The adsorption of three aromatic compounds on to an untreated carbon was investigated. The solution pH was lowered in all experiments so that all the solutes were in their molecular forms. It was shown that the difference in the maximum adsorption of the solutes was mainly a result of the difference in the sizes of the molecules and their functional groups. Further-more, it was illustrated that the packing arrangement was most likely edge-to-face (sorbate-sorbent) with various tilt angles. On the other hand, the affinity and heterogeneity of the adsorption systems were apparently related to the pK(a) values of the solutes.

The MUC1 mucin (CD227) is a cell surface mucin originally thought to be restricted to epithelial tissues. We report that CD227 is expressed on human blood dendritic cells (DC) and monocyte-derived DC following in vitro activation. Freshly isolated murine splenic DC had very low levels of CD227; however, all DC expressed CD227 following in vitro culture. In the mouse spleen, CD227 was seen on clusters within the red pulp and surrounding the marginal zone in the white pulp. Additionally, we confirm CD227 expression by activated human T cells and show for the first time that the CD227 cytoplasmic domain is tyrosine-phosphorylated in activated T cells and DC and is associated with other phosphoproteins, indicating a role in signaling. The function of CD227 on DC and T cells requires further elucidation.

Protease-activated receptors type 2 (PAR2) are activated by serine proteases like trypsin and mast cell tryptase. The function and physiological significance of PAR2 receptors is poorly understood, but recent studies suggest a role during inflammatory processes in both airways and intestine. PAR2 receptors are also likely to participate in the control of ion transport in these tissues. We demonstrate that stimulation of PAR2 in airways and intestine significantly enhanced ion transport. Trypsin induced CI- secretion in both airways and intestine when added to the basolateral but not to the luminal side of these tissues. In both airways and intestine, stimulation of ion transport was largely dependent on the increase in intracellular Ca2+. Effects of trypsin were largely reduced by basolateral bumetanide and barium and by trypsin inhibitor. Thrombin, an activator of proteinase-activated receptors types 1, 3, and 4 had no effects on equivalent short-circuit current in either airways or intestine. Expression of PAR2 in colon and airways was further confirmed by reverse transcription-polymerase chain reaction. We postulate that these receptors play a significant role in the regulation of electrolyte transport, which might be important during inflammatory diseases of airways and intestine.

Proteinase-activated receptor (PAR) type 2 (PAR-2) has been shown to mediate ion secretion in cultured epithelial cells and rat jejunum. With the use of a microUssing chamber, we demonstrate the role of PAR-2 for ion transport in native human colonic mucosa obtained from 30 normal individuals and 11 cystic fibrosis (CF) patients. Trypsin induced Cl- secretion when added to the basolateral but not luminal side of normal epithelia. Activation of Cl- secretion by trypsin was inhibited by indomethacin and was further increased by cAMP in normal tissues but was not present in CF colon, indicating the requirement of luminal CF transmembrane conductance regulator. Effects of trypsin were largely reduced by low Cl-,by basolateral bumetanide, and in the presence of barium or clotrimazole, but not by tetrodotoxin. Furthermore, trypsin-induced secretion was inhibited by the Ca2+-ATPase inhibitor cyclopiazonic acid and in low-Ca2+ buffer. The effects of trypsin were almost abolished by trypsin inhibitor. Thrombin, an activator of PAR types 1, 3, and 4, had no effects on equivalent short-circuit currents. The presence of PAR-2 in human colon epithelium was confirmed by RT-PCR and additional experiments with PAR-2-activating peptide. PAR-2-mediated intestinal electrolyte secretion by release of mast cell tryptase and potentiation of PAR-2 expression by tumor necrosis factor-alpha may contribute to the hypersecretion observed in inflammatory processes such as chronic inflammatory bowel disease.

Complete biological nutrient removal (BNR) in a single tank, sequencing batch reactor (SBR) process, is demonstrated here at full-scale on a typical domestic wastewater. The unique feature of the UniFed process is the introduction of the influent into the settled sludge blanket during the settling and decant periods of the SBR operation. This achieves suitable conditions for denitrification and anaerobic phosphate release which is critical to successful biological phosphorus removal, It also achieves a selector effect, which helps in generating a compact, well settling biomass in the reactor. The results of this demonstration show that it is possible to achieve well over 90% removal of GOD, nitrogen and phosphorus in such a process. Effluent quality achieved over a six-month operating period directly after commissioning was: 29 mg/l GOD, 0.5 mg/l NH4-N, 1.5 mg/l NOx-N and 1.5 mg/l PO4-P (50%-iles of daily samples). Lipid homeostasis is controlled by the peroxisome proliferator-activated receptors (PPARalpha, -beta/delta, and -gamma) that function as fatty acid-dependent DNA-binding proteins that regulate lipid metabolism. In vitro and in vivo genetic and pharmacological studies have demonstrated PPARalpha regulates lipid catabolism. In contrast, PPARgamma regulates the conflicting process of lipid storage. However, relatively little is known about PPARbeta/delta in the context of target tissues, target genes, lipid homeostasis, and functional overlap with PPARalpha and -gamma. PPARbeta/delta, a very low-density lipoprotein sensor, is abundantly expressed in skeletal muscle, a major mass peripheral tissue that accounts for approximately 40% of total body weight. Skeletal muscle is a metabolically active tissue, and a primary site of glucose metabolism, fatty acid oxidation, and cholesterol efflux. Consequently, it has a significant role in insulin sensitivity, the blood-lipid profile, and lipid homeostasis. Surprisingly, the role of PPARbeta/delta in skeletal muscle has not been investigated. We utilize selective PPARalpha, -beta/delta, -gamma, and liver X receptor agonists in skeletal muscle cells to understand the functional role of PPARbeta/delta, and the complementary and/or contrasting roles of PPARs in this major mass peripheral tissue. Activation of PPARbeta/delta by GW501516 in skeletal muscle cells induces the expression of genes involved in preferential lipid utilization, beta-oxidation, cholesterol efflux, and energy uncoupling. Furthermore, we show that treatment of muscle cells with GW501516 increases apolipoprotein-A1 specific efflux of intracellular cholesterol, thus identifying this tissue as an important target of PPARbeta/delta agonists. Interestingly, fenofibrate induces genes involved in fructose uptake, and glycogen formation. In contrast, rosiglitazone-mediated activation of PPARgamma induces gene expression associated with glucose uptake, fatty acid synthesis, and lipid storage. Furthermore, we show that the PPAR-dependent reporter in the muscle carnitine palmitoyltransferase-1 promoter is directly regulated by PPARbeta/delta, and not PPARalpha in skeletal muscle cells in a PPARgamma coactivator-1-dependent manner. This study demonstrates that PPARs have distinct roles in skeletal muscle cells with respect to the regulation of lipid, carbohydrate, and energy homeostasis. Moreover, we surmise that PPARgamma/delta agonists would increase fatty acid catabolism, cholesterol efflux, and energy expenditure in muscle, and speculate selective activators of PPARbeta/delta may have therapeutic utility in the treatment of hyperlipidemia, atherosclerosis, and obesity.

Conventional methods to determine surface diffusion of adsorbed molecules are proven to be inadequate for strongly adsorbing vapors on activated carbon. Knudsen diffusion permeability (B-k) for strongly adsorbing vapors cannot be directly estimated from that of inert gases such as helium. In this paper three models are considered to elucidate the mechanism of surface diffusion in activated carbon. The transport mechanism in all three models is a combination of Knudsen diffusion, viscous flow and surface diffusion. The collision reflection factor f (which is the fraction of molecules undergoing collision to the solid surface over reflection from the surface) of the Knudsen diffusivity is assumed to be a function of loading. It was found to be 1.79 in the limit of zero loading, and decreases as loading increases. The surface diffusion permeability increases sharply at very low pressures and then starts to decrease after it has reached a maximum (B(mum)s) at a threshold pressure. The initial rapid increase in the total permeability is mainly attributed to surface diffusion. Interestingly the B(mum)s for all adsorbates appear at the same volumetric adsorbed phase concentration, suggesting that the volume of adsorbed molecules may play an important role in the surface diffusion mechanism in activated carbon. An integrated mathematical model for the kinetics of multicomponent adsorption on microporous carbon was developed. Transport in this bidisperse solid is represented by balance equations in the macropore and micropore phases, in which gas-phase diffusion dominates the mass transfer in the macropores, with the phenomenological diffusivities represented by the generalized Maxwell-Stefan (GMS) formulation. Viscous flow also contributes to the macropore fluxes and is included in the MS expressions. Diffusion of the adsorbed phase controls the mass transfer in the micro ore phase, p which is also described in a similar way by the MS method. The adsorption isotherms are represented by a new heterogeneous modified vacancy solution theory formulation of adsorption, which has proved to be a robust method for adsorption on activated carbons. The model is applied to the coadsorption and codesorption of C2H6 and C3H8 on Ajax and Norit carbon, as well as the displacement on Ajax carbon. The effect of the viscous flow in the macropore phase is not significant for the cases studied. The model accurately predicts the overshoot behavior and rollup of C2H6 during coadsorption. The prediction for the heavier compound C3H8 is always satisfactory, though at higher C3H8 mole fraction, the overshoot extent of C2H6 is overpredicted, possibly due to neglect of heat effects.

The different methods of sewage sludge stabilization modify their physical chemical and biological properties, altering its efficiency when applied in agriculture. The objective of this study was to evaluate the nutrient levels in soil and the yield of sunflower fertilized with sewage sludge stabilized by different processes. The experiment was conducted in Cambisol, with the treatments: control (without fertilization), fertilization with sewage sludge solarized, composted, vermicomposted, limed and chemical fertilizer recommended for sunflower crop. The experimental design a randomized block with four replications. The different methods of sewage sludge treatment did not affect the yield; however, the application of sewage sludge, regardless the stabilization process adopted, was more effective than chemical fertilizer and the control treatment. Overall, fertilization with limed sewage sludge provided higher soil nutrients concentrations, while treatments with composted and vermicomposted sewage sludge showed higher levels of nutrients in the plant.

O presente trabalho teve como principais objectivos, estudar e optimizar o processo de tratamento do efluente proveniente das máquinas da unidade Cold-press da linha de produção da Empresa Swedwood, caracterizar a solução límpida obtida no tratamento e estudar a sua integração no processo, e por fim caracterizar o resíduo de pasta de cola obtido no tratamento e estudar a possível valorização energética deste resíduo. Após caracterização inicial do efluente e de acordo com os resultados de um estudo prévio solicitado pela Empresa Swedwood a uma empresa externa, decidiu-se iniciar o estudo de tratabilidade do efluente pelo processo físico-químico a coagulação/floculação. No processo de coagulação/floculação estudou-se a aplicabilidade, através de ensaios Jar-test, dos diferentes agentes de coagulação/floculação: a soda cáustica, a cal, o cloreto férrico e o sulfato de alumínio. Os melhores resultados neste processo foram obtidos com a adição de uma dose de cal de 500 mg/Lefluente, seguida da adição de 400 mg/Lefluente de sulfato de alumínio. Contudo, após este tratamento o clarificado obtido não possuía as características necessárias para a sua reintrodução no processo fabril nem para a sua descarga em meio hídrico. Deste modo procedeu-se ao estudo de tratamentos complementares. Nesta segunda fases de estudo testaram-se os seguintes os tratamentos: a oxidação química por Reagente de Fenton, o tratamento biológico por SBR (sequencing batch reactor) e o leito percolador. Da análise dos resultados obtidos nos diferentes tratamentos conclui-se que o tratamento mais eficaz foi o tratamento biológico por SBR com adição de carvão activado. Prevê-se que no final do processo de tratamento o clarificado obtido possa ser descarregado em meio hídrico ou reintroduzido no processo. Como o estudo apenas foi desenvolvido à escala laboratorial, seria útil poder validar os resultados numa escala piloto antes da sua implementação industrial. A partir dos resultados do estudo experimental, procedeu-se ao dimensionamento de uma unidade de tratamento físico-químico e biológico à escala industrial para o tratamento de 20 m3 de efluente produzido na fábrica, numa semana. Dimensionou-se ainda a unidade (leito de secagem) para tratamento das lamas produzidas. Na unidade de tratamento físico-químico (coagulação/floculação) os decantadores estáticos devem possuir o volume útil de 4,8 m3. Sendo necessários semanalmente 36 L da suspensão de cal (Neutrolac 300) e 12,3 L da solução de sulfato de alumínio a 8,3%. Os tanques de armazenamento destes compostos devem possuir 43,2 litros e 96 litros, respectivamente. Nesta unidade estimou-se que são produzidos diariamente 1,4 m3 de lamas. Na unidade de tratamento biológico o reactor biológico deve possuir um volume útil de 6 m3. Para que este processo seja eficaz é necessário fornecer diariamente 2,1 kg de oxigénio. Estima-se que neste processo será necessário efectuar a purga de 325 litros de lamas semanalmente. No final da purga repõe-se o carvão activado, que poderá ser arrastado juntamente com as lamas, adicionando-se 100 mg de carvão por litro de licor misto. De acordo com o volume de lamas produzidos em ambos os tratamentos a área mínima necessária para o leito de secagem é de cerca de 27 m2. A análise económica efectuada mostra que a aquisição do equipamento tem o custo de 22.079,50 euros, o custo dos reagentes necessários neste processo para um ano de funcionamento tem um custo total de 508,50 euros e as necessidades energéticas de 2.008,45 euros.

O presente trabalho experimental teve como objectivos estudar a tratabilidade dum efluente lácteo utilizando a técnica de coagulação/floculação e avaliar a possibilidade de utilização do carvão activado granulado (CAG) Aquasorb 2000 como adsorvente para a remoção de compostos orgânicos presentes nos efluentes lácteos pré–tratados por coagulação/floculação, funcionando como um tratamento de polimento. No estudo da tratabilidade do efluente por coagulação/floculação investigou-se a influência de determinadas variáveis como o tipo e dose de coagulante e pH, a fim de encontrar as melhores condições operatórias. A utilização da referida técnica visou a redução do valor de concentração de alguns parâmetros: carência química de oxigénio (CQO); fósforo total e turvação, tendo sido utilizados efluentes desta indústria recolhidos em diferentes datas e após tratamento biológico, designados por A, B, C e D apresentando características diferentes. Sendo que o efluente A apresentava valores de CQO, fósforo total e turvação de 500 mg O2/L;32 mg P/L e 40 NTU respectivamente, o efluente B 1400 mg O2/L; 120 mg P/L e 80 NTU respectivamente, o efluente C 12300 mg O2/L; 87 mg P/L e 350 NTU respectivamente e o efluente D 340 mg O2/L; 33 mg P/L e 42 NTU respectivamente. Os coagulantes estudados foram hidróxido de cálcio (HC), sulfato de alumínio (SA) e tricloreto de ferro (TF). Verificou-se que o coagulante com maior eficácia nos efluentes estudados foi o TF. As maiores remoções de CQO, fósforo total e turvação, 89%, 99,9% e 99%, respectivamente, foram obtidas para o efluente C, com uma dosagem de TF de 4 g/L e com um pH entre 6 e 7. Entre os efluentes estudados este era o que apresentava valores iniciais mais elevados para qualquer um destes parâmetros. As melhores percentagens de remoção obtidas com o HC, para a CQO, fósforo total e turvação foram de 59%, 99% e 91%, respectivamente, com uma dosagem de HC de 1 g/L e com um pH entre 10 e 11,5 foram conseguidas no tratamento do efluente D, com o qual se alcançaram também as melhores remoções de CQO, fósforo total e turvação de 65%, 99% e 87%, respectivamente, quando se utilizou o coagulante SA, com uma dosagem de 2 g/L e com um pH entre 7 e 7,5. Relativamente ao volume de lamas produzido neste processo pela utilização dos diferentes coagulantes no tratamento dos efluentes referidos concluiu-se que o coagulante que gera menor volume de lamas é o HC, sendo o SA aquele que origina um maior volume. Submeteu-se posteriormente o efluente D, pré-tratado por coagulação/floculação, a um processo de adsorção em batch utilizando o CAG Aquasorb 2000, onde se conseguiu uma remoção de CQO de 48%, alcançando para este parâmetro o valor de 63 mg O2/L, nas condições operatórias que correspondem a uma massa de CAG de 12,5 g/L e um tempo de contacto de 3 horas. Quanto aos custos associados com os coagulantes, o que menores custos apresenta é o HC (150 €/ton), seguido pelo TF (250 €/ton) e por ultimo o SA (340 €/ton). Sendo que o efluente quando tratado com TF e SA é necessário uma correcção do pH do meio para que estes coagulantes actuem eficazmente, em que essa correcção de pH é realizada com hidróxido de sódio (540 €/ton). Realizaram-se ainda estudos de equilíbrio de adsorção com o carvão activado referido e o azul-de-metileno usando diferentes concentrações deste (50 mg/L; 100 mg/L e 200 mg/L) e diferentes massas de CAG (0,1g; 0,2g; 0,3g; 0,4g e 0,5g). A temperatura a que se realizaram estes ensaios foi de 28,7ºC e o volume de azul-de-metileno foi de 200 mL. Verificou-se que os melhores resultados obtidos foram para uma concentração de adsorvato de 100 mg/L. Ajustaram-se os modelos de Langmuir e Freundlich às isotérmicas obtidas tendo correlações mais elevadas para a concentração de 100 mg/L de corante (azul de metileno), sendo o modelo de Freundlich aquele que melhor se ajustou apresentando uma correlação quadrática de 0,9744 e os seguintes parâmetros Kf = 6,59 e n = 5,33, enquanto que o de Langmuir apresentou uma correlação quadrática de 0,9583 e os seguintes parâmetros qmáx = 83,3 mg/g de adsorvente e K = 20 L/mg de adsorvato.. Verificou-se que a capacidade de adsorção promovida pelo CAG, em relação ao azul-demetileno, obtida experimentalmente, 83,3 mg/g, é muito inferior à capacidade de adsorção teoricamente prevista pela ficha técnica deste carvão, 280 mg/g a uma temperatura de 25ºC, o que pode indiciar que o carvão utilizado não estaria nas melhores condições.

Studies were undertaken to determine the adsorption behavior of α-cypermethrin [R)-α-cyano-3-phenoxybenzyl(1S)-cis- 3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropanecarboxylate, and (S)-α-cyano-3-phenoxybenzyl (1R)-cis-3-(2,2-dichlorovinyl)-2,2- dimethylcyclopropanecarboxylate] in solutions on granules of cork and activated carbon (GAC). The adsorption studies were carried out using a batch equilibrium technique. A gas chromatograph with an electron capture detector (GC-ECD) was used to analyze α-cypermethrin after solid phase extraction with C18 disks. Physical properties including real density, pore volume, surface area and pore diameter of cork were evaluated by mercury porosimetry. Characterization of cork particles showed variations thereby indicating the highly heterogeneous structure of the material. The average surface area of cork particles was lower than that of GAC. Kinetics adsorption studies allowed the determination of the equilibrium time—24 hours for both cork (1–2 mm and 3–4 mm) and GAC. For the studied α-cypermethrin concentration range, GAC revealed to be a better sorbent. However, adsorption parameters for equilibrium concentrations, obtained through the Langmuir and Freundlich models, showed that granulated cork 1–2 mm have the maximum amount of adsorbed α-cypermethrin (qm) (303 μg/g); followed by GAC (186 μg/g) and cork 3-4 mm (136 μg/g). The standard deviation (SD) values, demonstrate that Freundlich model better describes the α-cypermethrin adsorption phenomena on GAC, while α-cypermethrin adsorption on cork (1-2 mm and 3-4 mm) is better described by the Langmuir. In view of the adsorption results obtained in this study it appears that granulated cork may be a better and a cheaper alternative to GAC for removing α-cypermethrin from water.

Characteristics of tunable wavelength pi'n/pin filters based on a-SiC:H multilayered stacked cells are studied both experimentally and theoretically. Results show that the device combines the demultiplexing operation with the simultaneous photodetection and self amplification of the signal. An algorithm to decode the multiplex signal is established. A capacitive active band-pass filter model is presented and supported by an electrical simulation of the state variable filter circuit. Experimental and simulated results show that the device acts as a state variable filter. It combines the properties of active high-pass and low-pass filter sections into a capacitive active band-pass filter using a changing capacitance to control the power delivered to the load.
