Cerebellar Purkinje neurons receive two major excitatory inputs, the climbing fibers (CFs) and parallel fibers (PFs). Simultaneous, repeated activation of CFs and PFs results in the long-term depression (LTD) of the amplitude of PF-evoked synaptic currents. To induce LTD, activation of CFs may be substituted with depolarization of the Purkinje neuron to turn on voltage-activated calcium channels and increase the intracellular calcium concentration. The role of PFs in the induction of LTD, however, is less clear. PFs activate glutamate metabotropic receptors that increase phosphoinositide turnover and elevate cytosolic inositol 1,4,5-trisphosphate (InsP3). It has been proposed that calcium release from intracellular stores via InsP3 receptors may be important in the induction of LTD. We studied the role of InsP3 in the induction of LTD by photolytic release of InsP3 from its biologically inactive “caged” precursor in voltage-clamped Purkinje neurons in acutely prepared cerebellar slices. We find that InsP3-evoked calcium release is as effective in LTD induction as activation of PFs. InsP3-induced LTD was prevented by calcium chelator 1,2-bis(2-amino phenoxy)ethane-N,N,N′,N′-tetraacetic acid. LTD produced either by repeated activation of PFs combined with depolarization (PF+ΔV), or by InsP3 combined with depolarization (InsP3+ΔV) saturated at ≈50%. Maximal LTD induced by PF+ΔV could not be further increased by InsP3+ΔV and vice versa, which suggests that both protocols for induction of LTD share a common path. In addition to inducing LTD, photo-release of InsP3+ΔV resulted in the rebound potentiation of inhibitory synaptic currents. In the presence of heparin, an InsP3 receptor antagonist, repeated activation of PF+ΔV failed to induce LTD, suggesting that InsP3 receptors play an important role in LTD induction under physiological conditions.

The molecular reaction mechanism of the GTPase-activating protein (GAP)-catalyzed GTP hydrolysis by Ras was investigated by time resolved Fourier transform infrared (FTIR) difference spectroscopy using caged GTP (P3-1-(2-nitro)phenylethyl guanosine 5′-O-triphosphate) as photolabile trigger. This approach provides the complete GTPase reaction pathway with time resolution of milliseconds at the atomic level. Up to now, one structural model of the GAP⋅Ras⋅GDP⋅AlFx transition state analog is known, which represents a “snap shot” along the reaction-pathway. As now revealed, binding of GAP to Ras⋅GTP shifts negative charge from the γ to β phosphate. Such a shift was already identified by FTIR in GTP because of Ras binding and is now shown to be enhanced by GAP binding. Because the charge distribution of the GAP⋅Ras⋅GTP complex thus resembles a more dissociative-like transition state and is more like that in GDP, the activation free energy is reduced. An intermediate is observed on the reaction pathway that appears when the bond between β and γ phosphate is cleaved. In the intermediate, the released Pi is strongly bound to the protein and surprisingly shows bands typical of those seen for phosphorylated enzyme intermediates. All these results provide a mechanistic picture that is different from the intrinsic GTPase reaction of Ras. FTIR analysis reveals the release of Pi from the protein complex as the rate-limiting step for the GAP-catalyzed reaction. The approach presented allows the study not only of single proteins but of protein–protein interactions without intrinsic chromophores, in the non-crystalline state, in real time at the atomic level.

Photoactivation of caged fluorescent tubulin was used mark the microtubule (MT) lattice and monitor MT behavior in interphase cells. A broadening of the photoactivated region occurred as MTs moved bidirectionally. MT movement was not inhibited when MT assembly was suppressed with nocodazole or Taxol; MT movement was suppressed by inhibition of myosin light chain kinase with ML7 or by a peptide inhibitor. Conversely, MT movement was increased after inhibition of cytoplasmic dynein with the antibody 70.1. In addition, the half-time for MT turnover was decreased in cells treated with ML7. These results demonstrate that myosin II and cytoplasmic dynein contribute to a balance of forces that regulates MT organization, movement, and turnover in interphase cells.

The electrophoretic export of ATP against the import of ADP in mitochondria bridges the intra- versus extramitochondrial ATP potential gap. Here we report that the electrical nature of the ADP/ATP exchange by the mitochondrial ADP/ATP carrier (AAC) can be directly studied by measuring the electrical currents via capacitive coupling of AAC-containing vesicles on a planar lipid membrane. The currents were induced by the rapid liberation of ATP or ADP with UV flash photolysis from caged nucleotides. Six different transport modes of the AAC were studied: heteroexchange with either ADP or ATP inside the vesicles, initiated by photolysis of caged ATP or ADP; homoexchange with ADPex/ADPin or ATPex/ATPin; and caged ADP or ATP with unloaded vesicles. The heteroexchange produced the largest currents with the longest duration in line with the electrical charge difference ATP4- versus ADP3-. Surprisingly, also in the homoexchange and with unloaded vesicles, small currents were measured with shorter duration. In all three modes with caged ATP, a negative charge moved into the vesicles and with caged ADP it moved out of the vesicles. All currents were completely inhibited by a mixture of the inhibitors of the AAC, carboxyatractyloside and hongkrekate, which proves that the currents are exclusively due to AAC function. The observed charge movements in the heteroexchange system agree with the prediction from transport studies in mitochondria and reconstituted vesicles. The unexpected charge movements in the homoexchange or unloaded systems are interpreted to reveal transmembrane rearrangements of charged sites in the AAC when occupied with ADP or ATP. The results also indicate that not only ATP4- but also ADP3- contribute, albeit in opposite direction, to the electrical nature of the ADP/ATP exchange, which is at variance with former conclusions from biochemical transport studies. These measurements open up new avenues of studying the electrical interactions of ADP and ATP with the AAC.

Chemotactic signaling in Escherichia coli involves transmission of both negative and positive signals. In order to examine mechanisms of signal processing, behavioral responses to dual inputs have been measured by using photoactivable "caged" compounds, computer video analysis, and chemoreceptor deletion mutants. Signaling from Tar and Tsr, two receptors that sense amino acids and pH, was studied. In a Tar deletion mutant the photoactivated release of protons, a Tsr repellent, and of serine, a Tsr attractant, in separate experiments at pH 7.0 resulted in tumbling (negative) or smooth-swimming (positive) responses in ca. 50 and 140 ms, respectively. Simultaneous photorelease of protons and serine resulted in a single tumbling or smooth-swimming response, depending on the relative amounts of the two effectors. In contrast, in wild-type E. coli, proton release at pH 7.0 resulted in a biphasic response that was attributed to Tsr-mediated tumbling followed by Tar-mediated smooth-swimming. In wild-type E. coli at more alkaline pH values the Tar-mediated signal was stronger than the Tsr signal, resulting in a strong smooth-swimming response preceded by a diminished tumbling response. These observations imply that (i) a single receptor time-averages the binding of different chemotactic ligands generating a single response; (ii) ligand binding to different receptors can result in a nonintegrated response with the tumbling response preceding the smooth-swimming response; (iii) however, chemotactic signals of different intensities derived from different receptors can also result in an apparently integrated response; and (iv) the different chemotactic responses to protons at neutral and alkaline pH may contribute to E. coli migration toward neutrality.

Os principais objetivos deste trabalho são: 1) estudar o material malacofaunístico de novos jazigos da região de Rio Claro - Piracicaba (SP) e de testemunhos de sondagem de Conchas (SP), referentes à porção basal da Formação Corumbataí, termo superior do Grupo Passa Dois de idade permiana e 2) determinar sua distribuição bioestratigráfica. Incluiu-se um exame parcial da provável correlação entre as faunas das bases das Formações Corumbataí (São Paulo) e Estrada Nova (Paraná) e considerações de ordem paleoecológica da malacofauna. A análise taxonômica dos bivalves da porção basal da Formação Corumbataí no Estado, forneceu apreciável diversidade genérica e específica. Duas secções colunares, uma de superfície e outra de subsuperfície são apresentadas, mostrando a distribuição bioestratigráfica dessa malacofauna. Os bivalves identificados e descritos nos diversos afloramentos foram: Angatubia cf. A. cowperesioides Mendes, Anthraconaia ? mezzalirai sp. n., Barbosaia cf. B. angulata Mendes, Barbosaia roxoi sp n., Casterella cf. C. camargoi Beurlen, Ferrazia simplicicarinata Mezzalira, Ferrazia sp., Holdhausiella elongata Mendes, Holdhausiella sp., Kidodia cf. K. stockleyi Cox, Mendesia piracicabensis gen. et sp. n., Plesiocyprinella sp., Rioclaroa cf. R. lefrevei Mezzalira. Com base na composição observada, sugeriu-se reunir as Zonas Leinzia froezi Mendes e Barbosaia angulata Mendes propostas por MEZZALIRA (1980) em uma única designada Zona Leinzia froezi - Barbosaia angulata. Os quatro níveis fossilíferos reconhecidos em subsuperfície (Poço 1-IG-Conchas) com as espécies: Casterella cf. C. gratiosa, Ferrazia cardinalis Reed e Pinzonella cf. P. illusa, a julgar pela distribuição vertical em relação à base da formação, situar-se-iam na Zona III de MEZZALIRA (1980) considerada quando definida como afossilífera. Dentre as principais conclusões, notou-se a predominância de bivalves com carenas simples e a duplas, provavelmente caráter adaptativo ao ambiente que seria provavelmente circunscrito, mixo-halino, de águas rasas, turvas e mal oxigenadas em certos intervalos.

El presente trabajo muestra la caracterización en detalle de la secuencia estratigráfica de los depósitos miocenos de la isla de Nueva Tabarca, el empleo de estos materiales en el patrimonio arquitectónico de la isla y su comportamiento frente la degradación. En la serie miocena, a grandes rasgos, se definen tres tramos. El tramo inferior, rico en clastos dolomíticos, muestra una alternancia de arenitas y calcarenitas. El tramo intermedio corresponde a una sucesión de niveles de calcirruditas con laminación cruzada cuya parte superior está representada por un nivel de grandes bioclastos, rodolitos y arenitas de grano fino. El comienzo del último tramo está marcado por la presencia de varias superficies arenosas más litificadas sobre las que se sitúa un banco de arenitas de grano fino y laminación paralela. Si bien las canteras extraían sillares de todas estas litofacies, existe una mayor extracción de los últimos niveles de la serie. Estas rocas presentan una durabilidad moderada-baja, degradándose fácilmente mediante arenización, escamación y/o alveolización.

Malgré que le contenu des génomes mitochondriaux animaux soit dit bien conservé, des nouveaux gènes mitochondriaux ont été identifiés chez plusieurs espèces, surtout des invertébrés. Par exemple, les bivalves exhibant la double transmission uniparentale de leurs génomes mitochondriaux possèdent des nouveaux gènes spécifiques au sexe (M-ORF dans l’ADN de type M, F-ORF dans l’ADN de type F) qui ont été caractérisés in silico chez trois espèces de l’ordre Mytiloida, une espèce de Veneroida et une espèce de Unionoida par une précédente étude. Même si les séquences varient beaucoup entre ces trois ordres, cette étude à montré que des hélices transmembranaires ainsi que des peptides signaux sont conservés pour toutes les séquences. L’étude a aussi montré que les nouveaux gènes pourraient avoir des rôles dans la signalisation cellulaire, le cycle cellulaire et la réponse immunitaire et qu’ils pourraient être le résultat de l’endogénisation de l’ADN viral. Le projet présenté ici a pour but de mieux caractériser ces nouveaux gènes et leur origine potentielle, en plus d’étudier le H-ORF particulier aux hermaphrodites, en ciblant les espèces des unionidés. Les résultats montrent que les hélices transmembranaires et peptides signaux sont conservés chez les unionidés, les protéines semblent être associées à la membrane et être capables de lier des acides nucléiques et protéines, et les fonctions potentielles sont conservées. Les M-ORFs semblent avoir un rôle dans le transport et des processus cellulaires tels que la signalisation, le cycle cellulaire et la division, et l’organisation du cytosquelette. Les F-ORFs semblent être impliqués dans le trafic et transport cellulaire et la réponse immunitaire. Finalement, les H-ORFs semblent être des glycoprotéines structurales avec des rôles dans la signalisation, le transport et la transcription. Les résultats de ce projet pourraient supporter une origine virale ou mitochondriale pour ces gènes.

A new chemical procedure for cleaning marine carbonates was applied to remove detritus as well as metaloxide contaminations of marine shells from Eemian deposits and adjoining succession of a sediment core from Dagebüll, Schleswig- Holstein. Hence, one can significantly reduce the contamination with detrital uranium and thorium. Thermal ionisation mass spectrometry was applied to determine the uranium and thorium activities used for 230Th/U dating of these shells. At least ten samples of marine bivalves of five different core sections were analysed. Samples located below a five meter thick clay layer at 19-24 m yielded an average age of 132±18 ka. Shells located above the clays at 15-20 m depth were strongly influenced by percolating groundwaters of an open system. Therefore, a reliable dating of these samples was not possible.

A 0.25 m**2 United States Naval Electronics Laboratory box corer was used to take replicate samples from an oligotrophic bottom under the North Pacific Central Water Mass (~28°N, 155°W). The bottom is a red clay with manganese nodules at a depth of 5500-5800 m. Macrofaunal density ranges from 84 to 160 individuals per m**2 and is therefore much the same as in Northwest Atlantic Gyre waters. Of the macrofaunal taxa, polychaetes dominate (55 %), followed by tanaids (18 %), bivalves (7 %), and isopods (6 %). Meiofaunal taxa were only partially retained by the 297 µm screen used in washing. Even then, they are 1.5-3.9 times as abundant as the macrofaunal taxa, with nematodes being numerically dominant by far. Foraminifera seem to comprise an important portion of the community, but could not be assessed accurately because of the inability to discriminate living and dead tests. Remains of what are probably xenophyophoridans are also very important, but offer the same problem. Faunal diversity is extremely high, with deposit feeders comprising the overwhelming majority. Most species are rare, being encountered only once. The distributions of only three species show any significant deviation from randomness. The polychaete fauna from box cores collected from 90 miles to the north was not significantly different from that of the principal study locality. Concordance appeared at several taxonomic levels, from species through macrofaunal/meiofaunal relationships. As a result, the variation in total animal abundance shows aggregation among cores. We discuss Sokolova's concept of a deep-sea oligotrophic zone dominated by suspension feeders, and reconcile it with our present findings. The high diversity of the fauna combined with the low food level contradict theories that relate diversity directly with productivity.

The presence of gas hydrates on the Blake Ridge diapir, northeastern Atlantic Ocean, offers an opportunity to study the impact of methane seepage on the ecology and geochemistry of benthic foraminifera in the late Holocene. Three push cores, covering a time span of ~ 1000 yrs, were retrieved from three distinct microhabitats at the top of the diapir at a water depth of ~ 2150 m: (i) sediments away from seepage (control core), (ii) sediments overlain by clusters of methanotrophic and thiotrophic bivalves, and (iii) chemoautotrophic microbial mats. The foraminiferal assemblages at the two seep sites are marked by a reduction in benthic foraminiferal species diversity, coupled with a near-absence of agglutinated species. However, an opportunistic population rise in CH4- or H2S-tolerant calcareous species (e.g., Globocassidulina subglobosa and Cassidulina laevigata) that utilize the abundant trophic resources at the seeps has led to an increase in the overall assemblage density there. The delta18O and delta13C values of three species of benthic foraminifera - Gyroidinoides laevigatus, Globocassidulina subglobosa, and Uvigerina peregrina - and the planktonic species Globorotalia menardii were acquired from all three cores. The benthic species from methane seeps yield delta13C values of 0.1 to - 4.2 (per mil VPDB), that are distinctly more 13C-depleted relative to the delta13C of 0.4 to - 1.0 (per mil VPDB) at the control (off seep) site. The species from a mussel-bed site exhibit more negative delta13C values than those from microbial mats, possibly reflecting different food sources and higher rate of anaerobic oxidation of methane. The positive delta13C values in the paired planktonic species suggest that authigenic carbonate precipitation did not overprint the observed 13C depletions. Hence the probable cause of negative delta13C of benthic foraminifera is primary calcification from Dissolved Inorganic Carbon (DIC) containing mixed carbon fractions from (a) highly 13C-depleted, microbially-oxidized methane and (b) a seawater source.

The structure and distribution of the macrobenthic communities were studied in the southwestern Kara Sea. The material was collected in Baidaratskaya Bay in July 2007 and in a section running westward of the Yamal Peninsula in September 2007. The depths of the sampling stations ranged from 5 to 25 m in the Baidaratskaya Bay area and between 16 and 46 m in the Yamal section. A total of 212 benthic invertebrate species were recorded. In both areas, Bivalvia was the group with the highest biomass (54.88 g/m**2 in the Yamal section and 59.71 g/m**2 in the Baidaratskaya Bay area), while polychaetes were the group with the highest number of species (45 in the Yamal section and 64 the Baidaratskaya Bay area). Three major macrozoobenthic communities were recognized: the Astarte borealis community (20-46 m, the deepest sampling stations in both areas); the 'medium-depth' community (10-20 m, extremely mosaic, usually dominated by Serripes groenlandicus); and the Nephtys longosetosa community (depth smaller than 10 m, characterized by low biomass and the absence of large bivalves and echinoderms). The western Yamal shallow-water communities were shown to be generally similar to those of Baidaratskaya Bay. The comparison of these results with those of the benthos censuses performed in 1927-1945, 1975, and 1993 showed that the benthic communities in the southwestern Kara Sea remained relatively stable during the second half of the 20th century and the early 21st century.
