Zusammenfassung Die Rolle verschiedener Mitglieder der NFAT- Familie in der Entwicklung von T- Zellen und deren Funktion wird intensiv untersucht, wohingegen vergleichbare Untersuchungen in Mastzellen rar sind. Mastzellen exprimieren eine Vielzahl biologisch hochaktiver Mediatoren und sind auf diese Weise sowohl in angeborenen als auch adaptiven Immunantworten beteiligt. Die von Mastzellen produzierten Th2-Cytokine verstärken lokal Th2- Reaktionen und TNF-alpha ist ein wichtiger Initiator antimikrobieller Antworten. In dieser Arbeit wird gezeigt, dass die Transkriptionsfaktoren NFATc1 und NFATc2 eine bedeutende Rolle in der Regulation der Expression von TNF-alpha und IL-13 einnehmen, wohingegen NFATc3 hierbei keine Funktion zukommt. Murine „Bone marrow derived mast cells“ (BMMC) aus NFATc2- defizienten Mäusen, aktiviert entweder durch Kreuzvernetzung des IgE- Rezeptors oder Ionomycin, zeigen eine drastisch reduzierte Expression dieser Cytokine verglichen mit Mastzellen aus Wildtyp- Mäusen. Genauere Untersuchungen zeigen, dass sowohl NFATc2 als auch NFATc1 an der Expression von IL-13 und TNF-alpha beteiligt sind, wohingegen sie auf die Degranulation und die Expression von IL-6 keinen Einfluss nehmen. Zusammenfassend scheint eine hohe Aktivität von NFAT- Faktoren für die Induktion des IL-13 und TNF-alpha Promoters in Mastzellen erforderlich zu sein, unabhängig davon, ob diese durch NFATc2 oder NFATc1 oder eine Kombination beider Transkriptionsfaktoren bewerkstelligt wird.

In this report a new automated optical test for next generation of photonic integrated circuits (PICs) is provided by the test-bed design and assessment. After a briefly analysis of critical problems of actual optical tests, the main test features are defined: automation and flexibility, relaxed alignment procedure, speed up of entire test and data reliability. After studying varied solutions, the test-bed components are defined to be lens array, photo-detector array, and software controller. Each device is studied and calibrated, the spatial resolution, and reliability against interference at the photo-detector array are studied. The software is programmed in order to manage both PIC input, and photo-detector array output as well as data analysis. The test is validated by analysing state-of-art 16 ports PIC: the waveguide location, current versus power, and time-spatial power distribution are measured as well as the optical continuity of an entire path of PIC. Complexity, alignment tolerance, time of measurement are also discussed.

The main areas of research of this thesis are Interference Management and Link-Level Power Efficiency for Satellite Communications. The thesis is divided in two parts. Part I tackles the problem of interference environments in satellite communications, and interference mitigation strategies, not just in terms of avoidance of the interferers, but also in terms of actually exploiting the interference present in the system as a useful signal. The analysis follows a top-down approach across different levels of investigation, starting from system level consideration on interference management, down to link-level aspects and to intra-receiver design. Interference Management techniques are proposed at all the levels of investigation, with interesting results. Part II is related to efficiency in the power domain, for instance in terms of required Input Back-off at the power amplifiers, which can be an issue for waveform based on linear modulations, due to their varying envelope. To cope with such aspects, an analysis is carried out to compare linear modulation with waveforms based on constant envelope modulations. It is shown that in some scenarios, constant envelope waveforms, even if at lower spectral efficiency, outperform linear modulation waveform in terms of energy efficiency.

The development of next generation microwave technology for backhauling systems is driven by an increasing capacity demand. In order to provide higher data rates and throughputs over a point-to-point link, a cost-effective performance improvement is enabled by an enhanced energy-efficiency of the transmit power amplification stage, whereas a combination of spectrally efficient modulation formats and wider bandwidths is supported by amplifiers that fulfil strict constraints in terms of linearity. An optimal trade-off between these conflicting requirements can be achieved by resorting to flexible digital signal processing techniques at baseband. In such a scenario, the adaptive digital pre-distortion is a well-known linearization method, that comes up to be a potentially widely-used solution since it can be easily integrated into base stations. Its operation can effectively compensate for the inter-modulation distortion introduced by the power amplifier, keeping up with the frequency-dependent time-varying behaviour of the relative nonlinear characteristic. In particular, the impact of the memory effects become more relevant and their equalisation become more challenging as the input discrete signal feature a wider bandwidth and a faster envelope to pre-distort. This thesis project involves the research, design and simulation a pre-distorter implementation at RTL based on a novel polyphase architecture, which makes it capable of operating over very wideband signals at a sampling rate that complies with the actual available clock speed of current digital devices. The motivation behind this structure is to carry out a feasible pre-distortion for the multi-band spectrally efficient complex signals carrying multiple channels that are going to be transmitted in near future high capacity and reliability microwave backhaul links.

The theory on the intensities of 4f-4f transitions introduced by B.R. Judd and G.S. Ofelt in 1962 has become a center piece in rare-earth optical spectroscopy over the past five decades. Many fundamental studies have since explored the physical origins of the Judd–Ofelt theory and have proposed numerous extensions to the original model. A great number of studies have applied the Judd–Ofelt theory to a wide range of rare-earth doped materials, many of them with important applications in solid-state lasers, optical amplifiers, phosphors for displays and solid state lighting, upconversion and quantum-cutting materials, and fluorescent markers. This paper takes the view of the experimentalist who is interested in appreciating the basic concepts, implications, assumptions, and limitations of the Judd–Ofelt theory in order to properly apply it to practical problems. We first present the formalism for calculating the wavefunctions of 4f electronic states in a concise form and then show their application to the calculation and fitting of 4f-4f transition intensities. The potential, limitations and pitfalls of the theory are discussed, and a detailed case study of LaCl3:Er3+ is presented.

Recent improvements in the precision of mass spectrometric measurements have reduced the uncertainty of K-Ar and 39Ar-40Ar ages measured on geological materials. Now the major sources of uncertainty are the uncertainties on the 40K decay constant and the absolute abundance of 40K. In order to improve on this situation we determined the abundance of the 40K isotope in terrestrial standards. A ThermoFischer Triton+ thermal ionization mass spectrometer was used for K isotope ratio measurements of the NIST K standard reference materials SRM 918b and SRM 985. Ion beams were measured in Faraday cups with amplifiers equipped with 1E10, 1E11 and 1E12 Ω resistors. Three measurement protocols were used: (A) dynamic measurement with in-run fractionation correction by normalization to the IUPAC recommended isotope ratio 41K/39K = 0.0721677; (B) total evaporation; (C) a modified total evaporation with interblock baseline measurements. Different measurement protocols were combined with different loading procedures. The best results were obtained by loading samples on single tantalum filaments with 0.1M H3PO4. The total ion yields (ionization + transmission) were tested for the evaporation procedures (B) and (C) and ranged up to 48 %. The resulting best estimate for the 40K/39K ratio is 0.000 125 116 ± 57 (2σ), corresponding to 40K/K = (1.1668 ± 8; 2σ) x 10-4.

We present a power-scalable approach for yellow laser-light generation based on standard Ytterbium (Yb) doped fibers. To force the cavity to lase at 1154 nm, far above the gain-maximum, measures must be taken to fulfill lasing condition and to suppress competing amplified spontaneous emission (ASE) in the high-gain region. To prove the principle we built a fiber-laser cavity and a fiber-amplifier both at 1154 nm. In between cavity and amplifier we suppressed the ASE by 70 dB using a fiber Bragg grating (FBG) based filter. Finally we demonstrated efficient single pass frequency doubling to 577 nm with a periodically poled lithium niobate crystal (PPLN). With our linearly polarized 1154 nm master oscillator power fiber amplifier (MOFA) system we achieved slope efficiencies of more than 15 % inside the cavity and 24 % with the fiber-amplifier. The frequency doubling followed the predicted optimal efficiency achievable with a PPLN crystal. So far we generated 1.5 W at 1154nm and 90 mW at 577 nm. Our MOFA approach for generation of 1154 nm laser radiation is power-scalable by using multi-stage amplifiers and large mode-area fibers and is therefore very promising for building a high power yellow laser-light source of several tens of Watt.

We use the oxygen isotopic composition of planktonic foraminifera Globigerinoides ruber (white) from Ocean Drilling Program Site 1058 in the subtropical northwestern Atlantic to construct a high-resolution (~800 year) climate record spanning the mid-Pleistocene climate transition (~410 ka to 1350 ka). We investigate whether or not millennial-scale instabilities in the proxy record are associated with the extent of continental glaciation. G. ruber d18O values display high-frequency fluctuations throughout the record, but the amplitude about mean glacial and interglacial d18O values increases at marine isotope stage (MIS) 22 (880 ka) and is highest during MIS 12. These observations support that millennial-scale climate instabilities are associated with ice sheet size. Time series analysis illustrates that these variations have significant concentration of spectral power centered on periods of ~10-12 ka and ~5 ka. The timing of these fluctuations agrees well, or coincides with, the periodicities of the second and fourth harmonics, respectively, of precessional forcing at the equator. An insolation-based origin of the millennial-scale instabilities would be independent of ice volume and explains the presence of these fluctuations before the mid-Pleistocene climate transition as well as during interglacial intervals (e.g., MIS 37 and 17). Because the amplitude of the millennial-scale variations increases during the mid-Pleistocene transition, feedback mechanisms associated with the growth of large, 100-ka-paced, polar ice sheets may be important amplifiers of regional surface water hydrographic changes.

We will present calculations of opacities for matter under LTE conditions. Opacities are needed in radiation transport codes to study processes like Inertial Confinement Fusion and plasma amplifiers in X-ray secondary sources. For the calculations we use the code BiGBART, with either a hydrogenic approximation with j-splitting or self-consistent data generated with the atomic physics code FAC. We calculate the atomic structure, oscillator strengths, radiative transition energies, including UTA computations, and photoionization cross-sections. A DCA model determines the configurations considered in the computation of the opacities. The opacities obtained with these two models are compared with experimental measurements.

Amorphous samples with helical induced anisotropy show magnetization processes that can be controlled by applying a longitudinal magnetic field simultaneously with an alternating current flowing through the sample. By varying the current amplitude and the phase difference between current and applied field, a wide range of coercivity and susceptibility values can be achieved. This work shows that the apparent coercive field and the susceptibility can be controlled in amorphous ribbons with helical anisotropy. These characteristics make these samples very suitable for their application as sensor cores, magnetic amplifiers, variable reluctance transformer cores, etc

This paper analyses the noise and gain measurement of microwave differential amplifiers using two passive baluns. A model of the baluns that includes potential losses and unbalances has been considered. This analysis allows to de-embed the actual performance of the differential device from the single-ended measurements of the two-port cascaded system and the baluns. The method has been validated with measured results from a fully-differential amplifier prototype.

In this paper, an interleaved multiphase buck converter with minimum time control strategy for envelope amplifiers in high efficiency RF power amplifiers is proposed. The solution for the envelope amplifier is to combine the proposed converter with a linear regulator in series. High efficiency of envelope amplifier can be obtained through modulating the supply voltage of the linear regulator. Instead of tracking the envelope, the buck converter has discrete output voltage that corresponding to particular duty cycles which achieve total ripple cancellation. The transient model for minimum time control is explained, and the calculation of transient times that are pre-calculated and inserted into a lookup table is presented. The filter design trade-off that limits capability of envelope modulation is also discussed. The experimental results verify the fast voltage transient obtained with a 4-phase buck prototype.

This paper presents a theoretical analysis and an optimization method for envelope amplifier. Highly efficient envelope amplifiers based on a switching converter in parallel or series with a linear regulator have been analyzed and optimized. The results of the optimization process have been shown and these two architectures are compared regarding their complexity and efficiency. The optimization method that is proposed is based on the previous knowledge about the transmitted signal type (OFDM, WCDMA...) and it can be applied to any signal type as long as the envelope probability distribution is known. Finally, it is shown that the analyzed architectures have an inherent efficiency limit.

Classical linear amplifiers such as A, AB and B offer very good linearity suitable for RF power amplifiers. However, its inherent low efficiency limits its use especially in base-stations that manage tens or hundreds of Watts. The use of linearization techniques such as Envelope Elimination and Restoration (EER) allow an increase of efficiency keeping good linearity. This technique requires a very fast dc-dc power converter to provide variable voltage supply to the power amplifier. In this paper, several alternatives are analyzed to implement the envelope amplifier based on a cascade association of a switched dc-dc converter and a linear regulator. A simplified version of this approach is also suitable to operate with Envelope Tracking technique.
