O feijão-caupi Vigna unguiculata [L.] Walp. é a principal cultura de subsistência do semiárido brasileiro, sendo fonte de proteínas de baixo custo, notadamente, para populações carentes. A produção dessa cultura no Nordeste é baixa devido à não utilização de insumos agrícolas - entre eles, o fertilizante nitrogenado. Por outro lado, bactérias promotoras de crescimento em plantas (BPCPs) vêm sendo estudadas, de forma a maximizar a fixação de N2, disponibilizar nutrientes como P ou fito-hormônios e inibir doenças. Os objetivos deste trabalho foram verificar a viabilidade da coinoculação das sementes de feijão-caupi usando Paenibacillus, Brevibacillus e, ou, Bacillus e Bradyrhizobium-caupi; caracterizar as estirpes quanto à produção de ácido indol acético (AIA) e solubilização de fosfato; e avaliar o sinergismo entre os microrganismos como alternativa para otimizar a FBN. Os experimentos foram conduzidos em laboratório e em casa de vegetação do Instituto Agronômico de Pernambuco (IPA), utilizando o feijão-caupi cv. "IPA - 206". As estirpes utilizadas foram: Bacillus sp. - 449, 450, 451, 461 e ANBE 31; B. cereus - 440; B. subtilis - 438, 441, 454, 455 e 459; B. pumilus - 444, 445 e 448; B. megaterium - 462; Brevibacillus brevis - 447; Paenibacillus brasilensis - 24, 172 e 177; P. graminis - MC 04.21, MC 22.13 e BR 60106; P. polymyxa - S21; e P. durus - RBN4. Os microrganismos não apresentaram capacidade para produzir AIA nem solubilizar fosfato. Ocorreu sinergismo das estirpes de Bacillus sp. (449) e Bacillus pumilus (444) coinoculadas com a estirpe de Bradyrhizobium sp. (BR 3267) no feijão-caupi.

O uso de inoculantes contendo bactérias fixadoras de N2 aumenta anualmente na cultura do feijão-caupi nas regiões Norte, Nordeste e Centro-Oeste do Brasil. Entretanto, em muitas ocasiões vêm sendo utilizadas estirpes recomendadas para a cultura da soja, mesmo existindo estirpes próprias para o feijão-caupi. Este trabalho foi conduzido com o objetivo de avaliar a resposta das estirpes de Bradyrhizobium recomendadas para a soja na nodulação e no crescimento e desenvolvimento das plantas de feijão-caupi. Conduziu-se um experimento em condições controladas em casa de vegetação com a cultivar BRS Guariba inoculada com as estirpes recomendadas para a cultura da soja: SEMIA 5079 (= CPAC 15), SEMIA 5080 (= CPAC 7), SEMIA 587 e SEMIA 5019 (= BR 29); duas estirpes recomendadas para o feijão-caupi: SEMIA 6462 (= BR 3267) e SEMIA 6464 (= BR 3262); um tratamento com adubação nitrogenada; e um controle absoluto. Os resultados mostraram que a estirpe SEMIA 587 apresentou eficiência na FBN na cultura do feijão-caupi semelhante à das estirpes BR 3262 e BR 3267. Por outro lado, apesar de as estirpes CPAC 15 e CPAC 7 proporcionarem a formação de elevado número de nódulos no feijão-caupi, ocasionaram crescimento de plantas, eficiência nodular e acúmulo de N inferiores aos das estirpes BR 3262 e BR 3267, não devendo ser utilizadas como inoculante para a cultura.

Rhizoctonia-like fungi are the main mycorrhizal fungi in orchid roots. Morphological characterization and analysis of conserved sequences of genomic DNA are frequently employed in the identification and study of fungi diversity. However, phytopathogenic Rhizoctonia-like fungi have been reliably and accurately characterized and identified through the examination of the fatty acid composition. To evaluate the efficacy of fatty acid composition in characterizing and identifying Rhizoctonia-like mycorrhizal fungi in orchids, three Epulorhiza spp. mycorrhizal fungi from Epidendrum secundum, two unidentified fungi isolated from Epidendrum denticulatum, and a phytopathogenic fungus, Ceratorhiza sp. AGC, were grouped based on the profile of their fatty acids, which was assessed by the Euclidian and Mahalanobis distances and the UPGMA method. Dendrograms distinguished the phytopathogenical isolate of Ceratorhiza sp. AGC from the mycorrhizal fungi studied. The symbionts of E. secundum were grouped into two clades, one containing Epulorhiza sp.1 isolates and the other the Epulorhiza sp.2 isolate. The similarity between the symbionts of E. denticulatum and Epulorhiza spp. fungi suggests that symbionts found in E. denticulatum may be identified as Epulorhiza. These results were corroborated by the analysis of the rDNA ITS region. The dendrogram constructed based on the Mahalanobis distance differentiated the clades most clearly. Fatty acid composition analysis proved to be a useful tool for characterizing and identifying Rhizoctonia-like mycorrhizal fungi.

ABSTRACT Trichoderma species are non-pathogenic microorganisms that protect against fungal diseases and contribute to increased crop yields. However, not all Trichoderma species have the same effects on crop or a pathogen, whereby the characterization and identification of strains at the species level is the first step in the use of a microorganism. The aim of this study was the identification – at species level – of five strains of Trichoderma isolated from soil samples obtained from garlic and onion fields located in Costa Rica, through the analysis of the ITS1, 5.8S, and ITS2 ribosomal RNA regions; as well as the determination of their individual antagonistic ability over S. cepivorum Berkeley. In order to distinguish the strains, the amplified products were analyzed using MEGA v6.0 software, calculating the genetic distances through the Tamura-Nei model and building the phylogenetic tree using the Maximum Likelihood method. We established that the evaluated strains belonged to the species T. harzianum and T. asperellum; however it was not possible to identify one of the analyzed strains based on the species criterion. To evaluate their antagonistic ability, the dual culture technique, Bell’s scale, and the percentage inhibition of radial growth (PIRG) were used, evidencing that one of the T. asperellum isolates presented the best yields under standard, solid fermentation conditions.

Background: Molecular tools may help to uncover closely related and still diverging species from a wide variety of taxa and provide insight into the mechanisms, pace and geography of marine speciation. There is a certain controversy on the phylogeography and speciation modes of species-groups with an Eastern Atlantic-Western Indian Ocean distribution, with previous studies suggesting that older events (Miocene) and/or more recent (Pleistocene) oceanographic processes could have influenced the phylogeny of marine taxa. The spiny lobster genus Palinurus allows for testing among speciation hypotheses, since it has a particular distribution with two groups of three species each in the Northeastern Atlantic (P. elephas, P. mauritanicus and P. charlestoni) and Southeastern Atlantic and Southwestern Indian Oceans (P. gilchristi, P. delagoae and P. barbarae). In the present study, we obtain a more complete understanding of the phylogenetic relationships among these species through a combined dataset with both nuclear and mitochondrial markers, by testing alternative hypotheses on both the mutation rate and tree topology under the recently developed approximate Bayesian computation (ABC) methods. Results Our analyses support a North-to-South speciation pattern in Palinurus with all the South-African species forming a monophyletic clade nested within the Northern Hemisphere species. Coalescent-based ABC methods allowed us to reject the previously proposed hypothesis of a Middle Miocene speciation event related with the closure of the Tethyan Seaway. Instead, divergence times obtained for Palinurus species using the combined mtDNA-microsatellite dataset and standard mutation rates for mtDNA agree with known glaciation-related processes occurring during the last 2 my. Conclusion The Palinurus speciation pattern is a typical example of a series of rapid speciation events occurring within a group, with very short branches separating different species. Our results support the hypothesis that recent climate change-related oceanographic processes have influenced the phylogeny of marine taxa, with most Palinurus species originating during the last two million years. The present study highlights the value of new coalescent-based statistical methods such as ABC for testing different speciation hypotheses using molecular data.

Microbial mats are complex but stable, multi-layered and multi-functional biofilms, which are the most frequent bacterial formations in nature. The functional strategies and physiological versatility of the bacterial populations growing in microbial mats allow bacteria to resist changing conditions within their environment. One of these strategies is the accumulation of carbon- and energy-rich polymers that permit the recovery of metabolic activities when favorable conditions are restored. In the present study, we systematically screened microbial mats for bacteria able to accumulate large amounts of the ester carbon polymers polyhydroxyalkanoates (PHA). Several of these strains were isolated from Ebro Delta microbial mats and their ability to accumulate PHA up to 40-60 % of their dry weight was confirmed. According to two identification approaches (16S rRNA and ropD genes), these strains were identified as Halomonas alkaliphila (MAT-7, -13, -16), H. neptunia (MAT-17), and H. venusta (MAT-28). To determine the mode of growth yielding maximum PHA accumulation, these three different species were cultured in an artificial biofilm in which the cells were immobilized on alginate beads. PHA accumulation by cells that had detached from the biofilm was compared with that of their planktonic counterparts. Experiments in different culture media showed that PHA accumulation, measured as the relative fluorescence intensity after 48 h of incubation at 30 °C, was higher in immobilized than in planktonic cells, with the exception of cells growing in 5 % NaCl, in which PHA accumulation was drastically lower in both. Therefore, for obtaining high PHA concentrations, the use of immobilized cells may be a good alternative to the PHA accumulation by bacteria growing in the classical, planktonic mode. From the ecological point of view, increased PHA accumulation in detached cells from biofilms would be a natural strategy to improve bacterial dispersion capacity and, consequently, to increase survival in stressed environments.

Studies of hybrid zones can inform our understanding of reproductive isolation and speciation. Two species of brown lemur (Eulemur rufifrons and E. cinereiceps) form an apparently stable hybrid zone in the Andringitra region of south-eastern Madagascar. The aim of this study was to identify factors that contribute to this stability. We sampled animals at 11 sites along a 90-km transect through the hybrid zone and examined variation in 26 microsatellites, the D-loop region of mitochondrial DNA, six pelage and nine morphological traits; we also included samples collected in more distant allopatric sites. Clines in these traits were noncoincident, and there was no increase in either inbreeding coefficients or linkage disequilibrium at the centre of the zone. These results could suggest that the hybrid zone is maintained by weak selection against hybrids, conforming to either the tension zone or geographical selection-gradient model. However, a closer examination of clines in pelage and microsatellites indicates that these clines are not sigmoid or stepped in shape but instead plateau at their centre. Sites within the hybrid zone also occur in a distinct habitat, characterized by greater seasonality in precipitation and lower seasonality in temperature. Together, these findings suggest that the hybrid zone may follow the bounded superiority model, with exogenous selection favouring hybrids within the transitional zone. These findings are noteworthy, as examples supporting the bounded superiority model are rare and may indicate a process of ecologically driven speciation without geographical isolation.

Thyrinteina arnobia (Stoll, 1782) (Lepidoptera: Geometridae) é considerada uma das mais sérias pragas do eucalipto no Brasil. Este trabalho foi realizado com o objetivo de estudar a preferência alimentar de T. arnobia em seis espécies de eucalipto e a influência da idade foliar sobre a seleção hospedeira, utilizando-se folhas jovens e velhas de Eucalyptus grandis, Eucalyptus camaldulensis, Eucalyptus saligna, Eucalyptus citriodora, Eucalyptus robusta e Eucalyptus cloeziana. Lagartas de T. arnobia alimentadas na geração anterior com folhas de E. grandis preferiram folhas jovens de E. grandis e E.cloeziana, enquanto as alimentadas com E. saligna, na geração anterior, preferiram folhas velhas de E. grandis. A espécie preferida por lagartas de T. arnobia foi E. grandis, observando-se, também maior preferência por folhas jovens que por folhas velhas nas espécies utilizadas nos testes.

Ipomoea carnea spp. fistulosa, a native woody perennial, is capable of spreading rapidly over seasonally flooded grassland in the Brazilian Pantanal, South America's largest wetland, thus conflicting with the local cattle ranching. I. carnea is controlled by mowing at the onset of the rainy season, as close as possible before the seasonal flooding. Often, however, flooding begins after the plant has had enough time to re-sprout enabling it to survive. The objective of this study was to verify if Ipomoea carnea plant's production follows a seasonal cycle, and, if so, at which point in this cycle, the plant is most vulnerable to mechanical control measures. Seasonal dynamics of stem and leaf production of I. carnea were studied. The results showed that growth of I. carnea is fastest at the onset of the rainy season in November/December. Production declines when seasonal flooding commences in January/February and almost ceases towards the begin of the dry season in May/June. This leads to the proposal that I. carnea could be controlled more effectively if the weed were mown in the early dry season when its production and its capability to re-sprout is lowest, and if any new sprouts were cut by hand when the seasonal flooding starts.

O Bacillus subtilis pode favorecer o desempenho simbiótico do rizóbio, pelos efeitos na inibição de fitopatógenos ou pela exsudação de fitormônios. Com o objetivo de verificar a viabilidade da co-infecção de sementes de soja com Bradyrhizobium e Bacillus foram conduzidos três experimentos, no Paraná, em solos com população estabelecida de Bradyrhizobium, em que as estirpes de Bradyrhizobium SEMIA 5019 e SEMIA 5080 e suas variantes tolerantes aos metabólitos de Bacillus foram co-infectadas com duas estirpes de Bacillus (AP-3 e PRBS-1), ou seus metabólitos. Na safra 1993/94, em Londrina, o tratamento de co-inoculação de Bradyrhizobium com os metabólitos formulados de Bacillus incrementou, significativamente, em relação ao não-inoculado, o número de nódulos (59%, estádio V3), a ocupação dos nódulos pelas estirpes de Bradyrhizobium (76%, R2) e o rendimento de grãos (24%); em Ponta Grossa, esses incrementos foram de 60%, 145% e 22%, respectivamente. Nessa safra, em Londrina, a co-inoculação das variantes tolerantes com os metabólitos de Bacillus também aumentou o rendimento (26%) e N total (17%) dos grãos de soja e incrementos significativos foram constatados, na ocupação dos nódulos, pela co-inoculação das variantes tolerantes com as células de Bacillus (78%). Os resultados obtidos indicam a viabilidade da co-inoculação, em sementes de soja, de metabólitos brutos ou formulados ou, ainda, de células de Bacillus subtilis, para incrementar a contribuição do processo de fixação biológica do nitrogênio.

A eficiência das estirpes de Bradyrhizobium com características Hup+ (SR e USDA-110), Hup- (29W) e Hup hr (SEMIA-587) foi avaliada em caupi (Vigna unguiculata L.), cultivares IPA-202, BR-3 e VITA-4. Os resultados mostraram que VITA-4, em relação à nodulação, revelou-se superior às demais, e apresentou interação efetiva com as estirpes SEMIA-587 e USDA-110. Entretanto, quanto à eficiência nodular, a combinação IPA-202 x SEMIA-587 alcançou maior atividade da nitrogenase (ARA) com eficiência relativa próxima a 1,0. A ARA detectada nas estirpes SR e SEMIA-587 foi similar, porém, superior às estirpes USDA-110 e 29W, evidenciando que as estirpes Hup+ e Hup hr alcançaram maiores atividades enzimáticas. Os teores de leghemoglobina (Lb) detectados nas estirpes SR e USDA-110 foram positivamente relacionados com as respectivas ARA, contudo, a relação entre teor de Lb e ARA obtido para 29W-Hup- foi variável, sugerindo que, na ausência da hidrogenase, o sistema da nitrogenase fica afetado podendo influir no fluxo de Lb ao bacteróide. A avaliação do teor de N mostrou que não houve diferença entre cultivares, entretanto, foi detectada diferença significativa entre as estirpes. As estirpes Hup+ obtiveram maiores acúmulos de N, evidenciando que os sistemas simbióticos que menos liberam H2, acumulam mais N no hospedeiro.
Avaliação de linhagens, híbridos F1 e cultivares de pimentão quanto à resistência a Meloidogyne spp.

Com o objetivo de avaliar híbridos F1 de pimentão, juntamente com suas linhagens progenitoras e cultivares, quanto à resistência a Meloidogyne incognita (raças 1, 2, 3 e 4) e a M. javanica, foi instalado um experimento em casa de vegetação nas dependências da Pioneer Sementes Ltda, em Ijaci, MG. Foi utilizado o delineamento de blocos casualizados, em esquema de parcela subdividida, com cinco parcelas (compostas pelas quatro raças de M. incognita e mais a espécie M. javanica) e 48 subparcelas (compostas por 47 genótipos de pimentão e mais uma cultivar de tomate (Ângela Gigante I-5100), usada como testemunha padrão). Foram usadas cinco repetições e oito plantas em cada subparcela. A inoculação foi feita na concentração de 60 ovos/mL de substrato à base de vermiculita e casca de Pinus sp. (50%) e casca de arroz carbonizada (50%). Sessenta dias após a inoculação, procedeu-se às avaliações. Todas as cultivares e linhagens-padrão (Linha 004 e Linha 006) mostraram-se suscetíveis às raças 1, 2, 3 e 4 de M. incognita. Todos os genótipos de pimentão foram resistentes a M. javanica. Todas as linhagens experimentais mostraram-se resistentes às quatro raças de M. incognita; o mesmo ocorreu com a maioria dos híbridos F1 experimentais, apesar de o grau de resistência dos híbridos F1, em geral, ter sido inferior ao das respectivas linhagens. Os resultados indicaram que é viável a utilização de híbridos F1 entre linhagens resistentes vs. linhagens suscetíveis para fins de controle dos nematóides M. incognita e M. javanica, via resistência varietal.

Sessenta amostras de milho pós-colheita foram avaliadas quanto à contaminação fúngica endógena e o potencial toxígeno de espécies do gênero Aspergillus e seus teleomorfos. Quarenta grãos aparentemente sadios de cada amostra foram desinfestados em NaClO e incubados em câmara úmida a 25±1ºC para exteriorização dos fungos, que posteriormente foram isolados em ágar Czapek-Dox. Foram identificadas as espécies Aspergillus flavus, A. parasiticus, Eurotium amstelodami e E. chevalieri. O potencial toxígeno dos fungos A. flavus e A. parasiticus foi avaliado quanto à síntese de aflatoxinas em meio ágar-coco. Espécies do gênero Eurotium foram avaliadas quanto à síntese de esterigmatocistina, nos meios ágar-amendoim e trigo triturado. A porcentagem de grãos contaminados variou entre 0 e 100%, prevalecendo os gêneros Aspergillus, Penicillium e Fusarium. A espécie predominante foi a A. flavus (64%), seguida por E. amstelodami (19%), E. chevalieri (10%) e A. parasiticus (7%). A partir de 109 isolados de A. flavus, evidenciou-se que 73 isolados sintetizaram aflatoxinas B1 e B2, 20 sintetizaram B1, sete sintetizaram B1 e G1, três sintetizaram B1, B2 e G1 e em seis não foi detectada a síntese de aflatoxina. A síntese de esterigmatocistina pelas espécies E. amstelodami e E. chevalieri não foi detectada.
