Glutaredoxins (Grxs) are small (9-12 kDa) heat-stable proteins that are ubiquitously distributed. In Saccharomyces cerevisiae, seven Grx enzymes have been identified. Two of them (yGrx1 and yGrx2) are dithiolic, possessing a conserved Cys-Pro-Tyr-Cys motif. Here, we show that yGrx2 has a specific activity 15 times higher than that of yGrx1, although these two oxidoreductases share 64% identity and 85% similarity with respect to their amino acid sequences. Further characterization of the enzymatic activities through two-substrate kinetics analysis revealed that yGrx2 possesses a lower Km for glutathione and a higher turnover than yGrx1. To better comprehend these biochemical differences, the pK(a) of the N-terminal active-site cysteines (Cys27) of these two proteins and of the yGrx2-C30S mutant were determined. Since the pK(a) values of the yGrx1 and yGix2 Cys27 residues are very similar, these parameters cannot account for the difference observed between their specific activities. Therefore, crystal structures of yGrx2 in the oxidized form and with a glutathionyl mixed disulfide were determined at resolutions of 2.05 and 1.91 angstrom, respectively. Comparisons of yGrx2 structures with the recently determined structures of yGrx1 provided insights into their remarkable functional divergence. We hypothesize that the substitutions of Ser23 and Gln52 in yGrx1 by Ala23 and Glu52 in yGrx2 modify the capability of the active-site C-terminal cysteine to attack the mixed disulfide between the N-terminal active-site cysteine and the glutathione molecule. Mutagenesis studies supported this hypothesis. The observed structural and functional differences between yGrx1 and yGrx2 may reflect variations in substrate specificity. (C) 2008 Elsevier Ltd. Ce-基催化剂在汽车尾气净化,工业废气处理,烃类重整,烃类选择加氢等方面均有广泛的应用。近几十年来关于如何制备高活性、高稳定性的非贵金属复合氧化物催化剂一直是催化研究的重要课题之一。Ce-基催化剂主要都是通过其它金属氧化物M掺杂到CeO_2中形成Ce_(1-x)M_xO(M为掺杂物)固溶体。本文选用CeO_2作为主要研究对象,用柠檬酸法有目的的引入化学特性、离子半径不同的另一组分,用x值表示各元素间的化学计量比,优化催化剂的组成、结构,来调节所合成固溶体氧化物的物理化学性质。分别以碱土金属Ca、稀土金属La以及过渡金属Ni和Mn为掺杂物制备出含其它不同离子的Ce-基催化剂;将具有较高活性的Ni_(0.7)Ce_(0.3)O负载在ZrO_2上,以CH_4燃烧为模型反应,考察催化剂活性和氧化还原性的关系。1.Ce-Ca-La-O体系单独Ca或La分别加入到CeO_2中后催化剂的活性比单独的CeO_2的活性要高出很多,完全转化温度要下降近100℃,而且它们的H_2-TPR实验也证实了其氧化还原能力有很大的提高。将Ca和La同时引入到CeO_2的复合氧化物Ce-La-Ca-O材料,其活性比无La的Ce-Ca-O的活性没有明显的提高,而且反而要比Ce-La-O的活性低,且其HZ一TPR实验也显示出和复合氧化物Ce-Ca-O的轮廓一样。2.Ce-Ni-Mn-O体系对NiO、MnO_x、CeO_2三种金属氧化物,在优化两种金属氧化物最佳配比(组成)后,在复合氧化物中掺杂第三种金属氧化物以考察第三种金属对其甲烷燃烧活性的影响。(l)CeO_2-MnO_x体系中,在Ce_(0.8)Mn_(0.2)O掺杂NiO后,发现当Ni的摩尔量为-10%时,活性提高幅度的很大,完全燃烧的温度下降了近50℃,可在550℃将CH_4完全氧化到CO_2。(2)CeO_2-NiO体系中,Ce_(0.3)Ni_(0.7)O可在530℃将CH_4完全氧化到CO_2。向其中掺杂Mn后,复合氧化物的活性反而下降,要在550oC才能将CH4完全氧化到C02。这可能是阴离子缺陷减少所致。(3)NIO一Mnox体系中,Nio,IMn090掺杂Ce后,催化活性有大幅度提高,特别是Nio.ICeyMno90(0.3三y生0.8)中催化剂的活性更高,可在530oC体系中,其中y=0.5时更突出。3.Ni-Ce-O/ZrOZ体系(1)Ni1-x一Cex一O体系中,独立的CeOZ相促进了NIO的还原和表面积增加。(2)少量的CeOZ的掺杂明显改善了NIO对cH4完全氧化反应的活性。继续增加Ce的量催化活性弱有增加,然后下降。在Ce的掺杂量为30%时,即Nio7Ceo3O,催化活性最佳,此时甲烷完全转化的温度为530oC。(3)催化剂Ni07Ceo3O具有很好的稳定性,900oC下焙烧,还能在540oC将CH4完全氧化到COZ。(4)催化剂Pd/Ni07Ceo30的催化活性与Pd/A12O3的活性相当。(5)催化剂Ni07Ce03O负载在不同的载体上,发现ZrOZ作载体效果最佳,其次为5102,这可能是ZrOZ、5102对NIO、CeOZ相对惰性有关;而MgO、A1203虽表面积较大,但作为载体效果却不好,可能其易与NIO、CeOZ发生反应有关。(6)Nio7Ceo3O负载在ZrOZ上,提高了表面积同时促进了Nio7Ceo3O还原性,以负载量为50%时活性最好。结构分析发现有两个新相生成,Ni4Zro和CeZO3。(7)通过对比发现Nio7Ceo3O(50%)/ZrOZ体系高活性除了ZrOZ作为载体提高表面积外,Zr02和Ce在这里还起到助催化剂的作用。4.还探讨了Pr掺杂到CeO2,以及YSZ作为载体负载过渡金属氧化物在甲烷催化燃烧反应种的作用。

This is a summative evaluation of the Stronger Smarter Learning Communities (SSLC) project that examines whether and how the SSLC project had an impact on Australian state schools which adopted its models and approaches. Drawing from qualitative and quantitative data sets, it also presents the largest scale and most comprehensive analysis of Indigenous education practices and outcomes to date. It includes empirical findings on: success in changing school ethos and community engagement; challenges in progress at closure of the 'gap' in conventionally measured achievement and performance; schools' and principals' choices in curriculum and instruction; profiles of teachers' and principals' training and views on teacher education; and a strong emphasis on community and school Indigenoous voices and views on Indigenous education.

The structures of the anhydrous products from the interaction of 2-amino-5-(4-bromophenyl)-1,3,4-thiadiazole with (2-naphthoxy)acetic acid, the 1:1 adduct C8H6BrN3S . C12H10O3 (I) and 3,5-dinitrobenzoic acid, the salt C8H7BrN3S+ C7H3N2O6- (II) have been determined. In the adduct (I), a heterodimer is formed through a cyclic hydrogen-bonding motif [graph set R2/2(8)], involving carboxylic acid O-H...N(hetero)and amine N-H...O(carboxyl) interactions. The heterodimers are essentially planar with a thiadiazole to naphthyl ring dihedral angle of 15.9(2)deg. and the intramolecular thiadiazole to phenyl ring angle of 4.7(2)deg. An amine N-H...N(hetero) hydrogen bond between the heterodimers generates a one-dimensional chain structure extending down [001]. Also present are weak benzene-benzene and naphthalene-naphthalene pi-pi stacking interactions down the b axis [minimum ring centroid separation, 3.936(3) Ang.]. With the salt (II), the cation-anion association is also through a cyclic R2/2(8) motif but involving duplex N-H...O(carboxyl) hydrogen bonds, giving a heterodimer which is close to planar [dihedral angles between the thiadiazole ring and the two benzene rings, 5.00(16)deg. (intra) and 7.23(15)deg. (inter)]. A secondary centrosymmetric cyclic N-H...O(carboxyl) hydrogen-bonding association involving the second amino H-atom generates a heterotetramer. Also present in the crystal are weak pi-pi i-\p interactions between thiadiazolium rings [minimum ring centroid separation, 3.936(3)Ang.], as well as a short Br...O(nitro) interaction [3.314(4)Ang.]. The two structures reported here now provide a total of three crystallographically characterized examples of co-crystalline products from the interaction of 2-amino-5-(4-bromophenyl)-1,3,4-thiadiazole with carboxylic acids, of which only one involves proton-transfer.

In the title molecule, C13H15N3O2, the dihedral angle between the mean plane of the 1,3-dioxolane group and the 2-hydrazino-7-methylisoquinoline unit is 85.21 (5)degrees. The conformation of the molecule is influenced by bifurcated N-H center dot center dot center dot(O, O) and N-H center dot center dot center dot N intramolecular hydrogen bonds. In the crystal structure, molecules are linked via intermolecular N-H center dot center dot center dot O hydrogen bonds, forming extended chains along [001].

In the title moleclue, C19H21NO, the 4-piperidone ring adopts a chair conformation in which the two benzene rings and the methyl group attached to C atoms all have equatorial orientations. In the crystal structure, centrosymmetric dimers are formed through weak intermolecular C-H center dot center dot center dot O hydrogen bonds [the dihedral angle between the aromatic rings is 58.51 (5)degrees].

Infrared spectra of 1,3-dithiole-2-thione (DTT) and its four selenium analogues have been studied in the region 4000 to 20 cm�1. Assignment of all the fundamental frequencies was made by noting the band shifts on progressive selenation. Normal coordinate analysis procedures have been applied for both in-plane and out-of-plane vibrations to help the assignments. The Urey�Bradley force function supplemented with valence force constants for the out-of-plane vibrations was employed for coordinate calculations. A correlation of the infrared assignments of DTT with its different selenium analogues is accomplished. Further, the infrared assignments are compared with those of trithiocarbonate ion and its selenium analogues and other structurally related heterocyclic molecules.

This study aimed to examine the incidence of young adult-onset T1DM and T2DM among Finns, and to explore the possible risk factors for young adult-onset T1DM and T2DM that occur during the perinatal period and childhood. In the studies I-II, the incidence of diabetes was examined among 15-39-year-old Finns during the years 1992-2001. Information on the new diagnoses of diabetes was collected from four sources: standardized national reports filled in by diabetes nurses, the Hospital Discharge Register, the Drug Reimbursement Register, and the Drug Prescription Register. The type of diabetes was assigned using information obtained from these four data sources. The incidence of T1DM was 18 per 100,000/year, and there was a clear male predominance in the incidence of T1DM. The incidence of T1DM increased on average 3.9% per year during 1992-2001. The incidence of T2DM was 13 per 100,000/year, and it displayed an increase of 4.3% per year. In the studies III-V, the effects of perinatal exposures and childhood growth on the risk for young adult-onset T1DM and T2DM were explored in a case-control setting. Individuals diagnosed with T1DM (n=1,388) and T2DM (n=1,121) during the period 1992-1996 were chosen as the diabetes cases for the study, and two controls were chosen for each case from the National Population Register. Data on the study subjects parents and siblings was obtained from the National Population Register. The study subjects original birth records and child welfare clinic records were traced nationwide. The risk for young adult-onset T2DM was the lowest among the offspring of mothers aged about 30 years, whereas the risk for T2DM increased towards younger and older maternal ages. Birth orders second to fourth were found protective of T2DM. In addition, the risk for T2DM was observed to decrease with increasing birth weight until 4.2 kg, after which the risk began to increase. A high body mass index (BMI) at the BMI rebound between ages 3-11 years substantially increased the risk for T2DM, and the excess weight gain in individuals diagnosed with T2DM began in early childhood. Maternal age, birth order, or body size at birth had no effect on the risk for young adult-onset T1DM. Instead, individuals with T1DM were observed to have a higher maximum BMI before the age of 3 than their control subjects. In conclusion, the increasing trend in the development of both T1DM and T2DM among young Finnish adults is alarming. The high risk for T1DM among the Finnish population extends to at least 40 years of age, and at least 200-300 young Finnish adults are diagnosed with T2DM every year. Growth during the fetal period and childhood notably affects the risk for T2DM. T2DM prevention should also target childhood obesity. Rapid growth during the first years of life may be a risk factor for late-onset T1DM.

This publication lists the more important wood properties of commercial timbers used for construction in Queensland. It also provides requirements and conditions of use for these timbers to provide appropriate design service life in various construction applications. The correct specification of timber considers a range of timber properties including, but not limited to, stress grade; durability class; moisture content and insect resistance. For the specification of timber sizes and spans, relevant Australian Standards and design manuals should be consulted—e.g. Australian Standard AS 1684 series Residential timber—framed construction parts 2 and 3 (Standards Australia 2006a;b.) Book 1 explains the terms used; with reference to nomenclature; origin and timber properties presented under specific column headings in the schedules (Book 2). It also explains target design life; applications and decay hazard zones; presented in the Book 2 Schedules. Book 2 consists of reference tables; presented as schedules A; B and C: • Schedule A contains commercial mixtures of unidentified timbers and of some Australian and imported softwoods.
