This experimental study aimed the evaluation of the use of anthropomorphic phantoms in the analysis of the influence of the acquisition parameters in the contrast of radiographic images. The analyzed factors were the variation of the peak kilovoltage (kVp) and the product of the filament circuit by the time of exposition (mAs). The influence of these factors was verified for different anthropomorphic phantoms: foot, knee and chest. The image contrast behavior with the simulators was compared to values obtained with the use of an aluminum ladder being the behavior of this reference for analysis and discussion. To assure the reproducibility of images, quality control tests were made and evaluation of procedure conditions before the experiments. The results obtained are shown in a scale of images where it was possible to evaluate the impact in the darkness of images. Regions with different compositions were analyzed which were different in image, this way the values of optical density and contrast are represented as charts and graphics. We conclude that the use of anthropomorphic phantoms in the evaluation of the influence of tension of the tube and time of exposition in the contrast of the radiographic images is not indicated for a quantitative analysis through optical density, once they present incompatible results with the data as reference as the aluminum ladder even so these simulators present a great property in qualitative analysis regarding the differentiation of structures and subjective evaluations

Durante a infecção experimental por Trypanosoma cruzi, a resposta imune adaptativa mediada por células CD8+ é direcionada a poucos epítopos imunodominantes e subdominantes. Em estudos anteriores, foi demonstrado que, em diferentes linhagens de camundongos, a imunização com plasmídeos ou proteínas recombinantes, que expressam epítopos CD8 imunodominantes, antecipa a resposta imune após o desafio e proporciona resposta imune protetora contra infecções letais com T.cruzi. Em nosso estudo atual, desenvolvemos a hipótese que também é possível gerar resposta imune protetora a partir de imunizações com plasmídeos que expressam epítopos subdominantes. Neste caso, esta resposta poderia se somar a resposta imune dominante ampliando o espectro da resposta imuno-protetora. Para testar nossa hipótese, nós tivemos que construir diferentes plasmídeos contendo a ORF do gene da proteína 2 de superfície de amastigota (asp-2) de T.cruzi contendo mutações que: i) retiraram os epítopos imunodominantes CD8 (VNHRFTLV para H-2Kb ou TEWETGQI para H-2Kk) ;ii) trocaram o epítopo imunodominante por epítopos sub-dominantes (TsKb-18 ANYDFTLV ou TsKb-20 ANYKFTLV). Além disso, também foi construído um plasmídeo contendo somente a porção P4-P7 (261° a 500° aminoácidos) da asp-2. Após a construção destes plasmídeos, testamos a imunogencidade de dois deles, denominados pRE-P4P7 e o pIgSP-VNHRATLA, foram testados individualmente na vacinação de camundongos F1(C57BL/6 x B10.A) (H-2Kb e H- 2Kk) ou de CB10 (H-2Kb), respectivamente, e desafiados com tripomastigotas da cepa Y de T.cruzi. A vacinação de camundongos F1 (C57BL/6 x B10.A) com o plasmídeo pré- P4-P7 aumentou significativamente a resposta imune de células CD8+ específicas ao epítopo sub-dominante TEWETGQI, restrito ao MHC H-2kk, porém não diminuiu a resposta ...(Resumo completo, clicar acesso eletrônico abaixo)

The behavior and experience of pain are characterized by alertness and anxiety, which disappear soon after the healing process begins. Based on this area, the present study aimed to quantify the expression of c-fos in segments of the brain of rats after surgical stimulation in one rear hind paw (analgesia), using anesthesia with sodium thiopental and practices of acupuncture pre and post operative. The animals were stimulated with intraperitoneal injections of solution (NaCl) 0.2% (2ml), and divided into three groups, control, preoperative and postoperative. Each treatment was divided in manual acupuncture (AM) and electroacupuncture (EA). The animals were randomly distributed in each group. The c-fos expression was quantitated using immunohistochemistry for all situations. The results showed a great efficiency of all treatment compared to the control group (p <0.001), thus reenforcing data in the literature on the potential of acupuncture analgesia. Bacillus thuringiensis is an environmental bacteria that produces a group of crystallizable proteins (Cry) that are toxic for several insects and worms species. Recently, it was described a novel class of Cry proteins called parasporins (PS) that showed cytotoxic effects on animal and human tumor cells. Six types of PS have been described so far, PS1 to PS6, and their cytotoxic activity has been studied. However, the direct effect on tumor cells has been the current research focus, while the immunomodulatory role of the PS has not been studied yet. Therefore, this study aimed to verify whether PS of TC 2.3.1R6 B. thuringiensis strain has immunostimulatory activity on human lymphocytes and monocytes. We have evaluated the protein toxicity against human cells, the lymphoproliferative activity and the effects on peripheral blood monocytes. The PS-PK showed no toxic or stimulating activity on lymphocyte proliferation. However, it inhibited the spontaneous production of IL-10 as well as ConA-induced and the production of IFN-γ. PS-PK decreased the release of hydrogen peroxide and increased the production of TNF- α by monocytes. PS-PK performed inhibitory production of hydrogen peroxide and TNF-α by monocytes, whereas PS-Tp showed stimulation of the production of hydrogen and TNF-α

Diuron (3-(3,4-dichlorophenyl)-1,1-dimethylurea) is a substituted urea herbicide widely used in crops of sugar cane, cotton and soybeans. In 1997, this agent has been classified by the United States Environmental Protection Agency as known/likely human carcinogen because it induced tumors in the urinary bladder and renal pelvis of rats, and breast and skin of mice exposed to 2500 ppm for feed for two years. A previous study from our group demonstrated dose-response relationship in the gene expression profile associated with severe necrosis on bladder urothelium and increased incidence of simple hyperplasia in male Wistar rats treated with different concentrations of diuron for 20 weeks. To check how early the molecular changes occurs, rats were fed for 7 days with diets containing diuron at 0, 125, 500 or 2500 ppm. The main observations recorded were urothelium ultrastructural alterations and disruptions of molecular pathways associated with cell-cell interaction and the tissue organization maintenance. Particularly, the gene Glypican 3 (Gpc3), a surface proteoglycan related to cellular adhesion and apoptosis induction, was down regulated on urothelium exposed to 2500ppm diuron for 7 days and 20 weeks. The aim of this study was validate by quantitative RT-PCR real time, the reduced Gpc3 gene expression in epithelial cells of the urinary bladder of male Wistar rats treated with different concentrations of diuron for 7 days and 20 weeks. The endogenous control of the quantitative PCR real time technique was the β-actin gene and the target was the gene Gpc3. The relative quantification (RQ) was obtained by the method of relative quantification 2-ΔΔCt . Animals exposed to diuron for 7 days or for 20 weeks presented reduction of Gpc3 gene expression compared to the control group. This reduction was statistically significant only for the 7 days study. Tanto o BCG (Bacilo de Calmette-Guérin) quanto a DNAhsp65 são vacinas contra tuberculose, porém além da atividade protetora desejável ambas apresentam atividade imunomoduladora. Seguindo a linha de pesquisa de nosso laboratório que investiga a atividade da DNAhsp65 sobre doenças autoimunes, avaliamos o efeito da associação destas vacinas sob a forma de uma estratégia vacinal tipo prime-boost (priming com BCG seguida de booster com DNAhsp65), sobre o desenvolvimento da Encefalite Autoimune Experimental (EAE), que é modelo animal de esclerose múltipla. Ratos Lewis fêmeas foram divididos em grupos de 5 a 6 animais (salina, BCG, EAE, BCG/EAE, BCG/vetor/EAE e BCG/vacina/EAE) e imunizados em intervalos de 15 dias. Os animais receberam um primming com 2 - 10 x 105 unidades formadoras de colônias de BCG subcutâneo seguido de dois boosters com 300 g de pVAXhsp65 numa solução de sacarose 25% por via intramuscular. A EAE foi induzida 15 dias após a última imunização, por inoculação de 25 g de proteína básica de mielina (MBP) associada ao Adjuvante Completo de Freund contendo Mycobacterium butyricum. Aqueles que não receberam os inóculos descritos foram injetados com salina, exceto o grupo BCG/vetor/EAE que recebeu 300 g de pVAX (vetor plasmidial vazio) também em solução de sacarose. Os animais foram acompanhados diariamente até serem submetidos à eutanásia, 3 semanas mais tarde, durante a fase de recuperação da doença para avaliação da resposta imune, escore clínico e presença de infiltrado inflamatório no SNC. A evolução clínica da EAE foi similar entre animais vacinados e não vacinados, mas a análise histopatológica revelou que a imunização prévia com BCG ou BCG/DNAhsp65 diminui o processo inflamatório. Aquaporins (AQPs) are membrane protein channels expressed in a wide variety of cells and tissues that are involved in the transport of water. In the male excurrent duct, the movement of fluid and solutes across the epithelium is essential for establishing the proper luminal environment in which sperm mature and are stored. The initial aim of the present propose is to localize AQP 2 e 9 in epithelium of the initial segment, caput, corpus and cauda epididymidis in Mongolian gerbil during postnatal development (10, 30, 60, 120 and 300 days), observing possible changes in the pattern of expression of the AQPs along the postnatal development. The animals will be killed by inhalation of carbon dioxide. After euthanasia, the epididymis will be removed and the different regions will be isolated. The biological material obtained will be processed in accordance with the routines of histological techniques and imunnohistochemistry to study the expression and localization of the AQPs 2 and 9. The results obtained will be analyzed and photographed in conventional light microscope

Mesenchymal stem cells (MSCs) are adult multipotent cells with fibroblastoid morphology and adherent to plastic. Furthermore, they can be obtained from different sources. Besides bone marrow, these cells are taken from umbilical cord blood, umbilical vein, saphenous vein, peripheral blood, arteries, liver and fetal pancreas, placenta, dental pulp and adipose tissue. MSCs derived from adipose tissue are important because of the abundant number of cells that can be obtained from this tissue, easy access and little discomfort to the patient. This study compared two techniques for obtaining MSCs from adipose tissue: mechanical dissociation (MD) and enzymatic digestion (ED). We also analyzed the inter-species cross-reactions using commercial monoclonal antibodies directed against surface antigens of stem cells from different species: mouse, horse, rabbit, monkey and human. We found that MD technique is favorable in relation to ED within 15 days of culture, and ED is more efficient in the first days of culture. The data also showed that MD causes less damage to cellular DNA. About inter-species cross-reactions, the monoclonal antibody A69 directed against stem cells from rabbits, which can be used in veterinary medicine, particularly in research involving horses

This work will discuss how magnetic fields can be produced, either generated by magnets, natural, artificial, or even by an electric current going through a wire, as discovered by Oersted. Besides the theoretical content, experimental studies on magnetic induction and on the Laws of Faraday and Lenz will be performed. In the Magnetic Induction experiment, the electromotive force generated by varying the flow of the field B in a solenoid, depending on the variation of the current intensity and frequency associated with it will be measured; the experiment on the Laws of Faraday and Lenz the electromotive force produced by the relative movement of the magnet in relation to a coil. Os mecanismos pelos quais o hiper e hipotireoidismo afetam o osso durante a menopausa são pouco conhecidos. O objetivo deste estudo foi verificar a ação do hormônio triiodotironina (T3), em diferentes concentrações associado ao estrógeno (E2) em concentração infra-fisiológica (condição de menopausa), sobre a proliferação de células de rato ROS 17/2.8, através de uma curva de crescimento para obtenção do tempo de dobramento (TD). As células ROS 17/2.8 são consideradas modelo de células osteoblásticas e foram cultivadas em meio HAM F-12, e submetidas ao tratamento hormonal com as combinações das concentrações fisiológica, infrafisiológica e suprafisiológica de T3 e infra-fisiológica de E2. Utilizamos como grupo controle aquele tratado com as concentrações fisiológicas dos dois hormônios. Para cada concentração hormonal foram realizadas 5 repetições, e as células contadas manualmente nos dias 1, 3, 5 e 7, após o início da adição hormonal. A análise estatística utilizada foi a técnica da análise de variância não paramétrica, complementada com teste de comparações múltiplas. Baseando-se nos resultados obtidos, pode-se concluir que o crescimento das células ROS 17/2.8 é afetado pelos tratamentos hormonais, em especial quando a concentração infrafisiológica de E2 está associada à concentração suprafisiológica de T3, aumentando o tempo de dobramento celular. Este efeito poderia tornar o metabolismo do osso mais lento, prejudicando a neoformação óssea em pacientes menopausadas e hipertireoidéas

Adequate testosterone levels are necessary for the development, growth and maintenance of the male reproductive system. Testosterone deficiency is common in men with diabetes in whom it may contribute to impaired performance, with consequent reduction of the activity of the androgen regulated organs, such as the prostate. However, little attention has been given to the plasma dihydrotestosterone (DHT) level, the most potent androgen, nor to the expression of the androgen receptor (AR), insulin-like growth factor type I (IGF-1) and receptor (IGF-1R) in target tissues. Here, we investigated the effect of type I diabetes mellitus on DHT plasma levels and on prostate AR, IGF-1 and IGF-1R expression during rat pubertal growth. Diabetes was induced in prepubertal male rats through administration of streptozotocin (STZ; 40 mg/kg). Diabetic, diabetic treated with insulin, and age-matched control animals were killed by overdoses of pentobarbital. The ventral prostatic lobe (VP) was dissected, weighed and processed for immunohistochemistry for AR, IGF-1 e IGF-1R; plasma T and DHT levels were also determined. Hyperglycemia at puberty reduced VP weight gain to about 50% and plasma T level to about 80% of the control levels. In contrast there were no changes in plasma DHT levels. Insulin replacement restored the VP weight gain, but not the plasma T levels, which remained 90% below the ones of controls. Immunohistochemistry showed that AR, IGF-1 and IGF-1R expression in the prostate epithelial cells did not change with hyperglycemia or insulin replacement. Thus, the AR expression in the prostate epithelial cells appears to be regulated by DHT, and to a minor extent it also controls glandular growth

Os distúrbios do sistema nervoso central mais comuns relatados no mundo são a ansiedade e a depressão, com grande incidência de casos nos quais aparecem juntos. Os tratamentos utilizados na clínica além de não serem efetivos na totalidade dos casos podem, por vezes, apresentar diversos efeitos não desejados. Assim, na busca novas drogas mais efetivas e com menor incidência de efeitos adversos, a Cananga odorata (Lam.) Hook. f. & Thomson aparece como uma alternativa. Esta espécie vegetal é utilizada na aromaterapia como na prevenção do estresse, ansiedade e depressão (BIRD, 2003). A busca de novas ferramentas terapêuticas depende do uso de animais de experimentação, e com as discussões na sociedade científica sobre o uso de animais, há uma maior preocupação em se desenvolver novas técnicas experimentais onde se possa aplicar os conceitos dos 3Rs objetivando a redução (Replacement) do número de animais, sua substituição (Reduction) e o refinamento (Refinement) das técnicas experimentais. Neste trabalho avaliou-se a atividade ansiolítica e antidepressiva do óleo essencial (OE) de Cananga odorata, em animais que foram sucessivamente submetidos a testes para avaliar a atividade motora, ansiolítica e antidepressiva. Camundongos machos foram tratados por via oral com OE nas doses de 0,1; 1, 10 e 100 mg/kg, e após 30 minutos foram submetidos ao teste do Campo Aberto, da Caixa Claro-Escuro e Suspensão pela Cauda, nesta ordem, com um intervalo de 30 minutos entre eles. Os resultados indicaram que a seqüência de testes é adequada para a avaliação destas atividades com economia de tempo, animais e drogas

The use of pesticides in Brazil has intensified over the years. And since 2009 it was ranked as the largest market for pesticides worldwide. The consequential diffuse contamination of the environment is therefore due to the increasing number of chemicals of different classes, origins and modes of action. Little is known about the action of pesticides on human health in situations of co-exposure. Possible toxic effects are not restricted to agricultural and industrial workers, but also the general population that may be exposed continuously to its residues in food and water. Although these pesticides are mostly present in the environment at low doses, it must be considered that possible cumulative or synergistic effects may occur when there are concurrent or sustained exposure for two or more of these agents, which can lead to late manifestation of subclinical damages, sometimes irreversible. Thus, the specific objective of this study was to assess the effect of carcinogenesis promotion of a mixture of pesticides at low doses and analyze the phenomena of cell proliferation and apoptosis in rat liver. A total of 50 male Lewis rats was separated into 5 groups for 8 weeks in a medium term hepatocarcinogenesis model. The three different classes of pesticides (dieldrin, dicofol, endosulfan, dichlorvos and permethrin), whose residues were detected by ANVISA during the period from 2001 to 2005 in tomatoes cultures, were added to the feed of rats initiated to hepatocarcinogenesis with diethylnitrosamine (DEN- 200mg/kg ip). We used two different mixtures, one with no toxic effects at doses (MEX1) referring to the NOEL (no-observed-effect level) and another at doses LOEL / LEL / LOAEL (Lowest-observed-effect level / Lowest-effect level / Lowest -observed-adverse-effect level), to the installation of adverse effects (MEX2), derived from chronic studies. Experimental models are necessary to elucidate pathophysiological mechanisms not yet understood in humans. To evaluate the repercussions of the diabetes, considering two methodologies, on the pregnancy of Wistar rats and on the development of their offspring. In the 1st induction, female offspring were distributed into two experimental groups: Group streptozotocin (STZ, n=67): received the β-cytotoxic agent (100mg STZ/kg body weight - sc) on the 1st day of the life; and Non-diabetic Group (ND, n=14): received the vehicle in a similar time period. In the adult life, the animals were mated. After a positive diagnosis of pregnancy (0), female rats from group STZ presenting with lower glycemia than 120 mg/dL received more 20 mg STZ/kg (ip) at day 7 of pregnancy (2nd induction). The female rats with glycemia higher than 120mg/dL were discarded because they reproduced results already found in the literature. In the mornings of days 0, 7, 14 and 21 of the pregnancy glycemia was determined. At day 21 of pregnancy (at term), the female rats were anesthetized and killed for maternal reproductive performance and fetal development analysis. The data were analyzed using Student-Newman-Keuls, Chi-square and Zero-inflated Poisson (ZIP) Tests (p<0.05). STZ rats presented with increased rates of pre (STZ=22.0%; ND=5.1%) and post-implantation losses (STZ=26.1%; ND=5.7%), reduced rates of fetuses with appropriate weight for gestational age (STZ=66%; ND=93%) and reduced degree of development (ossification sites). Conclusion: Mild diabetes led a negative impact on maternal reproductive performance and caused intrauterine growth restriction and impaired fetal development
