The majority of patients with chronic hepatitis C fail to respond to antiviral therapy. The genetic basis of this resistance is unknown. The quasispecies nature of HCV may have an important implication concerning viral persistence and response to therapy. The HCV nonstructural 5A (NS5A) protein has been controversially implicated in the inherent resistance of HCV to interferon (IFN) antiviral therapy. To evaluate whether the NS5A quasispecies pre-treatment composition of HCV 1a/1b is related to responsiveness to combined pegylated interferon (PEG-IFN) and Ribavirin therapy, detailed analyses of the complete NS5A were performed. Fifteen full-length NS5A clones were sequenced from 11 pretreatment samples of patients infected with genotype 1 HCV (3 virological sustained responders, 4 non-responders, and 4 end-of-treatment responders). Our study could not show a significant correlation between the mean number of mutations in HCV NS5A before treatment and treatment outcome, and the phylogenetic construction of complete NS5A sequences obtained from all patients failed to show any clustering associated with a specific response pattern. No single amino acid position was associated with different responses to therapy in any of the NS5A regions analyzed, and mutations were clustered downstream the ISDR, primarily in the V3 region. We observed that the CRS and NLS regions of the NS5A protein were conflicting for some variables analyzed, although no significant differences were found. If these two regions can have antagonistic functions, it seems viable that they present different mutation profiles when compared with treatment response. The patient sample that presented the lowest genetic distance values also presented the smallest number of variants, and the most heterogeneous pattern was seen in the end-of-treatment patients. These results suggest that a detailed molecular analysis of the NS5A region on a larger sample size may be necessary for understanding its role in the therapy outcome of HCV 1a/1b infection. A proteólise do leite UAT/UHT durante a estocagem à temperatura ambiente é um dos fatores limitantes de sua vida de prateleira. Neste trabalho, dois lotes de leite cru contendo 10 amostras cada e, posteriormente ao processamento, dois lotes de leite UAT/UHT contendo 25 amostras cada foram colhidos em um laticínio para a contagem de microrganismos psicrotróficos (leite cru) e para o estudo do comportamento reológico e o índice proteolítico (leite UAT/UHT durante 120 dias de estocagem). Para a contagem de microrganismos psicrotróficos, foi utilizada a técnica da contagem padrão em placas. Para a determinação do índice proteolítico, foi determinada a presença de glicomacropeptídeo livre por espectrofotometria a 470 nm. A determinação dos parâmetros reológicos foi efetuada à temperatura ambiente, em quintuplicata em um reômetro de cone e placa. Houve aumento da proteólise no decorrer do armazenamento e aumento da viscosidade aparente após 60 dias de estocagem, provavelmente relacionados à presença de proteases de bactérias psicrotróficas do leite cru.

As características hematológicas do jundiá Rhamdia quelen Quoy & Gaimard, 1824 (Osteichthyes: Pimelodidae), oriundo de cativeiro foram estudadas. Determinou-se o valor médio da contagem de eritrócitos, taxa de hemoglobina, hematócrito, volume corpuscular médio (VCM), concentração de hemoglobina corpuscular média (CHCM) e distribuição percentual de células sangüíneas de defesa orgânica. As células sangüíneas de defesa orgânica (leucócitos e trombócitos) foram identificadas e caracterizadas morfologicamente. Trombócitos e linfócitos foram as células sangüíneas de defesa orgânica mais freqüente nas extensões sangüíneas do R. quelen. A análise de regressão mostrou correlação linear negativa entre o percentual de trombócitos e linfócitos. Os resultados deste estudo fornecem valores sangüíneos normais em R. quelen, em cultivo intensivo, que poderão servir de comparação com dados dessa espécie em outras situações de cultivo.

In the present work we study a long superconducting wire with a columnar defect in the presence of an applied magnetic field. The cross section of the cylinder is assumed to be circular. The field is taken uniform and parallel to the cylinder axis. We use the London theory to investigate the vortex lattice inside the wire. Although this theory is valid in the limit of low vortex density, that is, when the nearest neighbor vortex distance is much larger than the coherence length, we can obtain a reasonable qualitative description of lattice properties. We calculate: (1) the vortex lattice structure using the simulated annealing technique; (2) the magnetization curve as a function of the applied field.

Foram avaliadas três fontes protéicas (uréia - U, farinha penas - Fp e farelo soja - Fs) associadas a duas silagens de sorgo (silagens de sorgo corte alto - SCA e corte baixo - SCB) por intermédio do desempenho de bezerros de corte, confinados por 84 dias. Foram utilizados 36 bezerros mestiços (Charolês/RedAngus/Nelore), castrados, com cinco meses de idade e pesando em média 146,44 kg. Avaliaram-se o ganho de peso médio diário (GMD); o consumo de matéria seca expressos em kg/dia (CMS), percentagem de peso vivo (CMSPV) e peso metabólico (CMSPM); e a conversão alimentar (CA). O delineamento foi inteiramente casualizado por intermédio de um fatorial 3x2. Houve efeito significativo na interação silagem versus fonte protéica para a variável GMD, com o maior GMD para SCAFp (1,61 kg/animal/dia) versus a SCBFs (1,32 kg/animal/dia). Os demais parâmetros não diferiram estatisticamente quando analisada a interação ou isoladamente a silagem ou fonte protéica. A silagem de sorgo corte alto promoveu numericamente um maior GMD e CMS.

This system of equations, involving nonlinearity, dispersion, and dissipation, is a generalization of the usual Boussinesq system.

A single-phase superconducting Fault Current Limiter using a bifilar coil of BSCCO-2212 tube was tested in 220 V-60 Hz line during fault current between 1 kA to 4 kA, operating in 77 K. In this work are presented the critical current dependence as a function of an external magnetic field applied and the results can be used to predict the current limiter performance. The experimental setup is described and the test results are presented for the unit conducting a steady nominal AC current of 200 A, and also during the fault time (1 to 6 cycles). The performance of the bifilar coil to provide the limiting impedance associated with the dynamic resistance developed during the beginning of the fault was analyzed and compared with other types of superconducting current limiters.

A new shadow-ring device for measuring diffuse solar radiation at the surface is presented. In this device the seasonal variation of shadow is followed by moving the detector horizontally. This unique characteristic facilitates its application for long and continuous periods of time. The blocking effect caused by the ring and other related geometric properties are formulated considering the diffuse solar radiation isotropic. The correction factor, shadow size, and ring-detector distance are derived as a function of radius and width of the ring, sun position, and local latitude. The largest blocking occurs during summer, when the ring-detector distance and the shadow width are the smallest, and it is compensated by a smaller blocking effect in the winter period. The performance of the new device is verified comparing daily values of diffuse solar radiation measured simultaneously with a similar device from Kipp & Zonen, Inc. The results show a very good agreement (within 2.5%) between both devices. The new device was also able to reproduce the radiometric properties of the local atmosphere based on 3-yr-long measurements of direct solar radiation using a pyrheliometer. The new device can be applied to estimate daily values of diffuse solar radiation at the surface in the range of 30degreesN-30degreesS with results comparable to other similar apparatuses.
