Aims: A rapid and simple HPLC-MS method was developed for the simultaneousdetermination of antidementia drugs, including donepezil, galantamine, rivastigmineand its major metabolite NAP 226 - 90, and memantine, for TherapeuticDrug Monitoring (TDM). In the elderly population treated with antidementiadrugs, the presence of several comorbidities, drug interactions resulting frompolypharmacy, and variations in drug metabolism and elimination, are possiblefactors leading to the observed high interindividual variability in plasma levels.Although evidence for the benefit of TDM for antidementia drugs still remains tobe demonstrated, an individually adapted dosage through TDM might contributeto minimize the risk of adverse reactions and to increase the probability of efficienttherapeutic response. Methods: A solid-phase extraction procedure with amixed-mode cation exchange sorbent was used to isolate the drugs from 0.5 mL ofplasma. The compounds were analyzed on a reverse-phase column with a gradientelution consisting of an ammonium acetate buffer at pH 9.3 and acetonitrile anddetected by mass spectrometry in the single ion monitoring mode. Isotope-labeledinternal standards were used for quantification where possible. The validatedmethod was used to measure the plasma levels of antidementia drugs in 300patients treated with these drugs. Results: The method was validated accordingto international standards of validation, including the assessment of the trueness(-8 - 11 %), the imprecision (repeatability: 1-5%, intermediate imprecision:2 - 9 %), selectivity and matrix effects variability (less than 6 %). Furthermore,short and long-term stability of the analytes in plasma was ascertained. Themethod proved to be robust in the calibrated ranges of 1 - 300 ng/mL for rivastigmineand memantine and 2 - 300 mg/mL for donepezil, galantamine and NAP226 - 90. We recently published a full description of the method (1). We found ahigh interindividual variability in plasma levels of these drugs in a study populationof 300 patients. The plasma level measurements, with some preliminaryclinical and pharmacogenetic results, will be presented. Conclusion: A simpleLC-MS method was developed for plasma level determination of antidementiadrugs which was successfully used in a clinical study with 300 patients.

A simple and sensitive LC-MS method was developed and validated for the simultaneous quantification of aripiprazole (ARI), atomoxetine (ATO), duloxetine (DUL), clozapine (CLO), olanzapine (OLA), sertindole (STN), venlafaxine (VEN) and their active metabolites dehydroaripiprazole (DARI), norclozapine (NCLO), dehydrosertindole (DSTN) and O-desmethylvenlafaxine (OVEN) in human plasma. The above mentioned compounds and the internal standard (remoxipride) were extracted from 0.5 mL plasma by solid-phase extraction (mix mode support). The analytical separation was carried out on a reverse phase liquid chromatography at basic pH (pH 8.1) in gradient mode. All analytes were monitored by MS detection in the single ion monitoring mode and the method was validated covering the corresponding therapeutic range: 2-200 ng/mL for DUL, OLA, and STN, 4-200 ng/mL for DSTN, 5-1000 ng/mL for ARI, DARI and finally 2-1000 ng/mL for ATO, CLO, NCLO, VEN, OVEN. For all investigated compounds, good performance in terms of recoveries, selectivity, stability, repeatability, intermediate precision, trueness and accuracy, was obtained. Real patient plasma samples were then successfully analysed.

A method is reported for the quantification of isoorientin (using a standard addition method) and total flavonoids (expressed as rutin, using the external standard method) in passion fruit pulp (Passiflora edulis Sims f. flavicarpa Degener, Passifloraceae). Extraction of flavonoids was optimized by experimental design methodology, and quantitative analysis was performed by high-performance liquid chromatography with photo-diode array detection (HPLC-UV/DAD). The method was developed and validated according to ICH requirements for specificity, linearity, accuracy, precision (repeatability and intermediate precision). LOD and LOQ. Rutin was chosen as standard for the quantification of total flavonoids in order to propose a HPLC method feasible for routine analysis of the flavonoids in the passion fruit pulp. The passion fruit pulp contained 16.226 +/- 0.050 mg L(-1) of isoorientin and 158.037 +/- 0.602 mg L(-1) of total flavonoid, suggesting that P. edulis fruits may be comparable with other flavonoid food sources such as orange juice or sugarcane juice. Hoffmann La Roche Ltd., Switzerland) vs. generic (Lipiblock (R), EMS Sigma Pharma, a generic drug) were carried out with ethanol extracts of commercial samples. The generic formulation contained higher levels of common impurities as well as a considerable number of impurities not found in the reference product. The detected impurity profile of Lipiblock (R) revealed that it most likely is based on fermentation. Since the effect of the impurities is unknown, at this point fully synthetic Xenical (R) appears to offer a better safety margin than Lipiblock (R) which, however, compares quite well to other generic formulations.

The leaves of Myrcia DC. ex Guill species are used in traditional medicine and are also exploited commercially as herbal drugs for the treatment of diabetes mellitus. The present work aimed to assess the qualitative and quantitative profiles of M. bella hydroalcoholic extract, due to these uses, since the existing legislation in Brazil determines that a standard method must be developed in order to be used for quality control of raw plant materials. The current study identified eleven known flavonoid-O-glycosides and six acylated flavonoid derivatives of myricetin and quercetin, together with two kaempferol glycosides and phenolic acids such as caffeic acid, ethil galate, gallic acid and quinic acid. In total, 24 constituents were characterized, by means of extensive preparative chromatographic analyses, along with MS and NMR techniques. An HPLC-PAD-ESI-ITMS and FIA-ESI-IT-MSn method were developed for rapid identification of acylated flavonoids, flavonoid-O- glycosides derivatives of myricetin and quercetin and phenolic acids in the hydroalcoholic M. bella leaves extract. The FIA-ESI-IT-MS techinique is a powerful tool for direct and rapid identification of the constituents after isolation and NMR characterization. Thus, it could be used as an initial method for identification of authentic samples concerning quality control of Myrcia spp extracts. © 2013 by the authors.

The phenolic composition of heartwood extracts from Fraxinus excelsior L. and F. americana L., both before and after toasting in cooperage, was studied using LC-DAD/ESI-MS/MS. Low-molecular weight (LMW) phenolic compounds, secoiridoids, phenylethanoid glycosides, dilignols and oligolignols compounds were detected, and 48 were identified, or tentatively characterized, on the basis of their retention time, UV/Vis and MS spectra, and MS fragmentation patterns. Some LMW phenolic compounds like protocatechuic acid and aldehyde, hydroxytyrosol and tyrosol, were unlike to those for oak wood, while ellagic and gallic acid were not found. The toasting of wood resulted in a progressive increase in lignin degradation products with regard to toasting intensity. The levels of some of these compounds in medium-toasted ash woods were much higher than those normally detected in toasted oak, highlighting vanillin levels, thus a more pronounced vanilla character can be expected when using toasted ash wood in the aging wines. Moreover, in seasoned wood, we found a great variety of phenolic compounds which had not been found in oak wood, especially oleuropein, ligstroside and olivil, along with verbascoside and isoverbascoside in F. excelsior, and oleoside in F. americana. Toasting mainly provoked their degradation, thus in medium-toasted wood, only four of them were detected. This resulted in a minor differentiation between toasted ash and oak woods. The absence of tannins in ash wood, which are very important in oak wood, is another peculiar characteristic that should be taken into account when considering its use in cooperage. Die Erdatmosphäre besteht hauptsächlich aus Stickstoff (78%), Sauerstoff (21%) und Edelga¬sen. Obwohl Partikel weniger als 0,1% ausmachen, spielen sie eine entscheidende Rolle in der Chemie und Physik der Atmosphäre, da sie das Klima der Erde sowohl direkt als auch indirekt beeinflussen. Je nach Art der Bildung unterscheidet man zwischen primären und sekundären Partikeln, wobei primäre Partikel direkt in die Atmosphäre eingetragen werden. Sekundäre Partikel hingegen entstehen durch Kondensation von schwerflüchtigen Verbindungen aus der Gasphase, welche durch Reaktionen von gasförmigen Vorläufersubstanzen (volatile organic compounds, VOCs) mit atmosphärischen Oxidantien wie Ozon oder OH-Radikalen gebildet werden. Da die meisten Vorläufersubstanzen organischer Natur sind, wird das daraus gebil¬dete Aerosol als sekundäres organisches Aerosol (SOA) bezeichnet. Anders als die meisten primären Partikel stammen die VOCs überwiegend aus biogenen Quellen. Es handelt sich da¬bei um ungesättigte Kohlenwasserstoffe, die bei intensiver Sonneneinstrahlung und hohen Temperaturen von Pflanzen emittiert werden. Viele der leichtflüchtigen Vorläufersubstanzen sind chiral, sowohl die Vorläufer als auch die daraus gebildeten Partikel werden aber in den meisten Studien als eine Verbindung betrachtet und gemeinsam analysiert. Die mit Modellen berechneten SOA-Konzentrationen, welche auf dieser traditionellen Vorstellung der SOA-Bil¬dung beruhen, liegen deutlich unterhalb der in der Atmosphäre gefundenen, so dass neben diesem Bildungsweg auch noch andere SOA-Bildungsarten existieren müssen. Aus diesem Grund wird der Fokus der heutigen Forschung vermehrt auf die heterogene Chemie in der Partikelphase gerichtet. Glyoxal als Modellsubstanz kommt hierbei eine wichtige Rolle zu. Es handelt sich bei dieser Verbindung um ein Molekül mit einem hohen Dampfdruck, das auf Grund dieser Eigenschaft nur in der Gasphase zu finden sein sollte. Da es aber über zwei Alde¬hydgruppen verfügt, ist es sehr gut wasserlöslich und kann dadurch in die Partikelphase über¬gehen, wo es heterogenen chemischen Prozessen unterliegt. Unter anderem werden in An¬wesenheit von Ammoniumionen Imidazole gebildet, welche wegen der beiden Stickstoff-He¬teroatome lichtabsorbierende Eigenschaften besitzen. Die Verteilung von Glyoxal zwischen der Gas- und der Partikelphase wird durch das Henrysche Gesetz beschrieben, wobei die Gleichgewichtskonstante die sogenannte Henry-Konstante ist. Diese ist abhängig von der un¬tersuchten organischen Verbindung und den im Partikel vorhandenen anorganischen Salzen. Für die Untersuchung chiraler Verbindungen im SOA wurde zunächst eine Filterextraktions¬methode entwickelt und die erhaltenen Proben anschließend mittels chiraler Hochleistungs-Flüssigchromatographie, welche an ein Elektrospray-Massenspektrometer gekoppelt war, analysiert. Der Fokus lag hierbei auf dem am häufigsten emittierten Monoterpen α-Pinen und seinem Hauptprodukt, der Pinsäure. Da bei der Ozonolyse des α-Pinens das cyclische Grund¬gerüst erhalten bleibt, können trotz der beiden im Molekül vorhanden chiralen Zentren nur zwei Pinsäure Enantiomere gebildet werden. Als Extraktionsmittel wurde eine Mischung aus Methanol/Wasser 9/1 gewählt, mit welcher Extraktionseffizienzen von 65% für Pinsäure Enan¬tiomer 1 und 68% für Pinsäure Enantiomer 2 erreicht werden konnten. Des Weiteren wurden Experimente in einer Atmosphärensimulationskammer durchgeführt, um die Produkte der α-Pinen Ozonolyse eindeutig zu charakterisieren. Enantiomer 1 wurde demnach aus (+)-α-Pinen gebildet und Enantiomer 2 entstand aus (-)-α-Pinen. Auf Filterproben aus dem brasilianischen Regenwald konnte ausschließlich Pinsäure Enantiomer 2 gefunden werden. Enantiomer 1 lag dauerhaft unterhalb der Nachweisgrenze von 18,27 ng/mL. Im borealen Nadelwald war das Verhältnis umgekehrt und Pinsäure Enantiomer 1 überwog vor Pinsäure Enantiomer 2. Das Verhältnis betrug 56% Enantiomer 1 zu 44% Enantiomer 2. Saisonale Verläufe im tropischen Regenwald zeigten, dass die Konzentrationen zur Trockenzeit im August höher waren als wäh¬rend der Regenzeit im Februar. Auch im borealen Nadelwald wurden im Sommer höhere Kon¬zentrationen gemessen als im Winter. Die Verhältnisse der Enantiomere änderten sich nicht im jahreszeitlichen Verlauf. Die Bestimmung der Henry-Konstanten von Glyoxal bei verschiedenen Saataerosolen, nämlich Ammoniumsulfat, Natriumnitrat, Kaliumsulfat, Natriumchlorid und Ammoniumnitrat sowie die irreversible Produktbildung aus Glyoxal in Anwesenheit von Ammoniak waren Forschungs¬gegenstand einer Atmosphärensimulationskammer-Kampagne am Paul-Scherrer-Institut in Villigen, Schweiz. Hierzu wurde zunächst das zu untersuchende Saataerosol in der Kammer vorgelegt und dann aus photochemisch erzeugten OH-Radikalen und Acetylen Glyoxal er¬zeugt. Für die Bestimmung der Glyoxalkonzentration im Kammeraerosol wurde zunächst eine beste¬hende Filterextraktionsmethode modifiziert und die Analyse mittels hochauflösender Mas¬senspektrometrie realisiert. Als Extraktionsmittel kam 100% Acetonitril, ACN zum Einsatz wo¬bei die Extraktionseffizienz bei 85% lag. Für die anschließende Derivatisierung wurde 2,4-Di¬nitrophenylhydrazin, DNPH verwendet. Dieses musste zuvor drei Mal mittels Festphasenex¬traktion gereinigt werden um störende Blindwerte ausreichend zu minimieren. Die gefunde¬nen Henry-Konstanten für Ammoniumsulfat als Saataerosol stimmten gut mit in der Literatur gefundenen Werten überein. Die Werte für Natriumnitrat und Natriumchlorid als Saataerosol waren kleiner als die von Ammoniumsulfat aber größer als der Wert von reinem Wasser. Für Ammoniumnitrat und Kaliumsulfat konnten keine Konstanten berechnet werden. Alle drei Saataerosole führten zu einem „Salting-in“. Das bedeutet, dass bei Erhöhung der Salzmolalität auch die Glyoxalkonzentration im Partikel stieg. Diese Beobachtungen sind auch in der Litera¬tur beschrieben, wobei die Ergebnisse dort nicht auf der Durchführung von Kammerexperi¬menten beruhen, sondern mittels bulk-Experimenten generiert wurden. Für die Trennung der Imidazole wurde eine neue Filterextraktionsmethode entwickelt, wobei sich ein Gemisch aus mit HCl angesäuertem ACN/H2O im Verhältnis 9/1 als optimales Extrak¬tionsmittel herausstellte. Drei verschiedenen Imidazole konnten mit dieser Methode quanti¬fiziert werden, nämlich 1-H-Imidazol-4-carbaldehyd (IC), Imidazol (IM) und 2,2‘-Biimidazol (BI). Die Effizienzen lagen für BI bei 95%, für IC bei 58% und für IM bei 75%. Kammerexperimente unter Zugabe von Ammoniak zeigten höhere Imidazolkonzentrationen als solche ohne. Wurden die Experimente ohne Ammoniak in Anwesenheit von Ammoni¬umsulfat durchgeführt, wurden höhere Imidazol-Konzentrationen gefunden als ohne Ammo¬niumionen. Auch die relative Luftfeuchtigkeit spielte eine wichtige Rolle, da sowohl eine zu hohe als auch eine zu niedrige relative Luftfeuchtigkeit zu einer verminderten Imidazolbildung führte. Durch mit 13C-markiertem Kohlenstoff durchgeführte Experimente konnte eindeutig gezeigt werden, dass es sich bei den gebildeten Imidazolen und Glyoxalprodukte handelte. Außerdem konnte der in der Literatur beschriebene Bildungsmechanismus erfolgreich weiter¬entwickelt werden. Während der CYPHEX Kampagne in Zypern konnten erstmalig Imidazole in Feldproben nach¬gewiesen werden. Das Hauptprodukt IC zeigte einen tageszeitlichen Verlauf mit höheren Kon¬zentrationen während der Nacht und korrelierte signifikant aber schwach mit der Acidität und Ammoniumionenkonzentration des gefundenen Aerosols.

The role of the therapeutic drug monitoring laboratory in support of immunosuppressant drug therapy is well established, and the introduction of sirolimus (SRL) is a new direction in this field. The lack of an immunoassay for several years has restricted the availability of SRL assay services. The recent availability of a CEDIA (R) SRL assay has the potential to improve this situation. The present communication has compared the CEDIA (R) SRL method with 2 established chromatographic methods, HPLC-UV and HPLC-MS/MS. The CEDIA (R) method, run on a Hitachi 917 analyzer, showed acceptable validation criteria with within-assay precision of 9.1% and 3.3%, and bias of 17.1% and 5.8%, at SRL concentrations of 5.0 mu g/L and 20 mu g/L, respectively. The corresponding between-run precision values were 11.5% and 3.3% and bias of 7.1% and 2.9% at 5.0 mu g/L and 20 mu g/L, respectively, The lower limit of quantification was found to be 3.0 mu g/L. A series of 96 EDTA whole-blood samples predominantly from renal transplant recipients were assayed by the 3 methods for comparison. It was found that the CEDIA (R) method showed a Deming regression line of CEDIA = 1.20 X HPLC-MS/MS - 0.07 (r = 0.934, SEE = 1.47), with a mean bias of 20.4%. Serial blood samples from 8 patients included in this evaluation showed that the CEDIA (R) method reflected the clinical fluctuations in the chromatographic methods, albeit with the variable bias noted. The CEDIA (R) method on the H917 analyzer is therefore a useful adjunct to SRL dosage individualization in renal transplant recipients.

Therapeutic monitoring with dosage individualization of sirolimus drug therapy is standard clinical practice for organ transplant recipients. For several years sirolimus monitoring has been restricted as a result of lack of an immunoassay. The recent reintroduction of the microparticle enzyme immunoassay (MEIA (R)) for sirolimus on the IMx (R) analyser has the potential to address this situation. This Study, using patient samples, has compared the MEIA (R) sirolimus method with an established HPLC-tandem mass spectrometry method (HPLC-MS/MS). An established HPLC-UV assay was used for independent cross-validation. For quality control materials (5, 11, 22 mu g/L), the MEIA (R) showed acceptable validation criteria based on intra-and inter-run precision (CV) and accuracy (bias) of < 8% and < 13%, respectively. The lower limit of quantitation was found to be approximately 3 mu g/L. The performance of the immunoassay was compared with HPLC-MS/MS using EDTA whole-blood samples obtained from various types of organ transplant recipients (n = 116). The resultant Deming regression line was: MEIA = 1.3 x HPLC-MS/MS+ 1.3 (r = 0.967, s(y/x) = 1) with a mean bias of 49.2% +/- 23.1 % (range, -2.4% to 128%; P < 0.001). The reason for the large and variable bias was not explored in this study, but the sirolimus-metabolite cross-reactivity with the MEIA (R) antibody could be a substantive contributing factor. Whereas the MEIA (R) sirolimus method may be an adjunct to sirolimus dosage individualization in transplant recipients, users must consider the implications of the substantial and variable bias when interpreting results. In selected patients where difficult clinical issues arise, reference to a specific chromatographic method may be required.

Measurement of lipid peroxidation is a commonly used method of detecting oxidative damage to biological tissues, but the most frequently used methods, including MS, measure breakdown products and are therefore indirect. We have coupled reversed-phase HPLC with positive-ionization electrospray MS (LC-MS) to provide a method for separating and detecting intact oxidized phospholipids in oxidatively stressed mammalian cells without extensive sample preparation. The elution profile of phospholipid hydroperoxides and chlorohydrins was first characterized using individual phospholipids or a defined phospholipid mixture as a model system. The facility of detection of the oxidized species in complex mixtures was greatly improved compared with direct-injection MS analysis, as they eluted earlier than the native lipids, owing to the decrease in hydrophobicity. In U937 and HL60 cells treated in vitro with t-butylhydroperoxide plus Fe2+, lipid oxidation could not be observed by direct injection, but LC-MS allowed the detection of monohydroperoxides of palmitoyl-linoleoyl and stearoyl-linoleoyl phosphatidylcholines. The levels of hydroperoxides observed in U937 cells were found to depend on the duration and severity of the oxidative stress. In cells treated with HOCl, chlorohydrins of palmitoyloleoyl phosphatidylcholine were observed by LC-MS. The method was able to detect very small amounts of oxidized lipids compared with the levels of native lipids present. The membrane-lipid profiles of these cells were found to be quite resistant to damage until high concentrations of oxidants were used. This is the first report of direct detection by LC-MS of intact oxidized phospholipids induced in cultured cells subjected to oxidative stress.

A perceção, as opiniões e os desejos dos consumidores têm um enorme impacto na indústria alimentar. Na perceção visual, a cor torna-se um fator fundamental e, neste campo, os corantes alimentares assumem uma extrema importância. A cor pode ser considerada um dos atributos mais impressionantes dos géneros alimentícios, que influencia diretamente a preferência e a seleção dos consumidores[1]. Existem muitos corantes naturais utilizados na indústria alimentar, tais como carotenóides, antocianinas e betalaínas. As betalaínas incluem compostos com cores que vão do vermelho-violeta (betacianidinas) ao amarelo-laranja (betaxantinas). As betalaínas não têm sido tão extensamente estudadas como as antocianinas, mas possuem uma capacidade corante três-vezes maior. A única betalaína autorizada como corante natural deriva da beterraba(E-162)[2], mas existem outras fontes alternativas de betacianidinas ,como a que se apresenta neste trabalho: Gomphrenaglobosa L., vulgarmente designada por perpétua roxa.
