Hyperhomocysteinemia (HHcy) is a risk factor for vascular disease, but the underlying mechanisms remain incompletely defined. Reduced bioavailability of nitric oxide (NO) is a principal manifestation of underlying endothelial dysfunction, which is an initial event in vascular disease. Inhibition of cellular methylation reactions by S-adenosylhomocysteine (AdoHcy), which accumulates during HHcy, has been suggested to contribute to vascular dysfunction. However, thus far, the effect of intracellular AdoHcy accumulation on NO bioavailability has not yet been fully substantiated by experimental evidence. The present study was carried out to evaluate whether disturbances in cellular methylation status affect NO production by cultured human endothelial cells. Here, we show that a hypomethylating environment, induced by the accumulation of AdoHcy, impairs NO production. Consistent with this finding, we observed decreased eNOS expression and activity, but, by contrast, enhanced NOS3 transcription. Taken together, our data support the existence of regulatory post-transcriptional mechanisms modulated by cellular methylation potential leading to impaired NO production by cultured human endothelial cells. As such, our conclusions may have implications for the HHcy-mediated reductions in NO bioavailability and endothelial dysfunction.

Background: Poor nutritional status and worse health-related quality of life (QoL) have been reported in haemodialysis (HD) patients. The utilization of generic and disease specific QoL questionnaires in the same population may provide a better understanding of the significance of nutrition in QoL dimensions. Objective: To assess nutritional status by easy to use parameters and to evaluate the potential relationship with QoL measured by generic and disease specific questionnaires. Methods: Nutritional status was assessed by subjective global assessment adapted to renal patients (SGA), body mass index (BMI), nutritional intake and appetite. QoL was assessed by the generic EuroQoL and disease specific Kidney Disease Quality of Life-Short Form (KDQoL-SF) questionnaires. Results: The study comprised 130 patients of both genders, mean age 62.7 ± 14.7 years. The prevalence of undernutrition ranged from 3.1% by BMI ≤ 18.5 kg/m2 to 75.4% for patients below energy and protein intake recommendations. With the exception of BMI classification, undernourished patients had worse scores in nearly all QoL dimensions (EuroQoL and KDQoL-SF), a pattern which was dominantly maintained when adjusted for demographics and disease-related variables. Overweight/obese patients (BMI ≥ 25) also had worse scores in some QoL dimensions, but after adjustment the pattern was maintained only in the symptoms and problems dimension of KDQoL-SF (p = 0.011). Conclusion: Our study reveals that even in mildly undernourished HD patients, nutritional status has a significant impact in several QoL dimensions. The questionnaires used provided different, almost complementary perspectives, yet for daily practice EuroQoL is simpler. Assuring a good nutritional status, may positively influence QoL.

We examine volunteer satisfaction with HRM practices, namely recruitment, training and reward in NPOs and attitudes regarding the appropriateness of these practices. The participants in this study are 76 volunteers affiliated with four different NPOs, who work in hospitals and have direct contact with patients and their families. Analysing aggregate results we show that volunteers are more satisfied with training, and consider the training strategies to be very appropriate. After identifying differences between organisations we discover that in some organisations volunteers are satisfied with rewards but they have negative attitudes regarding the appropriateness of the recognition strategies. We also identify the volunteers who are the most and the least satisfied.

Anticancer activity of the new [Ru(eta(5)-C5H5)(PPh3)(Me(2)bpy)][CF3SO3] (Me(2)bpy = 4,4'-dimethyl-2,2'-bipyridine) complex was evaluated in vitro against several human cancer cell lines, namely A2780, A2780CisR, HT29, MCF7, MDAMB231 and PC3. Remarkably, the IC50 values, placed in the nanomolar and sub-micromolar range, largely exceeded the activity of cisplatin. Binding to human serum albumin, either HSA (human serum albumin) or HSA(faf) (fatty acid-free human serum albumin) does not affect the complex activity. Fluorescence studies revealed that the present ruthenium complex strongly quench the intrinsic fluorescence of albumin. Cell death by the [Ru(eta(5)-C5H5)(PPh3)(Me(2)bpy)][CF3SO3] complex was reduced in the presence of endocytosis modulators and at low temperature, suggesting an energy-dependent mechanism consistent with endocytosis. On the whole, the biological activity evaluated herein suggests that the complex could be a promising anticancer agent. (C) 2013 Elsevier Inc. All rights reserved.

Introdução: A cirurgia abdominal acarreta grande risco de complicações pulmonares pós-operatórias. As alterações pós-cirúrgicas abdominais, reflectem-se na dinâmica ventilatória, de modo particular nos volumes e capacidades pulmonares, e na capacidade de tosse. Objectivos: Compreender qual a variação dos volumes e capacidades pulmonares e da capacidade de tosse antes e depois da cirurgia abdominal (estômago e cólon), e qual a correlação dessa variação com o nível de dor percepcionada. Desenho do estudo: Unicêntrico, prospectivo e observacional. Amostra: 10 indivíduos, propostos para cirurgia abdominal – estômago e cólon. Metodologia: Dois momentos de avaliação: um nas 24h préoperatórias em que se mediu a capacidade vital forçada (CVF) e o volume expirado máximo no primeiro segundo (VEMS1) com espirometria, e do pico de fluxo de tosse (PCF); e um segundo momento nas 24h pós-operatórias onde se repetiram as medições do primeiro momento com o acréscimo da avaliação da dor. Resultados: No pós-operatorio imediato há uma diminuição significativa da CVF de 44,30%±17,24 (p=0,005), do VEMS1 de 35,50%±28,47 (p=0,009) e do PCF de 38,97%±38,66 (p=0,012). Não se verificou nenhuma relação entre a dor percepcionada na realização das manobras de espirometria e tosse com diminuição a da CVF e do VEMS1 e do PCF respectivamente. O sexo apresentou uma relação significativa com a variação da CRF e do VEMS1 (p=0,046 e p=0,008 respectivamente). A frequência respiratória apresentou um aumento significativo no pós-operatório de 10±11,22 cpm (p=0,019). A saturação periférica de oxigénio apresentou uma diminuição significativa no pós-operatório de 3,52±2,47 (p=0,011) Conclusão: No estudo efectuado fica demonstrado o impacto negativo da cirurgia abdominal na dinâmica respiratória. A diminuição dos valores da CVF, do VEMS1 e do PCF podem contribuir de forma significativa para o aumento do risco de complicações respiratória pós-operatórias. No entanto seria importante a realização deste estudo com uma amostra maior.

A great number of low-temperature geothermal fields occur in Northern-Portugal related to fractured rocks. The most important superficial manifestations of these hydrothermal systems appear in pull-apart tectonic basins and are strongly conditioned by the orientation of the main fault systems in the region. This work presents the interpretation of gravity gradient maps and 3D inversion model produced from a regional gravity survey. The horizontal gradients reveal a complex fault system. The obtained 3D model of density contrast puts into evidence the main fault zone in the region and the depth distribution of the granitic bodies. Their relationship with the hydrothermal systems supports the conceptual models elaborated from hydrochemical and isotopic water analyses. This work emphasizes the importance of the role of the gravity method and analysis to better understand the connection between hydrothermal systems and the fractured rock pattern and surrounding geology. This paper reports a novel application of microwave-assisted extraction (MAE) of polyphenols from brewer’s spent grains (BSG). A 24 orthogonal composite design was used to obtain the optimal conditions of MAE. The influence of the MAE operational parameters (extraction time, temperature, solvent volume and stirring speed) on the extraction yield of ferulic acid was investigated through response surface methodology. The results showed that the optimal conditions were 15 min extraction time, 100 °C extraction temperature, 20 mL of solvent, and maximum stirring speed. Under these conditions, the yield of ferulic acid was 1.31±0.04% (w/w), which was fivefold higher than that obtained with conventional solid–liquid extraction techniques. The developed new extraction method considerably reduces extraction time, energy and solvent consumption, while generating fewer wastes. HPLC-DADMS analysis indicated that other hydroxycinnamic acids and several ferulic acid dehydrodimers, as well as one dehydrotrimer were also present, confirming that BSG is a valuable source of antioxidant compounds.

Soil vapor extraction (SVE) is an efficient, well-known and widely applied soil remediation technology. However, under certain conditions it cannot achieve the defined cleanup goals, requiring further treatment, for example, through bioremediation (BR). The sequential application of these technologies is presented as a valid option but is not yet entirely studied. This work presents the study of the remediation of ethylbenzene (EB)-contaminated soils, with different soil water and natural organic matter (NOMC) contents, using sequential SVE and BR. The obtained results allow the conclusion that: (1) SVE was sufficient to reach the cleanup goals in 63% of the experiments (all the soils with NOMC below 4%), (2) higher NOMCs led to longer SVE remediation times, (3) BR showed to be a possible and cost-effective option when EB concentrations were lower than 335 mg kgsoil −1, and (4) concentrations of EB above 438 mg kgsoil −1 showed to be inhibitory for microbial activity.

OBJETIVO: Avaliar o consumo alimentar durante a gestação e pós-parto, segundo cor da pele. MÉTODOS: Estudo longitudinal prospectivo que incluiu 467 mulheres entre 15 e 45 anos no período pós-parto, no município do Rio de Janeiro, entre 1999 e 2001. Foi aplicado um questionário de freqüência de consumo de alimentos aos 15 dias pós-parto (consumo referente ao período da gestação) e aos seis meses (consumo referente ao período pós-parto). Foi utilizada análise de covariância para analisar diferenças no consumo alimentar, segundo cor da pele, controlada pela escolaridade. RESULTADOS: Durante a gestação, pretas e pardas apresentaram consumo de energia 13,4% e 9,1% (p=0,009 e p=0,028) e consumo de carboidrato 15,1% e 10,5% maior que brancas (p=0,005 e p=0,014), respectivamente. Mulheres pretas e brancas apresentaram consumo energético 34% e 20% acima das recomendações nutricionais, respectivamente (p=0,035). Durante o período pós-parto, as pretas apresentaram consumo de energia 7,7% maior e consumo de lipídios 14,8% maior que as brancas; consumo de ácidos graxos saturados 23,8% maior que brancas (p=0,003) e 13% maior que pardas (p=0,046). A adequação de consumo de lipídios e ácidos graxos saturados foi maior em pretas que em brancas (p=0,024 e p=0,011, respectivamente). CONCLUSÕES: Os resultados mostram ser necessário revisar estratégias de intervenção nutricional no pré-natal e implementar assistência nutricional no pós-parto, para ajustar o consumo alimentar a níveis adequados, considerando as diferenças por cor/raça identificadas.

OBJETIVO: Descrever os métodos utilizados no estudo longitudinal e acompanhamento das crianças nascidas em Pelotas (RS) em 1982. MÉTODOS: A coorte foi iniciada com um inquérito de saúde perinatal de todas as 6.011 crianças nascidas nas maternidades de Pelotas em 1982. As 5.914 crianças nascidas vivas foram incluídas nos estudos de acompanhamento. Até 2004-5 foram realizados oito acompanhamentos, com a aplicação de questionários às mães e/ou aos membros da coorte, conforme a faixa etária. Exames antropométricos e clínicos foram realizados nas visitas. Os participantes da coorte são descritos conforme variáveis demográficas, socioeconômicas e de saúde colhidas nos primeiros acompanhamentos, que são utilizadas como variáveis de exposição. RESULTADOS: A maior parte dos jovens da coorte foram acompanhados durante 23 anos de vida e distintas visitas. Os acompanhamentos que obtiveram maior êxito foram aqueles precedidos por um censo da cidade. Com este método foram localizados 87,2% em 1984 (idade média de 19 meses), 84,1% em 2006 (média 43 meses), e 77,4% em 2004-5 (média 23 anos). CONCLUSÕES: Estudos de coorte de nascimentos podem ser realizados com sucesso em países em desenvolvimento, e a metodologia empregada nesses estudos de ciclo vital permite estudar a influência de exposições precoces sobre a determinação das doenças da vida adulta.

OBJECTIVE: To evaluate metabolic changes associated with highly active antiretroviral therapy (HAART) in HIV-positive patients, and to identify risk factors associated. METHODS: Retrospective study that included 110 HIV-positive patients who where on HAART in the city of Porto Alegre (Southern Brazil) between January 2003 and March 2004. Data on demographic variables, cigarette smoking, diabetes mellitus, cholesterol and triglyceride levels, stage of HIV infection, antiretroviral therapy and HCV coinfection were collected. General linear models procedure for repeated measures was used to test the interaction between HAART and HCV coinfection or protease inhibitor treatment. RESULTS: Total cholesterol, triglycerides, and glucose levels significantly increased after receiving HAART (p<0.001 for all variables), but no interaction with protease inhibitors was seen for total cholesterol, glucose and triglyceride levels (interaction treatment*protease inhibitors p=0.741, p=0.784, and p=0.081, respectively). An association between total cholesterol levels and HCV coinfection was found both at baseline and follow-up (effect of HCV coinfection, p=0.011). Glucose levels were increased by HAART (treatment effect, p=0.036), but the effect was associated to HCV coinfection (treatment*HCV effect, p=0.018). Gender, smoking habit, intravenous drug use and age were not significantly associated with cholesterol, triglyceride and glucose changes. CONCLUSIONS: HCV-infected patients at baseline were significantly less likely to develop hypercholesterolemia. The results provide further evidence of the role of HAART for the development of metabolic disturbances.

Biphentrin, a known pyrethroid, was studied, aiming its removal from aqueous solutions by granulated cork sorption. Batch experiments, either for equilibrium or for kinetics, with two granulated cork sizes were performed and results were compared with those obtained with of activated carbon sorption. Langmuir and Freundlich adsorption isotherms were obtained both showing high linear correlations. Bifenthrin desorption was evaluated for cork and results varied with the granule size of sorbent. The results obtained in this work indicate that cork wastes may be used as a cheap natural sorbent for bifenthrin or similar compounds removal from wastewaters.

Huntington's disease (HD) is an incurable genetic neurodegenerative disorder that leads to motor and cognitive decline. It is caused by an expanded polyglutamine tract within the Huntingtin (HTT) gene, which translates into a toxic mutant HTT protein. Although no cure has yet been discovered, novel therapeutic strategies, such as RNA interference (RNAi), antisense oligonucleotides (ASO), ribozymes, DNA enzymes, and genome-editing approaches, aimed at silencing or repairing the mutant HTT gene hold great promise. Indeed, several preclinical studies have demonstrated the utility of such strategies to improve HD neuropathology and symptoms. In this review, we critically summarise the main advances and limitations of each gene-silencing technology as an effective therapeutic tool for the treatment of HD.
