Context: Iodide transport defect (ITD) is an autosomal recessive disorder caused by impaired Na(+)/I(-) symporter (NIS)-mediated active iodide accumulation into thyroid follicular cells. Clinical manifestations comprise a variable degree of congenital hypothyroidism and goiter, and low to absent radioiodide uptake, as determined by thyroid scintigraphy. Hereditary molecular defects in NIS have been shown to cause ITD. Objective: Our objective was to perform molecular studies on NIS in a patient with congenital hypothyroidism presenting a clinical ITD phenotype. Design: The genomic DNA encoding NIS was sequenced, and an in vitro functional study of a newly identified NIS mutation was performed. Results: The analysis revealed the presence of an undescribed homozygous C to T transition at nucleotide -54 (-54C>T) located in the 5`-untranslated region in the NIS sequence. Functional studies in vitro demonstrated that the mutation was associated with a substantial decrease in iodide uptake when transfected into Cos-7 cells. The mutation severely impaired NIS protein expression, although NIS mRNA levels remained similar to those in cells transfected with wild-type NIS, suggesting a translational deficiency elicited by the mutation. Polysome profile analysis demonstrated reduced levels of polyribosomes-associated mutant NIS mRNA, consistent with reduced translation efficiency. Conclusions: We described a novel mutation in the 5`-untranslated region of the NIS gene in a newborn with congenital hypothyroidism bearing a clinical ITD phenotype. Functional evaluation of the molecular mechanism responsible for impaired NIS-mediated iodide concentration in thyroid cells indicated that the identified mutation reduces NIS translation efficiency with a subsequent decrease in protein expression and function. Autosomal recessive spastic paraplegia with thinning of corpus callosum (ARHSP-TCC) is a complex form of HSP initially described in Japan but subsequently reported to have a worldwide distribution with a particular high frequency in multiple families from the Mediterranean basin. We recently showed that ARHSP-TCC is commonly associated with mutations in SPG11/KIAA1840 on chromosome 15q. We have now screened a collection of new patients mainly originating from Italy and Brazil, in order to further ascertain the spectrum of mutations in SPG11, enlarge the ethnic origin of SPG11 patients, determine the relative frequency at the level of single Countries (i.e., Italy), and establish whether there is one or more common mutation. In 25 index cases we identified 32 mutations; 22 are novel, including 9 nonsense, 3 small deletions, 4 insertions, 1 in/del, 1 small duplication, 1 missense, 2 splice-site, and for the first time a large genomic rearrangement. This brings the total number of SPG11 mutated patients in the SPATAX collection to 111 cases in 44 families and in 17 isolated cases, from 16 Countries, all assessed using homogeneous clinical criteria. While expanding the spectrum of mutations in SPG11, this larger series also corroborated the notion that even within apparently homogeneous population a molecular diagnosis cannot be achieved without full gene sequencing. There has been great interest in deciding whether a combinatorial structure satisfies some property, or in estimating the value of some numerical function associated with this combinatorial structure, by considering only a randomly chosen substructure of sufficiently large, but constant size. These problems are called property testing and parameter testing, where a property or parameter is said to be testable if it can be estimated accurately in this way. The algorithmic appeal is evident, as, conditional on sampling, this leads to reliable constant-time randomized estimators. Our paper addresses property testing and parameter testing for permutations in a subpermutation perspective; more precisely, we investigate permutation properties and parameters that can be well approximated based on a randomly chosen subpermutation of much smaller size. In this context, we use a theory of convergence of permutation sequences developed by the present authors [C. Hoppen, Y. Kohayakawa, C.G. Moreira, R.M. Sampaio, Limits of permutation sequences through permutation regularity, Manuscript, 2010, 34pp.] to characterize testable permutation parameters along the lines of the work of Borgs et al. [C. Borgs, J. Chayes, L Lovasz, V.T. Sos, B. Szegedy, K. Vesztergombi, Graph limits and parameter testing, in: STOC`06: Proceedings of the 38th Annual ACM Symposium on Theory of Computing, ACM, New York, 2006, pp. 261-270.] in the case of graphs. Moreover, we obtain a permutation result in the direction of a famous result of Alon and Shapira [N. Alon, A. Shapira, A characterization of the (natural) graph properties testable with one-sided error, SIAM J. Comput. 37 (6) (2008) 1703-1727.] stating that every hereditary graph property is testable. In metazoans, bone morphogenetic proteins (BMPS) direct a myriad of developmental and adult homeostatic evens through their heterotetrameric type I and type II receptor complexes. We examined 3 existing and 12 newly generated mutations in the Drosophila type I receptor gene, saxophone (sax), the ortholog of the human Activin Receptor-Like. Kinasel and -2 (ALK1/ACVR1 and ALK2/ACVR1) genes. Our genetic analyses identified two distinct classes of sax alleles. The first class consists of homozygous viable gain-of-function (GOF) alleles that exhibit (1) synthetic lethality in combination with mutations in BMP pathway components, and (2) significant maternal effect lethality that can be rescued by an increased dosage of the BMP encoding gene, dpp(+). In contrast, the second class consists of alleles that are recessive lethal and do not exhibit lethality in combination with mutations in other BMP pathway components. The alleles in this second class are clearly loss-of-function (LOF) with both complete and partial loss-of-function mutations represented. We find that one allele in the second class of recessive lethals exhibits dominant-negative behavior, albeit distinct from the GOF activity of the first class of viable alleles. On the basis of the fact that the first class of viable alleles can be reverted to lethality and on our ability to independently generate recessive lethal sat mutations, our analysis demonstrates that sax is an essential gene. Consistent with this conclusion, we find that a normal sax transcript is produced by sax(P), a viable allele previously reported to be mill, and that this allele can be reverted to lethality. Interestingly, we determine that two mutations in the first: class of sax alleles show the same amino acid substitutions as mutations in the human receptors ALK1/ACVR1-1 and ACVR1/ALK2, responsible for cases of hereditary hemorrhagic telangiectasia type 2 (HHT2) and fibrodysplasia ossificans progressiva (FOP), respectively. Finally, the data presented here identify different functional requirements for the Sax receptor, support the proposal that Sax participates in a heteromeric receptor complex, and provide a mechanistic framework for future investigations into disease states that arise from defects in BMP/TGF-beta signaling.

The deficiency of complement C5 is rare and frequently associated with severe and recurrent infections, especially caused by Neisseria spp. We observed the absence of component C5 in the serum of 3 siblings from a Brazilian family with history of consanguinity. The patients had suffered from recurrent episodes of meningitis and other less severe infections. Sera from these patients were unable to mediate hemolytic activity either by the classical or alternative pathways and presented extremely low levels of C5 protein (13, 0.9 and 1.0 mu g/ml-normal range: 45-190 mu g/ml). Hemolytic activity could be restored by the addition of purified C5 to deficient serum. Sequencing of sibling C5 cDNA revealed a homozygous 153 bp deletion that corresponds precisely to exon 30. The parents carried the same deletion but only in one allele. Sequencing of the corresponding region in the genomic DNA revealed a C to C substitution within intron 30 and, most significantly, the substitution of GAG(4028) for GAA(4028) at the 3` end of exon 30 which is most likely responsible for skipping of exon 30. The resulting in-frame deletion in the C5 mRNA codes for a mutant C5 protein lacking residues 1289-1339. These residues map to the CUB and C5d domains of the C5 alpha chain. This deletion is expected to produce a non-functional and unstable C5 protein which is more susceptible to degradation. (C) 2009 Published by Elsevier Ltd.

Neste trabalho apresentam-se pocedimentos para análise não linear de estruturas de materiais compostos laminados reforçados por fibras. A formulação é baseada em uma descrição cinemática incremental Lagrangeana Total, que permite o tratamento de deslocamentos arbitrariamente grandes com pequenas deformações, utilizando elementos finitos tridimensionais degenerados deduzidos para a análise de cascas. As estruturas são consideradas como submetidas a cargas mecânicas e a ações de temperatura e de umidade. O material é suposto elástico linear com propriedades dependentes, ou não, dos valores da temperatura e da concentração de umidade, ou viscoelástico linear com uma relação constitutiva em integral hereditária , e com comportamento higrotermo-reologicamente simples. As lâminas são consideradas como sujeitas a falhas, as quais são detectadas através de critérios macroscópicos, baseados em tensões ou em deformações. As equações não lineares de equilíbrio são resolvidas através de procedimentos iterativos e as deformações dependentes do tempo são avaliadas pelo método das variáveis de estado. Diversos exemplos numéricos de estruturas submetidas à flexão, flambagem elástica e viscoelástica e falhas são apresentados.

Objetivo: A trombose da veia porta é uma causa importante de hiper-tensão porta em crianças e adolescentes, porém, em uma proporção importante dos casos, não apresenta fator etiológico definido. O objetivo desse estudo é determinar a freqüência de deficiência das proteínas inibidoras da coagulação – proteínas C, S e antitrombina − e das mutações fator V Leiden, G20210A no gene da protrombina e C677T da metileno-tetraidrofolato redutase em crianças e adolescentes com trom-bose da veia porta, definir o padrão hereditário de uma eventual deficiência das pro-teínas inibidoras da coagulação nesses pacientes e avaliar a freqüência da deficiên-cia dessas proteínas em crianças e adolescentes com cirrose. Casuística e Métodos: Foi realizado um estudo prospectivo com 14 crianças e adolescentes com trombose da veia porta, seus pais (n = 25) e dois gru-pos controles pareados por idade, constituídos por um grupo controle sem hepato-patia (n = 28) e um com cirrose (n = 24). A trombose da veia porta foi diagnosticada por ultra-sonografia abdominal com Doppler e/ou fase venosa do angiograma celíaco seletivo. A dosagem da atividade das proteínas C, S e antitrombina foi determinada em todos os indivíduos e a pesquisa das mutações fator V Leiden, G20210A da pro-trombina e C677T da metileno-tetraidrofolato redutase, nas crianças e adolescentes com trombose da veia porta, nos pais, quando identificada a mutação na criança, e nos controles sem hepatopatia. Resultados: Foram avaliados 14 pacientes caucasóides, com uma média e desvio padrão de idade de 8 anos e 8 meses ± 4 anos e 5 meses e do diagnóstico de 3 anos e 8 meses ± 3 anos e seis meses. Metade dos pacientes pertenciam ao gênero masculino. O motivo da investigação da trombose da veia porta foi hemorra-gia digestiva alta em 9/14 (64,3%) e achado de esplenomegalia ao exame físico em 5/14 (35,7%). Anomalias congênitas extra-hepáticas foram identificadas em 3/14 (21,4%) e fatores de risco adquiridos em 5/14 (35,7%) dos pacientes. Nenhum pa-ciente tinha história familiar de consangüinidade ou trombose venosa. A deficiência das proteínas C, S e antitrombina foi constatada em 6/14 (42,9%) (p < 0,05 vs con-troles sem hepatopatia), 3/14 (21,4%) (p > 0,05) e 1/14 (7,1%) (p > 0,05) pacientes com trombose da veia porta, respectivamente. A deficiência dessas proteínas não foi identificada em nenhum dos pais ou controles sem hepatopatia. A mutação G20210A no gene da protrombina foi identificada em um paciente com trombose da veia porta e em um controle sem hepatopatia (p = 0,999), mas em nenhum desses foi identificado a mutação fator V Leiden. A mutação C677T da metileno-tetraidrofo-lato redutase foi observada na forma homozigota, em 3/14 (21,4%) dos pacientes com trombose da veia porta e em 5/28 (17,9%) controles sem hepatopatia (p = 0,356). A freqüência da deficiência das proteínas C, S e antitrombina nos pacientes com cir-rose foi de 14/24 (58,3%), 7/24 (29,2%) e 11/24 (45,8%), respectivamente (p < 0,05 vs controles sem hepatopatia), sendo mais freqüente nos pacientes do subgrupo Child-Pugh B ou C, que foi de 11/12 (91,7%), 5/12 (41,7%) e 9/12 (75%), respectivamente (p < 0,05 vs controles sem hepatopatia). Conclusões: A deficiência de proteína C foi freqüente nas crianças e adolescentes com trombose da veia porta e não parece ser de origem genética. A deficiência de proteína S, antitrombina e as presenças das mutações G20210A da protrombina e C677T da metileno-tetraidrofolato redutase foram observadas mas não apresentaram diferença estatística significativa em relação ao grupo controle sem hepatopatia. O fator V Leiden não foi identificado. Os resultados deste estudo sugerem que a deficiência da proteína C pode ocorre como conseqüência da hiper-tensão porta. Os distúrbios pró-trombóticos hereditários não parecem apresentar um papel importante em relação à trombose nas crianças e adolescentes estudadas.

Esta tese pretende levantar e problematizar a influência da televisão na investigação da agressividade infantil. Para esse objetivo selecionou-se dois grupos de crianças, um grupo experimental e outro de controle oriundos de dois estabelecimentos de ensino da cidade do Rio de Janeiro. As crianças que compuseram a população estudada variavam de idade entre cinco e sete anos. Projetaram-se dois filmes: ao grupo experimental, um filme de ação - aventura dado com frustração e ao outro grupo - grupo de controle - um filme neutro caracterizado pelo desenrolar de cenas da vida animal. A frustração ao grupo experimental foi introduzida através de três interrupções na projeção do filme, interrupções essas que, no contexto da programação infantil, seriam provenientes de anúncios, ou de quaisquer outros motivos. Os instrumentos utilizados foram questionários respondidos pelos professores e pelas crianças, anotados por dez auxiliares de pesquisa. O estímulo "diferencial foi introduzido através dos dois filmes. Seguiu-se a sessão lúdica onde predominaram brincadeiras de jogos e bola, sendo feito na ocasião um video-tape. As auxiliares de pesquisa anotavam também os itens que lhes foram solicitados.

A aquisição da linguagem é um processo invariavelmente dependente da exposição ao ambiente. Contudo, a mais famosa e controversa hipótese desenvolvida por Chomsky sustenta que a criança traz para o processo de aprendizagem da língua um conjunto de recursos específicos – a Gramática Universal – para superar a pobreza de estímulos do ambiente e construir uma gramática final adequada, composta por princípios universais e parâmetros fixados de acordo com a experiência na comunidade lingüística. A Gramática Universal é, assim, entendida como uma dotação genética, necessária, mas não suficiente, para a aquisição da linguagem. Para além da hipótese do inatismo – central à tradição gerativista – a relação entre genes e linguagem vem sendo investigada há muitos anos, com evidências significativas de transmissão hereditária e etiologia genética para diversos tipos de distúrbios de linguagem. Mais recentemente, em 2001, foi descoberto o primeiro caso de um gene, o FOXP2, implicado na habilidade de adquirir e desenvolver a fala e a linguagem. Este trabalho parte de uma introdução ao processo de aquisição da linguagem em uma perspectiva inatista (Capítulo I) para enfocar os aspectos centrais à organização biológica da faculdade da linguagem (Capítulo II), propondo, a partir da análise dos efeitos da mutação do gene FOXP2, uma discussão introdutória sobre como o estudo dos distúrbios do desenvolvimento da linguagem, avaliados a partir de suas características fenotípica e genotípica, pode contribuir para a exploração de questões fundamentais acerca do desenvolvimento lingüístico e cognitivo (Capítulo III).

Background. Gaucher Disease (GD) is a hereditary lysosomal storage disorder characterized by the accumulation of glucosylceramide, mainly in the cells of the reticuloendothelial system, due to a deficiency of the enzyme acid β-glucosidase (GBA). Diagnosis is usually based on measurement of GBA activity in peripheral leukocytes. The purpose of this study was to evaluate the ability of screening for GBA and chitotriosidase activity using Dried Blood Spots on Filter Paper (DBS-FP) to identify individuals at high risk for GD in high-risk populations such as that of Tabuleiro do Norte, a small town in Northeastern Brazil. Methods. Between June 1, 2007 and May 31, 2008, 740 consented residents and descendants of traditional families from Tabuleiro do Norte were submitted to screening with DBS-FP. Subjects with GBA activity <2.19 nmol/h/mL were referred to analysis of GBA and chitotriosidase activity in peripheral leukocytes and in plasma, respectively. Subjects at highest risk for GD (GBA activity in peripheral leukocytes <5.6 nmol/h/mg protein) were submitted to molecular analysis to confirm diagnosis. Results. Screening with DBS-FP identified 135 subjects (18.2%) with GBA activity <2.19 nmol/h/mL, 131 of whom remained in the study. In 10 of these (7.6%), GBA activity in leukocytes was 2.6 5.5 nmol/h/mg protein. Subsequent molecular analysis confirmed 6 cases of heterozygosity and 4 normals for GD. Conclusion. DBS-FP assay was shown to be an effective initial GD screening strategy for high-prevalence populations in developing regions. Diagnosis could not be established from GBA activity in leukocytes alone, but required confirmation with molecular analysis

Hereditary Hemochromatosis (HH) is a genetic disease caused by high iron absorption and deposition in several organs. This accumulation results in clinical disturbances such as cirrhosis, arthritis, cardiopathies, diabetes, sexual disorders and skin darkening. The H63D and C282Y mutations are well defined in the hemochromatosis etiology. The aim of this paper was that of identifying the H63D and C282Y genetical mutations in the hemochromatosis gene and the frequency assessment of these mutations in the HFE protein gene in patients with hyperferritin which are sent to the DNA Center laboratory in Natal, state of Rio Grande do Norte. This paper also evaluates the HH H63D and C282Y gene mutations genotype correlation with the serum ferritin concentration, glucose, alanine aminotransferasis, aspartato aminotransferasis, gama glutamil transferasis and with the clinical complications and also the interrelation with life habits including alcoholism and iron overload. The biochemical dosages and molecule analyses are done respectively by the enzymatic method and PCR with enzymatic restriction. Out of the 183 patients investigated, 51,4% showed no mutation and 48,6% showed some type of mutation: 5,0% were C282Y heterozygous mutation; 1,1%, C282Y homozygous mutation; 31%, H63D heterozygous mutation; 8,7%, H63D homozygous mutation; and 3,3%, heterozygous for the mutation in both genes. As to gender, we observed a greater percentage of cases with molecular alteration in men in relation to women in the two evaluated mutations. The individuals with negative results showed clinical and lab signs which indicate hemochromatosis that other genes could be involved in the iron metabolism. Due to the high prevalence of hemochromatosis and taking into account that hemochromatosis is considered a public health matter, its gravity being preventable and the loss treatment toxicity, the early genetic diagnosis is indicated, especially in patients with high ferritin, and this way it avoids serious clinical manifestations and increases patients' life expectation. Our findings show the importance of doing such genetic studies in individuals suspected of hereditary hemochromatosis due to the high incidence of such a hereditary disease in our region

Ferritin is a protein composed of heavy and light chains, non-covalently linked and which accommodates, in its core, thousands of atoms of iron. Furthermore, this protein represents the stock of iron in the body and it is characterized as an acute marker and predictor of diseases, such as iron deficiency anemia, hereditary hemochromatosis and others. Considering the variability of reference values and the analytical methods currently available, the aim of this work was to propose 95% confidence intervals for adults in the State of Rio Grande do Norte, Brazil, after determining the average concentration of serum ferritin for both sexes, beyond its correlation with the age. We analyzed 385 blood samples, collected by venipuncture from individuals residing in the State, after 12-14 hours of fast. The populational sample had 169 men and 216 women between 18-59 years old, which filled a questionnaire on socioeconomic, food habits and accounts about previous and current diseases. The sample collections were itinerant and the results of erythrogram, fasting glucose, alanine aminotransferase, aspartate aminotransferase, γ-glutamyl transferase, urea, creatinine, leukocyte count and platelets, beyond C-reactive protein, were issued to each participant, so that, after selection of the apparently healthy individuals, the dosage of serum ferritin was carried out. Statistical analysis was performed using the softwares SPSS 11.0 Windows version, Epi Info 3.3.2 and Graf instant pad (version 3.02), and the random population sample was single (finite population), for which the test of linear correlation and diagram of dispersion were also made. After selection of individuals and determination of serum ferritin, the most discrepant outliers were disregarded (N = 358, Men = 154/Women = 207) and the average value determined for the masculine sex individuals was 167,18 ng / dL; for the feminine sex individuals, the average value obtained was 81,55 ng / dL. Moreover, we found that 25% of men had values < 90,30 ng / dL; 50% ≤ 156,25 ng / dL and 75% ≤ 229,00 ng / dL. In the group of women, 25% had values < 38,80 ng / dL; 50% ≤ 65,00 ng / dL and 75% ≤ 119,00 ng / dL. Through the correlation coefficient (r = 0,23 with p = 0,003), it is possible to suggest the existence of positive linear correlation between age and serum ferritin for men. The correlation coefficient for women (r = 0,16 with p = 0,025) also confirms the existence of positive linear correlation between serum ferritin and age. Considering the analysis carried out and specific methods corroborating with the proposed benchmarks, we concluded that the average value found for men is higher than that found for women. Furthermore, this scenario rises with age for both sexes, and the 95% confidence intervals obtained were 74 ng/dL ≤ μ ≤ 89 ng/dL and 152ng/dL ≤ μ ≤183ng/dL for the feminine and masculine sex individuals respectively

Uma cadela da raça Yorkshire Terrier apresentou hematêmese, distensão abdominal, poliúria e polidipsia. Após o exame clínico do paciente, estabeleceu-se a suspeita clínica de nefropatia. Os resultados dos exames laboratoriais revelaram anemia normocítica normocrômica e concentrações séricas de uréia (306mg/dl) e de creatinina (3,6mg/dl) acima dos valores de referência. Ao ultra-som bidimensional observaram-se áreas císticas hipoecóicas e anecóicas, padrão renal hipercogênico e perda da relação corticomedular. À necropsia, a região medular apresentou grande quantidade de cistos pequenos em meio a tecido conjuntivo fibroso. A lesão tubulointersticial cortical foi a responsável pela insuficiência renal resultante. Firmou-se diagnóstico de nefrite tubulointersticial.

In this research, we reflect on Body and Poetry: for an Education of the senses. This piece of work has as its objective the comprehension of the body and poetry s interlacement, for an education of the senses, starting from the experience of the Being in the world. With this purpose, we search to understand the poetic creation as the corporal transubstantiation by means of the senses, which is realized in the creative expression and manifests itself in the body language, opening ways to an education felt in the aesthetic experience. In order to comprehend the process of poetic creation, we rely on the phenomenological method of the lived world from Merleau-Ponty, philosophy, always opening forests of questionings and sprouting new doubts in search of other comprehensions about Poetry, Body and Education. These three phenomena were investigated by means of a reflection on my own life trajectory as a poet and Physical Education professor; identification and analysis of poets/reciters; sensitive experiences experience lived in Oficinas de Poesias (Poetry Workshops), held in seven public governmental schools of the State of Rio Grande do Norte and through a permanent dialogue with the works of Merleau-Ponty, Severino Antonio, Paul Zumthor, Petrucia Nóbrega, among other authors. Phenomenology and the sources investigated have permitted us to conclude that poetic creation is not a product of a divine inspiration, of a spirit come from over yonder, nor a hereditary gift, but poetry is created from a dialogue between the poet and the organic and cultural world, revealing at each experience undergone a new world of senses and meanings. It was possible to comprehend the poetic language as a synesthetic and performable manifestation, which ontologically reveals itself and hides itself at each experience, bringing new expressive clarities of the Being in the world. This piece of research has revealed us that poetry as a sensitive experience of the poetic state makes it possible a construction of a sensitive, glad and ludic knowledge for an education of the senses

Foram utilizados 24 suínos desmamados, distribuídos em quatro tratamentos, com seis animais em cada. Os suínos dos tratamentos 1, 2, 3 e 4 receberam, durante 90 dias, ração com respectivamente: 0,0, 0,2, 0,4 e 0,6% de sementes de Crotalaria spectabilis. Os animais que morreram durante o período experimental foram necropsiados logo após a morte. Os sobreviventes foram sacrificados no último dia do experimento. Durante as necropsias, foram colhidos fragmentos do fígado, rim, pulmão e estômago, para realização de exame histopatológico. Os principais sintomas da intoxicação foram edemas subcutâneos, principalmente nos membros, na face e região do pescoço, mucosas oral e ocular pálidas, cerdas eriçadas, caquexia e apatia. O hidropericárdio foi a lesão macroscópica mais comum, acometendo vários animais que receberam a ração contaminada. As principais lesões microscópicas foram fibrose, proliferação de ductos biliares e megalocitose no fígado, broncopneumonia e bronquite crônicas, megalocitose de células epiteliais tubulares renais, nefrose e nefrite crônica, bem como gastrite e ulceração gástrica. Os resultados destes experimentos indicam que ração contaminada com sementes de C. spectabilis, nas proporções utilizadas, foi tóxica para suínos.
