The identification of subjects at high risk for Alzheimer’s disease is important for prognosis and early intervention. We investigated the polygenic architecture of Alzheimer’s disease and the accuracy of Alzheimer’s disease prediction models, including and excluding the polygenic component in the model. This study used genotype data from the powerful dataset comprising 17 008 cases and 37 154 controls obtained from the International Genomics of Alzheimer’s Project (IGAP). Polygenic score analysis tested whether the alleles identified to associate with disease in one sample set were significantly enriched in the cases relative to the controls in an independent sample. The disease prediction accuracy was investigated in a subset of the IGAP data, a sample of 3049 cases and 1554 controls (for whom APOE genotype data were available) by means of sensitivity, specificity, area under the receiver operating characteristic curve (AUC) and positive and negative predictive values. We observed significant evidence for a polygenic component enriched in Alzheimer’s disease (P = 4.9 × 10−26). This enrichment remained significant after APOE and other genome-wide associated regions were excluded (P = 3.4 × 10−19). The best prediction accuracy AUC = 78.2% (95% confidence interval 77–80%) was achieved by a logistic regression model with APOE, the polygenic score, sex and age as predictors. In conclusion, Alzheimer’s disease has a significant polygenic component, which has predictive utility for Alzheimer’s disease risk and could be a valuable research tool complementing experimental designs, including preventative clinical trials, stem cell selection and high/low risk clinical studies. In modelling a range of sample disease prevalences, we found that polygenic scores almost doubles case prediction from chance with increased prediction at polygenic extremes.

ABSTRACT - Meta-cinema can depict film viewers’ attitudes towards cinema and their type of devotion for films. One subcategory of viewers - which I call meta-spectators - is highly specialized in its type of consumption, bordering on obsession. I contend that there are two main varieties of meta-cinematic reception, not altogether incompatible with one another, despite their apparent differences. As both of them are depicted on meta-cinematic products, the films themselves are the best evidence of my typology. My categories of film viewers are the ‘cinephile’, an elite prone to artistic militancy and the adoration of filmic masters; and the ‘fan’, a low culture consumer keen on certain filmic universes and their respective figures and motifs. I will base my rationale on four films that portray such reception practices: Travelling Avant (Jean-Charles Tacchella, 1987, FRA), The Dreamers (Bernardo Bertolucci, 2003, UK/FRA/ITA); Free Enterprise (Robert Mayer Burnett, 1998, USA); Fanboys (2009, Kyle Newman, USA).

[…] À travers ces trois années passées auprès des alcooliques et leurs conjoints, nous avons constaté que la très grande majorité de ces individus avait un dénominateur commun: l'isolement. En effet, à la longue, l'alcoolisme fausse et brise les relations de l'alcoolique avec sa famille, son entourage, ses amis, son milieu de travail, son milieu de loisirs. Il y a bien les "amis" de taverne, mais de l'aveu même des alcooliques, il n'existe pas là de relations humaines interpersonnelles satisfaisantes, mais ce sont plutôt des relations d'intérêt, car ce qui réunit d'abord et avant tout ces gens, c'est leur besoin de boire. Et petit à petit, la famille de l'alcoolique se retrouve elle aussi isolée socialement.[…] Nous avons voulu, dans la présente étude, évaluer les résultats d'une expérience de thérapie de groupe avec des épouses d'alcooliques qui s'est déroulée à la clinique externe d'alcoologie du Centre Hospitalier St-Vincent de Paul. De façon plus spécifique, nous avons voulu, 1) mesurer les changements survenus au niveau des connaissances et des attitudes des épouses qui auraient participé à la thérapie de groupe, 2) voir jusqu'où les changements survenus étaient associés à certaines caractéristiques personnelles des participantes, et 3) voir s'il y avait une relation entre les changements survenus au niveau des connaissances et les changements survenus au niveau des attitudes. L'intérêt de la présente étude était double: d'une part, elle nous permettait d'évaluer une expérience de thérapie de groupe avec des épouses d'alcooliques selon les critères d'une recherche scientifique et de situer la thérapie de groupe en terme d'efficacité à l'intérieur des services offerts par la clinique où nous travaillions. D'autre part, étant donné la rareté de telles études dans ce domaine de l'alcoolisme (voir annexe A), la présente étude pouvait certes mieux faire connaître la thérapie de groupe avec des épouses d'alcooliques, illustrer la valeur de celle-ci et enfin, fournir des données susceptibles de contribuer au développement de ce mode d'intervention. This qualitative study examines five young Afro-Franco Caribbean males in the Diaspora and their experiences with systems of technology as a tool of oppression and liberation. The study utilized interpretive biography and participatory video research to examine the issues of identity, power/control, surveillance technology, love and freedom. The study made use of a number of data collection methods including interviews, round table discussions, and personal narratives. A hermeneutic theoretical framework is employed to develop an objective view of the problems facing Afro-Franco Caribbean males in the schools and community. The purpose of the study is to provide an environment and new media technology that Afro-Franco Caribbean males can use to engage and discuss their views on issues mentioned above and to ultimately develop a video project to share with the community. Moreover, the study sought to examine an epistemological approach (Creolization) that young black males, particularly Afro-Franco-Caribbean males, might use to communicate, document, and share their everyday experiences in the Diaspora. The findings in the study reveal that the participants are experiencing: (a) a lack of community involvement in the urban space they currently reside, (b) frustration with the perspective of their home country, Haiti, that is commonly shown in mainstream media, and (c) ridicule, shame, and violence in the spaces (school and community) that should be safe. The study provides the community (both local and scholarly) with an opportunity to hear the voices and concerns of youth in the urban space. In addition the study suggests a need for schools to create a critical pedagogical curriculum in which power can be democratically shared.

The current understanding of Arctic ecosystems is deeply rooted in the classical view of a bottom-up controlled system with strong physical forcing and seasonality in primary-production regimes. Consequently, the Arctic polar night is commonly disregarded as a time of year when biological activities are reduced to a minimum due to a reduced food supply. Here, based upon a multidisciplinary ecosystem-scale study from the polar night at 79 degrees N, we present an entirely different view. Instead of an ecosystem that has entered a resting state, we document a system with high activity levels and biological interactions across most trophic levels. In some habitats, biological diversity and presence of juvenile stages were elevated in winter months compared to the more productive and sunlit periods. Ultimately, our results suggest a different perspective regarding ecosystem function that will be of importance for future environmental management and decision making, especially at a time when Arctic regions are experiencing accelerated environmental change [1].
