This study investigated the effects of stocking density on the growth and fatty acid (FA) of Brycon insignis metabolism. Fingerlings (360) were distributed into eight ponds at two stocking densities (105 and 210 g/m(3)). The analysis of growth showed that the condition factor (K) and the coefficient of variation (CV) for body mass were not affected by stocking density. However, final body mass and length, specific growth rate (SGR), and weight gain (WG) were higher in the low stocking density group, which also presented a higher feed efficiency (FE) and survival (S). By contrast, muscle protein levels were higher in the high stocking density group. The plasma and muscle lipid content were not affected by stocking density, but fish reared at lower stocking density presented higher lipid concentration in the liver, with no differences in hepatosomatic index values. Even with the differences observed in metabolic and growth parameters, plasma cortisol was not affected by stocking density. The FA profile in the muscle and liver neutral fraction were not affected by stocking density, but the FA in the polar fractions differed between the two stocking densities. In the liver, total polyunsaturated fatty acids (PUFA) and PUFA n - 3 increased in higher stocking density, mainly due to an increase in docosahexaenoic acid (DHA). In addition, PUFA n - 6 were also increased in the higher stocking density group, mainly due to an increase in arachidonic acid (AA) and docosadienoic acid (22:2n - 6). In the muscle polar fraction, the saturated fatty acids (SFA) and monounsaturated fatty acids (MUFA) decreased in the animals from the higher stocking density group, and this reduction was compensated by an increase in PUFA n - 3 and PUFA n - 6, mainly the FA with 20-22 carbons (20:4n - 6: 22:4:n - 6; 22:5n - 6, 22:5n - 3, and 22:6n - 3). A different profile was observed for the C18 PUFAs, mainly 18:2n - 6 and 18:4n - 6, which were higher in the lower density stocking group. The data suggest that when living in high stocking density, B. insignis differentially utilizes the hepatic lipids as energy source and remodels the membrane fatty acids, with higher amounts of DHA in the polar muscle fraction compensated for by a decrease in MUFA. Fructose-1,6-bisphosphatase (FBPase) is a gluconeogenic enzyme that is upregulated in islets or pancreatic beta-cell lines exposed to high fat. However, whether specific beta-cell upregulation of FBPase can impair insulin secretory function is not known. The objective of this study therefore is to determine whether a specific increase in islet beta-cell FBPase can result in reduced glucose-mediated insulin secretion.

To test this hypothesis, we have generated three transgenic mouse lines overexpressing the human FBPase (huFBPase) gene specifically in pancreatic islet beta-cells. In addition, to investigate the biochemical mechanism by which elevated FBPase affects insulin secretion, we made two pancreatic beta-cell lines (MIN6) stably overexpressing huFBPase.

FBPase transgenic mice showed reduced insulin secretion in response to an intravenous glucose bolus. Compared with the untransfected parental MIN6, FBPase-overexpressing cells showed a decreased cell proliferation rate and significantly depressed glucose-induced insulin secretion. These defects were associated with a decrease in the rate of glucose utilization, resulting in reduced cellular ATP levels.

Taken together, these results suggest that upregulation of FBPase in pancreatic islet beta-cells, as occurs in states of lipid oversupply and type 2 diabetes, contributes to insulin secretory dysfunction.

As ataxias espinocerebelares (SCAs) constituem um grupo de doenças neurodegenerativas fatais que apresentam uma grande heterogeneidade clínica. A doença de Machado-Joseph (DMJ), ou ataxia espinocerebelar tipo 3 (SCA3), é causada por uma expansão de uma seqüência repetitiva CAG em um gene, denominado MJD1, localizado no braço longo do cromossomo 14, expansão codificadora de uma seqüência poliglutamínica constituinte da proteína ataxina 3. Indivíduos normais apresentam entre 12 a 41 repetições, enquanto indivíduos afetados apresentam 61 a 84 repetições CAGs neste gene. Este trabalho teve como objetivos principais a padronização de metodologias moleculares para o identificação e a quantificação do número de repetições CAG no gene responsável pela da DMJ. Um grupo de 112 pacientes, pertencentes a 77 famílias, com suspeita clínica de algum tipo de ataxia espinocerebelar foi avaliado no Hospital de Clínicas de Porto Alegre. Após a extração de DNA destes pacientes, este material foi amplificado por PCR utilizando oligonucleotídeos iniciadores específicos para a região de interesse e posterior transferência destes fragmentos (1) para uma membrana de nylon pelo método de Southern blot, visando ao estabelecimento de um protocolo não-radioativo para detectar a presença do alelo normal e/ou mutante; e (2) análise em gel de poliacrilamida para quantificação do número de repetições presentes no alelo mutante. As análises laboratoriais identificaram um total de 77 pacientes com uma expansão CAG no gene da MJD1. Considerando-se apenas indivíduos não relacionados, a freqüência encontrada foi de 61% (47 indivíduos). Os protocolos estabelecidos demonstraram-se bastante eficazes e sensíveis para o diagnóstico da DMJ e quantificação do alelo expandido da respectiva doença.

Orius insidiosus (SAY, 1832) is an important predator for biological pest control programs. The objective of this work was to evaluate the functional response, predation capacity and reproductive aspects of O. insidiosus when predating 1, 5, 10, 15, and 20 third/fourth instar nymphs of Aphis gossypii. The tests were conducted under laboratory conditions. Females of the predator were individually maintained in Petri dishes with a cotton leaf at each one of the aphid densities. The average rate of predation was of 0.93 [plus or minus] 0.13, 3.58 [plus or minus] 0.80, 7.31 [plus or minus] 1.45, 7.40 [plus or minus] 1.16, and 10.54 [plus or minus] 1.84 at the densities of 1, 5, 10, 15, and 20 aphids, respectively. The results showed an attack rate of 0.10 h-1 and 1.82 h of manipulation time. The predator spent 3.10 to 4.08 h feeding on leaf cotton glands; this behavior was not directly influenced by the prey density. The proportion of egg laying per female grew up to the density of 10 nymphs whereas the number of eggs per laying increased with the number of available preys.

The objective of this research was to determine the survival, reproduction and predaceous capacity of Orius insidiosus on cotton aphid Aphis gossypii. Cotton plants of Antares, CNPA7H and Acala 90 cultivars (respectively, without tricome, medium tricome density and high tricome density) were individualized and infested with 15 third/fourth instar nymphs of A. gossypii, and then first-instar nymphs of O. insidiosus were released on the plants. The evaluations were made daily, quantifying survival and nymphal development; the number of cotton aphids A. gossypii per day and total; the number of eggs and the population of the predator (number of insects by female); and the conversion efficiency of A. gossypii predaceous. The nymphal development did not differ according to the cotton cultivars. The Antares cultivar favored a higher daily predation rate for the 1st, 2nd and 4th instars and adults of O. insidiosus. The number of eggs and nymphs was smaller when O. insidiosus females were confined on the Acala 90 cultivar. According to the predation rate of O. insidiosus, the efficiency of alimentary conversion was determined for the Antares, CNPA7H and Acala 90 cultivars, being respectively, 4.28, 3.00 and 2.75 cotton aphid predation for each egg of the predator.

It was aimed to evaluate the influence of fungicides utilized in cucumber on the development of the aphid Aphis gossypii Glover under greenhouse conditions. The experimental design was completely randomized with four treatments and 45 replicates, being each plot consisting of a first instar nymph coming from an apterous female in early reproductive period. The fungicides utilized and their respective dosages at g i.a.L(-1) were 800 PM sulphur (1.6), 800 PM mancozeb (1.6) and 840 PM copper oxichloride (1.5). The control was made up of water only. The products did not influence the median time of life in each instar and the nymphal stage, being respectively 1 and 4 days. Copper oxychloride and mancozeb increased the median time of life in days and the total life cycle of the adult stage of this species.

Este trabalho teve como objetivo estudar os aspectos biológicos de A. gossypii em três cultivares de algodoeiro e em três espécies de plantas daninhas. O experimento foi conduzido no Laboratório de Controle Biológico, sob condições controladas de temperatura (25 ± 1ºC), umidade relativa do ar (70 ± 10%) e fotofase (12 horas). Foram utilizados seis tratamentos, correspondendo às cultivares de algodoeiro CNPA ITA 90, Coodetec 402 e Deltaopal e espécies de plantas daninhas: guanxuma (Sida santaremnensis), malva-preta (Sidastrum micranthum) e trapoeraba (Commelina benghalensis). As unidades experimentais (25 repetições) consistiram de placas de Petri contendo ágar-água a 1% solidificado. Nestas placas, os ofídeos foram mantidos individualmente sobre discos foliares (3cm de diâmetro) de seus respectivos tratamentos. Os menores valores, em dias, para a fase ninfal foram obtidos em CNPA ITA 90 (4,39 ± 0,74), Deltaopal (4,45 ± 0,75), Coodetec 402 (4,47 ± 0,75) e trapoeraba (4,75 ± 0,80). O período reprodutivo e a longevidade dos adultos variaram de 8,21 ± 0,85 a 15,52 ± 0,93 e 11,09 ± 0,97 a 24,33 ± 1,59 dias, respectivamente, sobre guanxuma e Deltaopal. As cultivares de algodoeiro são mais favoráveis a A. gossypii que as plantas daninhas e na ausência de plantas de algodoeiro, as plantas daninhas guanxuma, trapoeraba e malva-preta são hospedeiros potenciais para manutenção da população de A. gossypii.

The larval development of Pyromaia tuberculata (Lockington, 1877) is described from specimens reared in the laboratory. Larvae were obtained from ovigerous females collected by trawl in Ubatuba, São Paulo state, Brazil. Rearing was carried out at 24 +/- 1 degrees C in 35 parts per thousand S. The larvae pass through two zoeal stages before moulting to the megalopa. A comparison is made with previously studied majid species occurring in the southern and southeastern regions of Brazil.

This paper outlines the results of a study into the biological aspects of Aphis gossypii Glöver on colored cotton cultivars through the construction of life tables. In addition, we evaluate the influence of density of gossypol glands and trichomes of leaves on the biology of the aphid. Tests were conducted in a climate chamber at 25 ± 2°C, with relative humidity (RH) at 70 ± 10%, and a photophase of 12 hours, using the cultivars BRS Rubi, BRS Safira, and BRS Verde. Newly hatched nymphs were individually isolated in petri dishes containing leaf discs of cotton cultivars on a layer of water-agar (1%) of approximately 5 mm thick. An evaluation of the trichomes and gossypol gland densities of the cotton leaf plants was performed under a stereomicroscope, delimited to an area of 1 cm², and a count and identification of those on the surface was made. The feed substrates that were evaluated influenced the nymphal stage of A. gossypii. BRS Verde provided the shortest duration and BRS Safira was the longest during this phase. The cultivar BRS Verde, with the lowest density of trichomes, provided a large number of nymphs and led to a higher net reproductive rate (R0). Given these results, conclusions can be drawn that the colored cotton cultivars influence the duration of nymphal and adult stages of A. gossypii. The cultivar with a high density of trichomes on leaves (BRS Safira) adversely affects the fertility parameters of A. gossypii.

Mesoclemmys heliostemma (Testudines: Chelidae) was described based on five vouchered specimens and nine live specimens from the western Amazon basin. Some authors questioned its status as a valid species, suggesting that it represents a junior synonym of M. raniceps. Here, we report on eight additional specimens from eastern Peru and northern Brazil, and provide descriptive statistics of morphological characters for hatchlings, juveniles, and adults of M. heliostemma, M. raniceps, and M. gibba. We also test for group differences through univariate and multivariate statistical analyses, and discuss some advantages of this methodology. Our data suggest that all three taxa are morphologically divergent, and that M. heliostemma is a valid species.
