Opinnäytetyö on monimuototyö, jonka teososan muodostaa dokumentti myllystä ja nykypäivän mylläristä. Kyseinen mylly on yksi noin sadasta vielä toiminnassa olevasta myllystä. Myllyn uudelleen käyttöönottoa edelsi monta vaihetta koneiden puhdistamisesta jauhatusprosessin opettelemiseen. Mylläri viljelee itse ja jauhaa luomuviljan omassa myllyssään jauhoiksi. Vaimo leipoo niistä myllypuodissa myytävät leivät ja leivonnaiset. Dokumentin ajatus oli kuvata toimiva valssimylly mylläreineen, vielä kun niiden taltioiminen on mahdollista, tallentaa pala katoavaa kulttuurihistoriaa. Toimin dokumentin käsikirjoittajana, ohjaajana, tuottajana, toisena kuvaajana ja leikkaajana. Teososan tekeminen on ollut lähtökohtana kirjallisen osan pohdinnoille. Kirjallisessa osassa tutkin, kuinka tunteita tuodaan kuvallisesti ja leikkauksellisesi esille dokumenttielokuvassa. Tavoitteenani on selvittää, millä leikkausteknisillä keinoilla voidaan elokuvaan luoda lisää tunnetta ja mahdollisesti vaikuttaa katsojaan. Tarkastelen ja reflektoin aihetta ohjaaja-leikkaajan näkökulmasta. Käyn läpi Mylly-dokumentin tekoprosessin ja keskityn leikkauksen eri vaiheisiin. Tiedonhankinta perustui Myllyliitto ry:n edustajan ja myllärin haastatteluihin, kotimaisiin dokumenttielokuviin ja kirjallisuuteen, joka käsittelee pääasiassa leikkaustekniikoita. Lisäksi tuon esiin omia huomioitani dokumentin tekijänä. - Opinnäytetyöhön kuuluu teososa, dokumenttielokuva Mylly.

En septembre 2008, une vidéo est diffusée sur internet montrant Sarah Palin, alors candidate à la vice-présidence des États-Unis, en train de recevoir « une protection contre la sorcellerie » lors d'une célébration religieuse. La séquence est aussitôt reprise par les médias du monde entier qui s'interrogent et enquêtent sur les rapports entre ces images et l'engagement politique de la républicaine. Cet article suit la circulation de la vidéo au travers de différentes scènes de visibilité, allant des médias généralistes au site internet de l'Église où elle a été tournée, en passant par un blog qui a attiré l'attention publique sur ces images. L'enquête met au jour comment ce film a été compris dans ces différents contextes, et montre les effets de ce travail d'interprétation en 2011, suite à l'avènement du Tea Party et à l'horizon de la prochaine échéance présidentielle. La vidéo qui, en 2008, apparaissait dans les médias généralistes comme une illustration de la « bigoterie » de Palin a contribué à figurer, en 2011, une menace « théocratique » pour la démocratie américaine.

An alternative analytical method for nitrogen dioxide (NO2) in atmosphere was developed. The collection of NO2 is performed by a Sep-Pack C-18 cartridge impregnated with 11% (v/v) of triethanolamine plus 3,6 % (v/v) of ethylene glycol plus 25 % (v/v) of acetone combined solution. When the impregnating is used, NO2 is collected with good repeatibility (CV = 3,3 %). The NO2 absorbed in the sampler was stripped from the sorbent with a methanol 5% (v/v) aqueous solution and was determined by colorimetry as nitrite by using the Griess-Saltzman reagent. The detection limit of 1,4 ppb for 60 min sampling at 0,5 L min-1 flow rate was obtained. Preparation and conditioning procedures for TEA-C-18 cartridge, sampling flow rate, absorption capacity and interference of other species are discussed.

Caffeine extraction procedures from water soluble and water insoluble materials for preparing stimulating beverages are described. Water soluble materials used were instant tea and coffee and water insoluble materials were, among others, guaraná powder and maté leaves. The extraction of caffeine from water soluble materials, especially instant tea, is more suitable for an organic chemistry teaching laboratory than the classic experiment using tea leaves, due to the economy of time and a larger amount of extracted caffeine. The procedure is time-saving and requires only a four-hour period. The experiments illustrate the extraction process as used in undergraduate organic chemistry laboratories.

Chromium(III) at the ng L-1 level was extracted using partially silylated MCM-41 modified by a tetraazamacrocyclic compound (TAMC) and determined by inductively coupled plasma optical emision spectrometry (ICP OES). The extraction time and efficiency, pH and flow rate, type and minimum amount of stripping acid, and break- through volume were investigated. The method's enrichment factor and detection limit are 300 and 45.5 pg mL-1, respectively. The maximum capacity of the 10 mg of modified silylated MCM-41 was found to be 400.5±4.7 µg for Cr(III). The method was applied to the determination of Cr(III) and Cr(VI) in the wastewater of the chromium electroplating industry and in environmental and biological samples (black tea, hot and black pepper).

The objective of this paper was to determine nitrate and nitrite anions in toasted fish skin tea, which the population of southern Rio Grande do Sul believes to have pharmacological properties for trating asthma. Two methods were compared, an official and one described by Guozhen et al., that has the advantage of not needing reduction of nitrate to nitrite on a Cd column. The samples were the skin of the fish species Balistes capriscus (peixe porco) and Menticirrhus littoralis (papa-terra), both crude and toasted, as traditionally used. The method of Guozhen et al. was used due to its performance (detection limit 0.005 µmol/L NO3- and 0.02 µmol/L - NO2- and mean recuperation of 100 and 97% respectively for each anion) and results similar to those of the official methods under conditions used by the population, the estimated daily intake, of the teas is equivalent to 0.03 to 0.05 mg of nitrate and 0.007 to 0.014 mg of nitrite. These values are in accord with safety recommendations and don't represent a health risk.

Pectic substances are structural heteropolysaccharides that occur in the middle lamellae and primary cell walls of higher plants. They are composed of partially methyl-esterified galacturonic acid residues linked by alpha-1, 4-glycosidic bonds. Pectinolytic enzymes are complex enzymes that degrade pectic polymers and there are several classes of enzymes, which include pectin esterases, pectin and pectate lyases and polygalacturonases. Plants, filamentous fungi, bacteria and yeasts are able to produce pectinases. In the industrial world, pectinases are used in fruit juice clarification, in the production of wine, in the extraction of olive oil, fiber degumming and fermentation of tea, coffee and cocoa.

The aim of this work was to study the influence of green tea consumption on fluoride ingestion. The extraction conditions of fluorides from green tea infusions were defined and nine brands of green tea available in Portugal were analyzed. The quantification of fluorides in the green tea was preceded by the implementation and validation of the potentiometric method (commercial fluoride selective electrode). The concentration of fluorides in the samples ranged from 0. 8 to 2. 0 mg L-1.

In this work a new experiment using HPLC is proposed in order to explore the role of acidity and the organic modifiers in the determination of methylxanthines in tea and coffee. Multivariate and univariate optimizations of the experimental conditions were used.

Levels of Ca, Mg, Fe, Zn, Cu and Mn were determinated by FAAS in the tea and dry matter from senescent leaf from Montrichardia linifera, plant used in folk medicine Amazon. The content of these metals that are transferred of the leaf to the infusions have presented significant reductions, however, the Mn values in the infusion may exceed the tolerable daily intake (11 mg) if consumption of this tea is greater than 1.0 L per day. So the tea of senescent leaves of M. linifera may be considered as a toxic beverage and thus its use is not advised.

The goal of this study was to evaluate the feasibility of preparing nanocapsules and nanoemulsions using tea tree oil as oily phase aiming to protect its volatilization. The nanostructures presented nanometric mean size (160-220 nm) with a polydispersity index below 0.25 and negative zeta potential. The pH values were 6.43 ± 0.37 and 5.98 ± 0.00 for nanoemulsions and nanocapsules, respectively. The oil content after preparation was 96%. The inclusion of tea tree oil in nanocapsules showed higher protection against volatilization. The analysis of mean size and polydispersity index of formulations presented no significant alteration during the storage time.

A simple and sensitive method has been proposed for the determination of sibutramine-HCl in energy drinks, green tea and pharmaceutical formulations using differential pulse voltammetry performed on a hanging mercury drop electrode. In the chosen experimental condition (Mcllvaine pH 4.0 buffer, 50 mV pulse amplitude and 40 mV s-1 scan velocity), sibutramine-HCl presented a reversible behavior and a peak maximum at -80 mV. Detection limit was 0.4 mg L-1 and the working linear range extended up to 33.3 mg L-1 (r = 0.99). Analysis of real and fortified samples enabled recoveries between 91 and 102%. The electroanalytical method was compared with a HPLC method which indicated it accuracy.

The present study proposes a method for cloud point preconcentration of copper ions at pH 2.0 based on complexes formed with [4,5-dimercapto-1,3-dithyol-2-thionate] and subsequent determination by flame atomic absorption spectrometry (FAAS). Under optimal analytical conditions, the method provided limits of detection of 0.84 and 0.45 µg L-1, by preconcentrating 12.0 and 24.0 mL of sample, respectively. The method was applied for copper determination in water samples, synthetic saliva, guarana powder, tea samples and soft drinks and the accuracy was assessed by analyzing the certified reference materials Dogfish Liver (DOLT-4) and Lobster Hepatopancreas (TORT-2).

A new solid phase extraction (SPE) method has been developed for the selective separation and preconcentration of Cu (II) ions in food and water samples prior to its flame atomic absorption spectrometry determination. The method is based on the adsorption of the Cu(II) - 2-{[4-Amino-3-(4-methylphenyl-5-oxo-4,5-dihydro-1H-1,2,4-triazol-1-yl]acetyl}-N-phenyl hydrazinecarbothioamide complex on Amberlite XAD-8 resin. The metal complex retained on the resin was eluted with 7.5 mL of 2.0 mol L-1 HCl in acetone. The optimum conditions for the SPE of Cu(II) ions were investigated, and the method was subsequently applied to sea water, stream water, rice, tea, and tobacco samples for the determination of Cu(II) levels.
