasis (nonaffected areas). In contrast, skin specimens obtained from healthy individuals exhibited no HLA-G expression (P < 0 center dot 0001). The intensity of HLA-G expression was not associated with type I/II psoriasis, Psoriasis Area and Severity Index score or clinical forms.ConclusionsAs the HLA-G molecule was consistently expressed in affected and, to a lesser extent, in nonaffected areas of untreated patients with psoriasis, irrespective of the severity of the clinical variants, one may hypothesize that the presence of HLA-G may be responsible, at least in part, for the regulation of autoimmune effector cells.

Coffea canephora plants (clone INCAPER-99) were submitted to low N (LN) or high N (HN) applications and two watering regimes (daily irrigation and irrigation every 5 days for a month). Although water potential was not altered significantly by N, HN plants showed higher relative water content than did LN plants under water deficit. Only HN plants exhibited some ability for osmotic adjustment. Plants from both N treatments increased their cell wall rigidity under drought, with a more pronounced augmentation in HN plants. In well-watered plants, carbon assimilation rate increased with increasing N while stomatal conductance did not respond to N supply. Under drought conditions, carbon assimilation decreased by 68-80% compared to well-watered plants, whereas stomatal conductance and transpiration rate declined by 35% irrespective of the N applications. Stable carbon isotope analysis, combined with leaf gas exchange measurements, indicated that regardless of the watering treatments, N increased the long-term water use efficiency through changes in carbon assimilation with little or no effect on stomatal behaviour.

In mature and young leaves of sunflower (Helianthus annuus L. cv. Catissol-01) plants grown in the greenhouse, photosynthetic rate, stomatal conductance, and transpiration rate declined during water stress independently of leaf age and recovered after 24-h rehydration. The intercellular CO 2 concentration, chlorophyll (Chl) content, and photochemical activity were not affected by water stress. However, non-photochemical quenching increased in mature stressed leaves. Rehydration recovered the levels of non-photochemical quenching and increased the F v/F m in young leaves. Drought did not alter the total Chl content. However, the accumulation of proline under drought was dependent on leaf age: higher content of proline was found in young leaves. After 24 h of rehydration the content of proline returned to the same contents as in control plants.

DBMODELING is a relational database of annotated comparative protein structure models and their metabolic, pathway characterization. It is focused on enzymes identified in the genomes of Mycobacterium tuberculosis and Xylella fastidiosa. The main goal of the present database is to provide structural models to be used in docking simulations and drug design. However, since the accuracy of structural models is highly dependent on sequence identity between template and target, it is necessary to make clear to the user that only models which show high structural quality should be used in such efforts. Molecular modeling of these genomes generated a database, in which all structural models were built using alignments presenting more than 30% of sequence identity, generating models with medium and high accuracy. All models in the database are publicly accessible at http://www.biocristalografia.df.ibilce.unesp.br/tools. DBMODELING user interface provides users friendly menus, so that all information can be printed in one stop from any web browser. Furthermore, DBMODELING also provides a docking interface, which allows the user to carry out geometric docking simulation, against the molecular models available in the database. There are three other important homology model databases: MODBASE, SWISSMODEL, and GTOP. The main applications of these databases are described in the present article. © 2007 Bentham Science Publishers Ltd.
