A fibrinogen-clotting enzyme, Jararacussin-I, was purified from the venom of Bothrops jararacussu by a combination of ion exchange chromatography using Resource 15S resin and affinity chromatography using Benzamidine Sepharose 6B resin. Jararacussin-I displays a molecular mass of 28 kDa as estimated by sodium dodecyl sulphate-PAGE and possesses an isoetectric point of 5.0. The coagulant specific activity of the enzyme was determined to be 45.8 NIH U/mg using bovine fibrinogen as the substrate and the esterase specific activity was determined to be 258.7 U/mg. The protease inhibitors, benzamidine and DTT inhibited the esterase specific activity by 72.4 and 69.7%, respectively. The optimal temperature and pH for the degradation of both chains of fibrinogen and esterase specific activity were determined to be 37 degreesC and 7.4-8.0, respectively. The enzyme was inactivated at both 4 and 75 T. Single crystals of Jararacussin-I were obtained and complete three-dimensional X-ray diffraction data was collected at the Brazilian National Synchrotron Source (LNLS) to a resolution of 2.4 Angstrom. Na região tritícola sul-brasileira predominam invernos com temperatura baixa (mínima absoluta, em dias com geada, de até - 8,0ºC). No entanto, a incidência de elevada temperatura (máxima absoluta, em dias isolados entre outubro e novembro, de até 41,0ºC) pode ser encontrada durante todo o período de enchimento de grãos e na maturação fisiológica. Este trabalho teve por objetivos verificar a influência das temperaturas mínima e máxima na qualidade industrial e no rendimento de grãos. Foram usados dados de experimentos com trigo Empresa Brasileira de Pesquisa Agropecuária (EMBRAPA) 16, conduzidos nos anos de 1990 a 1998, em sete locais do Rio Grande do Sul e em quatro locais de Santa Catarina. A análise estatística realizada foi correlações múltiplas. Verificou-se que, nos diferentes períodos analisados: a) o aumento da temperatura máxima média resultou em acréscimo do peso de mil grãos, do rendimento de grãos, da força geral de glúten, da microssedimentação com dodecil sulfato de sódio e do número de queda: b) o peso do hectolitro (exceção feita ao período final de maturação fisiológica), o peso de mil grãos, o número de queda e a extração experimental de farinha foram influenciados negativamente pela temperatura mínima média; c) a temperatura mínima média influenciou positivamente a força geral de glúten, a relação P/L e a microssedimentação com dodecil sulfato de sódio.

A pair of primers directed to 16S-23S rDNA interspacer (ITS) was designed directed to Brucella genetic sequences in order to develop a polymerase chain reaction (PCR) putatively capable of amplifying DNA from any Brucella species. Nucleic acid extracts from whole-blood from naive dogs were spiked with decreasing amounts of Brucella canis RM6/66 DNA and the resulting solutions were tested by PCR. In addition, the ability of PCR to amplify Brucella spp. genetic sequences from naturally infected dogs was evaluated using 210 whole-blood samples of dogs from 19 kennels. The whole-blood samples collected were subjected to blood culture and PCR. Serodiagnosis was performed using the rapid slide agglutination test with and without 2-mercaptoethanol. The DNA from whole blood was extracted using proteinase-K, sodium dodecyl sulphate and cetyl trimethyl ammonium bromide followed by phenol-chloroform purification. The PCR was capable of detecting as little as 3.8 fg of Brucella DNA mixed with 450 ng of host DNA. Theoretically, 3.8 fg of Brucella DNA represents the total genomic mass of fewer than two bacterial cells. The PCR diagnostic sensitivity and specificity were 100%. From the results observed in the present study, we conclude that PCR could be used as confirmatory test for diagnosis of B. canis infection.

Intracellular and extracellular catalases of different species of Candida were investigated using different culture media. All the Candida strains produced intracellular catalase, whose enzymatic activity was detected by non-denaturating polyacrylamide gradient (4-30%) gel electrophoresis. The cell extracts presented a major 230 kDa catalase band and in some strains variants of catalase with different molecular weights were detected. Candida catalase activity was not affected by heating at 50degreesC and incubation with beta-mercaptoethanol, but treatment with sodium dodecyl sulphate inhibited or reduced enzymatic activity. Extracellular enzyme activity was not detected in any of the culture filtrate extracts tested.

This work describes a novel approach for the analysis of selected aldehydes (formaldehyde, acetaldehyde, propionaldehyde, and acrolein) and acetone in environmental samples using micellar electrokinetic chromatography (MEKC). The method is based on the reaction of carbonyl compounds with 3-methyl-2-benzothiazoline hydrazone (MBTH) that gives an azine intermediate with maximum absorbance at 216 nm. A systematic evaluation of sample dissolution medium was conducted as a means to enhancing sensitivity. In the best condition, samples were dissolved in 0.030 mol.L-1 tetraborate solution. This condition presented enhancement factors in the range of 35-54 for the aldehydes under investigation, computed as the improvement of the concentration limits of detection (LODs) with reference to the sample dissolved in pure water. The running buffer was 0.020 mol.L-1 tetraborate, pH 9.3, containing 0.050 mol-L-1 sodium dodecyly sulfate (SIDS). The overall methodology presented several advantages over established methods for aldehydes. Worthy mentioning that MBTH is available in high purity degree, dispensing laborious reagent purification procedures. A few method validation parameters were determined revealing good migration time repeatability (< 2.5% coefficient of variation, CV) and area repeatability (< 4% CV), excellent linearity (20-120 mug/L, r > 0.995) and adequate sensitivity for environmental applications. The LODs with respect to each single aldehyde were in the range of 0.54-4.0 mug.L-1 and 11 mug.L-1 for acetone. The methodology was applied to the determination of aldehydes indoors. Samples were collected in an impinger flask containing 0.05% MBTH solution, at a flow rate of 0.80 L.min(-1), during 2.5 h, at different times during the day. The most abundant carbonyls in the samples were acetone, followed by formaldehyde and acetaldehyde, with estimate peak concentrations of 452, 5.2 and 2.2 ppbv, respectively.

Objective-To determine whether plasma protein concentrations were altered in ponies with alimentary laminitis.Animals-12 adult ponies.Procedure-Acute laminitis was induced in 6 ponies by oral administration of carbohydrate (85% corn starch, 15% wood flour); the other 6 ponies were used as controls. A physical examination was performed and blood samples were collected immediately before and 4, 8, 12, 24, and 28 hours after administration of carbohydrate. Plasma protein concentrations were determined by means of sodium dodecyl sulphate-polyacrylamide gel electrophoresis.Results-19 plasma proteins ranging from a molecular weight of 24,000 to a molecular weight of 350,000 were identified in all 12 ponies. Plasma concentrations of proteins with molecular weights of 350,000 (fibrinogen), 130,000 (ceruloplasmin), 118,000 (c-reactive protein), 67,000 (alpha(1)-antitrypsin I), 65,000 (alpha(1)-antitrypsin II), 50,000 (haptoglobulin), and 45,000 (acid glycoprotein) were significantly increased in ponies with laminitis, compared with concentrations in control ponies.Conclusion-Changes in plasma protein concentrations are detectable within 4 hours after the onset of alimentary laminitis in ponies.Clinical Relevance-Measurement of plasma protein concentrations may be useful in monitoring the progression of laminitis in ponies.

Considered as one of the main agents of the tripanossomiases, Trypanosoma evansi causes a disease generically know as surra, with wide geographic occurence. This work has the aim to study the electrophoretic profile of the acute phase proteins of goats, experimentally infected with T. evansi. Ten crossbread female goats, around 4 months of age, clinically healthy and serum negative for the presence of antibodies anti-T. evansi (IFAT) were used. The animals were divided in two groups: six were inoculated (G1) intravenously with 2,38 × 10 6 tripomastigotes of T. evansi and four were kept as noninfected controls. The blood for serum was collected daily until the 14 days after inoculation (DAI), weekly up to the 98 th DAI and every two weeks up to the 364 th DAI. The serum proteins were separacted by sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDSPAGE). Twenty-one proteins were found in the serum of the goats, eight were nominally identified; phosphorylase, transferrin, albumin, antitrypsin, acid glicoprotein, haptoglobin, hemoglobin, and light chain immunoglobulin.

The microbiological control of moisturizing mask formulation added of hibiscus flowers, assai palm, black mulberry and papaw glycolic extracts, determining the number of viable microorganisms and possible presence of pathogenic. The moisturizing mask formulation was composed of zinc oxide (5. 0%) and moisturizing cream constituted of triceteareth-4 phosphate (and) cetyl alcohol (and) stearyl alcohol (and) sodium cetearyl sulfate (and) oleth-10 (qs 50g). To this formulation was added hibiscus flowers glycolic extract (2. 5%), assai palm glycolic extract (1. 5%), black mulberry glycolic extract (1. 5%) and papaw glycolic extract (2. 0%). The formulation was stored in aseptically clean recipients, away from humidity and light, in fresh and airy places. The results of the microbiological analysis on the counting of aerobic mesophilic microorganisms (bacteria and fungi), of the above mentioned formulation, revealed a bioburden < 10 CFU/mL in all samples. Such data indicate adequate microbiological quality of the tested products, according to official recommendations. Furthermore, it was not detected the presence of pathogenic microorganisms, assuring the harmlessness of the formulation. The results lead us to conclude that the formulation and raw materials analyzed did not present microbial contamination, evidenced for estimating the number of viable microorganisms (<10 UFC/g) and for researching pathogens.

Evidências experimentais sugerem que o alumínio é um agente neurotóxico com ações deletérias sobre os processos cognitivos. Neste estudo, investigou-se os efeitos comportamentais, histopatológicos e bioquímicos da intoxicação crônica com citrato de alumínio sobre o hipocampo de ratos adultos, e ao mesmo tempo aplicou-se uma terapia experimental de tratamento com magnésio para a reversão das alterações neuropatológicas encontradas. Utilizou-se 70 ratos Wistar machos de 200-250 g, divididos em grupos da seguinte maneira: controle, citrato de sódio (CNa), citrato de alumínio (CAl), citrato de alumínio + sulfato de magnésio (CAl+Mg), citrato de sódio + sulfato de magnésio (CNa+Mg). A dose usada de citrato de alumínio foi de 100 mg/kg e sulfato de magnésio 250 mg/kg durante 30 dias. Os animais foram submetidos aos testes comportamentais do campo aberto, Rota rod, Reconhecimento social e Labirinto em T elevado (LTE), análise bioquímica, histopatológica e imunoistoquímica para GFAP. Além disso, foram verificados os níveis de alumínio no plasma e no hipocampo dos animais em espectrômetro de absorção atômica em forno de grafite (GF AAS). Os resultados obtidos mostraram que o grupo CAl apresentou um aumento da atividade locomotora no teste do campo aberto em comparação ao grupo controle e já o grupo CAl+Mg apresentou uma diminuição (P<0.001). Nos testes de memória do LTE e de Reconhecimento social, os animais do grupo CAl apresentaram déficits no cognitivos em relação aos demais grupos enquanto os animais do grupo CAl+Mg apresentaram um bom desempenho nos teste (P<0.001). Os níveis de alumínio encontrados no hipocampo do grupo CAl foram consideravelmente elevados e nos demais grupos os níveis ficaram abaixo do limite de detecção. Na análise histopatológica e imunoistoquímica, os animais do grupo CAl apresentaram diminuição da densidade celular e reatividade astrocitária nas camadas CA1, CA3 e hilo do hipocampo. Estes resultados sugerem que a intoxicação experimental com citrato de alumínio induz déficits de aprendizado e memória e que a administração de sulfato de magnésio pode ter a capacidade de minimizar os danos causados pelo metal no hipocampo de animais intoxicados.

No nordeste do Estado do Pará predomina a agricultura familiar, a qual tem como principais produtos a mandioca e a malva. No processamento desses produtos, os agricultores costumam submergir fardos de mandioca e de malva em águas de igarapés, com a finalidade de amolecer a casca e remover as substâncias tóxicas, no caso da mandioca, ou proporcionar o desfibramento, no caso da malva. Os efeitos dessa prática sobre a qualidade da água fluvial são pouco conhecidos. Nesse contexto, a presente pesquisa objetivou avaliar os possíveis impactos dessas práticas sobre a hidrobiogeoquímica fluvial em igarapés dessa região. Adotou-se como estratégia coletar amostras de águas fluviais a montante do ponto de lavagem dos produtos, no local de lavagem e a jusante deste local. A fim de conhecer, com maior clareza, as possíveis alterações na química das águas, provocadas pelas lavagens de mandioca e de malva, somou-se à estratégia de campo a realização de experimentos em tanque com vazões de entrada e saída controladas. Em ambas abordagens foram monitoradas as seguintes variáveis hidrobiogeoquímicas no material dissolvido: pH, condutividade elétrica, oxigênio dissolvido, cálcio, magnésio, potássio, sódio, cloreto, sulfato, fosfato, nitrato, amônio, além de nitrogênio total e carbono orgânico e inorgânico dissolvidos. Os resultados obtidos evidenciaram que o processo de lavagem de raízes de mandioca e de plantas de malva contribuiu para alterar a hidrobiogeoquímica fluvial de pequenos igarapés apenas pontualmente. Porém, algumas das alterações observadas localmente permaneceram por até dez metros a jusante do local de lavagem dos produtos. Nos experimentos em tanque, as alterações foram mais evidentes e uma análise de cluster confirmou a hipótese de que o processamento dos produtos agrícolas enfocados colaborou para alterar a hidrobiogeoquímica fluvial nos igarapés monitorados. O estudo recomenda algumas precauções no tocante às práticas de processamento dos produtos em foco e também a adoção de alguns parâmetros para o monitoramento desses impactos.

Interfacial concentrations of chloride and bromide ions, with Li+, Na+, K+, Rb+, Cs+, trimethylammonium (TMA(+)), Ca2+, and Mg2+ as counterions, were determined by chemical trapping in micelles formed by two zwitterionic surfactants, namely N-hexadecyl-N,N-dimethyl-3-ammonio-1-propanesulfonate (HPS) and hexadecylphosphorylcholine (HDPC) micelles. Appropriate standard curves for the chemical trapping method were obtained by measuring the product yields of chloride and bromide salts with 2,4,6-trimethyl-benzenediazonium (BF4) in the presence of low molecular analogs (N,N,N-trimethyl-propane sulfonate and methyl-phosphorylcholine) of the employed surfactants. The experimentally determined values for the local Br- (Cl-) concentrations were modeled by fully integrated non-linear Poisson Boltzmann equations. The best fits to all experimental data were obtained by considering that ions at the interface are not fixed at an adsorption site but are free to move in the interfacial plane. In addition, the calculation of ion distribution allowed the estimation of the degree of ion coverage by using standard chemical potential differences accounting for ion specificity.
