Echovirus (Echo) 30 or human enterovirus B is the most frequent enterovirus associated with meningitis cases. Epidemics and outbreaks of this disease caused by Echo 30 have occurred in several countries. In Brazil, Echo 30 has been isolated from sporadic cases and outbreaks that occurred mainly in the south and southeast regions. We used RT-PCR to examine Echo 30 isolates from meningitis cases detected from March 2002 to December 2003 in Belém, state of Pará, in northern Brazil. The patients were attended in a Basic Health Unit (State Health Secretary of Pará), where cerebrospinal fluid (CSF) was collected and stored in liquid nitrogen. Weekly visits were made by technicians from Evandro Chagas Institute to the health unit and samples were stored at -70ºC in the laboratory until use. HEp-2 and RD cell lines were used for viral isolation and neutralization with specific antisera for viral identification. RNA extraction was made using Trizol reagent. The RT-PCR was made in one step, and the total mixture (50 µL) was composed of: RNA, reaction buffer, dNTP, primers, Rnase inhibitor, reverse transcriptase, Taq polymerase and water. The products were visualized in agarose gel stained with ethidium bromide, visualized under UV light. Among the 279 CSF samples examined, 30 (10.7%) were EV positive, 29 being Echo 30 and one was Cox B. Nineteen Echo 30 were examined with RT-PCR; 18 tested positive (762 and 494 base pairs). The use of this technique permitted viral identification in less time than usual, which benefits the patient and is of importance for public-health interventions.

O resíduo do refino da bauxita, comumente conhecido como lama vermelha, tem mostrado ser capaz de ligar metais traços tais como o cobre, cádmio, zinco, cromo, níquel e chumbo sob condições apropriadas. A neutralização da lama vermelha é necessária porque essa lama é tipicamente cáustica, com pH em torno de 13. Muitas pesquisas têm sido desenvolvidas envolvendo a utilização da lama vermelha, porque contem uma série de elementos valiosos. Assim a utilização da lama vermelha como material de construção e como adsorvente é atrativa porque ela é utilizada em grande quantidade. Nada pode ser feito sem descartar componentes valiosos como titânio, zircônio e principalmente ferro. A síntese de hidrotalcita conectada com a reciclagem de soluções alcalinas representa uma maneira efetiva de uso dessa solução considerando a forte demanda por proteção ambiental no mundo. Os compostos tipo hidrotalcita, precipitados durante a neutralização da lama, também removem oxi-ânions de metais de transição através da combinação de intercalações e adsorção de espécies aniônicas na sua superfície externa. Os hidróxidos duplos lamelares (HDL) vêm sendo investigados à muitos anos como materiais hospedeiros para uma variedade de reações de intercalação de anions. A estrutura lamelar dos HDLs pode ser usada para controlar a adição ou remoção de uma variedade de espécies químicas, tanto orgânicas quanto inorgânicas. Isto é alcançado através de sua habilidade de adaptar a separação das lamelas de hidróxidos, e da reatividade da região interlamelar. O material resultante adsorve anions quando colocado em soluções e reverte a estrutura da hidrotalcita. Avanços significativos têm sido alcançados recentemente na caracterização desses materiais, incluindo estudos estruturais sobre o mecanismo de intercalação. O objetivo desse trabalho foi a síntese e caracterização de HDLs a partir da lama vermelha, e testar esses materiais como adsorventes de metais pesados.

O beneficiamento de caulim para cobertura de papel produz grandes volumes de rejeito, constituído essencialmente do argilomineral caulinita, usado neste trabalho como material de partida para síntese de alúmen de amônio, visando a obtenção de alumina livre de sódio e de baixa granulometria para utilização cerâmica. O método de síntese desenvolvido para obtenção do alúmen de amônio constitui-se das etapas: calcinação do rejeito, lixiviação sulfúrica da metacaulinita, seguida da neutralização/cristalização da solução de Al2(SO4)3 com NH4OH concentrado. No processo, são estudadas as variáveis: temperatura de calcinação (650 °C, 700 °C, 750 °C) e tempo de calcinação do rejeito (30, 60 e 120 min), concentração de H2SO4, temperatura de lixiviação (70 °C, 80 °C e 90 °C) sobre a cinética de lixiviação do alumínio. Estudou-se também o efeito do pH na cristalização do alúmen de amônio, apresentando-se as análises químicas, DRX, ATD, ATG e granulometria dos materiais utilizados e sintetizados.

The efficiency of sources used for soil acidity correction depends on reactivity rate (RR) and neutralization power (NP), indicated by effective calcium carbonate (ECC). Few studies establish relative efficiency of reactivity (RER) for silicate particle-size fractions, therefore, the RER applied for lime are used. This study aimed to evaluate the reactivity of silicate materials affected by particle size throughout incubation periods in comparison to lime, and to calculate the RER for silicate particle-size fractions. Six correction sources were evaluated: three slags from distinct origins, dolomitic and calcitic lime separated into four particle-size fractions (2, 0.84, 0.30 and <0.30-mm sieves), and wollastonite, as an additional treatment. The treatments were applied to three soils with different texture classes. The dose of neutralizing material (calcium and magnesium oxides) was applied at equal quantities, and the only variation was the particle-size material. After a 90-day incubation period, the RER was calculated for each particle-size fraction, as well as the RR and ECC of each source. The neutralization of soil acidity of the same particle-size fraction for different sources showed distinct solubility and a distinct reaction between silicates and lime. The RER for slag were higher than the limits established by Brazilian legislation, indicating that the method used for limes should not be used for the slags studied here.

Envenomation via snakebites is an important public health problem in many tropical and subtropical countries that, in addition to mortality, can result in permanent sequelae as a consequence of local tissue damage, which represents a major challenge to antivenom therapy. Venom phospholipases A(2) (PLA(2)s) and PLA(2)-like proteins play a leading role in the complex pathogenesis of skeletal muscle necrosis, nevertheless their precise mechanism of action is only partially understood. Recently, detailed structural information has been obtained for more than twenty different members of the PLA(2)-like myotoxin subfamily. In this review, we integrate the available structural, biochemical and functional data on these toxins and present a comprehensive hypothesis for their myotoxic mechanism. This process involves an allosteric transition and the participation of two independent interaction sites for docking and disruption of the target membrane, respectively, leading to a five-step mechanism of action. Furthermore, recent functional and structural studies of these toxins complexed with ligands reveal diverse neutralization mechanisms that can be classified into at least three different groups. Therefore, the data summarized here for the PLA(2)-like myotoxins could provide a useful molecular basis for the search for novel neutralizing strategies to improve the treatment of envenomation by viperid snakes. A peptide-based indirect ELISA was developed to detect antibodies against Equine arteritis virus (EAV). Two peptides for epitope C of protein GP5 and fragment E of protein M were designed, synthesized, purified and used as antigens either alone or combined. Ninety-two serum samples obtained from the 2010 Equine viral arteritis outbreak, analyzed previously by virus neutralization, were evaluated by the ELISA here developed. The best resolution was obtained using peptide GP5. The analysis of the inter- and intraplate variability showed that the assay was robust. The results allow concluding that this peptide-based ELISA is a good alternative to the OIE-prescribed virus neutralization test because it can be standardized between laboratories, can serve as rapid screening, can improve the speed of diagnosis of EAV-negative horses and can be particularly useful for routine surveillance in large populations. The purpose of this study was to evaluate the antimicrobial activity of chlorhexidine gel 2% as auxiliary chemical substance on the biomechanical preparation (BMP) and medication intracanal (ICM) on C. albicans, E. faecalis, E. coli and their endotoxin in root canals. We used 48 single-rooted human teeth divided into four groups according to dressing ICM: 1) Ca(OH)2 + pyrogen-free saline solution; 2) 2% chlorhexidine gel (CLX); 3) Ca(OH)2 + CLX, and; 4) pyrogen-free saline solution (control group). Were collected the contents of root canals to confirm the presence of microorganisms (confirmation), immediately after instrumentation (1st collection), after 7 days of the BMP (2nd collection), after 14 days of the action of ICM (3rd Collection) and 7 days after removal of the ICM (4 th collection). Were performed: the evaluation of antimicrobial activity and the content analysis of endotoxins for all sampling tests. The results were statistically analyzed using Kruskall-Wallis and Dunn tests with a significance of 5%. It was found that the CLX as auxiliary chemical substance has significantly reduced microorganisms confirmation collection when compared. In relation to the neutralization of endotoxin, it was found that the 1st and 2nd collections presented a decrease of 92.03% and 98.10% in mean percentage respectively, when compared to the confirmation collection. In the 3rd and 4th samplings, the Ca (OH)2 + CLX group showed the best results. It was concluded that the BMP and the ICM were able to eliminate the tested microrganisms, however, they were not able to completely eliminate endotoxins root canal
