The market for table grapes is moving into mass production of specialty seed-less grapes in covered areas, aiming at obtaining premium prices with early or late production of high quality products. Production of quality seedless grapes is not straightforward since it is requires the correct combination of various independent characteristics, such as color, sugars, size and quantity at the right moment for successful harvesting and marketing. The present study was carried out at the two largest Portuguese producers located in Alentejo, and has the objective of studying the effect of irrigation management strategies and two different soils on the various relevant parameters for successful production and marketing. The management strategies were the application of ten day stress at the end of the cycle, in order to promote early maturing of the grapes. Three different timings of the stress were applied. Soil moisture, sap flow, bark thickness, as well as leaf water potential, stomatal conductance and chlorophyll content were measured regularly during the production season. The results indicate that the roots explore a rather large soil volume and the plants can successfully withstand reasonable periods of drought without significant changes to the plant physiology. Additionally late rains can mask the effect of any farmer applied drought and invalidate any farmer induced stress to the plants. Water-logged soils tend to cause early onset of maturity, but cause the ripening stage to extend over a longer period of time, and thus, in effect result in a delay in the harvest date. Topography also has some effect on the ripening, since hot air tends to accumulate under the plastic at the higher areas of the field. Em Portugal, as indústrias corticeira e de pasta de papel constituem um importante sector económico, contudo, gerando elevadas quantidades de subprodutos. Estes subprodutos poderiam ser explorados em aplicações de alto valor acrescentado, como fonte de compostos fenólicos, por exemplo, em vez de serem apenas queimados para produção de energia. Estes compostos são conhecidos pelas suas inúmeras propriedades, entre as quais, antioxidante, anti-inflamatória e anti-trombótica. Neste estudo as frações fenólicas da maior parte dos subprodutos gerados nas indústrias corticeira e de pasta de papel foram caracterizados em detalhe, com vista à sua valorização. A fração fenólica das cascas de Eucalyptus globulus, E. grandis, E. urograndis e E. maidenii, bem como da cortiça de Quercus suber e resíduos provenientes da sua exploração, nomeadamente, o pó de cortiça e os condensados negros, foi obtida por processos convencionais de extração sólido-líquido. No caso da casca de E. globulus, foi ainda avaliado o potencial de metodologias “verdes” no processo de extração de compostos fenólicos, usando extração com CO2 supercrítico. Esta técnica foi otimizada com recurso a metodologias de superfície de resposta. Na identificação e quantificação dos compostos fenólicos foi usada cromatografia líquida de alta resolução aliada a técnicas de espectrometria de massa. O teor de fenólicos totais foi ainda determinado pelo método de Folin- Ciocalteu, essencialmente para efeitos comparativos. A caracterização da fração fenólica de cada extrato foi ainda complementada com a análise da atividade antioxidante, usando o radical 2,2-difenil-1-picrilhidrazilo (DPPH). Foram identificados trinta compostos fenólicos na casca de E. globulus, 17 deles referenciados pela primeira vez como seus constituintes, nomeadamente os ácidos quínico, di-hidroxifenilacétic, cafeico e metil-elágico, bis-hexahidroxidifenoil( HHDP)-glucose, galoil- bis-HHDP-glucose, galoil-HHDPglucose, isoramnetina—hexosídeo, quercetina-hexosídeo, ácido metil-elágicopentosídeo, miricetina-ramnosídeo, isoramnetina-ramnosídeo, mearnsetina, floridzina, mearnsetina-hexosídeo, luteolina e uma proantocianidina B. Neste trabalho, foi estudada pela primeira vez a composição fenólica das cascas de E. grandis, E. urograndis e E. maidenii. Treze, doze e vinte e quatro compostos fenólicos foram identificados nas cascas de E. grandis, E. urograndis e E. maidenii, respetivamente. Entre estes compostos encontram-se os ácidos quínico, gálico, metilgálico, protocatequínico, clorogénico e elágico, catequina, galoil-bis-HHDP-glucose, digaloilglucose, epicatequina, quercetina-glucoronídeo, di-hidroxiisopropilcromona- hexosídeo, isoramnetina-hexosídeo, ácido elágicoramnosídeo, taxifolina, quercetina-hexosídeo, di-hidroxi- (metilpropil)isopropilcromona-hexosídeo, ácido metil-elágico-pentosídeo, miricetina-ramnosídeo, isoramnetina-ramnosídeo, aromadendrina-ramnosídeo, mearnsetina, mearnsetina-hexosídeo, eriodictiol, quercetina, isoramnetina e naringenina. A análise da fração fenólica da cortiça permitiu identificar vinte e dois compostos fenólicos, dez deles referenciados pela primeira vez como seus constituintes, nomeadamente, os ácidos quínico, salicílico, p-hidroxifenillático e metilgálico, ácido carboxílico da brevifolina, eriodictiol, naringenina, um éster isoprenílico do ácido cafeico, isoramnetina-ramnosídeo e isoramnetina. No pó de cortiça industrial foram identificados dezasseis compostos fenólicos, nomeadamente os ácidos quínico, gálico, protocatequínico, cafeico, ferúlico, elágico e metilgálico, esculetina, ácido carboxílico da brevifolina, coniferaldeído, um éster isoprenílico do ácido cafeico, uma dilactona do ácido valoneico, ácido elágico-pentosídeo, ácido elágico-ramnosídeo, isoramnetinaramnosídeo e isoramnetina. Destes, apenas o ácido elágico foi previamente referenciado como componente do pó de cortiça. Do mesmo modo, treze compostos fenólicos foram identificados no condensado negro, doze deles referenciados pela primeira vez como seus constituintes. São eles os ácidos quínico, gálico, p-hidroxifenil-láctico, protocatequínico, p-coumarico, cafeico e elágico, vanilina, esculetina, coniferaldeído, um éster isoprenílico do ácido cafeico e o eriodictiol. A extração supercrítica de compostos fenólicos da casca de eucalipto permitiu não só verificar os parâmetros que afetam a qualidade e quantidade finais dos extratos, como também obter os valores ótimos para estes parâmetros. Esta extração mostrou ainda ser bastante seletiva para determinados grupos de compostos fenólicos, como as flavanonas eriodictiol e naringenina e para o flavonol O-metilado isoramnetina. Este é também o primeiro estudo envolvendo a determinação da atividade antioxidante de extratos da cortiça e dos resíduos da sua exploração, bem como da casca de E. grandis, E. urograndis e E. maidenii. A vasta gama de compostos fenólicos identificados em cada extrato analisado, assim como as prominentes atividades antioxidantes, todas na mesma gama de valores do bem conhecido antioxidante comercial, ácido ascórbico, são claramente um grande contributo para a valorização destes subprodutos industriais.

Biodiversity continues to decline in the face of increasing anthropogenic pressures such as habitat destruction, exploitation, pollution and introduction of alien species. Existing global databases of species’ threat status or population time series are dominated by charismatic species. The collation of datasets with broad taxonomic and biogeographic extents, and that support computation of a range of biodiversity indicators, is necessary to enable better understanding of historical declines and to project – and avert – future declines. We describe and assess a new database of more than 1.6 million samples from 78 countries representing over 28,000 species, collated from existing spatial comparisons of local-scale biodiversity exposed to different intensities and types of anthropogenic pressures, from terrestrial sites around the world. The database contains measurements taken in 208 (of 814) ecoregions, 13 (of 14) biomes, 25 (of 35) biodiversity hotspots and 16 (of 17) megadiverse countries. The database contains more than 1% of the total number of all species described, and more than 1% of the described species within many taxonomic groups – including flowering plants, gymnosperms, birds, mammals, reptiles, amphibians, beetles, lepidopterans and hymenopterans. The dataset, which is still being added to, is therefore already considerably larger and more representative than those used by previous quantitative models of biodiversity trends and responses. The database is being assembled as part of the PREDICTS project (Projecting Responses of Ecological Diversity In Changing Terrestrial Systems – www.predicts.org.uk). We make site-level summary data available alongside this article. The full database will be publicly available in 2015.

O objectivo deste trabalho foi estudar a utilização de 5 compostos de resíduos de origem vegetal, como componentes de substratos hortícolas. Estudaram-se os processos de compostagem de bagaço de azeitona, bagaço de uva, polpa de alfarroba, casca de eucalipto e casca de pinheiro, caracterizaram-se os compostos obtidos e testou-se a sua aptidão agronómica, como substratos para viveiro de tomate (Lycopersicum esculentum Mill.) em placas alveoladas e cultura de pelargónio (Pelargonium x hortorum) em vaso. Após uma preparação prévia, que em alguns dos resíduos vegetais constou de moenda, suplemento de azoto e aumento do teor de humidade, efectuou-se a sua compostagem em caixas com 1200 L de capacidade, isoladas termicamente, com arejamento por volteio manual. Depois foram analisadas as suas características físicas e químicas e testados na sua aptidão como constituintes únicos de substratos e em misturas com turfa de sphagnum. A maioria dos processos de compostagem apresentou uma evolução habitual da temperatura, com a máxima aproximando-se ou ultrapassando 60 oC. Como excepções, verificou-se um período de compostagem muito longo em bagaço de azeitona; o atraso da fase termofílica em polpa de alfarroba e, na 1ª compostagem de casca de pinheiro, a temperatura não alcançou a zona termofílica. A maioria dos resíduos manifestaram uma acentuada resistência à degradação durante a compostagem, à excepção da polpa de alfarroba e da casca de eucalipto. Contudo, a nível físico-químico e químico registaram-se evoluções acentuadas, sobretudo nas variáveis relacionadas com alterações da superfície das partículas, como por exemplo, a capacidade de troca catiónica. Outras variáveis apresentaram uma evolução interessante para o acompanhamento do processo e o conhecimento do grau de estabilidade do composto final. Foi apreciável o efeito proporcionado pela adição de lamas de estação de tratamento de águas residuais urbanas, como suplemento azotado, a nível das propriedades físicas dos compostos, do aumento da velocidade do processo de compostagem e do enriquecimento dos compostos em macro e micronutrientes. A compostagem não conduziu à melhoria significativa das propriedades físicas dos materiais, excepto em casca de eucalipto. Os compostos caracterizaram-se fundamentalmente por uma reduzida capacidade de retenção de água e uma elevada capacidade de arejamento. Os compostos obtidos com incorporação de lamas apresentaram os valores mais baixos de capacidade de arejamento, mesmo assim dentro dos limites satisfatórios e, os valores mais altos de capacidade de retenção de água. Na cultura em placas alveoladas, o comportamento de muitas das misturas com compostos não se diferenciou do da testemunha, observando-se uma flutuação sazonal do comportamento das misturas nos dois viveiros de cada ano, associada às condições ambientais mais exigentes do ponto de vista da disponibilidade de água. Observou-se um maior número de correlações significativas entre as características dos substratos e as variáveis relativas ao crescimento das plantas nos viveiros efectuados mais tarde, sob condições de maior evapotranspiração. As plantas obtidas no segundo viveiro de tomate foram instaladas em cultura. As características das plantas do viveiro, apresentaram elevada correlação com as respectivas produções obtidas no campo, evidenciando a importância da qualidade das plantas de viveiro, na produção. Nos ensaios de pelargónio em vaso, também se observaram comportamentos idênticos ao da testemunha em muitas das misturas com compostos. Observou-se no 2º ensaio, um maior número de tratamentos com resultados idênticos aos da testemunha, resultante das condições de menor exigência hídrica, as quais fizeram sobressair menos as diferenças de propriedades físicas das misturas. Contudo, a diferença de crescimento obtido em algumas misturas, atribuiu-se também à interferência da continuidade capilar entre o vaso de papel com a estaca enraizada e a mistura em estudo. O trabalho presente mostrou que é possível obter compostos a partir de resíduos orgânicos, com boas características e a preço competitivo para utilização como substratos hortícolas. O processo de compostagem sendo um elemento chave, é fácil de controlar se houver uma regulação eficaz do arejamento e do teor de humidade durante o processo. Apesar de alguns resíduos, como a polpa de alfarroba, serem caros, se tomarmos em consideração o custo da sua eliminação, poderão conduzir a substratos comercialmente viáveis. Em condições ambientais que favoreçam uma maior evapotranspiração ou, em culturas com maior exigência hídrica, pode ser aconselhável a mistura dos compostos estudados com turfa de sphagnum pouco decomposta para melhorar a capacidade de retenção de água.

An efficient way of synthesizing the deuterium labelled analogues of three methoxypyrazine compounds: 2-d3-methoxy-3-isopropylpyrazine, 2-d3-methoxy-3- isobutylpyrazine, and 2-d3-methoxy-3-secbutylpyrazine, has been developed. To confirm that the deuterium labels had been incorporated into the expected positions in the molecules synthesized, the relevant characterization by NMR, HRMS and GC/MS analysis was conducted. Another part of this work involved quantitative determination of methoxypyrazines in water and wines. Solid-phase extraction (SPE) proved to be a suitable means for the sample separation and concentration prior to GC/MS analysis.Such factors as the presence of ethanol, salt, and acid have been investigated which can influence the recovery by SPE for the pyrazines from the water matrix. Significantly, in this work comparatively simple fractional distillation was attempted to replace the conventional steam distillation for pre-concentrating a sample with a relatively large volume prior to SPE. Finally, a real wine sample spiked with the relevant isotope-labelled methoxypyrazines was quantitatively analyzed, revealing that the wine with 10 beetles per litre contained 138 ppt of 2-methoxy-3-isopropylpyrazine. Interestingly, we have also found that 2-methoxy-3-secbutylpyrazine exhibits an extremely low detection limit in GC/MS analysis compared with the detection limit of the other two methoxypyrazines: 2- methoxy-3-isopropylpyrazine and 2-methoxy-3-isobutylpyrazine.

The Impact of the Multicolor Asian Lady Beetle (Harmonia axyridis) on Niagara Wine Quality The possible influence of Harmonia axyridis (the Multicolored Asian Lady Beetle) on the sensory properties of wine was investigated. H. axyridis beetles were added to white and red grape musts at a rate of 0, 1 or 10 per L, and a trained panel evaluated the finished wines using flavor-profiling techniques. Significant modification of both wine aroma and flavor characteristics were observed in the 10 beetlelL treatments, with smaller effects noted at the 1 beetlelL rate. Vinification in the presence of H. axyridis gave higher intensity scores for peanut, bell pepper and asparagus aromas and flavors in the white wines, and peanut, asparagus/bell pepper, and earthy/herbaceous aromas and flavors in the red wines. In addition, sweet, acid and bitter tastes were affected in red wines, and a general trend of decreasing fruit and floral intensities with increasing beetle rate was observed in both white and red wines. 15 ngIL Isopropylmethoxypyrazine was added to control wines and sensory profiles similar to high beetle treatments were obtained, supporting the hypothesis that methoxypyrazines from beetles are implicated in the taint. A trained panel evaluated the treated wines after 10 months of aging using the same sensory methods described above. Sensory profiles were very similar. Fennenting in the presence of Harmonia Axyridis (HA) had little influence on the chemical composition of the ftnished wine. The notable exception IS Isopropylmethoxypyrazine content, which was assessed usmg GC-MS analysis and showed increased concentration with increasing beetle nwnber for both white and red wmes. The influence of potential remedial treatments on the sensory properties of white and red wines tainted by Harmonia axyridis were also investigated. Bentonite, activated charcoal, oak chips, de-odorized oak chips, and UV or light irradiation were applied to tainted wine, and these wines evaluated chemically and sensorially. Both white and red wines treated with oak chips had strong oak characteristics, which masked the Harmonia axyridis-associated aroma and flavour attributes. In red wine, asparagus/bell pepper characteristics were decreased by bentonite and charcoal treatments. Only activated charcoal significantly decreased methoxypyrazine levels and only in white wine.

The Ontario Tender Fruit Marketing Board operates under the Farm Producers Marketing Act. It covers all tender fruit farmers who produce either fresh or canned products. Today the board has over 500 grower-members. Tender fruit in the Niagara region includes: peaches, pears, plums, grapes and cherries. The fruits are used in a number of different ways, from jams and jellies to desserts, sauces and wine. Peaches were first harvested along the Niagara river in 1779. Peter Secord (Laura Secord’s uncle) is thought to be the first farmer to plant fruit trees when he took a land grant near Niagara in the mid 1780s. Since the beginnings of Secord’s farm, peaches, pears and plums have been grown in the Niagara region ever since. However, none of the original varities of peach trees remain today. Peaches were often used for more than eating by early settlers. The leaves and bark of the tree was used to make teas for conditions such as chronic bronchitis, coughs and gastritis. Cherries have been known to have anti-inflammatory and pain relieving properties. Like peaches and cherries, pears had many uses for the early pioneers. The wood was used to make furniture. The juice made excellent ciders and the leaves provided yellow dyes. Plums have been around for centuries, not only in the Niagara region, but throughout the world. They have appeared in pre-historic writings and were present for the first Thanksgiving in 1621. The grape industry in Ontario has also been around for centuries. It began in 1798 when land was granted to Major David Secord (brother-in-law to Laura Secord) slightly east of St. David’s, on what is Highway No. 8 today. Major Secord’s son James was given a part of the land in 1818 and in 1857 passed it onto Porter Adams. Adams is known to be the first person to plant grapes in Ontario1. Tender fruits are best grown in warm temperate climates. The Niagara fruit belt, stretching 65km from Hamilton to Niagara on the Lake, provides the climate necessary for this fruit production. This belt produces 90% of Ontario’s annual tender fruit crop. It is one of the largest fruit producing regions in all of Canada.

Multicoloured Asian Lady Beetles (MALB) and 7-spot Lady Beetles that infect vineyards can secrete alkyl-methoxypyrazines when they are processed with the grapes, resulting in wines containing a taint. The main methoxypyrazine associated with this taint is 3-isopropyl-2-methoxypyrazine (IPMP). The wines are described as having aroma and flavours of peanut butter, peanut shells, asparagus and earthy which collectively, have become known as “ladybug taint”. To date, there are no known fining agents used commercially added to juice or wine that are effective in removing this taint. The goal of this project was to use previously identified proteins with an ability to bind to methoxypyrazines at low pH, and subsequently develop a binding assay to test the ability of these proteins to bind to and remove methoxypyrazines from grape juice. The piglet odorant binding protein (plOBP) and mouse major urinary protein (mMUP) were identified, cloned and expressed in the Pichia pastoris expression system. Protein expression was induced using methanol and the proteins were subsequently purified from the induction media using anion exchange chromatography. The purified proteins were freeze-dried and rehydrated prior to use in the methoxypyrazine removal assay. The expression and purification system resulted in yields of approximately 78% of purified plOBP and 62% of purified mMUP from expression to rehydration. Purified protein values were 87 mg of purified plOPB per litre of induction media and 19 mg of purified mMUP per litre of induction medium. In order to test the ability of the protein to bind to the MPs, an MP removal assay was developed. In the assay, the purified protein is incubated with either IPMP or 3-isobutyl-2-methoxypyrazine (IBMP) for two hours in either buffer or grape juice. Bentonite is then used to capture the protein-MP complex and the bentonite-protein-MP complex is then removed from solution by filtration. Residual MP is measured in solution following the MP removal assay and compared to that in the starting solution by Gas Chromatography Mass Spectrometry (GC/MS). GC/MS results indicated that the mMUP was capable of removing IBMP and IPMP from 300 ng/L in buffer pH 4.0, buffer pH 3.5 and Riesling Juice pH 3.5 down to the limit of quantification of the instrument, which is 6ng/L and 2ng/L for IBMP and IPMP, respectively. The results for the plOBP showed that although it could remove some IBMP, it was only approximately 50-70 ng/L more than bentonite treatment followed by filtration, resulting in approximately 100 ng/L of the MPs being left in solution. pIOBP was not able to remove IPMP in buffer pH 3.5 using this system above that removed by bentonite alone. As well, the pIOBP was not able to remove any additional MPs from Chardonnay juice pH 3.5 above that already removed by the bentonite and filtration alone. The mouse MUP was shown to be a better candidate protein for removal of MPs from juice using this system.

Methoxypyrazines are aroma active compounds found in many wine varietals. These compounds can be of either grape-derived nature or can be introduced into wines via Coccinellidae beetles. Regardless of their origin, methoxypyrazines can have either a beneficial role for wine quality, contributing to the specificity of certain wine varietals (Cabernet sauvignon, Cabernet franc, Sauvignon blanc) or a detrimental role, particularly at higher concentrations, resulting in overpowering green, unripe and herbaceous notes. When methoxypyrazines of exogenous nature are responsible for these unpleasant characteristics, wines are considered to be affected by what is generally known as Ladybug taint (LBT). This is work is a collection of studies seeking to create a sensitive analytical method for the detection and quantification of methoxypyrazines in wines; to investigate the role of different Coccinellidae species in the tainting of wines with LBT and identify the main compounds in ladybug tainted wines responsible for the typical green herbaceous characteristics; to determine the human detection threshold of 2,5-dimethyl-3-methoxypyrazine in wines as well as investigate its contribution to the aroma of wines; and finally to survey methoxypyrazine concentrations in a large set of wines from around the world. In the first study, an analytical method for the detection and quantitation of methoxypyrazines in wines was created and validated. The method employs multidimensional Gas Chromatography coupled with Mass Spectrometry to detect four different methoxypyrazines (2,5-dimethyl-3-methoxypyrazine, isobutyl methoxypyrazine, secbutyl methoxypyrazine and isopropyl methoxypyrazines) in wine. The low limits of detection for the compounds of interest, improved separation and isolation capabilities, good validation data, as well as the ease of use recommend this method as a good alternative to the existing analytical methods for methoxypyrazine detection in wine. In the second study the capacity of two Coccinellidae species, found in many wine regions – Harmonia axyridis and Coccinella septempunctata - to taint wines is evaluated. Coccinella septempunctata is shown to be as capable as causing LBT in wines as Harmonia axyridis. Dimethyl methoxypyrazine, previously thought to be of exogenous nature only (from Coccinellidae haemolymph), is also detected in control (untainted) wines. The main odor active compounds in LBT wines are investigated through Aroma Extract Dilution Assay. These compounds are identified as isopropyl methoxypyrazine, sec- and iso- butyl methoxypyrazine. In the third study, the human detection threshold for dimethyl methoxypyrazine in wine is established to be 31 ng/L in the orthonasal modality and 70 ng/L retronasally. After wines spiked with various amounts of dimethyl methoxypyrazine are evaluated sensorally, dimethyl methoxypyrazine causes significant detrimental effects to wine aroma at a concentration of 120 ng/L. The final study examines methoxypyrazine (dimethyl methoxypyrazine, isopropyl methoxypyrazine, secbutyl methoxypyrazine and isobutyl methoxypyrazine) concentrations in 187 wines from around the world. Dimethyl methoxypyrazine is detected in the majority of the red wines tested. Data are interpreted through statistical analyses. A new measure for predicting greenness/herbaceousness in wines - methoxypyrazine “total impact factor” is proposed.

Au Canada, nous remarquons une prédominance du diabète de type 2 au sein des communautés autochtones. Une approche ethnobotanique est utilisée en collaboration avec la Nation Crie de Eeyou Istchee afin de déterminer quels traitements à base de plantes peuvent être utilisés pour contrer les différentes conditions qui, collectivement, forment le diabète. Les pharmacopées de deux communautés cries, soit celles de Waskaganish et de Nemaska, ont été établies puis comparées à celles de étudiées antérieurement : communautés Whapmagoostui et Mistissini. Malgré les différences géographiques de ces groupes, leurs utilisations sont majoritairement semblables, avec pour seule exception le contraste entre les communautés de Nemaska et de Whapmagoostui. De plus, nous avons complété l’évaluation du taux cytoprotecteur des aiguilles, de l’écorce et des cônes de l’épinette noire (Picea mariana). Les extraits provenant de tous les organes des plantes démontrent une protection qui dépend de la concentration. La réponse spécifique d’organes peut varier selon l’habitat; ainsi, les plantes poussant dans les tourbières ou dans les forêts, sur le littoral ou à des terres l’intérieur démontrent des différences quant à leur efficacité. Bref, l’écorce démontre une relation dose-effet plus forte dans la forêt littorale, tandis que les aiguilles n’indiquent pas de changements significatifs selon leur environnement de croissance. La bioactivité observée démontre une corrélation avec le contenu phénolique et non avec l’activité de l’agent antioxydant. Ces résultats contribuent à péciser les activités antidiabétiques des plantes de la forêt boréale canadienne, telles qu’identifiées au niveau cellulaire par les guérisseurs Cries.
Resumo:
Nous avons utilisé une approche ethnobotanique pour identifier des espèces de plantes utilisées par les Cris afin de traiter les symptômes du diabète de type 2. Larix laricina du Roi (L. laricina) a récemment été identifiée comme une des meilleures plantes qui a stimulé le transport de glucose dans les cellules C2C12 et fortement potentialisé la différenciation des 3T3-L1 en indiquant une sensibilité potentiellement accrue à l’insuline. Ensuite, ces études de criblage ont été effectuées sur des extraits éthanolique (EE) en utilisant une série de bioessais in vitro. Cependant, les préparations traditionnelles des plantes sont souvent faites avec l’eau chaude. Le but de cette thèse de doctorat était d’isoler les principes actifs de L. laricina par un fractionnement guidé par l’adipogenèse; d’évaluer et de comparer l’activité et les mécanismes antidiabétiques des EE et des extraits aqueux (HWE) de ces 17 plantes. Pour le fractionnement de L. laricina, on a isolé plusieurs composés connus et identifié un nouveau composé actif cycloartane triterpene, qui a amélioré fortement l’adipogenèse et a été responsable en partie de l’activité adipogénique (potentiellement similaire à l’effet sensibilisateur à l’insuline des glitazone) de l’extrait éthanolique issu de l’écorce de L. laricina. Pour le métabolisme lipidique, nos résultats ont confirmé que 10 parmi les 17 EE ont augmenté la différenciation des adipocytes alors que 2 extraits seulement l’ont inhibée. Les HWE ont montré une faible activité adipogénique ou antiadipogénique. Les EE de R. groenlandicum et K. angustifolia ont le PPAR γ (peroxisome proliferator-activated receptor γ), le SREBP-1 (sterol regulatory element binding protein-1) et le C/EBP (CCAAT-enhancer binding proteins) α, alors que ceux de P. balsamifera et A. incana les ont inhibés. L’effet inhibiteur de P. balsamifera a également été prouvé d’avoir impliqué l’activation de la protéine kinase activée par l’AMP (AMPK). Les EE et HWE de R. groenlandicum ont stimulé les mêmes facteurs de transcription alors que les extraits aqueux d’autres plantes sélectionnées ont perdu ces effets en comparaison avec leurs extraits éthanoliques respectifs. L’analyse phytochimique a également identifié le groupe des espèces actives et inactives, notamment lorsque les espèces ont été séparées par famille de plante. Finalement concernant l’homéostasie de glucose, nos résultats ont confirmé que plusieurs EE ont stimulé le transport de glucose musculaire et inhibé l’activité de la glucose-6-phosphatase (G6Pase) hépatique. Certains des HWE ont partiellement ou complètement perdu ces activités antidiabétiques par rapport aux EE, tandis qu’une seule plante (R.groenlandicum) a juste conservé un potentiel similaire entre les EE et HWE dans les deux essais. Dans les cellules musculaires, les EE de R.groenlandicum, A. incana et S. purpurea ont stimulé le transport de glucose en activant la voie de signalisation de l’AMPK et en augmentant le niveau d’expression des GLUT4. En comparaison avec les EE, les HWE de R.groenlandicum ont montré des activités similaires; les HWE de A. incana ont complètement perdu leur effet sur tous les paramètres étudiés; les HWE de S. purpurea ont activé la voie de l’insuline au lieu de celle de l’AMPK pour augmenter le transport de glucose. Dans les cellules H4IIE, les EE et HWE des 5 plantes ont activé la voie de l’AMPK, et en plus les EE et HWE de 2 plantes ont activé la voie de l’insuline. La quercétine-3-O-galactoside et la quercétine 3-O-α-L-arabinopyranoside ont été identifiées comme des composés ayant un fort potentiel antidiabétique et donc responsables de l'activité biologique des plantes HWE actifs avec le transport du glucose. En conclusion, on a isolé plusieurs composés connus et identifié un nouveau triterpène actif à partir du fractionnement de L. laricina. Nous avons fourni également une preuve directe pour l'évaluation et la comparaison d'une action analogue à l'insuline ou insulino-sensibilisateur des EE et HWE de plantes médicinales Cris au niveau de muscle, de foie et de tissus adipeux. Une partie de leur action peut être liée à la stimulation des voies de signalisation intracellulaire insulino-dépendante et non-insulino-dépendante, ainsi que l’activation de PPARγ. Nos résultats indiquent que les espèces de plantes, les tissus ou les cellules cibles, ainsi que les méthodes d'extraction sont tous des déterminants significatifs de l'activité biologique de plantes médicinales Cris sur le métabolisme glucidique et lipidique.
