Hybrid stylos (Stylosanthes guianensis var. vulgaris x var. pauciflora) with durable, quantitative resistance to anthracnose, mid-season harvest maturity date (early-July), high DM and seed yields have been selected at the Embrapa Beef Cattle Research Center, Campo Grande, Brazil. The hybrids displayed improved forage traits in Brazil, the native habitat and major center of diversity of the species and its pathogen, as well as in vastly different ecosystems. Dry forage yields and anthracnose resistance of superior selections and their composites were equal, in some instances, significantly better, than those of cv. Mineir (a) over tildeo in multilocational trials situated in the Cerrados from lat. 6degrees S to lat. 20degrees S. Selected hybrids performed well in comparison with the highly successful CIAT 184 (cv.Reyan II) on Hainan Island, China. Composites have also shown good promise in seed multiplication plots in Queensland, Australia. A positive attribute of composite hybrids is their great genetic diversity in contrast to pure-line cultivars with a relatively narrow genetic base. These truly tropical forms of stylo are best adapted to regions with >1500mm average annual rainfall.

The objective of this study was to evaluate the occurrence of bovine mastitis by Staphylococcus sp., Streptococcus sp. and Candida sp. in a rural area of Indianopolis, Minas Gerais. It was realized the California Mastitis Test (CMT) in six collect, a total 671 of milk sample positive. Then the microbiological examination was performed, where the results revealed the presence of 137 milk samples with microbial multiplication. These, showed the presence of Staphylococcus aureus (45.2% of strains), other coagulase negative Staphylococcus (10.2%), Staphylococcus epidermidis (9.4%), Staphylococcus simulans (5.8%), other coagulase negative (15.3%), Streptococcus agalactiae (7.2%), other Streptococcus sp. (5.1%) and yeasts (1.4%). It was found that 100% of Staphylococcus were susceptible to rifampicin, gentamicin and ciprofloxacin; but, resistant to penicillin, tetracycline, and oxacillin. Regarding antimicrobial susceptibility Streptococcus, were employed, except to clindamycin, erythromycin and tetracycline. We conclude that there is a great necessity of proper hygiene practices and taking prophylactic measures taken in order to reduce the infection of animals caused by infectious microorganisms and resistance.

Calla lily (Zantedeschia aethiopica) is appreciated as cut flower and for the composition of gardens. However, many pathogens affect this species. By the traditional method of propagation, some units of new seedlings can only be produced annually. Tissue culture allows fast large-scale clonal propagation and provides healthy uniform plants. During the in vitro process, type and concentration of growth regulator could affect the growth of seedlings. Thus, the aim of this work was to determine sucrose and GA(3) concentrations to increase the efficiency of the in vitro multiplication of calla lily. After 60 days, the length of the above ground part and the roots, the number of sprouts, roots and leaves, above ground part and root fresh weight of seedlings were evaluated. The experimental design was entirely randomized with four replications. It was necessary the addition of 60.5 g L-1 sucrose associated to 5 mg L-1 GA(3) to obtain hight sprouts number. For higher length of the above ground part the addition of 45.3 g L-1 sucrose and 10 mg L-1 GA(3) was enough. Better results in the root length and number of roots were observed only in the sucrose presence, in concentrations in the range of 51.13 - 56.5 g L-1.

Nematodes cause extensive losses to sugarcane in Brazil and also in other producing regions. Meloidogyne incognita, M. javanica and Pratylenchus zeae are the key species for this culture worldwide. In the present study, the aggressiveness of M. javanica and M. incognita to sugarcane variety SP 911049 was evaluated comparatively,. The following parameters were compared: reproduction factor (RF) of these nematodes, effect of nematodes in the natural incidence of pests, and the influence on the development and technological characteristics of sugarcane. Considering the data of RF, biometrics, natural infestation of pests, mortality of plants, and technological variables, it was concluded that M. javanica was more aggressive to sugarcane, although its rate of multiplication was much smaller than the one of M. incognita.

Banana (Musa spp.) is one of the most consumed fruits in the world, and it is cultivated in many tropical countries. In Brazil, the productivity is low due to several factors such as the incidence of pests and diseases. The nematodes are among the most important problems found in the production of bananas. This research studied the reproduction of Meloidogyne incognita race 2, M. javanica and Pratylenchus coffeae in different cultivars of banana. The evaluation of the experiments with Meloidogyne spp. was carried out 120 days after the inoculation. The analyzed parameters were: number of gall, number of the external masses of eggs, number of eggs per gram of root, reproduction factor and number of juveniles in the soil. The evaluation of the experiment with P. coffeae was carried out 90 days after the inoculation; the final population was evaluated. The cultivars PV-0344, Maca, Grande Naine, FHIA 01, Thap Maeo and Prata Ana allowed greater multiplication of P. coffeae than Caipira, SH-3640 and FHIA-18. For M. javanica, the cultivars Calypso, Bucanneer, Grande Naine, PV-0344, Nanicao Magario, FHIA-02, SH-3640, Pacovan, and Prata Ana exhibited larger populations, decreasing in Maca. All cultivars allowed high multiplication rates of M. incognita.

The intra-generic inhibition of bacterial growth observed previously in vivo and in vitro with strains of Salmonella, Citrobacter and E. coli was studied in vitro using S. typhimurium strain F98. There was complete inhibition of multiplication of S. typhimurium when it was added to stationary-phase broth cultures of different Salmonella serotypes, but only partial inhibition when added to broth cultures of E. coli. The degree of inhibition between different mutants of F98 was affected by the numbers of bacteria of the inhibiting strain, but this was not the only factor, since exponential-phase bacterial cells were less inhibitory than stationary-phase cells. The inhibitory effect was produced at temperatures between 20°C and 40°C. The complete inhibition of growth observed between F98 mutants was abolished by ampicillin, rifampicin and streptomycin, but not by nalidixic acid. Inhibition was also prevented by separating the two cultures by a dialysis membrane. A Tnpho A Insertion mutant of F98 was produced which did not show inhibition in vitro but was still inhibitory in vivo. It is suggested that this complete inhibition of bacterial multiplication between organisms of the same genus, which is greater than that produced between organisms from different genera, is mediated by a cell surface protein.

Actions to overcome a disease are dependant, essentially, on what is known about it. Procedures followed in the past were sometimes bizarre, but justified because of how little was known about the disease. The tuberculosis prechemotherapeutic age was somber due to the high levels of fatalities and morbidity. With the arrival of the chemotherapeutic treatment its prognosis has changed. Tuberculosis declined in the 50's and stabilized in the 80's. Nevertheless, it is back increasing alarming its numbers more than ever; probably because of some factors, among them, the public health system lack of attention and the government's policies, increasing in the migration to and from the endemic areas, development of drug multi-resistant cepa and also to the HIV infection. An universal antimycobacteria chemotherapy treatment is not accepted, maybe because of the number of drugs that are available. Modern chemotherapy, however, has an attack and a maintenance phases with the aim to eliminate the bacillus of fast and slow multiplication, respectively. The treatment period is long, when compared with other infectious diseases, that leads to the lack of compliance. In spite of the available resources in the fight against tuberculosis they seem insufficient to restrain the disease. This has forced the search for new chemotherapy alternatives to avoid strong come back of tuberculosis to the point of being called the 'white plague' well into the 21'st century.

The mechanisms used by Paracoccidioides brasiliensis (Pb 18) to survive into monocytes are not clear. Cellular iron metabolism is of critical importance to the growth of several intracellular pathogens, including P. brasiliensis, whose capacity to multiply in mononuclear phagocytes is dependent on the availability of intracellular iron. Chloroquine, by virtue of its basic properties, has been shown to prevent release of iron from holotransferrin by raising endocytic and lysosomal pH, and thereby interfering with normal iron metabolism. Then, in view of this, we have studied the effects of CHLOR on P. brasiliensis multiplication in human monocytes and its effect on the murine paracoccidioidomycosis. CHLOR induced human monocytes to kill P. brasiliensis. The effect of CHLOR was reversed by FeNTA, an iron compound that is soluble at neutral to alkaline pH, but not by holotransferrin, which releases iron only in an acidic environment. CHLOR treatment of Pb 18-infected BALB/c mice significantly reduced the viable fungi recovery from lungs, during three different periods of evaluation, in a dose-dependent manner. This study demonstrates that iron is of critical importance to the survival of P. brasiliensis yeasts within human monocytes and the CHLOR treatment in vitro induces Pb 18 yeast-killing by monocytes by restricting the availability of intracellular iron. Besides, the CHLOR treatment in vivo significantly reduces the number of organisms in the lungs of Pb-infected mice protecting them from several infections. Thus, CHLOR was effective in the treatment of murine paracoccidioidomycosis, suggesting the potential use of this drug in patients' treatment.

The objective of this study is to use different in vitro culture systems of preantral follicles from Nelore breed bovine fetuses in the last gestation quarter. The evaluation of treatments considered the time of growth of isolated follicles. Preantral follicles were mechanically isolated and submitted to the individual culture, for 9 days, in media no supplemented or supplemented with fetal calf serum (FCS), bovine serum albumin (BSA) or synthetic defined supplement substitute of serum KnockoutSR (KNO). We have also evaluated the effects of collagen gel or fetal calf fibroblast monolayer as substratum for in vitro cultures. The increase on the follicular diameter was followed in the first day (0 h), at the 72 h, 144 h and 216 h. Considering cultures of isolated follicles, the results have shown that the association between media supplemented with FCS and collagen gel was significantly more efficient on the increase of the follicular diameter than other treatments. It is not still established a system of appropriate cultivation that sustains the differentiation and multiplication of the granular cells and that maintains the contact of the same ones with the oocyte to provide molecules and factors that supply the metabolic demand. We also understand that our results also represent another promising step on the search for the ultimate system of in vitro culture of preantral follicles from bovines.

Intestinal pathogens are exposed to various stress conditions during their infectious cycle. Anaerobiosis, one of such hostile condition, is offered by the host within gut and intestinal lumen, where survival, multiplication and entry into intestinal epithelial cells are priority for the invasion of the pathogen. The fumarate reductase (frdABCD), dimethyl sulfoxide (DMSO)-trimethylamine N-oxide (TMAO) reductase (dmsABC), and nitrate reductase (narGHIJ) operons in Salmonella Typhimurium (STM) encode enzymes involved in anaerobic respiration to the electron acceptors fumarate, DMSO, TMAO, and nitrate, respectively. They are regulated in response to nitrate and oxygen availability and changes in cell growth rate. Vitamin B12 (cobalamin) is synthesized by Salmonella Typhimurium only under anaerobic growth conditions used as a cofactor in four known reactions. The deletion of cobS and cbiA genes prevent any form of cobalamin production. In the present study we evaluate the infection of birds by mutants of STM, with the anaerobic respiratory system committed by mutations in the genes: narG, napA, cobS, cbiA, frdA, dmsA, and torC. Virulence was assessed by oral inoculation of groups of one-day-old broilers with 0.1 mL of culture contained 10 8 colony forming units (CFU)/mL or diluted at 10 -3 and 10 -2 of strains mutants of Salmonella Typhimurium. Clinical signs and mortality were recorded over a period of 21 days. In general, the symptoms of chickens infected with the mutant strains were similar to those presenting by control birds. Except for STMNalr cbiA, all showed reduced capacity to cause mortality in comparison with the original strain. The mortality of group of chickens infected with STMNal r △narG, STMNal r △frdA, STMNal r △dmsA and STMNal r △cobS△cbiA showed significant decrease in mortality compared to control group (p<0.05).
