Harmonia axyridis was detected for the first time in Brazil in 2002. Since then, it has been recorded from the South to the Midwest of the country. Until now, almost all the registered cases were associated with the presence of aphids, its preferred source of food. Due to the occurrence of this species in areas of fruit growing in the South and Southeast of Brazil, the aim of this study was to analyze the preference and use of three different cultivars of fruit. The tests were set at 25ºC ± 1ºC, RH 70% ± 10%, and in a photophase of 12 h and with apples (Gala and Fuji), grapes (Niágara and Rubi), and pears (Williams and Asian). In the undamaged fruit experiment, the insects did not cause any noticeable damage. In the damaged and undamaged fruit experiment, a higher and statistically significant percentage of H. axyridis adults were found in the three damaged fruits. In the different cultivar experiment the Niágara grape, the Gala apple, and the Williams pear were significantly preferred by H. axyridis adults. These results may help in the management of this insect, preventing damage, which have been observed in other places where H. axyridis was introduced.

Visando a dar suporte aos trabalhos de melhoramento genético do marmeleiro (Cydoniaoblonga Mill.), voltados para a seleção de cultivares altamente produtivas, aptas a serem cultivadas em regiões subtropicais e produtoras de doces de qualidade superior, objetivou-se ajustar o meio de cultura básico para a germinação de grãos de pólen de diferentes cultivares dessa espécie e quantificar o número de estames, número de grãos de pólen por antera e por flor. O pólen utilizado foi da cultivar Portugal, obtido de anteras provenientes de flores em estádio de balão. Em seguida, com auxílio de um pincel, os grãos de pólen foram espalhados sobre a superfície de placas de Petri, contendo 20 mL de meio de cultura, de acordo com os seguintes experimentos: 1) concentrações de ágar (4; 6; 8 e 10 g L-1) e valores de pH (3,5; 4,5; 5,5 e 6,5); 2) concentrações de sacarose (0; 30; 60 e 90 g L-1); 3) concentrações de nitrato de cálcio (0; 200; 400 e 800 mg L-1); 4) concentrações de ácido bórico (0; 400; 800 e 1.200 mg L-1); e 5) tempo de emissão do tubo polínico (0; 1; 2; 3; 4; 5 e 6 horas após a inoculação), os quais foram montados de forma sequencial. Após, avaliou-se a taxa de germinação dos grãos de pólen das 27 cultivares (Alaranjado, Alongado, Apple, BA29, Bereckzy, Champion, Cheldow, Constantinopla, CTS 207, Dangers, De Patras, De Vranja, Dulot, Fuller, Mendoza INTA 37, Kiakami, Lajeado, Meeck Profilic, Meliforme, Pineapple, Portugal, Provence, Radaelli, Rea's Mamouth, Smyrna, Van Deman e Zuquerineta), além do número de estames, número de grãos de pólen por antera e por flor. Realizando-se as leituras da porcentagem de germinação após cinco horas de incubação, conclui-se que o meio de cultura para a germinação de grãos de pólen do marmeleiro deve ser acrescido de 68 g L-1 de sacarose e 366 mg L-1 de ácido bórico, sendo o pH aferido para 5,8 e o meio solidificado com 10 g L-1 de ágar. Foram constatadas diferenças entre as cultivares quanto à quantidade de grãos de pólen e à capacidade germinativa dos mesmos, que variou de 37,83% a 82,23% entre as cultivares. Os grãos de pólen da cultivar Alaranjado apresentaram maior porcentagem de germinação, além da maior quantidade de grãos de pólen por flor.

As anonáceas compõem um grupo de plantas que se têm destacado em várias partes do mundo, principalmente por produzirem frutos de grande interesse comercial. Entre os mais destacados membros desta família, estão a cherimólia (Annona cherimola), a pinha (A. squamosa), a atemoia (híbrido A. squamosa x A. cherimola), e a graviola (A. muricata). No Brasil, pelas suas características de clima tropical e com baixas altitudes na maior parte de seu território, a cherimólia tem sido pouco cultivada devido a sua exigência em temperaturas baixas. As demais apresentam áreas cultivadas em diversas regiões do País para fazer frente à demanda dos mercados de frutas frescas (pinha e atemoia) e processadas (graviola). Todavia, o interesse neste grupo de plantas é mais abrangente, tendo em vista que inúmeras pesquisas comprovam que compostos secundários obtidos de várias anonáceas possuem atividade pesticida e antitumoral. Este trabalho objetiva apresentar um quadro atual da produção de anonáceas no Brasil, focando nas principais anonas cultivadas nas diferentes regiões do País. Para tanto, foram levantados dados de produção e principais tecnologias utilizadas nestas culturas. Os dados de produção obtidos serviram para estabelecer uma estimativa aproximada da realidade da produção de atemoia, pinha e graviola no Brasil. Os resultados demonstram um vigoroso crescimento na área total de cultivo, na produção e no uso de tecnologias modernas para as três espécies consideradas neste estudo. Apontam também para uma expansão dos cultivos em todas as regiões do Brasil, principalmente no Nordeste.

La carpocapsa o corc de la poma (Cydia pomonella L.) és la plaga clau de lesplantacions comercials de pomera de la zona de Girona. Per controlar-la amb eficàcia,anualment són necessaris diferents tractaments insecticides que a més del cost econòmicque representen, afavoreixen fenòmens de resistències, aparició de plagues induïdes iproblemes de toxicitat, residus i contaminació ambiental.La combinació de diferents metodologies per combatre aquesta plaga, algunes menysagressives amb la fauna auxiliar i el medi ambient, s’ha convertit en una pràcticaimprescindible per aconseguir-ne un control efectiu i sostenible. La confusió sexual ésuna tècnica de lluita biotecnològica que per les seves característiques d’especificitat,toxicologia i respecte al medi ambient, representa una alternativa a la problemàticagenerada pels productes químics tradicionals.Fructícola Empordà S.L., empresa agroalimentària situada al municipi de Sant PerePescador (Alt Empordà, Girona) dedicada a la producció i comercialització de fruitafresca, com a entitat participant del projecte Àrea Pilot de Reducció d’Insecticides enplantacions comercials de pomera de Girona (APRI) emmarcat en el programad’actuacions específiques de les Associacions de Defensa dels Vegetals (ADV’s) de lazona, introdueix per primera vegada la tècnica de la confusió sexual a l’estratègia decontrol de carpocapsa en determinades parcel•les comercials on la lluita químicaexclusiva no és suficient (2003). Durant el període 2003-2011, l’ús de la confusió sexual per al control de carpocapsa esgeneralitza progressivament a les finques dels agricultors de Fructícola Empordà S.L.amb l’objectiu principal de reduir el percentatge de dany per atac de carpocapsa en elsfruits a collita i, conseqüentment, disminuir la pressió del corc de la poma a la zona. Atal efecte es va plantejar un estudi amb l’objectiu d’avaluar l’eficàcia de l’estratègia decontrol de la carpocapsa basada en la combinació del mètode de la confusió sexual ambaplicacions fitosanitàries de reforç durant el període d’expansió de la tècnica (2003-2011) en les plantacions comercials de pomera de Fructícola Empordà S.L. així com,fer una valoració d’aquest respecte al sistema de control paral•lelament utilitzat a lazona, la lluita química assessorada

Certain strains of Pantoea are used as biocontrol agents for the suppression of plant diseases. However, their commercial registration is hampered in some countries because of biosafety concerns. This study compares clinical and plant-beneficial strains of P. agglomerans and related species using a phenotypic analysis approach in which plant-beneficial effects, adverse effects in nematode models, and toxicity were evaluated. Plant-beneficial effects were determined as the inhibition of apple fruit infection by Penicillium expansum and apple flower infection by Erwinia amylovora. Clinical strains had no general inhibitory activity against infection by the fungal or bacterial plant pathogens, as only one clinical strain inhibited P. expansum and three inhibited E. amylovora. By contrast, all biocontrol strains showed activity against at least one of the phytopathogens, and three strains were active against both. The adverse effects in animals were evaluated in the plant-parasitic nematode Meloidogyne javanica and the bacterial-feeding nematode Caenorhabditis elegans. Both models indicated adverse effects of the two clinical strains but not of any of the plant-beneficial strains. Toxicity was evaluated by means of hemolytic activity in blood, and genotoxicity with the Ames test. None of the strains, whether clinical or plant-beneficial, showed any evidence of toxicity

The Potentiometric Stripping Analysis (PSA) is described with emphasis on ultramicroelectrode applications with a laboratory developed computer based instrumentation. The technique potentialities as compared with the voltammetric approach are pointed out based on the current literature. Some results of trace metals analysis including zinc, cadmium, lead and copper in vinegar and canned food samples are presented. The mainly advantage found in our laboratory was the technique capability to analyse natural samples with minimum matrix interference and the low level of noise found in our determinations.

Crude extract of Tibouchina granulosa, Rhododendron simsii and Phaseolus vulgaris L. were prepared and used as alternative indicators in quantitative analysis teaching in standardization of NaOH solutions and in the determination of acetic acid contents in vinegar. Effect of using such natural extracts as indicators was very attractive to the students and the quantitative results were compared with conventional indicators with good agreement. Concepts of data statistics can successfully be discussed using the interest revived by the use of natural indicators.

To improve tannin assay in cashew apple, several parameters were examined, including (1) extraction solvents, (2) effects of water and boiling time on butanol acid reaction and (3) correlation between vanillin and butanol acid assay of tannin in cashew apples. The 50-70% acetone extracted the greatest amount of tannin from cashew apples. Concentrations of water in butanol reagents were adjusted and boiling time of butanol reaction was reduced at 15 min. Tannin of unripe cashew apples was purified on Sephadex LH-20, aiming to obtain tannin standard for butanol assay. The vanillin assay presented high correlation with the butanol acid assay.

A rapid and sensitive method is described for the determination of clofentezine residues in apple, papaya, mango and orange. The procedure is based on the extraction of the sample with a hexane:ethyl acetate mixture (1:1, v/v) and liquid chromatographic analysis using UV detection. Mean recoveries from 4 replicates of fortified fruit samples ranged from 81% to 96%, with coefficients of variation from 8.9% to 12.5%. The detection and quantification limits of the method were of 0.05 and 0.1 mg kg-1, respectively.

A simple and didactic experiment was developed for image monitoring of the browning of fruit tissues caused by the enzyme polyphenol oxidase. The procedure, easy and inexpensive, is a valuable tool to teach and demonstrate the redox reaction between the enzyme and the natural polyphenols. To obtain the browning percentage for apple, pear and banana, digital photographs were employed, and the images were analyzed by means of Monte Carlo methods and digital analysis programs. The effects of several experimental conditions were studied, such as pH, light, temperature and the presence of oxygen or anti-oxidants. It was observed that each fruit presented a different condition that better minimized the oxidation process. The absence of oxygen and the application of a bissulphite solution were sufficient to keep the quality of all fruits tested.

A solid phase extraction procedure using Amberlite XAD-1180/Pyrocatechol violet (PV) chelating resin for the determination of iron and lead ions in various environmental samples was established. The procedure is based on the sorption of lead(II) and iron(III) ions onto the resin at pH 9, followed by elution with 1 mol/L HNO3 and determination by flame atomic absorption spectrometry. The influence of alkaline, earth alkaline and some transition metals, as interferents, are discussed. The recoveries for the spiked analytes were greater than 95%. The detection limits for lead and iron by FAAS were 0.37 µg/L and 0.20 µg/L, respectively. Validation of the method described here was performed by using three certified reference materials (SRM 1515 Apple Leaves, SRM 2711 Montana Soil and NRCC-SLRS-4 Riverine Water). The procedure was successfully applied to natural waters and human hair.

The main purpose of this work is to describe the use of the technique Site-Specific Natural Isotopic Fractionation of hydrogen (SNIF-NMR), using ²H and ¹H NMR spectroscopy, to investigate the biosynthetic origin of acetic acid in commercial samples of Brazilian vinegar. This method is based on the deuterium to hydrogen ratio at a specific position (methyl group) of acetic acid obtained by fermentation, through different biosynthetic mechanisms, which result in different isotopic ratios. We measured the isotopic ratio of vinegars obtained through C3, C4, and CAM biosynthetic mechanisms, blends of C3 and C4 (agrins) and synthetic acetic acid.

Pantoea agglomerans strains are among the most promising biocontrol agents for avariety of bacterial and fungal plant diseases, particularly fire blight of apple and pear. However, commercial registration of P. agglomerans biocontrol products is hampered because this species is currently listed as a biosafety level 2 (BL2) organism due to clinical reports as an opportunistichuman pathogen. This study compares plant-origin and clinical strains in a search for discriminating genotypic/phenotypic markers using multi-locus phylogenetic analysis and fluorescent amplified fragment length polymorphisms (fAFLP) fingerprinting.Results: Majority of the clinical isolates from culture collections were found to be improperly designated as P. agglomerans after sequence analysis. The frequent taxonomic rearrangements underwent by the Enterobacter agglomerans/Erwinia herbicola complex may be a major problem in assessing clinical associations within P. agglomerans. In the P. agglomerans sensu stricto (in the stricter sense) group, there was no discrete clustering of clinical/biocontrol strains and no marker was identified that was uniquely associated to clinical strains. A putative biocontrol-specific fAFLP marker was identified only in biocontrol strains. The partial ORF located in this band corresponded to an ABC transporter that was found in all P. agglomerans strains. Conclusion: Taxonomic mischaracterization was identified as a major problem with P.agglomerans, and current techniques removed a majority of clinical strains from this species. Although clear discrimination between P. agglomerans plant and clinical strains was not obtained with phylogenetic analysis, a single marker characteristic of biocontrol strains was identified whichmay be of use in strain biosafety determinations. In addition, the lack of Koch's postulate fulfilment, rare retention of clinical strains for subsequent confirmation, and the polymicrobial nature of P. agglomerans clinical reports should be considered in biosafety assessment of beneficial strains in this species

The production of cashew apple wine has the purpose of minimizing the wastage in the Brazilian cashew production. Knowing that the cashew apple fermentation produces a good cashew wine, a study of alcoholic fermentation kinetics of the cashew apple and the physico-chemical characterization of the product were made. The cashew wine was produced in an stirred batch reactor. The results of the physico-chemical analysis of volatiles, residual sugars, total acidity and pH of cashew wine showed that their concentrations were within the standard limits established by the Brazilian legislation for fruit wines.
