A safe, effective, and inexpensive vaccine against typhoid and other Salmonella diseases is urgently needed. In order to address this need, we are developing a novel vaccine platform employing buoyant, self-adjuvanting gas vesicle nanoparticles (GVNPs) from the halophilic archaeon Halobacterium sp. NRC-1, bioengineered to display highly conserved Salmonella enterica antigens. As the initial antigen for testing, we selected SopB, a secreted inosine phosphate effector protein injected by pathogenic S. enterica bacteria during infection into the host cells. Two highly conserved sopB gene segments near the 3'- region, named sopB4 and sopB5, were each fused to the grIpC gene, and resulting SopB-GVNPs were purified by centrifugally accelerated flotation. Display of SopB4 and SopB5 antigenic epitopes on GVNPs was established by Western blotting analysis using antisera raised against short synthetic peptides of SopB. Immunostimulatory activities of the SopB4 and B5 nanoparticles were tested by intraperitoneal administration of SopB-GVNPs to BALB/c mice which had been immunized with S. enterica serovar Typhimurium 14028 ApmrG-H111-D (DV-STM-07), a live attenuated vaccine strain. Proinflammatory cytokines IFN-y, IL-2, and IL-9 were significantly induced in mice boosted with SopB5-GVNPs, consistent with a robust Thl response. After challenge with virulent S. enterica serovar Typhimurium 14028, bacterial burden was found to be diminished in spleen of mice boosted with SopB4-GVNPs and absent or significantly diminished in liver, mesenteric lymph node, and spleen of mice boosted with SopB5GVNPs, indicating that the C-terminal portions of SopB displayed on GVNPs elicit a protective response to Salmonella infection in mice. SopB antigen-GVNPs were also found to be stable at elevated temperatures for extended periods without refrigeration. The results show that bioengineered GVNPs are likely to represent a valuable platform for antigen delivery and development of improved vaccines against Salmonella and other diseases.

A new monoclinic polymorph, form II (P2(1)/c, Z = 4), has been isolated for 3,4-dimethoxycinnamic acid (DMCA). Its solid-state 2 + 2 photoreaction to the corresponding alpha-truxillic acid is different from that of the first polymorph, the triclinic form I (P (1) over bar, Z = 4) that was reported in 1984. The crystal structures of the two forms are rather different. The two polymorphs also exhibit different photomechanical properties. Form I exhibits photosalient behavior but this effect is absent in form II. These properties can be explained on the basis of the crystal packing in the two forms. The nanoindentation technique is used to shed further insights into these structure-property relationships. A faster photoreaction in form I and a higher yield in form II are rationalized on the basis of the mechanical properties of the individual crystal forms. It is suggested that both Schmidt-type and Kaupp-type topochemistry are applicable for the solid-state trans-cinnamic acid photodimerization reaction. Form I of DMCA is more plastic and seems to react under Kaupp-type conditions with maximum molecular movements. Form II is more brittle, and its interlocked structure seems to favor Schmidt-type topochemistry with minimum molecular movement.

People living under $2 income per day, referred as Base of the Pyramid (BoP), face undesired situations like lack of nutrition, health, education etc. Design as a process of changing current undesired situation to a desired situation has failed. A crucial reason behind these failures is lack of normative basis to identify and understand the absent or unsatisfied stakeholder. Currently stakeholder analysis in the design is heuristic. This paper uses a normative framework of Capability Approach (CA) for the stakeholder analysis. A brief discussion on stakeholder theory and analysis is used to identify gaps in the literature. The constructs of the CA are discussed for its suitability to the purpose. Along with methodological details, data generated from the stakeholder interviews, focus groups in a case study of dissemination of improved cook-stoves is used to interlink the theory with the practice. The scope of this work is in identifying and investigating the motives of the stakeholders in the involvement in the product. Though a lot of insights to discern and manage crucial stakeholders is inbuilt in the methodology, this work does not claim explicit coverage of these aspects.

We use numerical dynamo models with heterogeneous core-mantle boundary (CMB) heat flux to show that lower mantle lateral thermal variability may help support a dynamo under weak thermal convection. In our reference models with homogeneous CMB heat flux, convection is either marginally supercritical or absent, always below the threshold for dynamo onset. We find that lateral CMB heat flux variations organize the flow in the core into patterns that favour the growth of an early magnetic field. Heat flux patterns symmetric about the equator produce non-reversing magnetic fields, whereas anti-symmetric patterns produce polarity reversals. Our results may explain the existence of the geodynamo prior to inner core nucleation under a tight energy budget. Furthermore, in order to sustain a strong geomagnetic field, the lower mantle thermal distribution was likely dominantly symmetric about the equator. (C) 2015 Elsevier B.V. All rights reserved.

Maximum, spreading of liquid drops impacting on solid surfaces textured with unidirectional parallel grooves is studied for drop Weber number in the range 1-100 focusing on the role of texture geometry and wettability. The maximum spread factor of impacting drops measured perpendicular to grooves; beta(m,perpendicular to) is seen to be less than, that:measured parallel to grooves, beta(m,perpendicular to).The difference between beta(m,perpendicular to), and beta(m,parallel to) increases with drop impact velocity. This deviation of beta(m,perpendicular to) from beta(m,parallel to) is analyzed by considering the possible mechanisms, correspond, ing to experimental observations (1) impregnation of drop into the grooves, (2) convex shape of liquid vapor interface near contact line at maximum spreading, and (3) contact line pinning of spreading drop at the pillar edges by incorporating them into an energy conservation-based model. The analysis reveals that contact line pinning offers a physically meaningful justification of the observed: deviation of beta(m,perpendicular to) from beta(m,parallel to) compared to other possible candidates. A unified model, incorporating all the above-mentioned mechanisms, is formulated, which predicts beta(m,perpendicular to) on several groove-textured surfaces made of intrinsically hydrophilic and hydrophobic materials with an average error of 8.3%. The effect of groove-texture geometrical parameters,on maximum drop spreading is explained using this unified model. A special case of the unified model, with contact line pinning, absent, predicts beta(m,parallel to) with an average error of 6.3%.

Collective cell migrations are essential in several physiological processes and are driven by both chemical and mechanical cues. The roles of substrate stiffness and confinement on collective migrations have been investigated in recent years, however few studies have addressed how geometric shapes influence collective cell migrations. Here, we address the hypothesis that the relative position of a cell within the confinement influences its motility. Monolayers of two types of epithelial cells-MCF7, a breast epithelial cancer cell line, and MDCK, a control epithelial cell line-were confined within circular, square, and cross-shaped stencils and their migration velocities were quantified upon release of the constraint using particle image velocimetry. The choice of stencil geometry allowed us to investigate individual cell motility within convex, straight and concave boundaries. Cells located in sharp, convex boundaries migrated at slower rates than those in concave or straight edges in both cell types. The overall cluster migration occurred in three phases: an initial linear increase with time, followed by a plateau region and a subsequent decrease in cluster speeds. An acto-myosin contractile ring, present in the MDCK but absent in MCF7 monolayer, was a prominent feature in the emergence of leader cells from the MDCK clusters which occurred every similar to 125 mu m from the vertex of the cross. Further, coordinated cell movements displayed vorticity patterns in MDCK which were absent in MCF7 clusters. We also used cytoskeletal inhibitors to show the importance of acto-myosin bounding cables in collective migrations through translation of local movements to create long range coordinated movements and the creation of leader cells within ensembles. To our knowledge, this is the first demonstration of how bounding shapes influence long-term migratory behaviours of epithelial cell monolayers. These results are important for tissue engineering and may also enhance our understanding of cell movements during developmental patterning and cancer metastasis.

The Vi capsular polysaccharide is a virulence-associated factor expressed by Salmonella enterica serotype Typhi but absent from virtually all other Salmonella serotypes. In order to study this determinant in vivo, we characterised a Vi-positive S. Typhimurium (C5.507 Vi(+)), harbouring the Salmonella pathogenicity island (SPI)-7, which encodes the Vi locus. S. Typhimurium C5.507 Vi(+) colonised and persisted in mice at similar levels compared to the parent strain, S. Typhimurium C5. However, the innate immune response to infection with C5.507 Vi(+) and SGB1, an isogenic derivative not expressing Vi, differed markedly. Infection with C5.507 Vi(+) resulted in a significant reduction in cellular trafficking of innate immune cells, including PMN and NK cells, compared to SGB1 Vi(-) infected animals. C5.507 Vi(+) infection stimulated reduced numbers of TNF-α, MIP-2 and perforin producing cells compared to SGB1 Vi(-). The modulating effect associated with Vi was not observed in MyD88(-/-) and was reduced in TLR4(-/-) mice. The presence of the Vi capsule also correlated with induction of the anti-inflammatory cytokine IL-10 in vivo, a factor that impacted on chemotaxis and the activation of immune cells in vitro.

By solving numerically the full set of hydrodynamic equations governing the pulsation of a bubble,we show that shock waves are often absent in a stable sonoluminescing bubble. Nevertheless, for a wide range of physical parameters, a continuous compressional wave emerges and heats up the bubble, and the resulting black-body radiations have pulse heights and widths that agree with experimental data. Shock waves, being much less robust, are not essential for stable single-bubble sonoluminescence.

The fracture toughness and interfacial adhesion properties of a coating on its substrate are considered to be crucial intrinsic parameters determining performance and reliability of coating-substrate system. In this work, the fracture toughness and interfacial shear strength of a hard and brittle Cr coating on a normal medium carbon steel substrate were investigated by means of a tensile test. The normal medium carbon steel substrate electroplated with a hard and brittle Cr coating was quasi-statically stretched to induce an array of parallel cracks in the coating. An optical microscope was used to observe the cracking of the coating and the interfacial decohesion between the coating and the substrate during the loading. It was found that the cracking of the coating initiated at critical strain, and then the number of the cracks of the coating per unit axial distance increased with the increase in the tensile strain. At another critical strain, the number of the cracks of the coating became saturated, i.e. the number of cracks per unit axial distance became a constant after this critical strain. Based on the experiment result, the fracture toughness of the brittle coating can be determined using a mechanical model. Interestingly, even when the whole specimen fractured completely under an extreme strain of the substrate, the interfacial decohesion or buckling of the coating on its substrate was completely absent. The test result is different from that appeared in the literature though the identical test method and the brittle coating/ductile metal substrate system are taken. It was found that this difference can be attributed to an important mechanism that the Cr coating on the steel substrate has a good adhesion, and the ultimate interfacial shear strength between the Cr coating and the steel substrate has exceeded the maximum shear flow strength level of the steel substrate. This result also indicates that the maximum shear flow strength level of the ductile steel substrate can be only taken as a lower bound estimate on the ultimate shear strength of the interface. This estimation of the ultimate interfacial shear strength is consistent with the theoretical analysis and prediction presented in the literature.

The Argentine Republic is situated in the southernmost portion of the American continent, occupying over 2,785,600 km2 not including the Antarctic territory. The country ranges from subtropical areas (21º46’S) to subantarctic regions (55º03’S), extending latitudinally over about 4,000 km. It possesses significant latitudinal and altitudinal variation (33º of latitudinal range, and heights from Bajo de San Julián in Santa Cruz province at 105 m below sea level, up to Mt. Aconcagua, 6,959 m over sea level), as well as two gradients of physical variability, extending in north-south and east-west directions. Owing to these features, the country presents a wide range of climates and soil types, being one of the countries with greatest diversity of biogeographical units (Lean et al., 1990, In: Bertonatti & Corcuera, 2000). There are four main hydrographic systems: Río de la Plata basin, the Atlantic and Pacific drainages, and several endorrheic systems. Within these basins, the ichthyofaunistic assemblage is well represented, with different magnitude in accordance with the different taxonomic groupings and regions considered. From an ichthyogeographic standpoint, and according to the works of Ringuelet (1975) and Arratia et al. (1983), Argentina is included in the Brasilic and Austral Subregions. The first of these is represented by two domains: the Andean Domain, comprising the southernmost portion of Titicaca Province, and the Paranensean Domain, including part of Alto Paraná and Paranoplatensean Provinces. The Austral Subregion is represented in Argentina by the Subandean-Cuyan and Patagonian Provinces. The present survey indicates that there are about 441 fish species in Argentina, distributed throughout the country; this number represents less than 10% of the total fish species occurring in the Neotropical Region. There is a recognizable trend of faunal impoverishment, both in North-South and East-West direction, reaching its maximum expression in the provinces of Tierra del Fuego (situated at approximately 52º30’S to 55ºS, and 65ºS to 68º50’W) and San Juan (approximately 28º50’S and 67ºW to 70º45’W), which have 4 and 5 fish species respectively. In north-south direction, one of the regional indicators of this phenomenon is the Salado river basin in Buenos Aires province, which constitutes the southern distributional boundary for the majority of the paranoplatensean ichthyofauna; 12 of the families occurring in the Paraná-Plata system are absent from this pauperized paranensean ichthyofaunal assemblage. Most of the continental fish fauna of Argentina belongs to the primary division of Myers (1949), while some elements are included in the secondary division and others in an amphibiotic or ‘marine penetration’ category. This ichthyofaunistic scope encompasses a wide range of morphological, biological, ecological and ethological types (benthic and pelagic, migrating and sedentary, haematophagous or parasites, annual species, inhabitants of plains or heights, estivation-adapted, etc.) inhabiting different regions within the national territory.

On 15-16 January 2005, three offshore species of cetaceans (33 short-finned pilot whales, Globicephala macrorhynchus, one minke whale, Balaenoptera acutorostrata, and two dwarf sperm whales, Kogia sima) stranded alive on the beaches of North Carolina. The pilot whales stranded near Oregon Inlet, the minke whale in northern North Carolina, and the dwarf sperm whales near Cape Hatteras. Live strandings of three species in one weekend was unique in North Carolina and qualified as an Unusual Mortality Event. Gross necropsies were conducted on 16-17 January 2005 on 27 pilot whales, two dwarf sperm whales, and the minke whale. Samples were collected for clinical pathology, parasitology, gross pathology, histopathology, microbiology and serology. There was variation in the number of animals sampled for each collection type, however, due to carcasses washing off the beach or degradation in carcass condition during the course of the response. Comprehensive histologic examination was conducted on 16 pilot whales, both dwarf sperm whales, and the minke whale. Limited organ or only head tissue suites were obtained from nine pilot whales. Histologic examination of tissues began in February 2005 and concluded in December 2005 when final sampling was concluded. Neither the pilot whales nor dwarf sperm whales were emaciated although none had recently ingested prey in their stomachs. The minke whale was emaciated; it was likely a dependent calf that became separated from the female. Most serum biochemistry abnormalities appear to have resulted from the stranding and indicated deteriorating condition from being on land for an extended period. Three pilot whales had clinical evidence of pre-existing systemic inflammation, which was supported by histopathologic findings. Although gross and histologic lesions involving all organ systems were noted, consistent lesions were not observed across species. Verminous pterygoid sinusitis and healed fishery interactions were seen in pilot whales but neither of these changes were causes of debilitation or death. In three pilot whales and one dwarf sperm whale there was evidence of clinically significant disease in postcranial tissues which led to chronic debilitation. Cardiovascular disease was present in one pilot whale and one dwarf sperm whale; musculoskeletal disease and intra-abdominal granulomas were present in two pilot whales. These lesions were possible, but not definitive, causal factors in the stranding. Remaining lesions were incidental or post-stranding. The minke whale and three of five tested pilot whales had positive morbillivirus titers (≥1:8 with one at >1:256), but there was no histologic evidence of active viral infection. Parasites (nematodes, cestodes, and trematodes) were collected from 26 pilot whales and two dwarf sperm whales. Sites of collection included stomach, nasal/pterygoid, peribullar sinuses, blubber, and abdominal cavity. Parasite species, locations and loads were within normal limits for free-ranging cetaceans and were not considered causative for the stranding event. Gas emboli lesions which were considered consistent with or diagnostic of sonarassociated strandings of beaked whales or small cetaceans were not found in the whales stranded as part of UMESE0501Sp. Twenty-five heads were examined with nine specific anatomic locations of interest: extramandibular fat, intramandibular fat, auditory meatus, peribullar acoustic fat, peribullar soft tissue, peribullar sinus, pterygoid sinus, melon, and brain. The common finding in all examined heads was verminous pterygoid sinusitis. Intramandibular adipose tissue reddening, typically adjacent to the vascular plexus, was observed in some individuals and could represent localized hemorrhage resulting from vascular rete rupture, hypostatic congestion, or erythrocyte rupture during the freeze/thaw cycle. One cetacean had peracute to acute subdural hemorrhage that likely occurred from thrashing on the beach post-stranding, although its occurrence prior to stranding cannot be excluded. Information provided to NMFS by the U.S. Navy indicated routine tactical mid-frequency sonar operations from individual surface vessels over relatively short durations and small spatial scales within the area and time period investigated. No marine mammals were detected by marine mammal observers on operational vessels; standard operating procedure for surface naval vessels operating mid-frequency sonar is the use of trained visual lookouts using high-powered binoculars. Sound propagation modeling using information provided to NMFS indicated that acoustic conditions in the vicinity likely depended heavily on position of the receivers (e.g., range, bearing, depth) relative to that of the sources. Absent explicit information on the location of animals meant that it was not possible to estimate received acoustic exposures from active sonar transmissions. Nonetheless, the event was associated in time and space with naval activity using mid-frequency active sonar. It also had a number of features in common (e.g., the “atypical” distribution of strandings involving multiple offshore species, all stranding alive, and without evidence of common infectious or other disease process) with other sonar-related cetacean mass stranding events. Given that this event was the only stranding of offshore species to occur within a 2-3 day period in the region on record (i.e., a very rare event), and given the occurrence of the event simultaneously in time and space with a naval exercise using active sonar, the association between the naval sonar activity and the location and timing of the event could be a causal rather than a coincidental relationship. However, evidence supporting a definitive association is lacking, and, in particular, there are differences in operational/environmental characteristics between this event and previous events where sonar has apparently played a role in marine mammal strandings. This does not preclude behavorial avoidance of noise exposure. No harmful algal blooms were present along the Atlantic coast south of the Chesapeake Bay during the months prior to the event. Environmental conditions, including strong winds, changes in upwelling- to downwelling-favorable conditions, and gently sloping bathymetry, were consistent with conditions which have been correlated with other mass strandings. In summary, we did not find commonality in gross and histologic lesions that would indicate a single cause for this stranding event. Three pilot whales and one dwarf sperm whale had debilitating conditions identified that could have contributed to stranding, one pilot whale had a debilitating condition (subdural hemorrhage) that could have been present prior to or resulting from stranding. While the pilot and dwarf sperm whale strandings may have had a common cause, the minke whale stranding was probably just coincidental. On the basis of examination of physical evidence in the affected whales, however, we cannot definitively conclude that there was or was not a causal link between anthropogenic sonar activity or environmental conditions (or a combination of these factors) and the strandings. The three areas in Rookery Bay, near Marco Island and Fakahatchee Bay were sampled from July 1971 through July 1972, and 1,006,640 individual animals were collected, of which the majority (55%) came from the Marco area. The large disparity between the catches at Marco and the remaining study areas was due mainly to the appearance of high numbers of species of polychaetes and echinoderms that were of very minor importance or absent from the catches in Rookery Bay and Fakahatchee Bay. When only the major classes of animals in the catch are considered (i.e., crustaceans, fish and mollusks) the total counts for Fakahatchee (298,830) and Marco (275,075) are quite comparable but both exceed Rookery Bay (119,388) by a considerable margin. The effects of the red tide outbreak in the summer of 1971 were apparently restricted to the Rookery Bay Sanctuary and may account for some of the observed differences. For the purposes of making controlled comparisons between the study areas, three common habitats were selected in each area so that a mud bottom habitat, a sand-shell bottom habitat and a vegetated bottom habitat were located in each of the study areas. Total catches by habitat types for crustaceans, fish and mollusks and certain of the more abundant species show clearly the overwhelming importance of the vegetated bottom as a habitat for animals. By habitat the vegetated areas had the most "indicator species" with five, the mud habitat was next with three and the sand-shell habitat third with two. Thus the vegetated habitat would be the best choice if a single habitat were to be used to detect environmental changes between study areas. (PDF contains 137 pages)

[ES] La historia del teatro traducido en la España del siglo XX está aún por escribirse. Este segmento de nuestra cultura traducida ha sido tradicionalmente ignorado en las historias del teatro español. Por suerte, lo que hace sólo veinte años se describía como un páramo investigador es hoy un terreno mucho mejor abonado y roturado. Las investigaciones sobre teatro traducido que han visto la luz progresivamente en estos años nos permitirán en breve escribir y documentar la historia del teatro traducido. Se ofrece en este artículo una visión del modo en que podría acometerse esa tarea,partiendo de lo ya investigado en el proyecto TRACE desde la perspectiva de lo archivado por la censura (de teatro) en la época de Franco.

We present data on ichthyoplankton distribution, abundance, and seasonality and supporting environmental information for four species of coastal pelagics from the family Clupeidae: round herring Etrumeus teres, scaled sardine Harengula jaguana, Atlantic thread herring Opisthonema oglinum, and Spanish sardine Sardinella aurita. Data are from 1982 and 1983 cruises across the northern Gulf of Mexico sponsored by the Southeastern Area Monitoring and Assessment Program (SEAMAP). This is the first such examination for these species on a multiyear and gulfwide scale. Bioproflles on reproductive biology, early life history, meristics, adult distribution, and fisheries characteristics are also presented for these species. During the summer, larval Atlantic thread herring and scaled and Spanish sardines were abundant on the inner shelf <40 m depth), but were rare or absent in deeper waters. Scaled sardine and thread herring were found virtually everywhere inner-shelf waters were sampled, but Spanish sardines were rare in the north-central Gulf. During 1982, larval Atlantic thread herring were the most abundant of the four target c1upeid species, whereas Spanish sardine were the most abundant during 1983. On the west Florida shelf, Spanish sardine dominated larval c1upeid populations both years. Scaled sardine larvae were the least abundant of the four species both years, but were still captured in 25% of inner-shelf bongo net collections. Round herring larvae, collected February-early June (primarily March-April), were abundant on the outer shelf (40-182 m depth) and especially off Louisiana. Over the 2-year period, outer-shelf mean abundance for round herring was 40.2 larvae/10 m2; inner-shelf mean abundances for scaled sardine, Atlantic thread herring, and Spanish sardine were 14.9, 39.2, and 41.9 larvae/l0 m2, respectively. Fish larvae surveys provide information on the species composition in an area of investigation and leads to a better understanding of the entire fish community. Since 2000 every year in April/May an ichthyoplankton survey in ICES-Subdivision 22 and 24 has been performed to sample the plankton community in the western Baltic Sea. One objective was to acquire indicators of possible changes in the natural structures of the fish community. The time series derived from the Bongo-Net samples is too short to show a trend in larval densities. Up to now samples in the western Baltic Sea yielded only low mean densities. According to the reproductive biology the fish species were divided in three major groups: -Fish species with a long developmental phase in the plankton community -Small short living species with benthic eggs and a reduced planktonic phase -Guests without local spawning populations Species were identified, which were absent in the plankton of the western Baltic Sea in the 1960s and 1970s.
