Este livro é resultado da pesquisa conduzida no âmbito do “Núcleo de Justiça e Constituição” da Escola de Direito de São Paulo da Fundação Getulio Vargas – FGV DIREITO SP. A pesquisa foi realizada por integrantes do Núcleo e com financiamento da Fapesp entre março de 2012 e fevereiro de 2013. Teve como objetivo analisar a atuação do Supremo Tribunal Federal (STF) enquanto Corte recursal, função exercida especialmente por meio do julgamento de recursos extraordinários. O nosso estudo examina o processo de tomada de decisão do STF nos recursos extraordinários julgados em Plenário, de 5 de setembro de 2007 a 1º de setembro de 2009. Esse é o maior período no qual a composição do Tribunal permaneceu estável desde 2000. Nesse intervalo de tempo, integravam o STF as Ministras e os Ministros: Celso de Mello, Marco Aurélio, Gilmar Mendes, Ellen Gracie, Carlos Britto, Cezar Peluso, Eros Grau, Joaquim Barbosa, Ricardo Lewandowski, Cármen Lúcia e Menezes Direito. Optou-se por investigar somente as decisões tomadas em sede de Plenário, por sua relevância qualitativa.

O objetivo deste estudo é avaliar a propagação de choques econômicos de alguns países sobre o crescimento econômico brasileiro, com principal destaque para China, Estados Unidos da América (EUA) e Argentina, que são os principais parceiros comerciais do Brasil. O aumento do comércio com a China tornou o Brasil muito mais vulnerável a choques no PIB chinês e menos vulnerável, do que no passado recente, a choques no PIB americano, enquanto que a influência da Argentina manteve-se estável. Foi aplicada a metodologia Vetor Autorregressivo Global (Global Var – GVAR), introduzida por Pesaran, Schuermann e Weiner (2004), Garratt, Lee, Pesaran e Shin (2006) e Dées, Di Mauro, Pesaran e Smith (2007), para analisar os canais de comércio e a transmissão de choques entre o resto do mundo e o Brasil. Usando dados trimestrais a partir de 1990 até o final de 2013, foi possível constatar que o aumento da relevância da economia Chinesa na balança comercial Brasileira exerce pressão sobre o crescimento econômico do Brasil. Em suma, a China tornou-se mais relevante para o crescimento econômico do Brasil do que os EUA e a Argentina.

Neste trabalho investigamos a formação de network considerando agentes cautelosos. O modelo consiste em duas regiões com (n/2) bancos em cada, onde a interligação entre eles ocorre através e depósitos interbancários. Cada banco está sujeito a corrida bancária, ou devido a um choque negativo de agentes impacientes, ou devido a contaminação da corrida de um banco pertencente a infraestrutura bancária. Os bancos podem tentar eliminar a possibilidade de contágio ao fazer um número alto de inter-ligações. Para isso, é necessário uma coordenação entre todos os bancos. Se um banco não se prevenir de um contágio, ele impõe a todos os outros a possibilidade de contágio no pior cenário. Há duas regiões bem definidas de equilíbrio de nash simétrico com network estável, uma na qual todos os bancos se previnem do cenário de contágio no pior cenário e a outra na qual nenhum banco se previne. Devido ao problema de coordenação, o equilíbrio com contágio no pior cenário pode ocorrer mesmo sendo pareto dominado pelo equilíbrio sem contágio. Sob certas condições, o equilíbrio com contágio ocorre com um network pareto eficiente. Neste caso o network eficiente é diferente do network mais resiliente ao contágio.

Esse trabalho busca analisar empiricamente a persistência inflacionária de um grupo de dez países da América do Sul e verificar se a persistência ficou estável durante o período analisado e se persistência inflacionária é mais alta em países que apresentaram alta inflação no seu passado recente. Os dados são trimestrais, tendo início no primeiro trimestre de 2000, e contém 60 observações. Os resultados do trabalho foram obtidos por meio da estimação dos seguintes modelos: modelo com defasagens de inflação com e sem o hiato do PIB; curva de Phillips Novo-Keynesiana com taxa de câmbio; e a forma reduzida do modelo estrutural de Blanchard e Gali (2005), que incorpora a rigidez de salários. Os resultados mostraram que a persistência inflacionária ficou estável durante o período analisado e que seu nível ficou abaixo de 1, na média, no grupo de países que apresentaram alta inflação no passado recente e no grupo de países que não apresentaram. Além disso, os resultados mostraram que, na amostra selecionada, a persistência inflacionária é mais alta nos países que apresentaram alta inflação no seu passado recente. Também foi verificado que, com 5% de significância, não é possível afirmar que a persistência inflacionária de todos os países que apresentaram alta inflação no passado recente seja diferente das economias que não apresentaram.

This thesis contains three chapters. The first chapter uses a general equilibrium framework to simulate and compare the long run effects of the Patient Protection and Affordable Care Act (PPACA) and of health care costs reduction policies on macroeconomic variables, government budget, and welfare of individuals. We found that all policies were able to reduce uninsured population, with the PPACA being more effective than cost reductions. The PPACA increased public deficit mainly due to the Medicaid expansion, forcing tax hikes. On the other hand, cost reductions alleviated the fiscal burden of public insurance, reducing public deficit and taxes. Regarding welfare effects, the PPACA as a whole and cost reductions are welfare improving. High welfare gains would be achieved if the U.S. medical costs followed the same trend of OECD countries. Besides, feasible cost reductions are more welfare improving than most of the PPACA components, proving to be a good alternative. The second chapter documents that life cycle general equilibrium models with heterogeneous agents have a very hard time reproducing the American wealth distribution. A common assumption made in this literature is that all young adults enter the economy with no initial assets. In this chapter, we relax this assumption – not supported by the data – and evaluate the ability of an otherwise standard life cycle model to account for the U.S. wealth inequality. The new feature of the model is that agents enter the economy with assets drawn from an initial distribution of assets. We found that heterogeneity with respect to initial wealth is key for this class of models to replicate the data. According to our results, American inequality can be explained almost entirely by the fact that some individuals are lucky enough to be born into wealth, while others are born with few or no assets. The third chapter documents that a common assumption adopted in life cycle general equilibrium models is that the population is stable at steady state, that is, its relative age distribution becomes constant over time. An open question is whether the demographic assumptions commonly adopted in these models in fact imply that the population becomes stable. In this chapter we prove the existence of a stable population in a demographic environment where both the age-specific mortality rates and the population growth rate are constant over time, the setup commonly adopted in life cycle general equilibrium models. Hence, the stability of the population do not need to be taken as assumption in these models.

A terapia genética tem se revelado uma ferramenta potente na Medicina, na tentativa de revolucionar o tratamento de várias doenças hereditárias e adquiridas. A introdução de genes em células pretende a expressão estável e prolongada de proteínas com efeitos terapêuticos. O silenciamento de genes, através da terapia genética que faz uso de oligonucleótidos antisense, pequenos RNA de interferência (siRNA) ou ribozimas, visa o decréscimo ou anulação do funcionamento de um gene cuja expressão amplificada, por algum motivo, leva ao desenvolvimento de umapatologia. A internalização de material genético nas células, usualmente, carece de métodos e/ou sistemas de entrega (vectores). Estes podem pertencer a duas categorias, designadamente, métodos virais e métodos não-virais. O primeiro é considerado o mais eficiente, apresentando porém, sérias desvantagens como o risco de carcinogénese. A solução é a utilização de métodos não virais,que podem ser físicos ou químicos. O objectivo principal desta dissertação foi a utilização de dendrímeros para o silenciamento do gene da proteína fluorescente optimizada (EGFP) em células HeLa, previamente modificadas para expressarem esta proteína. Dendrímeros poli(amidoamina) geração 5 (PAMAM G5) modificados com 4 ou 8 moléculas de ácidos gordos de diferentes comprimentos foram complexados com oligonucleótidos antisense. A vantagem que estes apresentam em relação aos dendrímeros nativos é que são capazes de interagir com os lípidos da membrana celular, esperando-se, por isso, uma melhor eficiência de transfecção e efeitos antisense. Isto foi efectivamente verificado, sendo que o nível de silenciamento do gene da EGFP obtido, está directamente relacionado com o aumento da razão NP, o número e o comprimento das cadeias hidrofóbicas. O silencimento de genes tem sofrido grandes avanços, havendo actualmente uma série de ensaios clínicos para a sua utilização no tratamento de doenças como cancros de origem hereditária ou viral, prevendo-se que venha para ficar, juntamente com o silenciamento mediado por siRNA.

Este estudo avalia as dimensões do engagement que os professores utilizam para lidar com o seu trabalho docente. Para a avaliação das dimensões utilizou-se a Utrecht Work Engagement Scale (UWES) elaborada por Schaufeli e Bakker (2003), distinguindo-se as variáveis vigor, dedicação e absorção. Os resultados obtidos nesta investigação (N=54) coincidem com estudos anteriores quanto à consistência interna das três subescalas, que é estável e satisfatória. Revelam também que a UWES apresenta-se como um instrumento válido e adequado para ser utilizado em investigações futuras.

The consumption of energy on the planet is currently based on fossil fuels. They are responsible for adverse effects on the environment. Renewables propose solutions for this scenario, but must face issues related to the capacity of the power supply. Wind energy offshore emerging as a promising alternative. The speed and stability are greater winds over oceans, but the variability of these may cause inconvenience to the generation of electric power fluctuations. To reduce this, a combination of wind farms geographically distributed was proposed. The greater the distance between them, the lower the correlation between the wind velocity, increasing the likelihood that together achieve more stable power system with less fluctuations in power generation. The efficient use of production capacity of the wind park however, depends on their distribution in marine environments. The objective of this research was to analyze the optimal allocation of wind farms offshore on the east coast of the U.S. by Modern Portfolio Theory. The Modern Portfolio Theory was used so that the process of building portfolios of wind energy offshore contemplate the particularity of intermittency of wind, through calculations of return and risk of the production of wind farms. The research was conducted with 25.934 observations of energy produced by wind farms 11 hypothetical offshore, from the installation of 01 simulated ocean turbine with a capacity of 5 MW. The data show hourly time resolution and covers the period between January 1, 1998 until December 31, 2002. Through the Matlab R software, six were calculated minimum variance portfolios, each for a period of time distinct. Given the inequality of the variability of wind over time, set up four strategies rebalancing to evaluate the performance of the related portfolios, which enabled us to identify the most beneficial to the stability of the wind energy production offshore. The results showed that the production of wind energy for 1998, 1999, 2000 and 2001 should be considered by the portfolio weights calculated for the same periods, respectively. Energy data for 2002 should use the weights derived from the portfolio calculated in the previous time period. Finally, the production of wind energy in the period 1998-2002 should also be weighted by 1/11. It follows therefore that the portfolios found failed to show reduced levels of variability when compared to the individual production of wind farms hypothetical offshore

Four different sponge species were screened using Ouchterlony agarose gel and immunodiffusion tests to identify cross-reactivity with the polyclonal antibody IgG anti-deglicosilated CvL, a lectin from Cliona varians. Crude extract from the sponge Cinachyrella apion showed cross-reactivity and also a strong haemmaglutinating activity towards human erythrocytes of all ABO groups. Thus, it was submitted to acetone fractionation, IgG anti-deglicosilated CvL Sepharose affinity chromatography, and Fast Protein Liquid Chromatography (FPLC-AKTA) gel filtration on a Superose 6 10 300 column to purify a novel lectin. C. apion lectin (CaL) agglutinated all types of human erythrocytes with preference for papainized type A and O erythrocytes. The haemagglutinating activity is independent of Ca2+, Mg2+ and Mn2+ ions, and it was strongly inhibited by the disaccharide D-lactose, up to a minimum concentration of 6.25 mM. CaL molecular mass determined by FPLC-AKTA gel filtration on a Superose 12 10 300 column and SDS gel electrophoresis was approximately 124 kDa, consisting of eight subunits of 15.5 kDa, assembled by hydrophobic interactions. The lectin was relatively heat- and pH-stable. Leishmania chagasi romastigotes were agglutinated by CaL, indicating that lactose receptors could be presented in this parasite stage. These findings are indicative of the physiological defense roles of CaL and its possible use in the antibiosis of pathogenic protozoa

A proteinaceous trypsin inhibitor was purified from Crotalaria pallida seeds by ammonium sulphate fractionation, affinity chromatography on immobilized Trypsin-Sepharose and TCA precipitation. The trypsin inhibitor, named ITC, had Mr of 32.5 kDa by SDS-PAGE and was composed by two subunits with 27.7 and 5.6 kDa linked by disulphide bridges, a typical characteristic of Kunitz-Inhibitor family. ITC was stable until 50°C, and at 100°C its residual activity was of about 60%. Also, ITC was stable at pHs 2 to 12. The inhibition of trypsin by ITC was non-competitive, with a Ki of 8,8 x 10-7M. ITC inhibits weakly other serine proteinases such as chymotrypsin and elastase. The inhibition of papain (44% of inhibition), a cysteine proteinase was an indicative of the bi-functionality of ITC. In vitro assays against digestive proteinases from several Lepdoptera, Diptera and Coleoptera pests were made. ITC inhibited in 100% digestive enzymes of Ceratitis capitata (fruit fly), Spodoptera frugiperda and Alabama argillacea, the last one being a cotton pest. It also inhibited in 74.4% Callosobruchus maculatus (bean weevil) digestive enzymes, a Coleoptera pest. ITC, when added in artificial diet models, affected weakly the development of C. capitata larvae and it had a WD50 of 2.65% to C. maculatus larvae

One Kunitz-type trypsin inhibitors (PmTI) was purified from Piptadenia moniliformis seeds, a tree of the sub-family Mimosoideae, by TCA precipitation, affinity chromatography on immobilized trypsin-Sepharose, DEAE cellulose (ion exchange) and Superose 12 (molecular exclusion) column FPLC/AKTA. The inhibitor has Mr of 25 kDa by SDS-PAGE and chromatography molecular exclusion. The N-terminal sequence of this inhibitor showed high homology with other family Kunitz inhibitors. This also stable variations in temperature and pH and showed a small decrease in its activity when incubated with DDT in the concentration of 100mM for 120 minutes. The inhibition of trypsin by PmTI was competitive, with Ki of 1.57 x10-11 M. The activity of trypsin was effectively inhibited by percentage of inhibition of 100%, among enzymes tested, was not detected inhibition for the bromelain, was weak inhibitor of pancreatic elastase (3.17% of inhibition) and inhibited by 76.42% elastase of neutrophils, and inhibited in a moderate, chymotrypsin and papain with percentage of inhibition of 42.96% and 23.10% respectively. In vitro assays against digestive proteinases from Lepidoptera, Diptera and Coleoptera pests were carried out. Several degrees of inhibition were found. For Anthonomus grandis and Ceratitis capitata the inhibition was 89.93% and 70.52%, respectively, and the enzymes of Zabrotes subfasciatus and Callosobruchus maculatus were inhibited by 5.96% and 9.41%, respectively, and the enzymes of Plodia. interpunctella and Castnia licus were inhibited by 59.94% and 23.67, respectively. In vivo assays, was observed reduction in the development of larvae in 4rd instar of C. capitata, when PmTI was added to the artificial diet, getting WD50 and LD50 of 0.30% and 0.33%, respectively. A chymotrypsin inhibitor was purified from Erythrina velutina seeds by ammonium sulphate fractionation, affinities chromatographies on Trypsin-Sepharose, Quimotrypsin-Sepharose and reversed phase C-18 FPLC/AKTA system. The inhibitor, named EvCI, shown molecular mass of 17 kDa, as determined by SDSPAGE. 2D-PAGE showed four isoinhibitors with pI values of 4,42, 4,63, 4,83 and 5,06, with molecular mass of 17 kDa each. The aminoacid sequence of EvCI was determined by MALDI-TOF-MS and showed a high similarity with other Kunitz-type inhibitor of Erythrina variegata. EvCI competitively inhibited chymotrypsin, with Ki of 4 x10-8 M, but did not inhibited trypsin, pancreatic elastase, bromelain and papain. The inhibitory activity of EvCI was stable over wide pH and temperature ranges. In the presence of DTT 100 mM for 120 min, EvCI lost 50 % of activity. Cytotoxicity was studied in HeLa, MDA, HepG2, K562 and PC3 cells after 72-h incubation period. EvCl inhibited HeLa cells growth with an IC50 value of 50 μg/ml. Subsequent studies in HeLa cells analysis of cell death by annexin V/PI double-staining and cell cycle, using flow cytometry. The results provide evidence for a cytostatic activity of EvCl and support further studies on potential application of this inhibitors as an antiproliferative agent in combined therapy against cervical cancer

A 140,0 kDa lectin was purified and characterized from the mushroom Clavaria cristata. The purification procedures from the crude extract of the mushroom comprised gel filtration chromatography on Sephacryl s200 and ion exchange on Resource Q column. The purified lectin agglutinated all types of human erythrocytes with preference for trypsinized type O erythrocytes. The haemagglutinating activity is dependent of Ca 2+ ions and was strongly inhibited by the glycoprotein bovine submaxillary mucin (BSM) up to the concentration of 0, 125 mg/mL. The C. cristata lectin (CcL) was stable in the pH range of 2,5-11,5 and termostable up to 80 °C. CcL molecular mass determined by gel filtration on a Superose 6 10 300 column was approximately 140,3 kDa. SDS polyacrilamide gel electrophoresis revealed a single band with a molecular mass of approximately 14,5 kDa, when the lectin was heated at 100 ⁰C in the presence or absence of β-mercaptoethanol. CcL induced activation of murine peritoneal macrophages in vitro resulting in the release of nitric oxide (NO), reaching the maximum production at 24 h. In experimental paw oedema model in mice, CcL showed proinflammatory activity being able to induce oedema formation. Cell viability of HepG2, MDA 435 e 3T3 cell lines was examined after 72 h of incubation with CcL in different concentrations (0,5-50 μg/mL). CcL inhibited HepG2 cells growth with an IC50 value of 50 μg/mL. In the present work, the observed immunomodulatory and antiproliferative effects indicate CcL as a possible immunomodulator compound, interfering in the macrophages immune response, taking possible anti-parasitic, anti-tumoral effects or diagnostic and/or therapeutic
