Acetylcholinesterase (AChE) activity was measured in Daphnia magna that had been exposed to four organophosphates (OPs; parathion, chlorpyrifos, malathion, and acephate) and one carbamate (propoxur) for 48 h. These results were related to acute toxicity (median effective concentration [EC50] for immobility). For the four OPs, the EC50s were 7.03 pM, 3.17 pM, 10.56 pM, and 309.82 muM, respectively. The EC50 for propoxur was 449.90 pM. Reduction in AChE activity was directly related to an increase in immobility in all chemicals tested. However, the ratio between the EC50 and the AChE median inhibiting concentration ranged from 0.31 to 0.90. A 50% reduction in AChE activity generally was associated with detrimental effects on mobility. However, for acephate, high levels of AChE inhibition (70%) were observed in very low concentrations and were not associated with immobility. In addition, increasing the concentration of acephate further had a slight negative effect oil AChE activity but a Strong detrimental effect on mobility. Binding sites other than AChE possibly are involved in acephate toxicity to D. magna. Our findings demonstrate different associations between AChE inhibition and toxicity when different chemicals are compared. Therefore, the value of using AChE activity as a biomarker in D. magna will be dependent on the chemical tested.

In this paper sequential importance sampling is used to assess the impact of observations on a ensemble prediction for the decadal path transitions of the Kuroshio Extension (KE). This particle filtering approach gives access to the probability density of the state vector, which allows us to determine the predictive power — an entropy based measure — of the ensemble prediction. The proposed set-up makes use of an ensemble that, at each time, samples the climatological probability distribution. Then, in a post-processing step, the impact of different sets of observations is measured by the increase in predictive power of the ensemble over the climatological signal during one-year. The method is applied in an identical-twin experiment for the Kuroshio Extension using a reduced-gravity shallow water model. We investigate the impact of assimilating velocity observations from different locations during the elongated and the contracted meandering state of the KE. Optimal observations location correspond to regions with strong potential vorticity gradients. For the elongated state the optimal location is in the first meander of the KE. During the contracted state of the KE it is located south of Japan, where the Kuroshio separates from the coast.

Until December 2011, Hungary had the most stable electoral system in Eastern Europe: except for an increase in the electoral threshold in 1994, no significant change had taken place since the transition from communism in 1989. On 23rd December 2011, however, Parliament passed a wholly new electoral law, which will first be used in the elections due in 2014. The purposes of this article are twofold. The first is to outline the change of electoral system that took place. The previous electoral law was, famously, one of the most complex in the world. The new law is somewhat simpler, but nevertheless retains many of the old law’s features. The second purpose is to analyse the origins and implications of the new law. Did it simply reflect the interests of those in power or did it also respond to public concerns? Can it be expected to enhance or detract from the quality of Hungarian democracy?

This paper describes the methodology used to compile a corpus called MorphoQuantics that contains a comprehensive set of 17,943 complex word types extracted from the spoken component of the British National Corpus (BNC). The categorisation of these complex words was derived primarily from the classification of Prefixes, Suffixes and Combining Forms proposed by Stein (2007). The MorphoQuantics corpus has been made available on a website of the same name; it lists 554 word-initial and 281 word-final morphemes in English, their etymology and meaning, and records the type and token frequencies of all the associated complex words containing these morphemes from the spoken element of the BNC, together with their Part of Speech. The results show that, although the number of word-initial affixes is nearly double that of word-final affixes, the relative number of each observed in the BNC is very similar; however, word-final affixes are more productive in that, on average, the frequency with which they attach to different bases is three times that of word-initial affixes. Finally, this paper considers how linguists, psycholinguists and psychologists may use MorphoQuantics to support their empirical work in first and second language acquisition, and clinical and educational research.

This study explores the role of the International Accounting Standards Board’s (IASB) due process in developing its International Financial Reporting Standards for Small and Medium-sized Entities (IFRS or SMEs)standard. There were tensions between the IASB’s desire to minimise divergence from full IFRS and preserve recognition and measurement principles, and the primary reasons for undertaking the project – to meet the needs of users of financial statements of SMEs and to reduce the financial reporting burden on SMEs. Examination of events during the development of the project reveals much that was not apparent from material in the public domain. Most significantly, the IASB recognised that the final title of the standard, IFRS for SMEs, does not necessarily describe the scope of the standard. This paper also shows that the due process followed in the case of the IFRS for SMEs project barely reflected the ‘will of people’ but was more inclined towards acting as a communicative function for the IASB without any commitment to change its stance on the SME standard.

Purpose: To evaluate in vitro the influence of water flow rate on shear bond strength of a resin composite to enamel and dentin after Er:YAG cavity preparation. Methods: Ten bovine incisors were selected and roots removed. Crowns were sectioned in four pieces, resulting in 40 samples that were individually embedded in polyester resin (n=10), and ground to plane the enamel and expose the dentin. The bonding site was delimited and samples were randomly assigned according to cavity preparation: (1) Er:YAG/1.0 mL/minute; (2) Er:YAG/1.5 mL/minute; (3) Er:YAG/2.0 mL/minute and (4) High speed handpiece/bur (control group). Samples were fixed to a metallic device, where composite resin cylinders were prepared. Subsequently, they were stored for 24 hours and subjected to a shear bond strength test (500N at 0.5 mm/minute). Results: Means (MPa) were: enamel: 1: 12.8; 2: 16.8; 3: 17.5; 4: 36.0 and Dentin: 1: 13.6; 2: 18.7; 3: 12.1; 4: 21.3. Data were submitted to ANOVA and Tukey`s test. Adhesion to enamel was more efficient than for dentin. The cavities prepared with conventional bur (control) presented higher statistically significant bond strength values (P<0.05) than for Er:YAG laser for both enamel and dentin. No significant differences were observed between water flow rates employed during enamel ablation. For dentin, the shear bond strength of 2.0 mL/minute water flow rate was lower than for 1.5 mL/minute and 1.0 mL/minute rates. The Er:YAG laser adversely affected shear bond strength of resin composite to both enamel and dentin, regardless of the water flow rate used.

Benzodiazepínicos são amplamente utilizados em animais e humanos, inclusive durante a gestação. Benzodiazepínicos são utilizados como ansiolíticos, anticonvulsivantes, relaxantes musculares e sedativos. Este trabalho investiga o potencial teratogênico dos benzodiazepínicos diazepam e maleato de midazolam em ratos Wistar. Os animais foram divididos em três grupos “experimentais”, um grupo controle que recebeu água destilada, um grupo que recebeu diazepam na dose de 10 mg/kg/dia, e um grupo que recebeu 5 mg/kg/dia de midazolam. Os animais receberam os tratamentos por administração via oral. As fêmeas gestantes foram tratadas durante o período de organogênese, que no rato é do 6° ao 15° dia da gestação. Durante a gestação as fêmeas foram monitoradas quanto ao desenvolvimento corporal, ingesta de água e ração. No 21° dia de gestação as progenitoras foram sacrificadas e avaliadas quanto à toxicidade materna sistêmica. Os fetos foram avaliados quanto a embriotoxicidade e alterações esqueléticas. Os resultados demonstraram que as fêmeas dos três grupos tiveram ganho de peso, consumo de água e consumo de ração semelhante, assim como peso dos órgãos internos, sugerindo não haver toxicidade materna. Nos grupos diazepam e maleto de midazolam houve uma diminuição do número de fetos por ninhada (9,2 ± 3,17 e 9,29 ± 3,29) comparado ao grupo controle (10,76 ± 2,62), também houve diminuição no peso dos fetos nos grupos diazepam e midazolam (4,73 ± 0,59 e 4,79 ± 0,64) em comparação ao grupo controle (5,02 ± 0,36). Houve um aumento no número de abortos precoces nos grupos diazepam e midazolam As perdas pós-implantação foram de 1,47% no grupo controle, 10,76% no grupo diazepam e 13,30% no grupo midazolam. A taxa de viabilidade fetal foi semelhante nos três grupos. As avaliações das anormalidades esqueléticas demonstraram um retardo de desenvolvimento assim como houve evidências de malformações nos grupos diazepam e midazolam, sendo que o grupo tratado com maleato de midazolam demonstrou um maior percentual de anormalidades esqueléticas que os fetos do grupo tratado com diazepam. Este estudo sugere efeitos teratogênicos dos medicamentos diazepam e midazolam quando utilizados nas doses testadas.

Objectivo: Proceder à validação da equação de Slaugther e col., (1988), na estimação da percentagem de massa gorda (%MG), em crianças com 9 anos de idade, tendo a DXA como método de referência. Metodologia: A avaliação da composição corporal foi realizada em 450 crianças, das quais 219 eram raparigas (idade: 9.74 ± 0.33 anos; altura: 136.88 ± 6.8 cm; peso: 33.77 ± 8.16 kg; índice de massa corporal (IMC): 17.85 ± 3.16 kg/m2) e 231 eram rapazes (idade: 9.75 ± 0.33 anos; altura: 137.17 ± 6.97cm; peso: 34.3 ± 8.09 kg; IMC: 18.09 ± 3.17 kg/m2), pela DXA (QDR – 1500: Hologic, Waltham, MA, pencil beam mode, software version 5.67 anhanced whole body analisis) e pelas pregas adiposas subcutâneas, cujo os valores das pregas adiposas tricipital e geminal foram utilizados na equação desenvolvida por Slaugther e col., (1988). Na análise estatística, foram utilizadas a comparação de médias, a regressão linear e a concordância entre os métodos. Resultados: A %MG obtida por ambos os métodos apresentou diferenças significativas (p<0.05) entre os géneros, sendo as raparigas as que apresentam, em média, maiores valores de gordura corporal. Tanto para os rapazes como para as raparigas a %MGDXA é superior à %MGSKF. Na predição dos valores de %MG, a equação de Slaugther e col., (1988) tem para ambos os sexos (raparigas: r=0.94; EPE=3.2 e rapazes: r=0.96; EPE=2.7) uma correlação elevada com reduzido erro padrão de estimação com a DXA. Na análise de concordância entre os métodos, os rapazes parecem apresentar uma maior concordância entre o método alternativo e o método de referência, com os limites de concordância a variarem entre -9.26 e 1.57, enquanto nas raparigas variam entre -11.19 e 3.16. Em ambos os sexos é visível a subestimação, em valor médio, do método de referência, com uma média da diferença a situar-se nos -3.8% e -4.0%, respectivamente para rapazes e raparigas. Discussão: A equação de Slaugther e col., (1988) explica, para as raparigas, 87.5% da variância do método de referência, enquanto nos rapazes, 91.3% da variância é explicada.A diferença entre os métodos alternativo e de referência está dependente do nível de adiposidade, sendo que o método alternativo tende a sobrestimar a %MG nas crianças mais magras e a subestimar nas mais gordas. A equação de Slaugther e col., (1988) apresentou uma validade aceitável na avaliação da composição corporal num grupo de crianças, constituindo uma alternativa rápida na estimação da composição corporal numa avaliação inicial em escolas, clubes e estudos de larga escala. Contudo a sua utilidade, para uma correcta intervenção na saúde da criança, pode apresentar uma validade limitada, pelo que pode ser justificada a utilização de um método mais válido em termos clínicos, como a DXA.

One Kunitz-type trypsin inhibitors (PmTI) was purified from Piptadenia moniliformis seeds, a tree of the sub-family Mimosoideae, by TCA precipitation, affinity chromatography on immobilized trypsin-Sepharose, DEAE cellulose (ion exchange) and Superose 12 (molecular exclusion) column FPLC/AKTA. The inhibitor has Mr of 25 kDa by SDS-PAGE and chromatography molecular exclusion. The N-terminal sequence of this inhibitor showed high homology with other family Kunitz inhibitors. This also stable variations in temperature and pH and showed a small decrease in its activity when incubated with DDT in the concentration of 100mM for 120 minutes. The inhibition of trypsin by PmTI was competitive, with Ki of 1.57 x10-11 M. The activity of trypsin was effectively inhibited by percentage of inhibition of 100%, among enzymes tested, was not detected inhibition for the bromelain, was weak inhibitor of pancreatic elastase (3.17% of inhibition) and inhibited by 76.42% elastase of neutrophils, and inhibited in a moderate, chymotrypsin and papain with percentage of inhibition of 42.96% and 23.10% respectively. In vitro assays against digestive proteinases from Lepidoptera, Diptera and Coleoptera pests were carried out. Several degrees of inhibition were found. For Anthonomus grandis and Ceratitis capitata the inhibition was 89.93% and 70.52%, respectively, and the enzymes of Zabrotes subfasciatus and Callosobruchus maculatus were inhibited by 5.96% and 9.41%, respectively, and the enzymes of Plodia. interpunctella and Castnia licus were inhibited by 59.94% and 23.67, respectively. In vivo assays, was observed reduction in the development of larvae in 4rd instar of C. capitata, when PmTI was added to the artificial diet, getting WD50 and LD50 of 0.30% and 0.33%, respectively. The aging process if characterizes for a complex events network, from multidimensional nature, that encloses biological, social, psychic and functional aspects. The alteration of one or more aspects can speed up the aging process, anticipating limitations and until the death in the aged. For an adjusted confrontation of this question is necessary an interdisciplinary vision, in which the some areas of the knowledge can interact and with this to intervenes of the best possible form. Then, information derived from studies of aspects related to incidence, morbidity-mortality and transition patterns, involved in the health-illness process can more accurately identify risk groups thereby establishing links between social factors, illness, incapacity and death. Thus, this study aimed to identify, by a multidimensional vision, the risk factors of mortality in a coorth of elderly in a city in the interior of the state of Rio Grande do Norte (RN), Brazil. A prospective study carried out in Santa Cruz RN, where 310 elderly were randomly selected to form a baseline. The follow-up was 53 months. The predictive variables were divided into sociodemographic, physical health, neuropsychiatric and functional capacity. The statistical analysis carried out by bivariate analysis, survival analysis, followed by binary logistic regression and Cox regression, in the multivariate analysis, considering significant levels p < 0.05 and confidence interval (CI) of 95%. A total of 60 (19.3%) elderly died during the follow-up, where cardiovascular disease was the main cause. The survival was approximately 24.8 months. The study of general survival showed, at 12, 24, 36, and 48 months of observation, a survival rate of 97%, 54%, 31%, and 5% respectively, with a statistical difference in survival only observed for the variables of cognitive function and Basic Activities of Daily Living. In the logistic regression analysis, the risk factors identified were cognitive deficits (OR = 8.74), poor perception of health (OR = 3.89) and dependence for Basic Activities of Daily Living (OR = 3.96). In the Cox analysis, as well as dependence for Basic Activities of Daily Living (HR = 3.17), cognitive deficit (HR = 4.30) and stroke (CVA) (HR = 3.49) continued as independent risk factors for death. The risk factors found in the study can be interpreted as the primary predictors for death among elderly members of the community. Therefore, improvements in health conditions, with actions towards sustaining an autonomous life with special attention for elderly with cognitive impairment, could mean additional healthy quality of life, resulting in the reduction of premature mortality in this population

O objetivo deste estudo foi avaliar o efeito do horário, taxa e freqüência de alimentação no desempenho do matrinxã (Brycon amazonicus) em tanques de cultivo. O trabalho foi realizado no Centro de Aqüicultura da UNESP, Jaboticabal, SP, no período de outubro de 1997 a janeiro de 1998 sendo realizados três ensaios, em tanques de 200m² subdivididos em 4 de 50 m². No ensaio I foram medidos em 3 períodos (manhã-m; meio do dia-md e tarde-t) o consumo de ração, índice de ingestão, tempo de saciação e velocidade de ingestão em peixes com peso médio de 232,13 g, alimentados com ração extrusada (32% de PB). Não foi observada diferença significativa nos parâmetros analisados. No ensaio II, em peixes com peso médio de 233,98 g, foi medido o consumo médio de ração, em intervalos de duas horas, das 07 às 19 horas. O maior consumo ocorreu quando o matrinxã foi alimentado às 17 horas. No ensaio III, durante 57 dias, os peixes foram alimentados uma vez ao dia (m); uma vez ao dia (t); duas vezes ao dia(m/t) e três vezes ao dia (m,md,t). Peixes com peso médio inicial de 322,25 g receberam ração com 32% de PB, na quantidade de 2% do PV. Não foram observadas diferenças significativas no ganho de peso diário (3,17; 2,80; 3,04 e 2,81 g) e na conversão alimentar aparente (2,11; 2,48; 2,16 e 2,31:1). Concluiu-se que a freqüência de alimentação de uma vez ao dia, em qualquer horário, mostrou ser suficiente.

A melancia é uma espécie tradicionalmente conduzida em campo no sistema rasteiro. As cultivares de frutos pequenos (1 a 3 kg), que adquirem melhores preços de mercado, vêm sendo cultivadas também em ambiente protegido, onde são conduzidas no sistema vertical, com poda de ramos e raleio de frutos. Essas práticas possibilitam aumentar o adensamento das plantas, a qualidade e a produtividade de frutos em comparação ao sistema rasteiro. Objetivou-se com este trabalho avaliar a influência de três alturas de condução (1,7; 2,2 e 2,7 m) e duas densidades de plantas (3,17 e 4,76 plantas m-2) sobre as características produtivas e qualitativas da mini melancia Smile cultivada em ambiente protegido. A poda da haste principal foi realizada aos 43, 55 e 66 dias após o transplante (DAT) para as alturas de condução de 1,7; 2,2 e 2,7 m, respectivamente. A massa seca dos ramos, dos pecíolos, das folhas e total foram afetados pela altura de condução, cujos maiores valores foram obtidos para as plantas conduzidas a 2,2 e 2,7 m de altura. A área foliar, a área foliar específica e o índice de área foliar não foram influenciados pela altura de condução das plantas. A altura de condução de 2,7 m elevou a produtividade total. Entretanto, a produtividade comercial, a massa média dos frutos e todas as características qualitativas não foram significativamente diferentes das obtidos pela altura de poda de 2,2 m. em relação à densidade de plantas, a melhor opção foi a de 4,76 plantas m-2, pois elevou a produtividade comercial em 37,4% sem reduzir a massa média dos frutos.
