De uma plantação de almeirão (Chicorium intybus c.v. Folha Larga) foram coletadas 30 plantas com 4 repetições aos 25, 35, 45 e 55 dias após a emergência. O material coletado foi dividido em folhas e raízes e analisados por N, P, K, Ca, Mg, S, B, Cu, Fe, Mn, Zn, pelos métodos convencionais de laboratório. O elemento extraído em maior quantidade foi o nitrogênio, seguido de potássio, cálcio, fósforo, magnésio e enxofre. Dentre os micronutrientes o elemento extraído em maiores quantidades foi o ferro, seguido do manganês, zinco, boro e finalmente pelo cobre. Estabelece-se o nível crítico para os nutrientes os seguintes valores: 4,39% N, 0,47% P, 2,93 K, 1,00 % Ca, 0,35% Mg, 0,20% S, 59 ppm B, 15 ppm Cu, 2,926 ppm Fe, 117 ppm Mn e 80 ppm Zn. A extração total por folhas e raízes foi de 5.000 plantas/100 m² e de 92,3 g N, 13,3 g P, 83,2 g K, 28,8 g Ca, 8,6 g Mg, 5,45 g S, 154 mg B, 38 mg Cu, 7,979 mg Fe, 301 mg Mn e 214 mg Zn.

Eleven species of tanagers in Ibitipoca State Park, Minas Gerais, southeastern Brazil, were studied for 12 months and the birds diet was described. Fruits of 52 plant species were the main item consumed for all tanagers, representing 59.70% of all items in 1330 events, although different proportions in the use of this resource have been found among the bird species. The main plant families found were Myrtaceae with eight species accounting for 22.29% (n=177), and Melastomataceae with seven species accounting for 22.29% (n=177) of fruit intake. The most frequent plant species in the diet was Cecropia glaziovi Snethl. (Cecropiaceae), which made up 17.76% (n=141) of fruit intake of all species of tanagers. Arthropod consumption came second with 22.63% (n=301), but flowers, leaves, nectar, food remains left by tourists and galls were also represented. Several environmental features influenced diet composition. An Asteraceae, Vanillosmopsis erythropappa Schultz, very common in the park, was important for some birds as its flowers attracted arthropods and its branches and leaves were used as a substrate for insectivory. Fruit intake rate, some bird-plant interaction and features of food-handling were also discussed, pointing to the importance of these birds for the structure of the heterogeneous local landscape, possibly through the spread of seeds of different plant species.

Quatro frangos e três pombos (1ª e 2ª experiências) não se infectaram com o "T. (S.) cruzi" (amostra Y), injetados na pele com doses de 9,8 x10*5 a 3,8 x 10*6 parasitos (mistura de formas de cultivo tripomastigotos sangüíneos). Em outros 2 frangos (6ª experiência), a refratariedade não foi rompida com, respectivamente, 5 injeções peritoneais em dias alternados, de 2,8 x 10*6 a 6,1 x 10*6 parasitos, e 5 injeções peritoneais diárias do conteúdo de um cultivo suspenso em salina, adicionado de tripamastigotos sangüíneos, obtidos de 3 camundongos jovens infectados. E, ainda em outros 2 frangos (7ª experiência), a refratariedade não foi rompida associando-se a doses peritoneais de 8,1 x 10*6 e 3,5 x 10*7 parasitos, respectivamente, a ministração de dexametasona, seja com o início da droga no mesmo dia da inoculação (0,4 mg/8 doses/10 dias), seja com o ínicio da droga 3dias antes do dia da inoculação (0,4 mg/10 doses/12 dias). Nessas 11 aves a refratariedade foi comprovada pela inoculação do sangue em camundongos albinos recém-nascidos (total de 54 inoculaçoes negativas em 301 camundongos); e por 52 xenodiagnósticos negativos (total de 175 larvas e adultos de Triatoma infestans e Panstrongylys megistus). As inoculações e os xenodiagnósticos foram feitos entre 1 hora (h) e 96 h e aos 10, 20 e 30 dias depois da inoculação das aves. (Quadro 1). Portanto, além da refratariedade, ficou demonstrado que os parasitos não passam para o sangue circulante das aves. em 10 frangos (3ª, 4ª e 5ª experiências) inoculados na pele com, 1,9 x 10*6 a 3,7 x 10*6 parasitos, verificou-se a duração da viabilidade dos parasitos nas áreas injetadas. Estas foram puncionadas de 1/2 com 1/2 hora, e o material obtido microscopado e inoculado em camundongos recém-nascidos. De 17 inoculações feitas até às 8:30 h, doze foram positivas, inclusive a última; e de 13 inoculações feitas entre 9 e 90 h (as 4 primeiras ainda de 1/2 em 1/2 h), todas foram negativas. Por outro lado, de 10 xenodiagnósticos feitos nas áreas injetadas, entre 1/2h e 9 h após a inoculação das aves, apenas 2 foram positivos; e de 10 xenodiagnósticos feitos entre 9 e 90 h, todos foram negativos. (Quadro 2). Em 4 frangos (8ª experiência), 10 xenodiagnósticos foram feitos entre 1 e 10 h, sendo 2 positivos (7 h e 8 h). Apesar de dificuldades operacionais na execução dos xenodiagnósticos, nas condições experimentais deste trabalho, eles se mostraram inferiores ás inoculações em camundongos recém-nascidos, como prova diagnóstica laboratorial. Portanto, verificou-se que ainda são encontrados parasitos viáveis nas áreas injetadas, pelo menos até ás 8:30 h depois inoculação. todavia, a partir de 2 h após a inoculação os parasitos diminuem de número progressivamente nas áreas injetadas, ao mesmo tempo que mostram sensíveis alterações morfológicas, encontrando-se também muito deles mortos. Ao exame direto, não mais foram vistos depois das 5:30 h. As inoculações provocam na pele dos frangos...

Metalworking fluid-associated hypersensitivity pneumonitis (MWF-HP) is a pulmonary disease caused by inhaling microorganisms present in the metalworking fluids used in the industrial sector. Mycobacterium immunogenum is the main etiological agent. Among the clinical, radiological and biological tools used for diagnosis, serological tests are important. The aim of this study was to identify immunogenic proteins in M. immunogenum and to use recombinant antigens for serological diagnosis of MWF-HP. Immunogenic proteins were detected by two-dimensional Western blot and candidate proteins were identified by mass spectrometry. Recombinant antigens were expressed in Escherichia coli and tested by enzyme-linked immunosorbent assay (ELISA) with the sera of 14 subjects with MWF-HP and 12 asymptomatic controls exposed to M. immunogenum. From the 350 spots visualized by two-dimensional gel electrophoresis with M. immunogenum extract, 6 immunogenic proteins were selected to be expressed as recombinant antigens. Acyl-CoA dehydrogenase antigen allowed for the best discrimination of MWF-HP cases against controls with an area under the receiver operating characteristics (ROC) curve of 0.930 (95% CI=0.820-1), a sensitivity of 100% and a specificity of 83% for the optimum threshold. Other recombinant antigens correspond to acyl-CoA dehydrogenase FadE, cytosol aminopeptidase, dihydrolipoyl dehydrogenase, serine hydroxymethyltransferase and superoxide dismutase. This is the first time that recombinant antigens have been used for the serodiagnosis of hypersensitivity pneumonitis. The availability of recombinant antigens makes it possible to develop standardized serological tests which in turn could simplify diagnosis, thus making it less invasive.

Lung cancer mortality in young women in the European Union (EU) has steadily increased until the mid 1990 s and has levelled off thereafter, but trends have been heterogeneous in various countries. We analyzed therefore age-standardized trends in lung cancer mortality in young women (20-44) for the 6 major European countries, using joinpoint regression. In the early 1970s the highest lung cancer mortality in young women was in the UK (2.1/100,000). UK rates, however, steadily declined and in 2000-2004 they were the lowest of all 6 major EU countries (1.2/100,000). The second lowest rate in 2000-2002 was in Italy, whose rates remained around 1.1/100,000 between 1970 and 1994, and increased to 1.4 thereafter. In Germany and Poland, lung cancer rates in young women rose from 0.8-1.0/100,000 in the early 1970s to 1.7-1.9 in the mid 1990 s and levelled off during the last decade. Major rises over recent years were observed in France (from 0.8/100,000 in 1985-1989 to 2.2 in 2000-2003) and in Spain (from 0.8 in the 1985-1989 to 1.7 in 2000-2004). Thus, France showed both the highest rate observed over the last 3 decades and the largest rise over the last 2 decades. Since recent trends in the young give relevant information to the likely future trends in middle age, the female lung cancer epidemic is likely to expand in southern Europe from the current rates of 5.0/100,000 in Spain and 7.7 in France to approach 20/100,000 within the next 2-3 decades. Urgent interventions for smoking cessation in women are therefore required.

The epidermal growth factor receptor (EGFR) plays a central role in cell life by controlling processes such as growth or proliferation. This receptor is commonly overexpressed in a number of epithelial malignancies and its upregulation is often associated with an aggressive phenotype of the tumor. Thus, targeting of EGFR represents a very promising challenge in oncology, and antibodies raised against this receptor have been investigated as potential antitumor agents. Various putative mechanisms of action were proposed for such antibodies, including decreased proliferation, induction of apoptosis, stimulation of the immunological response against targeted cancer cells or combinations thereof. We report here the development of an alternative high affinity molecule that is directed against EGFR. Production of this pentameric protein, named peptabody-EGF, includes expression in a bacterial expression system and subsequent refolding and multimerization of peptabody monomers. The protein complex contains 5 human EGF ligand domains, which confer specific binding towards the extracellular portion of EGFR. Receptor binding of the peptabody-EGF had a strong antiproliferative effect on different cancer cell lines overexpressing EGFR. However, cells expressing constitutive levels of the target receptor were barely affected. Peptabody-EGF treated cancer cells exhibited typical characteristics of apoptosis, which was found to be induced within 30 min after the addition of the peptabody-EGF. In vitro experiments demonstrated a significantly higher binding activity for peptabody-EGF than for the therapeutic monoclonal EGFR antibody Mab-425. Furthermore, the antitumor action provoked by the peptabody-EGF was greatly superior than antibody mediated effects when tested on EGFR overexpressing cancer cell lines. These findings suggest a potential application of this high affinity molecule as a novel tool for anti-EGFR therapy.

A procedure was devised for the identification and specific cloning of functionally rearranged variable region immunoglobulin (Ig) gene segments from genomic DNA of a murine hybridoma cell line which produces a high-affinity monoclonal antibody (MAb) directed against human carcinoembryonic antigen (CEA). The cloned, functionally-rearranged murine Ig H-chain and L-chain variable region gene segments were incorporated into plasmid vectors capable of directing the expression of a chimaeric mouse-human antibody molecule with human (gamma 4, kappa) constant region sequences. Expression plasmids were transfected into a mouse myeloma cell line by electroporation and transfectomas secreting functional chimaeric antibody selected. Chimaeric antibody generated by transfectomas was analysed and shown to compete effectively with its murine counterpart for binding to the CEA epitope, and to have an equivalent antigen-binding affinity. This anti-CEA recombinant antibody should find application in in vivo diagnosis by immunoscintigraphy of human colonic carcinoma, and possibly also in therapy of the disease, overcoming some of the difficulties associated with the repeated use of non-human immunoglobulins in human patients.

Issues. Numerous studies have reported that brief interventions delivered in primary care are effective in reducing excessive drinking. However, much of this work has been criticised for being clinically unrepresentative. This review aimed to assess the effectiveness of brief interventions in primary care and determine if outcomes differ between efficacy and effectiveness trials. Approach. A pre-specified search strategy was used to search all relevant electronic databases up to 2006. We also hand-searched the reference lists of key articles and reviews. We included randomised controlled trials (RCT) involving patients in primary care who were not seeking alcohol treatment and who received brief intervention. Two authors independently abstracted data and assessed trial quality. Random effects meta-analyses, subgroup and sensitivity analyses and meta-regression were conducted. Key Findings. The primary meta-analysis included 22 RCT and evaluated outcomes in over 5800 patients. At 1 year follow up, patients receiving brief intervention had a significant reduction in alcohol consumption compared with controls [mean difference: -38 g week(-1), 95%CI (confidence interval): -54 to -23], although there was substantial heterogeneity between trials (I(2) = 57%). Subgroup analysis confirmed the benefit of brief intervention in men but not in women. Extended intervention was associated with a non-significantly increased reduction in alcohol consumption compared with brief intervention. There was no significant difference in effect sizes for efficacy and effectiveness trials. Conclusions. Brief interventions can reduce alcohol consumption in men, with benefit at a year after intervention, but they are unproven in women for whom there is insufficient research data. Longer counselling has little additional effect over brief intervention. The lack of differences in outcomes between efficacy and effectiveness trials suggests that the current literature is relevant to routine primary care.

Erythrokeratodermia variabilis (EKV) is an autosomal dominant keratinization disorder characterized by migratory erythematous lesions and fixed keratotic plaques. All families with EKV show mapping to chromosome 1p34-p35, and mutations in the gene for connexin 31 (Cx31) have been reported in some but not all families. We studied eight affected and three healthy subjects in an Israeli family, of Kurdish origin, with EKV. After having mapped the disorder to chromosome 1p34-p35, we found no mutations in the genes for Cx31, Cx31.1, and Cx37. Further investigation revealed a heterozygous T-->C transition leading to the missense mutation (F137L) in the human gene for Cx30.3 that colocalizes on chromosome 1p34-p35. This nucleotide change cosegregated with the disease and was not found in 200 alleles from normal individuals. This mutation concerns a highly conserved phenylalanine, in the third transmembrane region of the Cx30.3 molecule, known to be implicated in the wall formation of the gap-junction pore. Our results show that mutations in the gene for Cx30.3 can be causally involved in EKV and point to genetic heterogeneity of this disorder. Furthermore, we suggest that our family presents a new type of EKV because of the hitherto unreported association with erythema gyratum repens.

This study presents a classification criteria for two-class Cannabis seedlings. As the cultivation of drug type cannabis is forbidden in Switzerland, law enforcement authorities regularly ask laboratories to determine cannabis plant's chemotype from seized material in order to ascertain that the plantation is legal or not. In this study, the classification analysis is based on data obtained from the relative proportion of three major leaf compounds measured by gas-chromatography interfaced with mass spectrometry (GC-MS). The aim is to discriminate between drug type (illegal) and fiber type (legal) cannabis at an early stage of the growth. A Bayesian procedure is proposed: a Bayes factor is computed and classification is performed on the basis of the decision maker specifications (i.e. prior probability distributions on cannabis type and consequences of classification measured by losses). Classification rates are computed with two statistical models and results are compared. Sensitivity analysis is then performed to analyze the robustness of classification criteria.
