This study developed and validated a method for moisture determination in artisanal Minas cheese, using near-infrared spectroscopy and partial-least-squares. The model robustness was assured by broad sample diversity, real conditions of routine analysis, variable selection, outlier detection and analytical validation. The model was built from 28.5-55.5% w/w, with a root-mean-square-error-of-prediction of 1.6%. After its adoption, the method stability was confirmed over a period of two years through the development of a control chart. Besides this specific method, the present study sought to provide an example multivariate metrological methodology with potential for application in several areas, including new aspects, such as more stringent evaluation of the linearity of multivariate methods.

Several studies have described the benefits of nanoscience and nanotechnology (N&N) in different sectors such as agriculture, energy, environmental preservation, and public health. The rapid evolution of N&N can be shown through a panoramic analysis of scientific papers and patents. In the area of public health, it is estimated that the global market for nanotechnology products will expand to 160 billion U.S. dollars in 2015. The Brazilian government has also strengthened its innovative potential in N&N through economic subsidies, as observed for other countries. This review is focused on the current landscape of N&N in a therapeutic context, highlighting the development of nanotech-products produced with biocompatible and biodegradable materials that are already commercially available or under investigation. Most studies under investigation are focused on the development of nanotechnology-based formulations intended for treatment of cancer, inflammatory, cardiovascular, and neurological diseases. Although there are several advantages of N&N in healthcare, many challenges have to be conquered to increase the availability of nanotechnology products in toxicological, preclinical and clinical studies, scale-up, regulatory, and private investments.

The use of veterinary drugs in food producing animals may result in the presence of residues in foods, including milk and eggs. Immunoassay or microbiological tests are used to screen for residues, but chromatographic methods are needed to confirm positive results. In most methods, the sample is extracted with acetonitrile, submitted to clean up or directly analyzed by LC-MS/MS. Results of the Brazilian governmental monitoring programs from 2006 to 2011 have shown that the antiparasitic ivermectin was the drug most frequently found in milk. Residues in eggs are only monitored by one of the programs, and few studies have reported the incidence of veterinary drugs in this matrix in Brazil.

The chemist must master a number of abilities. These include a solid knowledge of theory and the capacity to devise theoretical problems. The chemist is also expected to solve problems. In order to improve these abilities, this work aimed to evaluate several tools, such as the use of contextualization, problem-based learning and the interdisciplinary approach. In the methodology developed, undergraduate students had to investigate and propose a hypothesis, suggesting how physical changes took place in inappropriately stored drugs. The interdisciplinary approach is achieved by connecting public interest, chemistry, biology and pharmacology concepts. The tools can improve these abilities required for a well-prepared chemist.

An experiment was proposed applying the Chemometric approach of Multivariate Analysis for inclusion in undergraduate Chemistry courses to promote and expand the use of this analytical-statistical tool. The experiment entails the determination of the acid dissociation constant of dyes via UV-Vis electronic spectrophotometry. The dyes used show from simple equilibrium to very complex systems involving up to four protolytic species with high spectral overlap. The Chemometric methodology was more efficient than univariate methods. For use in classes, it is up to the teacher to decide which systems should be utilized given the time constraints and laboratory conditions.

Multivariate Curve Resolution with Alternating Least Squares (MCR-ALS) is a resolution method that has been efficiently applied in many different fields, such as process analysis, environmental data and, more recently, hyperspectral image analysis. When applied to second order data (or to three-way data) arrays, recovery of the underlying basis vectors in both measurement orders (i.e. signal and concentration orders) from the data matrix can be achieved without ambiguities if the trilinear model constraint is considered during the ALS optimization. This work summarizes different protocols of MCR-ALS application, presenting a case study: near-infrared image spectroscopy.

The conventional curriculum of Analytical Chemistry undergraduate courses emphasizes the introduction of techniques, methods and procedures used for instrumental analysis. All these concepts must be integrated into a sound conceptual framework to allow students to make appropriate decisions. Method calibration is one of the most critical parameters that has to be grasped since most analytical techniques depend on it for quantitative analysis. The conceptual understanding of calibration is not trivial for undergraduate students. External calibration is widely discussed during instrumental analysis courses. However, the understanding of the limitations of external calibration to correct some systematic errors is not directly derived from laboratory examples. The conceptual understanding of other calibration methods (standard addition, matrix matching, and internal standard) is imperative. The aim of this work is to present a simple experiment using grains (beans, corn and chickpeas) to explore different types of calibration methods.

The binding of [RuCl2(L)] (L = N,N-bis(7-methyl-2-pyridylmethylene)-1,3-diiminopropane) to bovine and human serum albumin was investigated by the fluorescence quenching technique. The comparison of the quenching effect of serum albumin fluorescence by ruthenium complex allowed the estimation of subdomain IB in BSA and subdomain IIA in HSA as the binding sites for this complex. The results of fluorescence titration revealed that ruthenium complex quenches the intrinsic fluorescence of BSA through a dynamic quenching mechanism, while HSA has a static quenching mechanism. The thermodynamic parameters indicated that hydrophobic forces played a major role in the binding of ruthenium complex to proteins. The process of binding was a spontaneous process in which Gibbs free energy change was negative.

Wood is an extremely complex biological material, which can show macroscopic similarities that make it difficult to discriminate between species. Discrimination between similar wood species can be achieved by either anatomic or instrumental methods, such as near infrared spectroscopy (NIR). Although different spectroscopy methods are currently available, few studies have applied them to discriminate between wood species. In this study, we applied a partial least squares-discriminant analysis (PLS-DA) model to evaluate the viability of using direct fluorescence measurements for discriminating between Eucalyptus grandis, Eucalyptus urograndis, and Cedrela odorata. The results show that molecular fluorescence is an efficient technique for discriminating between these visually similar wood species. With respect to calibration and the validation samples, we observed no misclassifications or outliers.

Two simple and efficient procedures have been developed for the rapid simultaneous determination of compounds with mutual spectral interference (rifampicin (RIF) and isoniazid (INH)). The first method was based on the UV–Vis spectral signal (190–600 nm) of synthetic RIF and INH aqueous solutions, whereas the second method involved the visible spectral signal registered between 350 and 800 nm after the reaction of INH with a Cu2+/neocuproine complex. Both multivariate spectrophotometric methods show excellent prevision capacity, providing results that are statistically equivalent with those provided by the standard chromatographic procedure. The methods were validated according to criteria established by ANVISA, showing precision, accuracy and robustness compatible with the requirements for new analytical methods, additionally allowing the reduction of waste generation.

No presente trabalho desenvolveu-se metodologia simplificada para análise de hexametilenotetramina em medicamentos que apresentam corantes em sua formulação (azul de metileno, Ponceau Red, Bordeaux Red, acriflavina) seguida por titulação ácido-base usando indicador. O método foi aplicado com sucesso a amostras comerciais e os resultados obtidos foram comparados com aqueles encontrados por titulação potenciométrica, apresentando muito boa concordância. Recobrimentos situaram-se na faixa de 101,3 - 104,2%, com desvios padrão de 0,1 - 2,1.

Neste estudo foi desenvolvido uma metodologia para determinação de Ni(II) em amostras de água usando a extração em fase sólida (SPE) em um sistema por injeção em fluxo (FI) e detecção por espectrometria de absorção atômica em chama (F AAS). O adsorvente utilizado para a extração e pré-concentração do Ni(II) foi a sílica gel modificada com óxido de nióbio(V). Variáveis químicas e de fluxo do sistema em linha foram otimizadas usando planejamento fatorial completo (N = 2k + 3). As condições iniciais do sistema FI-F AAS foram volume de amostra de 10 mL e concentração de Ni(II) de 100 µg L-1. O tampão Sörensen foi selecionado neste estudo. A resposta analítica utilizada foi absorvância integrada. Após a otimização foram obtidos os parâmetros analíticos de mérito: faixa linear de trabalho estudada de 5-100 µg L-1; R = 0.9999; RSD = 1,5% (35 µg L-1, n = 7); limite de detecção de 0,8 µg L-1; limite de quantificação de 2,7 µg L-1 e fator de enriquecimento de 92,25. Foram analisadas amostras de água do rio Araranguá e a da rede de abastecimento da cidade de Florianópolis, ambas do estado de Santa Catarina. As duas amostras não apresentaram concentração de níquel acima do limite de detecção e após fortificação das mesmas os valores de recuperação foram na faixa de 100,2 a 103,8%.
