The design of therapeutic compounds targeting transthyretin (TTR) is challenging due to the low specificity of interaction in the hormone binding site. Such feature is highlighted by the interactions of TTR with diclofenac, a compound with high affinity for TTR, in two dissimilar modes, as evidenced by crystal structure of the complex. We report here structural analysis of the interactions of TTR with two small molecules, 1-amino-5-naphthalene sulfonate (1,5-AmNS) and 1-anilino-8-naphthalene sulfonate (1,8-ANS). Crystal structure of TTR: 1,8-ANS complex reveals a peculiar interaction, through the stacking of the naphthalene ring between the side-chain of Lys15 and Leu17. The sulfonate moiety provides additional interaction with Lys15` and a water-mediated hydrogen bond with Thr119`. The uniqueness of this mode of ligand recognition is corroborated by the crystal structure of TTR in complex with the weak analogue 1,5-AmNS, the binding of which is driven mainly by hydrophobic partition and one electrostatic interaction between the sulfonate group and the Lys15. The ligand binding motif unraveled by 1,8-ANS may open new possibilities to treat TTR amyloid diseases by the elucidation of novel candidates for a more specific pharmacophoric pattern. Pure O-methyl N-methoxycarbonyl thiocarbamate CH(3)OC(S)N(H)C(O)OCH(3) (I) and O-ethyl N-methoxycarbonyl thiocarbamate, CH(3)CH(2)OC(S)N(H)C(O)OCH(3) (II), are quantitatively prepared by the addition reaction between the CH(3)OC(O)NCS and the corresponding alcohols. The compounds are characterized by multinuclear ((1)H and (13)C) and bi-dimensional ((13)C HSQC) NMR, GC-MS and FTIR spectroscopy techniques. Structural and conformational properties are analyzed using a combined approach involving crystallographic data, vibration spectra and theoretical calculations. The low-temperature (150 K) crystal structure of II was determined by X-ray diffraction methods. The substance crystallizes in the monoclinic space group P2(1)/n with a = 4.088(1)angstrom. b = 22.346(1)angstrom, c = 8.284(1)angstrom, beta = 100.687(3)degrees and Z = 4 molecules per unit cell. The conformation adopted by the thiocarbamate group -OC(S)N(H)- is syn (C=S double bond in synperiplanar orientation with respect to the N-H single bond), while the methoxycarbonyl C=O double bond is in antiperiplanar orientation with respect to the N-H bond. The non-H atoms in II are essentially coplanar and the molecules are arranged in the crystal lattice as centro-symmetric dimeric units held by N-H center dot center dot center dot S=C hydrogen bonds Id(N center dot center dot center dot S) = 3.387(1)angstrom, <(N-H center dot center dot center dot S) = 166.4(2)degrees]. The electrochemical oxidation of anodic metal (cobalt, nickel, copper, zinc and cadmium) in an acetonitrile solution of the Schiff-base ligand 2-(tosylamino)-N-[2-(tosylamino)-benzylidene] aniline (H(2)L) afforded the homoleptic compounds [ML]. The addition of 1,1-diphenylphosphanylmethane (dppm), 2,2`-bipyridine (bipy) or 1,10-phenanthroline (phen) to the electrolytic phase gave the heteroleptic complexes [NiL(dppm)], [ML(bipy)] and [ML(phen)]. The crystal structures of H(2)L (1), [NiL] (2), [CuL] (3), [NiL(dppm)] (4), [CoL(phen)] (5), [CuL(bipy)] (6) and [Zn(Lphen)] (7) were determined by X-ray diffraction. The homoleptic compounds [NiL] and [CuL] are mononuclear with a distorted square planar [MN(3)O] geometry with the Schiff base acting as a dianionic (N(amide)N(amide)N(imine)O(tosyl)) tetradentate ligand. Both compounds exhibit an unusual pi-pi stacking interaction be-tween a six-membered chelate ring containing the metal and a phenylic ring of the ligand. In the heteroleptic complex [NiL(dppm)], the nickel atom is in a distorted tetrahedral [NiN(3)P] environment defined by the imine, two amide nitrogen atoms of the L(2-) dianionic tridentate ligand and one of the phosphorus atoms of the dppm molecule. In the other heteroleptic complexes, [CoL(phen)], [CuL(bipy)] and [ZnL(phen)], the metal atom is in a five-coordinate environment defined by the imine, two amide nitrogen atoms of the dianionic tridentate ligand and the two bipyridine or phenanthroline nitrogen atoms. The compounds were characterized by microanalysis, IR and UV/Vis (Co, Ni and Cu complexes) spectroscopy, FAB mass spectrometry and (1)H NMR ([NiL] and Zn and Cd complexes) and EPR spectroscopy (Cu complexes).

The structural, spectroscopic and theoretical study of cyclocreatine (1-carboxymethyl-2-iminoimidazolidine, CyCre) has been performed prompted by the biological relevance of the molecule and its potential role as a ligand in biometalic compounds. The crystal structure of CyCre has been determined by X-ray diffraction methods. The compound crystallizes as a zwitterion in the monoclinic system, space group P2(1)/c. The crystal is further stabilized by a network of N-H center dot center dot center dot O bonds. Infrared and Raman spectra of the solid, electronic spectra of aqueous solutions at different pH values and (1)H and (13)C NMR spectra have been recorded and analyzed. Band assignments were accomplished with the help of theoretical calculations. Optimized molecular geometries, harmonic vibrational frequencies and molecular electrostatic potentials were calculated using methods based on the density functional theory. (C) 2010 Elsevier B.V. For the first time, crystals of suitable size for X-ray diffractometry structure determination (Dian important anti-HI V drug were prepared under solvothermal conditions. In this study, the crystal structure of didanosine (2`,3`-dideoxyinosine, ddI) in the form of a hydrate was determined using single-crystal X-ray diffractometry. Powder X-ray diffraction analysis revealed that the solid-state phase of the drug incorporated into pharmaceutical solid dosage forms is isostructural to the solvothermally prepared ddI material, even though they do not exhibit an identical chemical composition due to different water fractions occupying hydrophobic channels formed within the crystal lattice. Two ddI conformers are present in the structure, in agreement with a previous structure elucidation attempt. Concerning the keto enol equilibrium of ddI, our crystal data and vibrational characterizations by Fourier transform infrared (FTIR) and FT-Raman spectroscopy techniques were conclusive to state that both conformers exist in the keto form, contrary to solid-state NMR spectroscopic assignments that suggested ddI molecules occur as enol tautomers. In addition, characterizations by thermal (differential scanning calorimetry) and spectroscopic techniques allowed us to understand the structural similarities and the differences related to the hydration pattern of the nonstoichiometric hydrates.

In the present work, the thermal behavior of prednicarbate was studied using DSC and TG/DTG. The solid product remaining at the first decomposition step of the drug was isolated by TG, in air and N(2) atmospheres and was characterized using LC-MS/MS, NMR, and IR spectroscopy. It was found that the product at the first thermal decomposition step of prednicarbate corresponds to the elimination of the carbonate group bonding to C(17), and a consequent formation of double bond between C(17) and C(16). Structure elucidation of this degradation product by spectral data has been discussed in detail.

The bioactivity-guided fractionation of the crude extracts from leaves of Brazilian species Piper aduncum and Piper hostmannianum by means of bioautography using the fungi Cladosporium cladosporioides and C. sphaerospermum afforded prenylated methyl benzoate, chromenes, and dihydrobenzopyran derivatives as antifungal compounds. The isolation and structural elucidation of a new compound methyl 4-hydroxy-3-(2`-hydroperoxy-3`-methyl-3`-butenyl) benzoate were performed by application of chromatographic techniques and spectroscopic analyses. Five tetrahydrofuran lignans and two known flavones were isolated from the aerial parts of Peperomia blanda. The structures of the isolated lignans were elucidated by interpretation of their spectroscopic data, including by gHMQC and gHMBC. The relative and absolute configurations of the isolates were determined from NOESY interactions and optical properties, respectively. Four of the lignans were diastercomeric whilst one was of mixed biosynthetic origin. All but one of the lignans exhibited high in vitro trypanocidal activity when assayed against epimastigotes of Trypanosoma cruzi strain Y. (C) 2007 Elsevier Ltd. All rights reserved.

Mixed-ligand complexes of technetium(V) or rhenium(V) containing tridentate N-[(dialkylamino)(thiocarbonyl)]benzamidine (H(2)L(1)) and bidentate N,N-dialkyl-N`-benzoylthiourea (HL(2)) ligands were formed in high yields when (NBu(4))[MOCl(4)] (M = Tc or Re) or [ReOCl(3)(PPh(3))(2)] was treated with mixtures of the proligands. Other approaches for the synthesis of the products are reactions of [MOCl(L(1))] complexes with HL(2) or compounds of the-composition [ReOCl(2)(PPh(3))(L(2))] with H(2)L(1). The resulting air-stable [MO(L(1))(L(2))] complexes possess potential for the development of metal-based radiopharmaceuticals. [TcO(L(1))(L(2))] complexes are readily reduced by PPh3 with formation of [Tc(L(1))(L(2))(PPh(3))]. Introdução A mastocitose abrange um grupo heterogêneo de condições crônicas caracterizado pela proliferação excessiva de mastócitos nos tecidos. Os sinais e sintomas clínicos são decorrentes da distribuição anatômica dos mastócitos e do efeito funcional dos mediadores produzidos e liberados por estas células. Na infância, a doença é considerada uma condição benigna na maioria dos casos, cujo comprometimento característico é o cutâneo. As mais freqüentes manifestações na pele são os mastocitomas e a urticária pigmentosa. Lesões cutâneas bolhosas podem manifestar-se e acompanhar todas as formas de mastocitose e quando esta apresentação é a predominante, é denominada de mastocitose bolhosa. O diagnóstico de mastocitose é suspeitado clinicamente e confirmado pela histologia. A demonstração do aumento do número de mastócitos nas lesões cutâneas características se constitui no principal critério diagnóstico. Contudo, este método tem dificuldades técnicas que impedem a adequada reprodutibilidade dos achados, dificultando a elucidação de casos duvidosos e retardando seu tratamento. Considerando as propriedades imunológicas e a importância clínica dos mastócitos reveste-se de maior importância compreender o papel destas células nas doenças, sendo indispensável identificá-las e enumerá-las com acurácia nos tecidos. Objetivos Quantificar o número de mastócitos marcados com anticorpo monoclonal antitriptase, através de técnica imuno-histoquímica e análise de imagem, em biópsias cutâneas de crianças, com diagnóstico clínico de mastocitose. Descrever os achados histológicos; quantificar o número de mastócitos marcados com o anticorpo antitriptase entre as diferentes expressões clínicas da mastocitose cutânea; comparar o número de mastócitos entre os casos de mastocitose cutânea e mastocitose associada à sintomas sistêmicos e correlacionar as contagens de mastócitos entre os dois diferentes métodos (coloração por Giemsa com contagem manual e marcação com anticorpo antitriptase e análise digital). Material e Método Foram incluídas no estudo biópsias cutâneas de crianças de 0 a 14 anos, com diagnóstico clínico e histológico de mastocitose. Os casos foram classificados de acordo com a apresentação clínica cutânea em mastocitoma, urticária pigmentosa ou mastocitose bolhosa e assinalada a presença de sintomas sistêmicos associados. Os fragmentos de pele fixados em formalina e emblocados em parafina foram cortados e utilizados para diagnóstico histopatológico convencional, corados com hematoxilina-eosina e Giemsa, e para análise imuno-histoquímica com estreptavidina peroxidase marcados com anticorpo antitriptase. A densidade de mastócitos (número de células por área) foi realizada por um único observador na técnica histológica e através de um sistema de análise de imagem de vídeo no método imuno-histoquímico. Resultados Foram avaliados 33 casos de mastocitose, sendo 21 do sexo masculino. Dez casos (30,3%) apresentavam mastocitoma, 21 (63,6%) urticária pigmentosa e 2 (6,1%) mastocitose bolhosa. Todos os casos da amostra foram classificados como tendo mastocitose incipiente e em 6 (18,8%) pacientes pôde ser identificada a associação com sintomas sistêmicos. Prurido foi o sintoma mais freqüente, sendo relatado em 21 casos. Em 21 dos 33 casos foi identificada a infiltração de mastócitos na derme havendo predominância pela região perivascular (p=0,001, teste exato de Fisher). Não houve diferenças significativas entre a presença de infiltrado mastocitário e as várias formas cutâneas de mastocitose ou a mastocitose sistêmica. A presença de eosinófilos foi identificada em 15 casos (45,5%) e em 10 casos associadamente ao infiltrado perivascular de mastócitos. A densidade de mastócitos na técnica histológica, incluindo-se todos os casos, foi 50,00 células/mm2. Não houve diferença significativa das contagens entre os pacientes com mastocitoma e aqueles com urticária pigmentosa, assim como entre os pacientes com e sem sintomas sistêmicos associados aos cutâneos. A densidade de mastócitos encontrada com a técnica imuno-histoquímica e contagem por análise de imagem foi 158,85 células/mm2. Não houve diferença significativa das contagens entre os pacientes com mastocitoma e aqueles com urticária pigmentosa, assim como entre aqueles com e sem sintomas sistêmicos. Comparando-se a contagem dos mastócitos por área (densidade) entre a histologia e a imuno-histoquímica houve uma diferença significativa (p=0,0001 teste não-paramétrico de Wilcoxon). A média da diferença entre as contagens foi 199,98 células/mm2 (±365,31 DP). Também não houve semelhança, entre os dois métodos, nos grupos mastocitoma e urticária pigmentosa (p=0,005 e p=0,01, respectivamente, teste não-paramétrico de Wilcoxon). Puderam ser identificados 518% a mais de mastócitos com a técnica imunohistoquímica quando comparada com a histológica. Conclusões O presente estudo permite concluir que: 1) a localização preferencial da infiltração de mastócitos é dérmica e perivascular, não sendo possível identificar diferenças histológicas entre casos de urticária pigmentosa e mastocitoma; 2) o número de mastócitos marcados com o anticorpo monoclonal antitriptase e contados com análise digital de imagem, em biópsia de pele de crianças com diagnóstico clínico de mastocitose, foi 159 células por milímetro quadrado; 3) a densidade de mastócitos, foi semelhante entre os casos de urticária pigmentosa e mastocitoma e entre os casos com e sem sintomas sistêmicos associados nas duas diferentes técnicas empregadas; 4) o número de mastócitos por milímetro quadrado com a técnica imuno-histoquímica e a contagem através de análise de imagem foi significativamente maior quando comparada com a coloração através de Giemsa e a contagem manual, com uma diferença média entre os dois métodos de 200 células por milímetro quadrado; 5) a densidade de mastócitos com a técnica imunohistoquímica foi significativamente maior tanto nos casos com urticária pigmentosa quanto nos com mastocitoma, quando comparada com a técnica empregada rotineiramente e 6) com a técnica imuno-histoquímica e a contagem através de análise de imagem foi possível identificar 518% a mais de mastócitos quando comparada com a técnica histológica.

A ansiedade é um dos conceitos mais importantes dentro da Psicologia. Uma grande quantidade de trabalhos experimentais tratando sobre os efeitos da ansiedade-de-teste, tem sido conduzida. A discussão sobre ansiedade-de-teste e sua relação com o desempenho acadêmico deve ser precedida por uma elucidação do conceito de ansiedade. Este conceito tem sido interpretado de diferentes formas pelas várias escolas de pensamento na Psicologia e Filosofia. Os dois primeiros capítulos deste trabalho são dedicados à uma apresentação dos vários conceitos de ansiedade e de teorias que consideramos importantes que abordam o assunto. A seguir, passamos a descrever a análise experimental que foi realizada com o objetivo de detectar os efeitos da ansiedade-de-teste no desempenho e verificar os resultados da manipulação da variável tempo limitado e da magnitude do "stress", na relação estudada. Grande parte da literatura sobre ansiedade tem sublinhado que a influência desta sobre o comportamento é bastante complexa. A pesquisa aqui reportada se baseia nas investigações de Spielberger que focalizam a relação entre o desempenho do estudante e seu nível de ansiedade. Como Spielberger, julgamos ser necessário controlar as variáveis dificuldade da tarefa e inteligência dos sujeitos. Com base nos resultados encontrados, concluímos que uma série de outros aspectos devem ser considerados, tais como elaboração da prova, matéria envolvida, experiências anteriores dos alunos, imagem do professor etc. Busca-se conhecer a natureza da ansiedade-de-teste e encontrar formas efetivas para controlar este tipo específico de ansiedade, de maneira a aparelhar melhor os alunos para lidarem com ele. É necessário também que os educadores tomem conhecimento das características e implicações da ansiedade-de-teste, de forma que sejam capazes de tirar partido dela e consigam minimizar seus efeitos negativos no processo ensino-aprendizagem.

The purpose of this study was to identify whether activity modeling framework supports problem analysis and provides a traceable and tangible connection from the problem identification up to solution modeling. Methodology validation relied on a real problem from a Portuguese teaching syndicate (ASPE), regarding courses development and management. The study was carried out with a perspective to elaborate a complete tutorial of how to apply activity modeling framework to a real world problem. Within each step of activity modeling, we provided a summary elucidation of the relevant elements required to perform it, pointed out some improvements and applied it to ASPE’s real problem. It was found that activity modeling potentiates well structured problem analysis as well as provides a guiding thread between problem and solution modeling. It was concluded that activity-based task modeling is key to shorten the gap between problem and solution. The results revealed that the solution obtained using activity modeling framework solved the core concerns of our customer and allowed them to enhance the quality of their courses development and management. The principal conclusion was that activity modeling is a properly defined methodology that supports software engineers in problem analysis, keeping a traceable guide among problem and solution.

The mobilization of food reserves in storage tissues and allocation of their hydrolysis products in the growing axis are critical processes for the establishment of seedlings after germination. Therefore, it is crucial for mobilization of reserves to be synchronized with the growing axis, so that photosynthetic activity can be started before depletion of reserves. For this, integrative approaches involving different reserves, different hydrolysis products and interaction between storage and growing axis tissues, either through hormones or metabolites with signaling role, can contribute greatly to the elucidation of the regulation mechanisms for reserve mobilization. In this study, was hypothesized that hormones and metabolites have different actions on reserve mobilization, and there must be a crossed effect of sugars on the mobilization of proteins and amino acids on lipids and starch mobilization in sunflower seedlings. This study was conducted with seeds of sunflower (Helianthus annuus L.) hybrid Helio 253 using in vitro culture system. Seeds were germinated on Germitest® paper and grown on agar-water 4 g/L without addition of nutrients during 9 days after imbibition (DAI) for growth curve. To verify the effect of metabolites and hormones, seedlings were transferred in the 2nd DAI to agar-water 4 g/L supplemented with increasing concentrations of sucrose or L-glutamine, abscisic acid, gibberellic acid or indolebutyric acid. The results of this study confirm that the mobilization of lipids and storage proteins occurs in a coordinated manner during post-germination growth in sunflower, corroborating the hypothesis that the application of external carbon (sucrose) and nitrogen (L-glutamine) sources can delay the mobilization of these reserves in a crossed way. Moreover, considering the changes in the patterns of reserve mobilization and partition of their products in seedlings treated with different growth regulators, it is evident that the effects of metabolites and hormones must involve, at least in part, distinct mechanisms of action

Sulfated polysaccharides comprise a complex group of macromolecules with a range of several biological activities, including antiviral activity, anticoagulant, antiproliferative, antiherpética, antitumor, anti-inflammatory and antioxidant. These anionic polymers are widely distributed in tissues of vertebrates, invertebrates and algae. Seaweeds are the most abundant sources of sulfated polysaccharides in nature. The green algal sulfated polysaccharides are homo or heteropolysaccharides comprised of galactose, glucose, arabinose and/or glucuronic acid. They are described as anticoagulant, anti-inflammatory, antiviral, anti-angiogenic, antitumor compounds. However, there are few studies about elucidation and evaluation of biological/pharmacological effects of sulfated polysaccharides obtained from green algae, for example, there is only one paper reporting the antinociceptive activity of sulfated polysaccharides of these algae. Therefore this study aimed to obtain sulfated polysaccharides of green seaweed Codium isthmocladum and evaluates them as potential antinociceptive agents. Thus, in this study, the total extract of polysaccharides of green alga C. isthmocladum was obtained by proteolytic digestion, followed by fractionation resulting in five fractions (F0.3, F0.5, F0.7, F0.9 and F1.2) by sequential precipitation with acetone. Using the test of abdominal contractions we observed that the fraction F0.9 was the most potent antinociceptive aompound. F0.9 consists mainly of a sulfated heterogalactana. More specific tests showed that Fo.9 effect is dose and time dependent, reaching a maximum at 90 after administration (10 mg / kg of animal). F0.9 is associated with TRPV1 and TRPA1 receptors and inhibits painful sensation in animals. Furthermore, F0.9 inhibits the migration of lymphocytes induced peritonitis test. On the other hand, stimulates the release of NO and TNF-α. These results suggest that F0.9 has the potential to be used as a source of sulfated galactan antinociceptive and anti-inflammatory

Industrial activities, oil spills and its derivatives, as well as the incomplete combustion of fossil fuels have caused a great accumulation of hydrocarbons in the environment. The number of microorganisms on the planet is estimated at 1030 and prokaryotes the most abundant. They colonized diverse environments for thousands of years, including those considered extreme and represent an untapped source of metabolic and genetic diversity with a large biotechnological potential. It is also known that certain microorganisms have the enzymatic capacity to degrade petroleum hydrocarbons and, in many ecosystems, there is an indigenous community capable of performing this function. The metagenomic has revolutionized the microbiology allowing access uncultured microbial communities, being a powerful tool for elucidation of their ecological functions and metabolic profiles, as well as for identification of new biomolecules. Thus, this study applied metagenomic approaches not only for functional selection of genes involved in biodegradation and emulsification processes of the petroleum-derived hydrocarbons, but also to describe the taxonomic and metabolic composition of two metagenomes from aquatic microbiome. We analyzed 123.116 (365 ± 118 bp) and 127.563 sequences (352 ± 120 bp) of marine and estuarine metagenomes, respectively. Eight clones were found, four involved in the petroleum biodegradation and four were able to emulsify kerosene indicating their abilities in biosurfactants synthesis. Therefore, the metagenomic analyses performed were efficient not only in the search of bioproducts of biotechnological interest and in the analysis of the functional and taxonomic profile of the metagenomes studied as well
