The use of morphological data obtained from field (plot test) and glasshouse trials to identify and discriminate among four Iranian and two New Zealand lucerne (Medicago sativa L.) cultivars was investigated, following guidelines established by the International Union for the Protection of New Varieties of Plants (UPOV) for cultivar registration and the Organisation for Economic Co-operation and Development (OECD) for seed certification. Data were collected for terminal leaflet length, width and ratio, angle of stem growth, date of first flowering, stem height at first flowering, flower colour, cutting recovery height, and disease scores. None of these characters were sufficient to identify or discriminate among the six cultivars. The results indicate a need to find cost-effective and efficient laboratory techniques to enhance the assessment of distinctness of lucerne cultivars (UPOV) and for determining cultivar purity for lucerne seed certification (OECD).

The effect of soil puddling on growth of lowland rice (Oryza sativa) and post-rice mungbean (Vigna radiata) was investigated using mini rice beds under controlled glasshouse conditions. Each mini rice bed was approximately 1 m(3) in size. Three different soil types were used: a well-drained, permeable loam; a hardsetting, structurally unstable silty loam; and a medium clay. Rice yields were reduced by low puddling compared with high puddling intensity on the loam but not affected on the heavier textured soils (silty loam and clay). Yield of mungbean was reduced on highly puddle, structurally unstable soil, indicating that puddling should be reduced on structurally unstable soils. Under glasshouse condition where crop establishment was not a limiting factor and plant available water in 0.65 m of soil was 100 mm, mungbean yields of >1 t/ha were achieved. However, under conditions where subsoil water reserves were depleted for the production of vegetative biomass during initial optimal growing condition, grain yield remained well below 1 t/ha.

We have mapped and identifed DNA markers linked to morphology, yield, and yield components of lucerne, using a backcross population derived from winter-active parents. The high-yielding and recurrent parent, D, produced individual markers that accounted for up to 18% of total yield over 6 harvests, at Gatton, south-eastern Queensland. The same marker, AC/TT8, was consistently identified at each individual harvest, and in individual harvests accounted for up to 26% of the phenotypic variation for yield. This marker was located in linkage group 2 of the D map, and several other markers positively associated with yield were consistently identified in this linkage group. Similarly, markers negatively associated with yield were consistently identified in the W116 map, W116 being the low-yielding parent. Highly significant positive correlations were observed between total yield and yield for harvests 1-6, and between total yield and stem length, tiller number, leaf yield/plant, leaf yield/5 stems, stem yield/plant, and stem yield/5 stems. Highly significant QTL were located for all these characters as well as for leaf shape and pubescence.

An experiment was conducted in 2013 and 2014 with three newly introduced cultivars of apricot (Prunus armeniaca L.), namely “Antonio Errani”, “Tirynthos” and “Ninfa” to study their performance and adaptability under Egyptian conditions. Results indicated that calculating the chilling hours temperature at or below 15°C was more suitable than temperatures at or below 7.2°C and 10°C. The cultivar with a low chilling requirement started with the opening of vegetative and flower buds earlier when compared to other cultivars. Furthermore, the cultivar Ninfa required less heat units as compared to the other two cultivars. Thus, the accumulated growing degree-days (GDDs) from the time of the flower bud break l until fruit maturity was low in early matured Ninfa cultivar. However, Antonio Errani and Tirynthos cultivars were late in the date of fruit ripening. Meanwhile, there was no significant difference in the opening percentage of vegetative and flower buds, trunk circumference, fruit drop, fruit number and yield weight among cultivars during the two seasons. Conversely, the leaf drop of Antonio Errani cultivar was earlier while Ninfa cultivar started it’s leaf drop later in the two seasons. Tirynthos gave the highest fruit weight, fruit size and fruit surface lightness. Meanwhile, the Antonio Errani cultivar was the highest in fruit firmness and total soluble solids. The appearance and behavior of cultivars under the study varied from one season to another with shoot length, leaf area, percentage of fruit set and acidity. It can be recommended from the present study that, Antonio Errani, Tirynthos and Ninfa cultivars are well adapted under Egyptian conditions. Further, fruits from the cultivars mature early and late in the season and can fulfill the demands of the market.

Il controllo degli artropodi infestanti le derrate in post-raccolta si basa principalmente sull’utilizzo di molecole chimiche di sintesi. I loro residui – spesso presenti nei prodotti alimentari finiti – hanno destato nell’opinione pubblica crescenti preoccupazioni di ordine igienico sanitario. Anche la legislazione Europea – dal canto suo – impone limiti restrittivi alla loro presenza nei cibi e al loro uso. Nell’ambito delle tecniche alternative ai pesticidi di sintesi, gli oli essenziali mostrano interessanti potenzialità per il controllo degli insetti. Il lavoro svolto in questa tesi si è proposto di valutare lefficacia insetticida di due oli essenziali commerciali, estratti da Cannabis sativa L. e Coridothymus capitatus (L.) Rchb. f., nel controllo di Tribolium castaneum Herbst, uno degli insetti più dannosi delle derrate. La loro caratterizzazione chimica ha permesso di individuare alcune molecole che presumibilmente sono responsabili dell’azione tossica nei confronti del coleottero. I risultati ottenuti mostrano un’attività insetticida di entrambi gli oli essenziali in condizione di temperatura modificata. All’interno di un moderno protocollo di lotta integrata agli insetti, gli oli essenziali potranno effettivamente rappresentare uno tra i diversi ausili tecnici alternativi agli insetticidi di sintesi, che puntino a ridurre al minimo la presenza di residui chimici nei prodotti alimentari.

We studied the effect of Silicon (Si) on Casparian band (CB) development, chemical composition of the exodermal CB and Si deposition across the root in the Si accumulators rice and maize and the Si non-accumulator onion. Plants were cultivated in nutrient solution with and without Si supply. The CB development was determined in stained root cross-sections. The outer part of the roots containing the exodermis was isolated after enzymatic treatment. The exodermal suberin was transesterified with MeOH/BF3 and the chemical composition was measured using gas chromatography-mass spectroscopy (GC-MS) and flame ionization detector (GC-FID). Laser ablation-inductively coupled plasma-mass spectroscopy (LA-ICP-MS) was used to determine the Si deposition across root cross sections. Si promoted CB formation in the roots of Si-accumulator and Si non-accumulator species. The exodermal suberin was decreased in rice and maize due to decreased amounts of aromatic suberin fractions. Si did not affect the concentration of lignin and lignin-like polymers in the outer part of rice, maize and onion roots. The highest Si depositions were found in the tissues containing CB. These data along with literature were used to suggest a mechanism how Si promotes the CB development by forming complexes with phenols.

During surveys of wild and cultivated rice in northern Queensland in 2014 and 2015, 92 fungal isolates were obtained from plants that were afflicted by foliar diseases, including the rice blast pathogen, Pyricularia oryzae, and the brown spot pathogen, Bipolaris oryzae. Seven species of Curvularia were found, viz. Curvularia aeria, C. alcornii, C. asianensis, C. clavata, C. lunata, C. muehlenbeckiae and an undescribed species. To remove uncertainty about the identity of the host plants from which the fungi were isolated, a DNA barcoding strategy was developed using regions of the chloroplast genome. Pathogenicity tests using wild rice isolates of P. oryzae indicated that many local rice varieties are susceptible to infection.

Se presentan los resultados de las fases de evaluación - selección de lneas avanzadas de arroz con alto contenido de zinc a nivel de vivero, evaluación de lneas avanzadas y evaluación multi - ambiental. Estos ensayos se establecieron en condiciones de riego y secano del 2014 y 2015, en el Centro Experimental de Arroz “TAINIC” y en diferentes regiones de secano del sub ciclo invierno del 2015. El Diseño empleado fue de Bloques Incompletos No Balanceados (Alpha Latice ). Se midieron datos de comportamiento agronómico, calidad de grano y contenido de zinc en el grano pulido mediante la metodología de fluorescencia de rayos X (FRX). De los genotipos evaluados se seleccionaron cuatro que combinan características agronómicas, industriales y nu tricionales deseables . Se estableció una relación positiva entre la cantidad de zinc disponible en el suelo y su influencia en la cantidad de zinc encontrada en grano pulido. Se determinó diferencias significativas entre los Genotipos, Ambientes e Interacc ión Genotipo*Ambiente. Se seleccionaron genotipos que presentan un comportamiento adaptativo amplio y genotipos con adaptación específica para cada condición agroclimática y de manejo . Se identificaron dos Mega-ambientes con genotipos en común. En el primero formado por los sitios de Pantasma y Jalapa destaca el genotipo IR31917-45-3-2-1-1SR-5-M(T4). El segundo Mega-ambiente formado por los sitios de Masatepe y San Carlos destaca el genotipo CT18504-4-5-3Vi-2-3P(T14). En los ambientes únicos de Posoltega y Darío los mejores genotipos fueron IR75870-5-8-5-B-2-B-1SR-8-M (T6) y CT19298-(100)-1-2-3-1-4MP (T1) respectivamente. Las variables que más influenciaron en la producción son en primer lugar días a madures, confirmando la tendencia de que a ciclo más extenso del cultivo habrá mayor rendimiento; en segundo lugar, la variable más influyente es longitud de panícula.

Olive (Olea europaea L.), one of the main crops in the Mediterranean basin, is mainly propagated by cuttings, a classical propagation method that relies on the ability of the cuttings to form adventitious roots. While some cultivars are easily propagated by this technique, some of the most interesting olive cultivars are considered difficult-to-root which poses a challenge for their preservation and commercialization. Therefore, increasing the current knowledge on adventitious root formation is extremely important for species like olive. This research focuses on evaluating the role of free auxins and oxidative enzymes on adventitious root formation of two olive cultivars with different rooting ability - ‘Galega vulgar’ (difficult-to-root) and ‘Cobrançosa’ (easy-to-root). In this context, free auxin levels and enzyme activities were determined in in vitro-cultured ‘Galega vulgar’ microshoots and in semi-hardwood cuttings of cvs. ‘Galega vulgar’ and ‘Cobrançosa’. To attain this goal, an analytical method for the quantification of free indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) was developed, which is based on dispersive liquid-liquid microextraction followed by microwave derivatization (DLLME-MAD) and gas chromatography-mass spectrometry (GC/MS) analysis. The developed method was validated in terms of linearity, recovery, limit of detection (LOD) and limit of quantification (LOQ) and proved to be useful in the analysis of two very different types of plant tissues. The results from auxin quantification in olive samples point at a relationship between free auxin levels and rooting ability of both microshoots and semihardwood cuttings. A defective IBA-IAA conversion, resulting in a peak of free IAA during initiation phase, seems to be associated with low rooting ability. Likewise, differences in the activity of oxidative enzymes also appear to be related with rooting ability. Higher polyphenol oxidases (PPO) activity is likely related with an easyto- root behavior, while the opposite is true for peroxidases (POX) (including IAA oxidase (IAAox)) activity. A possible hypothesis for adventitious root formation in olive microcuttings is presented herein for the first time. Free auxins, oxidative enzymes, alternative oxidase (AOX) and reactive oxygen species (ROS) are some of the factors that may be involved in this highly complex physiological process. Interestingly, while temporal changes in auxin levels were similar between microshoots and semihardwood cuttings, the conclusions obtained from enzyme activity results in microshoots didn’t translate to semi-hardwood tissues, showing the emerging need for adaptation of classical agronomical research studies to modern techniques; Resumo: Procurando compreender o papel das auxinas e enzimas oxidativas na formação de raízes adventícias em cultivares de oliveira (Olea europaea L.) A oliveira (Olea europaea L.) é uma das principais culturas da bacia Mediterrânica e é propagada maioritariamente por estacaria, um processo altamente dependente da capacidade das estacas para formar raízes adventícias. Enquanto algumas cultivares são fáceis de propagar desta forma, algumas das cultivares de oliveira mais interessantes são consideradas difíceis de enraizar, o que dificulta a sua preservação e comercialização e torna extremamente importante aprofundar o conhecimento sobre o enraizamento adventício desta espécie. Este trabalho foca-se na avaliação do papel das auxinas livres e das enzimas oxidativas na formação de raízes adventícias em duas cultivares de oliveira com diferente capacidade de enraizamento - ‘Galega vulgar’ (difícil de enraizar) e ‘Cobrançosa’ (fácil de enraizar). Neste contexto, determinaram-se os níveis de auxinas livres e as actividades de enzimas oxidativas em microestacas de ‘Galega vulgar’ cultivadas in vitro bem como em estacas semi-lenhosas das cvs. ‘Galega vulgar’ e ‘Cobrançosa’. Para tal foi necessário desenvolver uma metodologia analtica para a quantificação de ácido indol-3-acético (IAA) e ácido indol-3-butírico (IBA), baseada em microextracção dispersiva lquido-lquido (DLLME) seguida de derivatização em microondas (MAD) e análise por cromatografia gasosa acoplada a espectrometria de massa (GC/MS). O método desenvolvido foi validado em termos de linearidade, recuperação, limite de detecção (LOD) e limite de quantificação (LOQ), e mostrou-se eficaz na análise de dois tipos de tecidos vegetais bastante diferentes. Os resultados da análise de auxinas em amostras de oliveira apontam para uma possível relação entre os níveis de auxinas livres e a capacidade de enraizamento, tanto em microestacas como em estacas semi-lenhosas. Uma conversão IBA-IAA deficiente, que resulta num pico de IAA durante a fase de iniciação, parece estar associada à baixa capacidade de enraizamento. Por outro lado, a capacidade de enraizamento também parece estar relacionada com diferenças na actividade de enzimas oxidativas. Comportamentos fáceis de enraizar estão associados a actividade mais elevada das polifenoloxidases (PPO), enquanto o oposto é verdade para a actividade das peroxidases (POX) (incluindo a IAA oxidase (IAAox)). Neste trabalho propõe-se pela primeira vez uma possível explicação para o enraizamento adventício em microestacas de oliveira. Auxinas livres, enzimas oxidativas, oxidase alternativa (AOX) e espécies reactivas de oxigénio (ROS) são alguns dos factores envolvidos neste processo fisiolgico altamente complexo. Curiosamente, enquanto as alterações temporais nos níveis de auxinas foram semelhantes entre microestacas e estacas semi-lenhosas, o mesmo não se observou relativamente à actividade enzimática, o que mostra a necessidade de adaptação dos estudos agronómicos tradicionais às técnicas correntes.

Background: The present study was undertaken towards the development of SSR markers and assessing genetic relationships among 32 date palm ( Phoenix dactylifera L.) representing common cultivars grown in different geographical regions in Saudi Arabia. Results: Ninety-three novel simple sequence repeat markers were developed and screened for their ability to detect polymorphism in date palm. Around 71% of genomic SSRs were dinucleotide, 25% tri, 3% tetra and 1% penta nucleotide motives. Twenty-two primers generated a total of 91 alleles with a mean of 4.14 alleles per locus and 100% polymorphism percentage. A 0.595 average polymorphic information content and 0.662 primer discrimination power values were recorded. The expected and observed heterozygosities were 0.676 and 0.763 respectively. Pair-wise similarity values ranged from 0.06 to 0.89 and the overall cultivars averaged 0.41. The UPGMA cluster analysis recovered by principal coordinate analysis illustrated that cultivars tend to group according to their class of maturity, region of cultivation, and fruit color. Analysis of molecular variations (AMOVA) revealed that genetic variation among and within cultivars were 27% and 73%, respectively according to geographical distribution of cultivars. Conclusions: The developed microsatellite markers are additional values to date palm characterization tools that can be used by researchers in population genetics, cultivar identification as well as genetic resource exploration and management. The tested cultivars exhibited a significant amount of genetic diversity and could be suitable for successful breeding program. O objetivo deste trabalho foi avaliar o efeito de diferentes dosagens de vermicomposto sobre a produção de alface, cultivar Baba de Verão. O ensaio foi conduzido em Latossolo Amarelo Distrófico de textura média fase caatinga litorânea, na Embrapa Meio-Norte, UEP - Parnaíba, de setembro a outubro de 2013. O delineamento experimental adotado foi o de blocos casualizados, com cinco tratamentos e quatro repetições.

In Europe, the current demand for vegetable oils and the need to find alternative crops for the regions most affected by climate change (i.e., Mediterranean basin) may be a launchpad for camelina [Camelina sativa (L.) Crantz] to be steadily introduced in European cropping systems. Camelina is mainly known for the unique composition of its oil, with a fatty acids profile including more than 50% content of essential linoleic and linolenic fatty acids, and a high tocopherol content. Being tocopherols part of the vitamin E family of antioxidants, the added value of growing camelina in harsh environments could be the enhancement of tocopherol content in camelina oil, thus having a more stable and nutritionally valuable product. With the final purpose of fully valorize camelina as a tolerant, valuable-oil producing crop for the Mediterranean basin, the main aim of this study was to investigate whether and how sowing date, cultivar choice, and abiotic stresses can affect tocopherol content and composition in camelina oil. The results showed that cultivar choice and growing conditions influenced total tocopherol, γ-tocopherol, and α-tocopherol contents. Moreover, heat stress trial revealed that high temperature increased α-tocopherol content, while no effect was observed in total tocopherols and in γ-tocopherol content. Finally, drought increased total tocopherols in camelina, and in drought-sensitive lines an increase in α-tocopherol was observed. This study allowed to acquire awareness on camelina resistance to abiotic stresses, coupled with a better knowledge on tocopherol content and composition in relation to cultivar, sowing date, and abiotic stresses. This will have an impact for the introduction of camelina as an alternative crop in harsher environments, such as the Mediterranean basin, to produce an oil suitable for food, feed, and industrial applications.
