Alternative splicing (AS) has the potential to greatly expand the functional repertoire of mammalian transcriptomes. However, few variant transcripts have been characterized functionally, making it difficult to assess the contribution of AS to the generation of phenotypic complexity and to study the evolution of splicing patterns. We have compared the AS of 309 protein-coding genes in the human ENCODE pilot regions against their mouse orthologs in unprecedented detail, utilizing traditional transcriptomic and RNAseq data. The conservation status of every transcript has been investigated, and each functionally categorized as coding (separated into coding sequence [CDS] or nonsense-mediated decay [NMD] linked) or noncoding. In total, 36.7% of human and 19.3% of mouse coding transcripts are species specific, and we observe a 3.6 times excess of human NMD transcripts compared with mouse; in contrast to previous studies, the majority of species-specific AS is unlinked to transposable elements. We observe one conserved CDS variant and one conserved NMD variant per 2.3 and 11.4 genes, respectively. Subsequently, we identify and characterize equivalent AS patterns for 22.9% of these CDS or NMD-linked events in nonmammalian vertebrate genomes, and our data indicate that functional NMD-linked AS is more widespread and ancient than previously thought. Furthermore, although we observe an association between conserved AS and elevated sequence conservation, as previously reported, we emphasize that 30% of conserved AS exons display sequence conservation below the average score for constitutive exons. In conclusion, we demonstrate the value of detailed comparative annotation in generating a comprehensive set of AS transcripts, increasing our understanding of AS evolution in vertebrates. Our data supports a model whereby the acquisition of functional AS has occurred throughout vertebrate evolution and is considered alongside amino acid change as a key mechanism in gene evolution.

Työn tarkoitus on löytää paperitehdasintegraatin tuorevesien käsittelyyn kloorikaasun korvaava desinfiointimenetelmä Stora Enso Publication Papers Oy Ltd.:n Anjalan tehtailla. Tehtaalla on ympäristöluvassaan velvoite selvittää mahdollisuudet tuoreveden desinfiointiin käytettävän kaasumaisen kloorin korvaamiseen hypokloriitilla. Kloorikaasun käyttö tehtaalla on työturvallisuusriski, ja jäännösklooripitoisuuksien hallittavuus kloorikaasua käytettäessä on heikko. Työssä tutkittava vesijae on kemiallisesti puhdistettu vesi. Koeajossa käytettävät kemikaalit ovat 10 % natriumhypokloriitti (NaOCl) ja kantaja-aine eli bromikloori- di-metyyli-hydantoiini (BCDMH). Tuloksista voidaan päätellä, että NaOCl:lla ja NaOCl:lla yhdessä BCDMH:n kanssa saavutetaan veden käyttökohteessa korkeammat aktiivisen kloorin jäännöspitoisuudet kuin kloorikaasulla. NaOCl:n ja kaksikomponenttisysteemin käyttökustannukset ovat kuitenkin yli kuusinkertaiset kloorikaasuun verrattuna. NaOCl annosteltuna yhteen pisteeseen olisi vaivattomin menetelmä kloorikaasun korvaamiseen. NaOCl:n annostelulla kahteen pisteeseen voitaisiin saavuttaa korkeampia jäännöspitoisuuksia kuin yhden pisteen annostelulla, mutta menetelmä vaatii lisätutkimuksia. Kaksikomponenttisysteemillä saavutetaan korkeimmat aktiivisen kloorin jäännöspitoisuudet, mutta sen seurantamenetelmien luotettavuudesta tulee varmistua ennen menetelmän käyttöönottoa. Korroosion ehkäisyn ja työturvallisuuden kannalta sekä NaOCl että NaOCl yhdessä BCDMH:n kanssa ovat turvallisempia vaihtoehtoja kuin kaasukloori.

RATIONALE:We investigated the impact of canakinumab, a fully human anti-interleukin-1b monoclonal antibody on inflammation and HRQoL in gouty arthritis patients.METHODS: In this 8-week, single-blind, dose-ranging study, patients with acute gouty arthritis flares, unresponsive/intolerant or contraindicated to NSAIDs and/or colchicine were randomized to single subcutaneous canakinumab (10, 25, 50, 90, or 150mg, N5143) or single intramuscular triamcinolone acetonide (TA, 40mg, N557). Patients assessed pain (Likert scale), physicians assessed clinical signs of joint inflammation, and HRQoL was recorded using SF-36.RESULTS: At baseline, 98% patients had moderate-to-extreme pain, 85% had moderate/severe joint swelling, 64-79% had elevated inflammatory markers and HRQoL scores indicated impaired physical function. Percentage of patients with no/mild pain was numerically greater in most canakinumab groups vs. TA, 24-72h post-dose; difference significant for 150mg group at these time-points (P<0.05). Canakinumab 150mg was associated with significantly lower Likert scores for tenderness [OR, 3.2; 95% CI, 1.27-7.89; P50.014] and swelling (OR, 2.7; 95% CI, 1.09-6.50, P50.032) at 72h vs. TA; erythema was not different. Median CRP and SAA levels normalized by 7 days post-dose in most canakinumab groups, but remained elevated in TA. Physical function improved at 7 days postdose in all groups, highest improvement for canakinumab 150mg. SF-36 scores for physical functioning and bodily pain with canakinumab 150mg approached US general population scores by 7 days post-dose and exceeded normal values by 8 weeks post-dose.CONCLUSION: Canakinumab 150mg produced significantly greater and rapid pain-relief and improvements in HRQoL vs. TAin acute gouty arthritis patients.

Valssilangan valmistuksessa päättymättömällä valssausprosessilla voidaan parantaa tuotannon tuottavuutta ja joustavuutta. Menetelmän ansiosta myös valssilankakieppien kokoa voidaan vaihdella asiakkaan tarpeiden mukaan. Prosessin joustavuus ja tuottavuus perustuvat liikkuvaan leimuhitsauslaitteistoon, jolla austeniittialueelle kuumennetut teelmät hitsataan toisiinsa ennen valssausta. Koska leimuhitsausliitoksen hyvä laatu on valmistusprosessin kannalta erityisen tärkeää, tutkittiin tässä työssä hitsausparametrien vaikutusta liitoksen laatuun. Hitsausparametreista tärkeimpiä ovat leimutusaika ja -jännite sekä tyssäyksen voima ja määrä. Parametrien ollessa liian suuria muodostuvat liitospinnat liian plastisiksi ja sopivan tyssäysvoiman ylläpitäminen on vaikeaa. Liian pienillä parametreilla liitospintojen plastisoituminen jää kesken ja tyssäys jää puutteelliseksi. Leimuhitsauksen laatuun vaikuttaa myös liitospinta-alan koko. Kaikki nämä tekijät heikentävät liitoksia, joten parametrien optimaalisuus on liitoksen hyvän laadun kannalta välttämätön. Parametrikokeina hitsattujen koekappaleiden rakennetta ja ominaisuuksiatutkittiin ainetta rikkovilla kokeilla. Lyhyemmillä leimutusajoilla hitsatuistakoekappaleista löytyi virheitä, jotka heikensivät huomattavasti liitoksien laatua. Menetelmäkokeiden perusteella koekappaleet eivät kaikilta osin täyttäneet liitosten laatuvaatimuksia.

The global economic and financial crisis is a challenge for all governments, but particularly for federal states because divided and/or shared territorial powers make federations susceptible to coordination problems in fiscal policy making. This article explores the effects of the ongoing crisis on federal relations. Three kinds of problems that may become the cause of federal tensions and conflicts are evoked: opportunism of subgovernments, centralisation and erosion of solidarity among members of the federation. Our analysis of fiscal policies and federal conflicts of 11 federations between 2007 and the present reveals three kinds of coordination problems: shirking in the use of federal government grants, rent-seeking in equalisation payments, and over-borrowing and over-spending. Our results show that shirking remained limited to few cases and occurred only in the first part of the crisis. However, rent-seeking and over-borrowing and over-spending led to a reduction of solidarity among subgovernments and to increased regulation of the fiscal discretion of the members of the federation. Subsequently, tensions in federal relations increased - although only in one case did this challenged the federal order.

Religion is alive and well all over the world, especially in times of personal, political, and social crisis. Even in Europe, long regarded the most "secular" continent, religion has taken centre stage in how people respond to the crises associated with modernity, or how they interact with the nation-state. In this book, scholars working in and on Europe offer fresh perspectives on how religion provides answers to existential crisis, how crisis increases the salience of religious identities and cultural polarization, and how religion is contributing to changes in the modern world in Europe and beyond. Cases from Poland to Pakistan and from Ireland to Zimbabwe, among others, demonstrate the complexity and ambivalence of religion's role in the contemporary world.

Introduction Our institution (University hospital) is encouraging physical activities for health through various popular sporting events in the city of Lausanne, the biggest of which is a road race of 2, 4, 10 and 20km. Objective To create an efficient and sustainable training program in preparation of the race for a group of motivated hospital employees without any prior experience with structured training and to identifying the benefits and limitations encountered.. Methods Subjects of various fitness levels were recruited by add and agreed to undergo lab and field testing before a 12-week 3 times/week running program, based on maximal aerobic speed (MAS-30/30 sec intervals), running technique exercises and endurance training. The interval session was the only one supervised. Their goal was the 10km (11 subjects) and the 20km (6 subjects). Results A group of 17 subjects (7 male and 10 female), mean age 36.6±7.3 years, VO2max 44.0±5.5 ml/kg/min, filed test interval MAS 15.1±2.4 km/h started the program. 2 were lost because of injury (while skiing). Adherence to interval sessions was excellent, although 3 weekly training sessions proved to be difficult for most of the subjects. Performance in the race was satisfying for all of them, 6/7 subjects having improved their running time from the previous year, the others participated for the first time and 7/8 completed the race satisfyingly, one DNF-ed because of sinusitis. Repeat MAS field test was available for 6 subjects, who improved by 5.9% (p<0.01). Subjectively, all of the participants were very satisfied with improvement, interaction with colleagues from various professions, and with self achievement and confidence. Conclusions Implementation of a structured training program for recreational or non-athletes can be very successful in creating a better self-confidence, a better working environment inside a hospital facility and obviously in improvement of physical fitness and athletic performance. Above all, it can only encourage health institutions to promote the health of their own employees through physical activity, which can allow people to connect through sports. As a result, subjects in this study tend to encourage other employees to be more active and are hungry for more advice and continued offers for physical activities benefiting both them and the institution through better efficiency at work and less absenteeism common to more active people.

Gas-liquid mass transfer is an important issue in the design and operation of many chemical unit operations. Despite its importance, the evaluation of gas-liquid mass transfer is not straightforward due to the complex nature of the phenomena involved. In this thesis gas-liquid mass transfer was evaluated in three different gas-liquid reactors in a traditional way by measuring the volumetric mass transfer coefficient (kLa). The studied reactors were a bubble column with a T-junction two-phase nozzle for gas dispersion, an industrial scale bubble column reactor for the oxidation of tetrahydroanthrahydroquinone and a concurrent downflow structured bed.The main drawback of this approach is that the obtained correlations give only the average volumetric mass transfer coefficient, which is dependent on average conditions. Moreover, the obtained correlations are valid only for the studied geometry and for the chemical system used in the measurements. In principle, a more fundamental approach is to estimate the interfacial area available for mass transfer from bubble size distributions obtained by solution of population balance equations. This approach has been used in this thesis by developing a population balance model for a bubble column together with phenomenological models for bubble breakage and coalescence. The parameters of the bubble breakage rate and coalescence rate models were estimated by comparing the measured and calculated bubble sizes. The coalescence models always have at least one experimental parameter. This is because the bubble coalescence depends on liquid composition in a way which is difficult to evaluate using known physical properties. The coalescence properties of some model solutions were evaluated by measuring the time that a bubble rests at the free liquid-gas interface before coalescing (the so-calledpersistence time or rest time). The measured persistence times range from 10 msup to 15 s depending on the solution. The coalescence was never found to be instantaneous. The bubble oscillates up and down at the interface at least a coupleof times before coalescence takes place. The measured persistence times were compared to coalescence times obtained by parameter fitting using measured bubble size distributions in a bubble column and a bubble column population balance model. For short persistence times, the persistence and coalescence times are in good agreement. For longer persistence times, however, the persistence times are at least an order of magnitude longer than the corresponding coalescence times from parameter fitting. This discrepancy may be attributed to the uncertainties concerning the estimation of energy dissipation rates, collision rates and mechanisms and contact times of the bubbles.
