Aquest projecte s'ha realitzat al Servei de Microscòpia de la Universitat Autònoma de Barcelona, i ha tingut una durada de dos anys (2007-2009). Els objectius concrets d'aquest projecte assolits per a les diferents assignatures ha estat l'anàlisi de característiques citològiques, característiques morfològiques i anatòmiques de les estructures vegetatives dels diferents nivells d’organització, característiques morfològiques i anatòmiques de les estructures reproductores. El material docent obtingut en aquest projecte està sent utilitzat en les assignatures següents: Botànica Farmacèutica, Biodiversitat vegetal marina, Recursos vegetals aquàtics: les algues, Plantes d'ús alimentari, Conserv. i gestió de poblacions i comunitats marines, Usos i cultius de recursos algals marins, B. i diversitat de criptògames, Biologia II, Tècniques Exp. de biologia II, Botànica, Biologia animal i vegetal. Per a la realització de l’estudi, s'ha obtingut una col·lecció d’imatges al MLC, MER i MET per aclarir els criteris sistemàtics. Durant el primer any s’han adquirit totes les imatges necessàries, per tal de poder elaborar el material didàctic en suport digital. Durant el segon any s’ha implementat aquest material a les respectives classes pràctiques i s’ha avaluat la seva importància en la millora de la formació dels alumnes i del suport al professor. L’aplicació del material elaborat a la docència pràctica durant el tot segon any ha permès fer un primer estudi per valorar la seva eficàcia i fer-hi les correccions oportunes. Durant tot el projecte, el treball conjunt de professors i tècnics ha estat bàsic per a l’obtenció de material didàctic adequat a la realitat dels estudis actuals i a la docència pràctica.

Molecular and stable carbon isotope compositions of source-specific hydrocarbons have been used to reconstruct palaeoenvironmental conditions during deposition of the Middle Hettangian to Upper Sinemurian sediments on the northern epicontinental Tethys margin, Frick Swiss Jura. Increasing algal, cyanobacterial and phytoplanktonic (i.e., dinoflagellate) contributions associated with the C-13-enrichment of cyanobacteria derivatives (i.e., hopanes and monomethylalkanes) suggest enhanced primary productivity upsection. This is related to the C-13-enrichment of dissolved CO2 in the upper layers and the progressive increase of depth and oxygenation of the water column. In the Middle Hettangian shallow-water environments (lagoon), the occurrence of green sulfur bacteria (Chlorobiaceae) derivatives indicates that the lower part of the water column was strictly anoxic and rich in H2S. Since these bacteria require very low light intensity to grow, these euxinic conditions may be extended up to the photic zone, allowing for anaerobic photosynthesis. Light penetration depth is most likely reduced by high productivity and/or turbidity in the photic zone. In these sediments, C-13-depleted hopanoids (-39.5 parts per thousand) are most likely associated with phototrophic purple sulfur bacteria utilizing isotopically light organic carbon at the base of the aerobic zone. These purple sulfur bacteria may have consumed the H2S used by Chlorobiaceae in the deeper layers and thus, sustained the algae and cyanobacteria productivity in the upper layers. The C-13-depleted carbonate (-13.3 parts per thousand) may be partially related to the anaerobic oxidation of the organic matter during bacterial sulfate-reduction. (c) 2006 Elsevier Ltd. Hydrocarbon distributions and stable isotope ratios of carbonates (delta(13)C(car), delta(18)O(car)), kerogen (delta(13)C(ker)), extractable organic matter (delta(13)C(EOM)) and individual hydrocarbons of Liassic black shale samples from a prograde metamorphic sequence in the Swiss Alps were used to identify the major organic reactions with increasing metamorphic grade. The studied samples range from the diagenetic zone (< 100 degrees C) to amphibolite facies (similar to 550 degrees C). The samples within the diagenetic zones (< 100 and 150 degrees C) are characterized by the dominance of C-< 20 n-alkanes, suggesting an origin related with marine and/or bacterial inputs. The metamorphic samples (200 to 550 degrees C) have distributions significantly dominated by C-12 and C-13 n-alkanes, C-14, C-16 and C-18 n-alkylcyclopentanes and to a lesser extend C-15, C-17 and C-21 n-alkylcyclohexanes. The progressive C-13-enrichment (up to 3.9 parts per thousand) with metamorphism of the C-> 17 n-alkanes suggests the occurrence of cracking reactions of high molecular weight compounds. The isotopically heavier (up to 5.6 parts per thousand) C-< 17 n-alkanes in metamorphic samples are likely originated by thermal degradation of long-chain homologous with preferential release of isotopically light C-1 and C-2 radicals. The dominance of specific even C-number n-alkylcyclopentanes suggests an origin related to direct cyclization mechanism (without decarboxylation step) of algal or bacterial fatty acids occurring in reducing aqueous metamorphic fluid conditions. The regular increase of the concentrations of n-alkylcycloalkanes vs. C-> 13 n-alkanes with metamorphism suggests progressive thermal release of kerogen-linked fatty acid precursors and degradation of n-alkanes. Changes of the steroid and terpenoid distributions are clearly related to increasing metamorphic temperatures. The absence of 18 alpha(H)-22,29,30-trisnorneohopane (Ts), the occurrence of 17 beta(H)-trisnorhopane, 17 beta(H), 21 alpha(H)-hopanes in the C-29 to C-31 range and 5 alpha(H),14 alpha(H),17 alpha(H)-20R C-27, C-29 steranes in the low diagenetic samples (< 100 degrees C) are characteristic of immature bitumens. The higher thermal stress within the upper diagenetic zone (150 degrees C) is marked by the presence of Ts, the disappearance of 17 beta(H)-trisnorhopane and thermodynamic equilibrium of the 22S/(22S + 22R) homohopane ratios. The increase of the alpha alpha alpha-sterane 20S/(20S + 20R) and 20R beta beta/(beta beta + alpha alpha) ratios (from 0.0 to 0.55 and from 0.0 to 0.40, respectively) in the upper diagenetic zone indicates the occurrence of isomerization reactions already at < 150 degrees C. However, the isomerization at C-20 (R -> S) reaches thermodynamic equilibrium values already at the upper diagenesis (similar to 150 degrees C) whereas the epimerisation at C-14 and C-17 (alpha alpha ->beta beta) arrives to constant values in the lower anchizone (similar to 200 degrees C). The ratios Ts vs. 17 alpha(H)-22,29,30-trisnorneohopane [(Ts/(Ts + Tm)] and 18 alpha(H)-30-norneohopane (C29Ts) vs. 17 alpha(H),21 beta(H)-30-norhopane [C29Ts/(C29Ts + C-29)] increase until the medium anchizone (200 to 250 degrees C) from 0.0 to 0.96 and from 0.0 to 0.44, respectively. An opposite trend owards lower values is observed in the higher metamorphic samples. The occurrence of specific hydrocarbons (e.g., n-alkylcyclopentanes, cadalene, hydrogenated aromatic compounds) in metamorphic samples points to kerogen degradation reactions most probably occurring in the presence of water and under reducing conditions. The changes of hydrocarbon distributions and carbon isotopic compositions of n-alkanes related to metamorphism suggest that the organic geochemistry may help to evaluate the lowest grades of prograde metamorphism. S’ha detectat un problema de terbolesa de les aigües de l’estany del Cortalet del PNAE. El sòl d’aquest estany és de tipus sòdic, si pateix alguna alteració és possible que es presenti una desmembració deguda a l’acció dispersant del sodi. Alguns estudis demostren que la vegetació actua com a agent estructurador d’aquest sòl i li dóna estabilitat al sediment. Segons això, podria ser que l’origen de la terbolesa de l’aigua fos causa d’aquesta acció dispersant del sodi en sòls mancats de vegetació. Per analitzar si aquesta és la causa de la terbolesa, s’ha dut a terme un assaig experimental in situ de les causes físiques de la terbolesa de l’aigua, on s’ha tractat d’aïllar una zona amb vegetació, d’una sense vegetació. Els resultats demostren que la terbolesa era major a les zones vegetades i descarten la hipòtesi de l’acció dispersant del sodi en el sediment en sòls mancats de vegetació. Els resultats en canvi suggereixen que les causes de la terbolesa podrien ser d’origen biològic. L'objectiu general del projecte de tesi és el d'estudiar la resposta de les espècies algals a canvis en la disponibilitat de nutrients (i.e. NH4+/NO3-/PO43-). La hipòtesi de partida és que les alteracions en les concentracions absolutes i relatives de nutrients provoquen canvis en l'activitat de les espècies algals que seran més o menys intensos en funció de l’espècie i que són fruit de les característiques eco-evolutives intrínseques de cadascuna. Els objectius concrets es diferencien per l’escala temporal a la que s’observa la resposta, que va des de minuts fins a setmanes: Objectiu 1. Analitzar canvis a curt termini en l’activitat de les espècies degut a augments puntuals en la concentració absoluta i relativa de nutrients (NH4+/NO3-/ PO43-). S'ha vist que algunes espècies redueixen l’activitat fotosintètica a curt termini per destinar tots els recursos a l’assimilació de nutrients (Elrifi i Turpin 1986). S’estudiarà si aquesta pauta es troba de forma generalitzada i si respon a patrons evolutius, funcionals (mida, forma...) o ecològics (disponibilitat de nutrients). Objectiu 2. Analitzar, a mig termini, l’activitat de les espècies algals a diferents concentracions de NH4+/NO3-/ PO43-. Les respostes s’intentaran modelar, de manera que els paràmetres es puguin considerar trets funcionals característics de les espècies. S'estudiarà si la variabilitat en aquests trets respon a patrons evolutius o a d’altres trets funcionals com per exemple la mida. Objectiu 3. Analitzar com afecta, a llarg termini, la disponibilitat de nutrients en la composició d’espècies. Es detectarà quines espècies són més sensibles a les condicions de NH4+:NO3- i N:P del medi, i com es relaciona això amb les respostes específiques trobades en els objectius anteriors.

The development, validation, comparison and evaluation of analytical methods for marine toxins rely on the availability of toxic material. Within the project JACUMAR PSP, our interest is mainly focused on autochthonous bivalve species with the toxic profile of Alexandrium minutum, since this is the principal species involved regionally in PSP outbreaks. Mussels and oysters were exposed during few days in the harbor of Vilanova i la Geltrú, to blooms reaching a maximum A. minutum concentration of 200,000 cells L-1 in 2008, and 40,000 and 800,000 cells L-1, in 2009. Mussels, oysters and clams were exposed to one bloom of 22,000 cells L-1 in the harbor of Cambrils in 2009. In all situations higher toxic levels analyzed by HPLC-FD with postcolumn oxidation were observed in mussels (i.e. 1,200-2,500 μg eq. STX kg-1) than in oysters (i.e. 60-800 μg eq. STX kg-1) exposed to the same bloom. Blooms with higher concentrations of A. minutum did not correspond to higher levels of PSP toxins in bivalves. These differences may be explained by differences in A. minutum population dynamics, toxin production or in the physiological state or behaviour of shellfish. These results confirm that mussels concentrate more PSP toxins from A. minutum than oysters and clams.

Pseudomonas fluorescens EPS62e was selected during a screening procedure for its high efficacy in controlling infections by Erwinia amylovora, the causal agent of fire blight disease, on different plant materials. In field trials carried out in pear trees during bloom, EPS62e colonized flowers until the carrying capacity, providing a moderate efficacy of fire-blight control. The putative mechanisms of EPS62e antagonism against E. amylovora were studied. EPS62e did not produce antimicrobial compounds described in P. fluorescens species and only developed antagonism in King’s B medium, where it produced siderophores. Interaction experiments in culture plate wells including a membrane filter, which physically separated the cultures, confirmed that inhibition of E. amylovora requires cell-to-cell contact. The spectrum of nutrient assimilation indicated that EPS62e used significantly more or different carbon sources than the pathogen. The maximum growth rate and affinity for nutrients in immature fruit extract were higher in EPS62e than in E. amylovora, but the cell yield was similar. The fitness of EPS62e and E. amylovora was studied upon inoculation in immature pear fruit wounds and hypanthia of intact flowers under controlled-environment conditions. When inoculated separately, EPS62e grew faster in flowers, whereas E. amylovora grew faster in fruit wounds because of its rapid spread to adjacent tissues. However, in preventive inoculations of EPS62e, subsequent growth of EPS101 was significantly inhibited. It is concluded that cell-to-cell interference as well as differences in growth potential and the spectrum and efficiency of nutrient use are mechanisms of antagonism of EPS62e against E. amylovora

This review paper reports the consensus of a technical workshop hosted by the European network, NanoImpactNet (NIN). The workshop aimed to review the collective experience of working at the bench with manufactured nanomaterials (MNMs), and to recommend modifications to existing experimental methods and OECD protocols. Current procedures for cleaning glassware are appropriate for most MNMs, although interference with electrodes may occur. Maintaining exposure is more difficult with MNMs compared to conventional chemicals. A metal salt control is recommended for experiments with metallic MNMs that may release free metal ions. Dispersing agents should be avoided, but if they must be used, then natural or synthetic dispersing agents are possible, and dispersion controls essential. Time constraints and technology gaps indicate that full characterisation of test media during ecotoxicity tests is currently not practical. Details of electron microscopy, dark-field microscopy, a range of spectroscopic methods (EDX, XRD, XANES, EXAFS), light scattering techniques (DLS, SLS) and chromatography are discussed. The development of user-friendly software to predict particle behaviour in test media according to DLVO theory is in progress, and simple optical methods are available to estimate the settling behaviour of suspensions during experiments. However, for soil matrices such simple approaches may not be applicable. Alternatively, a Critical Body Residue approach may be taken in which body concentrations in organisms are related to effects, and toxicity thresholds derived. For microbial assays, the cell wall is a formidable barrier to MNMs and end points that rely on the test substance penetrating the cell may be insensitive. Instead assays based on the cell envelope should be developed for MNMs. In algal growth tests, the abiotic factors that promote particle aggregation in the media (e.g. ionic strength) are also important in providing nutrients, and manipulation of the media to control the dispersion may also inhibit growth. Controls to quantify shading effects, and precise details of lighting regimes, shaking or mixing should be reported in algal tests. Photosynthesis may be more sensitive than traditional growth end points for algae and plants. Tests with invertebrates should consider non-chemical toxicity from particle adherence to the organisms. The use of semi-static exposure methods with fish can reduce the logistical issues of waste water disposal and facilitate aspects of animal husbandry relevant to MMNs. There are concerns that the existing bioaccumulation tests are conceptually flawed for MNMs and that new test(s) are required. In vitro testing strategies, as exemplified by genotoxicity assays, can be modified for MNMs, but the risk of false negatives in some assays is highlighted. In conclusion, most protocols will require some modifications and recommendations are made to aid the researcher at the bench. [Authors]

The amino acid sequence of mouse brain beta spectrin (beta fodrin), deduced from the nucleotide sequence of complementary DNA clones, reveals that this non-erythroid beta spectrin comprises 2363 residues, with a molecular weight of 274,449 Da. Brain beta spectrin contains three structural domains and we suggest the position of several functional domains including f-actin, synapsin I, ankyrin and spectrin self association sites. Analysis of deduced amino acid sequences indicated striking homology and similar structural characteristics of brain beta spectrin repeats beta 11 and beta 12 to globins. In vitro analysis has demonstrated that heme is capable of specific attachment to brain spectrin, suggesting possible new functions in electron transfer, oxygen binding, nitric oxide binding or heme scavenging.

Para el estudio de la microalga Dunaliella salina Teodoresco, se colectaron muestras de 2 lagunas hipersalinas; las Salinas de Chimus y las Salinas de Chilca. La metodología usada fue la técnica de micropipeta, tratamiento con antibiótico y la técnica de sedimentación algal, obteniendo cultivos unialgales y axénicos. El medio usado fue medio Johnson modificado en agua de mar, utilizado concentraciones de NaCl (1 – 5M), éste permitió evaluar el crecimiento, densidad y velocidad algal a través de conteo en cámara de Newbahuer y lecturas de absorvancia en espectrofotómetro. La relación entre los métodos de conteo, se realizó con el análisis de regresión potencial. Las mejores densidades algales se observaron en cultivo de concentraciones 1M, 1,5M y 3,5M de NaCl, para las cepas de las salinas de Chimus y Chilca. La mayor densidad algal ( 4,603 x 106 cels. ml-1 equivalente al 56.4 % para la cepa de Chimus) se observó en el cultivo de 1,5M de NaCl. La velocidad de crecimiento durante la fase exponencial para ambas cepas, estuvieron entre 0,56-0,83 div. día-1, con un Td de 29-43 horas, en cámara de Newbahuer como en espectrofotómetro y a las mismas concentraciones de cultivo.

El cultivo masivo de microalgas en biorreactores presenta una alta tasa de productividad frente a los cultivos abiertos, esta ventaja, aunada a determinados factores ambientales, se relaciona directamente con la cantidad y calidad de biomasa algal obtenida como producto final en el proceso de cultivo. El presente trabajo centró sus investigaciones en la evaluación de los parámetros físicos del ambiente (temperatura e intensidad lumínica), así como, físico-químicos (temperatura, pH y CO2) en la capacidad de carga de los cultivos. La investigación se desarrolló en el invernadero del Área de Biotecnología Acuática, del Instituto del Mar del Perú, dentro del marco del Proyecto IMARPE-EEP-FINCyT, denominado “Determinación de la Biomasa Microalgal potencialmente acumuladora de lípidos para la obtención de combustibles”, según contrato Nº025-FINCYT-PIBAP-2007. Los cultivos se realizaron empleando la cepa IMP-LBA-009, correspondiente a la microalga Nannochloropsis spp., con un flujo de producción semi-continuo por 48 horas, en un periodo de 6 meses, entre julio a diciembre, Bajo condiciones de iluminación (22,003.93 Lux.m-2.seg-1 ±10640.94), temperatura ambiental (31.7°C±2.8), temperatura de cultivo (25.7°C±1.2), CO2 (0.2 g.L-1) y pH (8.36 ±0.18); los resultados mostraron que la producción de biomasa húmeda fue 0.48g.L-1, equivalente a 0.13 g.L-1 de biomasa seca, durante los meses de octubre y noviembre, con una razón de conversión BS/BH del 28%. El impacto lumino-térmico, al interior del invernadero, favoreció el incremento progresivo de la concentración celular del cultivo en biorreactores, por ende, la producción de biomasa húmeda y seca, durante el periodo de trabajo.

Se presenta un breve diagnóstico de los aspectos ambientales, sociales y económicos del área marino costera de Pisco Paracas, cuya bahía forma parte de la zona de amortiguamiento de la Reserva Nacional de Paracas. A pesar de la importancia y fragilidad del ecosistema, y su proximidad en la zona de amortiguamiento de la Reserva, se ha permitido el asentamiento y desarrollo de diversas actividades extractivas, productivas con el consiguiente incremento en el tráfico marítimo. Diversos programas de vigilancia y seguimiento de entidades públicas o multisectoriales se han realizado en la zona, pero sin un enfoque más integral sobre las interacciones que se dan en un medio tan sui géneris como el medio marino Pisco-Paracas . Se realiza un análisis de la data histórica de los parámetros físico químicos de calidad acuática del período 2000-2010, obtenido principalmente de los programas de seguimiento o evaluación que realiza IMARPE y la empresa PLUS PETROL, contrastándolo con los valores ECA – Categoría 4 (DS 02-2008 – MINAM) a fin de determinar si ellos constituyen indicadores apropiados para determinar el estado real del ecosistema marino costero de Pisco – Paracas. La actual legislación de los recursos hídricos, no es muy clara ni precisa sobre la aplicabilidad para el control de los ECA‟ s en el ámbito marino; esta falta de precisión no contribuye a una vigilancia más eficiente. Con relación a ello, se alcanza una propuesta del sistema de indicadores que incluye parámetro biológico, como la determinación de las floraciones algales nocivas (FAN), episodios recurrentes en Pisco – Paracas; así mismo se introduce el tema de indicadores que midan el cambio en el ecosistema marino, con un enfoque más integral en la zona de Pisco – Paracas tan sujeta a cambios de origen climático

Durante el 2010, se capturó 55 ejemplares de tortuga verde Chelonia mydas en La Aguada (13°51’S y 76°15’W) al sureste de la bahía de Paracas; el número promedio de tortugas capturadas por kilómetro de red tendida fue 3,08±2,5; el tamaño promedio de la LCC fue 60,3±10,5cm; el 78% de los ejemplares presentaron el patrón 5c, 4d, 4i y 11d, 11i, para los escudos centrales, costales y marginales, respectivamente. La TSM donde se capturaron varió entre 15,2 y 20,9 °C, la mayor ocurrencia de tortugas se registró de 18,5 a 20 °C. Los epibiontes más representativos fueron Platylepas hexastylos (56,8%), Conchoderma virgatum (26,9%) y Chelonibia testudinaria (13,3%); la ocurrencia de los ítems alimenticios: Clorophyta (78%), Rhodophyta (30%), Cnidaria (43%), Crustacea (43%), Polichaeta (17%), Mollusca (17%), arena (26%) y plástico (17%); el 72% de las tortugas presentaron cobertura algal, de las cuales el 65% fue el alga verde Enteromorpha sp.

Parasites have to survive in their vertebrate host during a sufficiently prolonged period of time to achieve their life cycle through successful transmission via insect vectors. In their vertebrate hosts, parasites are often confronted by vigorous effector immune responses that they have to subvert somehow to be able to outlast and be successfully transmitted.

At this time, about 3,000 different viruses are recognized, but metagenomic studies suggest that these viruses are a small fraction of the viruses that exist in nature. We have explored viral diversity by deep sequencing nucleic acids obtained from virion populations enriched from raw sewage. We identified 234 known viruses, including 17 that infect humans. Plant, insect, and algal viruses as well as bacteriophages were also present. These viruses represented 26 taxonomic families and included viruses with single-stranded DNA (ssDNA), double-stranded DNA (dsDNA), positive-sense ssRNA [ssRNA(¿)], and dsRNA genomes. Novel viruses that could be placed in specific taxa represented 51 different families, making untreated wastewater the most diverse viral metagenome (genetic material recovered directly from environmental samples) examined thus far. However, the vast majority of sequence reads bore little or no sequence relation to known viruses and thus could not be placed into specific taxa. These results show that the vast majority of the viruses on Earth have not yet been characterized. Untreated wastewater provides a rich matrix for identifying novel viruses and for studying virus diversity.
