Introduction: Scientific evidence indicates that neonatal exposure to ototoxic drugs cause hearing loss in newborns. Objective: To characterize the use of ototoxic antibiotics in newborns (NB), treated in the Neonatal Intensive Care Unit (NICU) and evaluate possible hearing modifications. Methods: A descriptive cross-sectional quantitative approach, using data from medical records of infants who were at some time in the NICU and used antibiotics, including ototoxic, from January to June 2004 as much as 2010, and the data were compared and analyzed. Parents/guardians of infants born in 2004 were contacted and applied a questionnaire containing questions about the children’s hearing. These children were submitted to audiological evaluation. Results: There was significant reduction in the time of use, the amount of antibiotics prescribed to newborns and Vancomycin prescription in 2010 compared to 2004. The hearing tests of 13 born in 2004 showed: sensorineural hearing loss in only 2 (one with moderate hearing loss and descending configuration in pure tone audiometry and the other with bilateral cochlear impairment); audiometric thresholds within the normal range in 11 patients, and the presence of otoacoustic emissions in 9. In Evoked Auditory Brainstem Response (ABR) no changes were observed. Conclusion: The reduction in the time of use, the amount and types of antibiotics observed may be related to the adoption of a Protocol in 2008, by the service. In contrast, auditory alterations may be related to a neonatal exposure to antibiotics in 2004.

Due to the complexity and instability of clinical conditions of ICU patients, the drug therapy applied in this type of environment requires a combination of several prescribed drugs, which is a favorable condition for drug interaction, toxic synergism and possible iatrogenia. In the possible universe of ICU occurrences, this study aimed at identifying and evaluating the incidence of adverse events in drug therapy at the Intensive Care Therapy Service (SETI) in wards I and II. It is a cross-sectional, descriptive, prospective and quantitative study conducted from August to September, 2011 in the Intensive Care Service of the Botucatu School of Medicine University Hospital - UNESP. The population consisted of fifteen clinical nurses, including those in the Improvement and Volunteer Internship Programs, who contributed to the investigation after signing an informed consent form and according to approval by the Research Ethics Committee number 10711/CE - FMB. The data were entered on a form and analyzed. Results showed that, on average, 8.9% of events/day occurred, and the highest frequency was observed on August 04, 2011. 63% and 22% were respectively observed in the morning and afternoon shifts, and 15% in the night shift. 48% of these were due to administration time errors, followed by drug prescription and dispensation errors, with percentages of 22% and 18%, respectively. Antibiotics showed the highest frequency of adverse events - 18%, which was followed by 13% for anticoagulant, 11% for antiemetic and 10% for antiulcerative drugs. As regards the occurrence of adverse events related to hospitalization time, the highest frequency occurred in patients who were hospitalized for 10 days. Photodynamic therapy, term introduced by von Tapeiner in 1900, can be defined as the administration of a non toxic drug, i.e., a dye, known as photosensitizer (FS), which subsequently will be illuminated with light of specific wavelength. PDT is based on the interaction among FS, oxygen and light, which through photochemical reactions cause cell death. The FS molecules must have a high probability to form the singlet state after the excitation, which can induce chemical changes in the neighborhood in two ways, called reactions type I and type II. The type II reaction is based on the exchange of energy to molecular oxygen, exciting it to its state of higher energy (singlet), which is highly reactive. The proposed mechanisms for cell death are linked to damage to the DNA, mitochondria and to the cytoplasmic membrane. Several pre‐clinical and clinical trials have been carried out and the PDT is already used in many countries for treatment mainly against certain types of cancer. The therapy also has been gaining strength in antimicrobial control, since the microorganisms have appeared increasingly resistant to current antibiotics. Another attempt to use the PDT is for the inactivation of macro‐organisms, such as micro‐crustaceans and mosquitoes. To this end I tested whether the photosensitizers methylene blue, rose Bengal and the chlorophyll a has insecticidal activity against the yellow fever and dengue vector mosquito, Aedes aegypti. Since these diseases have no effective treatments, its control is linked to the vector control, which has shown resistance to chemical pesticides used. Based on this, this work shows its importance, because it is a new type of mosquito control since all the photosensitizers used are low cost, do not generate toxic products at the concentrations used and showed good results in mortality. Atualmente o uso indiscriminado e não criterioso de antibióticos está causando um aumento alarmante de resistência bacteriana chegando a ser considerado um problema de saúde pública. Nesse contexto, a utilização do peptídeo antimicrobiano (PAM) Hylina-a1 proveniente de uma rã da fauna brasileira está sendo estudado. Sua atuação na membrana celular dificulta a ocorrência de resistência bacteriana. No entanto, o uso de PAMs ainda apresenta problemas principalmente relacionados à disponibilidade, eficácia e toxicidade. A fim de contornar esses problemas, este trabalho teve como objetivo principal avaliar a incorporação deste peptídeo a um sistema líquido cristalino nanoestruturado com capacidade de regular a liberação e proteger os peptídeos da degradação química e física mantendo a bioatividade. O sistema líquido cristalino foi preparado com fosfatidilcolina de soja (FS), oleato de sódio (OS) e Tween 20 (TW), colesterol (CHO) e tampão fosfato, sendo que alguns pontos apresentaram fases líquido-cristalinas do tipo lamelar. Para os estudos posteriores foi escolhido um ponto com características visuais de viscosidade apropriadas ao uso tópico e a este se incorporou o peptídeo. A obtenção do peptídeo por meio da SPFS foi viável, obtendo-se materiais com alto índice de pureza. No ensaio de reologia a formulação apresentou-se como fluido pseudoplástico não–Newtoniano, com a presença de tixotropia; sendo possível também confirmar a incorporação do peptídeo ao sistema. A espectroscopia UV e a CLAE comprovaram que a maior parte do peptídeo é incorporada à fase líquido-cristalina do sistema. Os ensaios biológicos mostraram que o sistema líquido-cristalino possui atividade contra bactérias muito menor que o peptídeo livre. Este fato foi atribuído à baixa liberação do peptídeo pelo sistema

As formigas da tribo Attini apresentam reconhecida simbiose com microrganismos: fungos mutualistas basidiomicetos nas famílias Lepiotaceae e Pterulaceae cultivados pelas formigas, bactérias actinomicetas ou no gênero Burkholderia produtoras de antibióticos contra fungos entomopatogênicos, fungos ascomicetos do gênero Escovopsis micófagos para o mutualista, e comensais como fungos, leveduras e bactérias que degradam celulose. Um novo mutualista é descrito no presente trabalho: uma espécie de bactéria no gênero Mesoplasma, detectada via PCR para 16S no DNA genômico de Attini derivadas (gêneros Acromyrmex, Atta, Serycomyrmex e Cyphomyrmex), intermediárias (Trachymyrmex) e basais (Apterostigma, Mycetarotes, Mycocepurus e Mycetagroicos). Uma única linhagem de Mesoplasma com baixíssima diversidade 16S esteve presente nas Attini, mas não em formigas de tribos filogeneticamente próximas a Attini, como Dolichoderini (Tapinoma sp), Camponotini (Camponotus sp), Cephalotini (Cephalotes sp), Crematogastrini (Crematogaster sp), Pheidolini (Pheidole sp) e Ponerini (Pachycondyla sp), indicando uma simbiose Mesoplasma-Attini recente e específica.

The catalytic function of extended-spectrum β-lactamases can result in high degrees of bacterial resistance to β-lactamic antimicrobials and in the emergence of ESBL among the members of Enterobacteriaceae family, especially Klebsiella pneumoniae and Escherichia coli. This occurs due to the dissemination and emergence of new variants of these enzymes caused by the high utilization of antibiotics like broad-spectrum cephalosporins. The ESBL are β-lactamases capable of conferring bacterial resistance to the penicillins, 1st, 2nd and 3rd generation cephalosporins, and aztreonam (but not cephamycins and carbapenems) through the hydrolysis of these antibiotics. In view of this phenomenon, the exact screening and detection of the producers of ESBL are essential for the appropriate selection of the antimicrobial therapy. The purposes of this study were to evaluate the best antimicrobial for the selection of ESBL producers and to determine the best method for the detection of such microorganisms. We evaluated 200 sequential bacterial samples including the species Klebsiella pneumoniae (56.5%), Escherichia coli (34%), Proteus mirabilis (8.5%) and Klebsiella oxytoca (1%), previously characterized as ESBL producers between February and September 2008 in the Laboratory of Microbiology, Botucatu Medical School - UNESP, Botucatu, São Paulo State, Brazil. To select the ESBL-producer bacteria, we used the disks recommended by CLSI 2008, aztreonam (ATM), cefpodoxime (CPD), ceftriaxone (CRO), cefotaxime (CTX) and ceftazidime (CAZ), besides cefepime (FEP). ESBL production was confirmed by three methods: double disk screening, ESBL Etest®, and Vitek® automated system. A teicoplanina é um complexo antibiótico glicopeptídico derivado do Actinoplanes teichomyceticus, ativo contra bactérias Gram-positivas resistentes a outros antibióticos. Seu espectro de ação é similar ao da vancomicina, sendo, porém mais ativo para Streptococcus faecalis e Clostridium difficile. Seu uso é indicado para profilaxia de endocardite, peritonite, osteomielite e para septicemia estafilocócica. No Brasil, a teicoplanina é comercializada sob a forma farmacêutica de pó liofilizado, que deve ser reconstituído antes da administração. O medicamento de referência é o Targocid®, produzido pelo laboratório Sanofi-Aventis, em duas apresentações, 66 mg/mL e 133 mg/mL. A teicoplanina, bem como a vancomicina, inibe a síntese da parede celular bacteriana, pois a molécula se liga ao precursor da parede D-alanil-D-alanina, formando um complexo, impedindo a ligação à porção terminal do peptidoglicano, que é o alvo das enzimas transglicolase e transpeptidase. Desse modo, não há incorporação de aminoácidos aos glicopeptídeos integrantes da parede celular das bactérias Gram-positivas. No estudo de validação, foram aplicados os parâmetros de linearidade, intervalo, precisão, sensibilidade, especificidade, exatidão e robustez. O método desenvolvido e validado para a quantificação de teicoplanina pó liofilizado foi: Ensaio microbiológico por turbidimetria na faixa de concentração de 20,0 a 80,0 μg/mL, utilizando o micro-organismo Staphylococcus epidermidis ATCC 12228 IAL 2150. Os parâmetros estudados para a validação do método turbidimétrico atenderam a todas as especificações para a adequada quantificação de teicoplanina na forma farmacêutica pó liofilizado

The increase of the antimicrobial resistance and its propagation around the world are the biggest threats to the public health care and to the treatment of diseases caused by microorganisms. Nowadays the antimicrobial resistance has increased abruptly. The essential oils are volatile and aromatic compounds derived from parts of plants as flowers, leafs, fruits, seeds, roots, sprouts, among others. The activity of extracts and essential oils of several plant species have been recognized and studied by empirical methods since a long time, but its antimicrobial activities were confirmed recently. Medicinal plants are used in folk medicine as medicines, antibiotic, analgesic, sedative and anti-inflammatory. The use of medicinal plants like source of medicines is an alternative of therapeutics for diseases treatment. In Brazil, studies with this goal are very important, once medicinal plants have been used as a choice of treatment and prevention of infections and diseases in health areas. Considering the fact that some products from medicinal plants have antimicrobial properties it is expected that using screening programs, new potential medicaments could be developed. Otherwise, scientific researches focused on determining therapeutic potential of plants are limited, there are lack of scientific studies which confirms the potential antibiotics properties of a large number of plants. The aim of the present study is determinate the antimicrobial activity of 10 medicinal species belonging to CPMA - Collection of Medicinal and Aromatic Plants from CPQBA/UNICAMP. The minimal inhibitory (MIC) and minimal bactericidal or fungicidal concentration (MBC) will be determined against the bacteria Escherichia coli, Pseudomonas aeruginosa, Salmonella choleraesuis, Staphylococcus aureus and the yeast Candida albicans. Study the semi-quantitative and quantitative technique in the diagnosis of catheter-related infections in newborns and to determine oxacillin resistance in Staphylococcus isolated. It was analyzed 353 catheter tips from 273 newborns in the Neonatal Unit of Hospital FMB. To confirm the diagnosis of infection, were analyzed the clinical data of newborns, the presence of at least one positive blood culture and growth of ≥ 1000 CFU/mL on quantitative culture and/or ³15 UFC on semiquantitative culture, with the same microorganism isolation (species and drug sensitivity) in blood culture and no other focus of infection except the catheter. The disk diffusion technique was used to check similarity of strains and resistance to oxacillin. Of the 353 tips analyzed, 39 were included in this study as the inclusion criteria. The semiquantitative culture was positive in 26 (66.7%) catheters and quantitative culture was positive in 24 (61.5%). Of 273 patients, 19 (6.9%) had a diagnosis of catheter-related bloodstream Infection (CR-BSI). Of the 19 episodes of CR-BSI, S. epidermidis was the predominant etiological agent (84.2%). The resistance to the antibiotic methicillin was found in 14 (73.7%) strains of Staphylococcus. The semiquantitative method was more sensitive (79%) compared with the quantitative method (63%). The use of antibiotics may have influenced the sensitivity of the quantitative method as the microorganisms present in the lumen are exposed to higher concentrations of antibiotics administered via the catheter

Pneumonia is a respiratory disease that most affects the cattle, especially calves, which according to their anatomical and physiological characteristics are more susceptible to respiratory diseases compared to other large mammals. Therefore, prevention of pneumonia is a key factor to minimize the possible economic losses generated by the early involvement of a calf by these diseases, which can cause a decrease in animal development. Despite the multifactorial etiology and classification on the types of pneumonia is still subject of discussion, most pneumonias are in three categories: bronchopneumonia, interstitial pneumonia and metastatic pneumonia, the first being the most important among them. Clinical signs vary with the cause, but are classified as medical conditions ranging since subclinical to clinically irreversible. Treatment depends on the clinical experience and can associate drugs for alleviating the symptoms with anti-inflammatories and antibiotics. Thus, premature diagnosis is directly linked to prognosis, treatment costs and spread of the disease

As abelhas da espécie Apis mellifera são conhecidas por sua organização social, comportamento devotado e aparente complexidade comportamental, sendo também consideradas excelentes sistemas-modelo para estudos de comunicação, aprendizado e formação de memória. Uma vez que o comportamento social dos insetos é resultante de interações complexas em diferentes níveis de organização biológica, torna-se importante estudar os peptídeos envolvidos nesse processo, já que estes são indispensáveis em muitos processos fisiológicos, funcionando como neurotransmissores, hormônios, toxinas, antibióticos, defensinas, neuromoduladores e neurohormônios. Desta forma, o objetivo do presente trabalho foi padronizar uma metodologia analítica visando à determinação do perfil peptídico do cérebro de abelhas da espécie Apis mellifera utilizando como ferramentas a espectrometria de massas e a cromatografia líquida. Foram coletadas operárias de Apis mellifera com 20 dias de idade, cujos cérebros foram dissecados e devidamente processados visando a posterior separação e identificação dos peptídeos. Foram testadas duas estratégias distintas para a separação e identificação dos peptídeos, denominadas on-line e off-line. Ambas baseavam-se na utilização de um sistema de cromatografia líquida sob fase reversa e um espectrômetro de massas do tipo electrospray/Ion Trap-Time of Flight, mas na primeira essas ferramentas estavam acopladas, enquanto na segunda os dois processos foram realizados separadamente. Tais procedimentos permitiram identificar 19 peptídeos pertencentes às principais famílias de neuropeptídeos de cérebros de abelhas já descritos na literatura, entre elas as Alatostatina, Apidaecina, Corazonina e Taquicininas. O estudo de novas alternativas terapêuticas no tratamento de infecções microbianas tornou-se crescente no cenário científico devido à grande variação genética desses micro-organismos, que resultou na resistência aos antimicrobianos existentes. A grande diversidade na flora brasileira e a ampla utilização das plantas como medicamentos pela população justificam os estudos e o crescente interesse pela descoberta de novos compostos bioativos isolados dos vegetais. Plantas usualmente utilizadas na agricultura, apenas como adubação verde, por exemplo, são atualmente alvo de estudos científicos com potenciais promissores de atuação como de produtos terapêuticos. Plantas da família Leguminosae, amplamente conhecidas e utilizadas como fornecedoras de nitrogênio ao solo, vem sendo estudadas por diversas áreas da saúde para comprovarem a ação de compostos isolados entre estes os alcalóides pirrolidizínicos como antiinflamatórios, antibióticos e até como veneno para pragas. O objetivo do presente estudo foi determinar, a partir de extrativos de Crotalaria pallida, a atividade antimicrobiana in vitro utilizando cepas padrões de: Staphylococccus aureus, Escherichia coli, Salmonella sp e da levedura Candida albicans. Para determinação dessa atividade foi utilizada a técnica de diluição em microplaca que possibilitou o estudo da atividade do extrativo vegetal e da concentração inibitória mínima, isto é, concentração bactericida e/ou bacteriostática mínima e concentração fungiostática e/ou fungicida mínima. A Crotalaria pallida, não apresentou atividade frente aos micro-organismos testados nas condições padronizadas neste estudo
