The study of protein fold is a central problem in life science, leading in the last years to several attempts for improving our knowledge of the protein structures. In this thesis this challenging problem is tackled by means of molecular dynamics, chirality and NMR studies. In the last decades, many algorithms were designed for the protein secondary structure assignment, which reveals the local protein shape adopted by segments of amino acids. In this regard, the use of local chirality for the protein secondary structure assignment was demonstreted, trying to correlate as well the propensity of a given amino acid for a particular secondary structure. The protein fold can be studied also by Nuclear Magnetic Resonance (NMR) investigations, finding the average structure adopted from a protein. In this context, the effect of Residual Dipolar Couplings (RDCs) in the structure refinement was shown, revealing a strong improvement of structure resolution. A wide extent of this thesis is devoted to the study of avian prion protein. Prion protein is the main responsible of a vast class of neurodegenerative diseases, known as Bovine Spongiform Encephalopathy (BSE), present in mammals, but not in avian species and it is caused from the conversion of cellular prion protein to the pathogenic misfolded isoform, accumulating in the brain in form of amiloyd plaques. In particular, the N-terminal region, namely the initial part of the protein, is quite different between mammal and avian species but both of them contain multimeric sequences called Repeats, octameric in mammals and hexameric in avians. However, such repeat regions show differences in the contained amino acids, in particular only avian hexarepeats contain tyrosine residues. The chirality analysis of avian prion protein configurations obtained from molecular dynamics reveals a high stiffness of the avian protein, which tends to preserve its regular secondary structure. This is due to the presence of prolines, histidines and especially tyrosines, which form a hydrogen bond network in the hexarepeat region, only possible in the avian protein, and thus probably hampering the aggregation.

Die Messung der 1s Hyperfeinstruktur (HFS) wasserstoffähn-licher Ionen bei hoher Kernladung Z erlaubt den Test der QED in Gegenwart starker elektrischer und magnetischer Felder durchzuführen. Aufgrund der Z^3-Abhängigkeit der magnetischen Wechselwirkung liegt die 1s-HFS bei hohem Z im optischen Spektrum und kann mit der Laserspektroskopie untersucht werden.In der vorliegenden Arbeit wurde die Grundzustands-HFS an 207Pb81+ bestimmt. Die Experimente wurden am Speicherring ESR der Gesellschaft für Schwerionenforschung mbH am Elek-tronen-gekühlten Bleistrahl durchgeführt.Ein besonderer Schwerpunkt dieser Arbeit war die Entwick-lung neuer Experimentiertechniken, die es erlauben, lang-lebigen Strahlungsübergänge im nahen infraroten Spektral-bereich mit Hilfe der Fluoreszenz-Laserspektroskopie am Speicherring zu untersuchen. Der Ionenstrahl wurde in kollinearer Geometrie mit einem Nd:YAG-Laser angeregt und die Resonanz durch Doppler-Abstimmung gemessen. Der M1-Übergang der 1s HFS an 207Pb81+ liegt bei 1019,7(2) nm. Die QED-Korrekturen sind damit auf wenige Prozent genau bestimmt. Die theoretische Berechnung der QED-Korrekturen benutzt die gemessenen bzw. den Kernmodellen entnommenen Verteilungen der Kernladung und der Kernmagnetisierung. Die Unsicherheit der Kenntnis dieser Verteilungen spielt nur eine untergeordnete Rolle für die Fehler in der gerechneten QED-Korrektur. Allerdings existieren für das magnetische Moment von 207Pb zwei widersprüchliche Literaturwerte. Die Diskussion der Ergebnisse wird im Rahmen dieses Sachver-haltes geführt.Die natürliche Lebensdauer des oberen Hyperfein-Niveaus wurde mit 49,5(6,5) ms gemessen und ist mit dem Theoriewert von 52,3(2) ms verträglich. Durch präzise Lebensdauermes-sungen können QED-Korrekturen zum g-Faktor des gebundenen Elektrons getestet werden.

In Drosophila melanogaster wird das Nervensystem von neuralen Vorläuferzellen, den Neuroblasten (NB) gebildet. Diese teilen sich im Stammzellmodus und bringen bei jeder Teilung eine Ganglienmutterzelle (GMZ) hervor. GMZ teilen sich einmal und generieren zwei Zellen, die ein neuronales und/oder gliales Schicksal annehmen. Die Reihenfolge in der ein NB GMZ generiert, ist vermutlich ein zellautonomer Prozess, der an die transiente und sequenzielle Expression der Transkriptionsfaktoren Hunchback (Hb), Krüppel (Kr), Pdm, Castor (Cas) und Grainy head (Grh) gekoppelt ist. Hb ist der erste Faktor dieser Genkaskade. Damit der NB zur nächsten zeitlichen Identität übergehen kann, die in der Regel von Kr bestimmt wird, muss die Hb-Expression im NB beendet werden. Das Abschalten wird transkriptionell reguliert und ist von einer vorhergehenden Mitose abhängig. Im Gegensatz zum NB wird in der GMZ, die bei dieser Teilung entsteht, die Hb-Expression beibehalten. In der vorliegenden Arbeit wurde der Mitose-abhängige Mechanismus aufgeklärt, durch den eine selektive Abschaltung von hb im NB erfolgt, nicht aber in der GMZ. Der Transkriptionsfaktor Seven-up (Svp) beendet die hb-Expression im NB. svp wird vor der entscheidenden Zellteilung im NB transkribiert, aber kaum translatiert. Nach erfolgter Zellteilung wird Svp verstärkt translatiert und sowohl im NB als auch in der neu entstandenen GMZ exprimiert. Im NB führt die Svp-Funktion zur Abschaltung von Hb. In der ebenfalls Svp-positiven GMZ verhindert jedoch Prospero (Pros), das während der Teilung in diese Zelle segregiert ist, die Aktivität von Svp. Um zu überprüfen, ob Svp und Pros die Transkription von Hb entweder direkt oder indirekt regulieren, wurde eine Enhancerfragment-Analyse durchgeführt. Durch diese Unter-suchungen sollten relevante regulatorische Bereiche des hb-Gens identifiziert werden. Zusätzlich zu den bereits bekannten Regionen im 5´-Bereich von hb, konnten 3´ des transkribierten Bereiches weitere nervensystemspezifische Enhancerelemente gefunden werden. Neben der Spezifizierung von früh geborenen Nachkommen von NB hat Hb im Nervensystem die Funktion, den multipotenten Zustand junger NB aufrecht zu erhalten. Um den Beitrag einzelner Domänen des Hb-Proteins an diesen Aufgaben zu bestimmen, wurden verschiedene hb-Varianten, in denen spezifische Domänen deletiert bzw. mutiert waren, im NB7-1 überexprimiert und auf die Induktion von „frühem“ Schicksal getestet. In einem zweiten Ansatz wurden mutante hb-Allele analysiert. Es stellte sich heraus, dass der D-Box eine entscheidende Rolle bei der Spezifizierung von „frühem“ Schicksal zukommt. Die terminalen Zinkfinger sind vermutlich in die Aufrechterhaltung der Kompetenz von NB involviert. Eine Deletion der A-Box erhöht möglicherweise die Aktivität des Hb-Proteins.

Malignant pleural mesothelioma (MPM) is a lethal cancer of the mesothelium with high chemotherapeutic resistance via unknown mechanisms. A prevailing hypothesis states that cancer stem cells (CSCs) persist in tumors causing relapse after chemotherapy, thus, rendering these cells as critical targets responsible for tumor resistance and recurrence. We selected candidate CSC markers based on expansion under hypoxic conditions, a hallmark for the selection of chemoresistant cells; and investigated the expression of CSC markers: CD133, Bmi-1, uPAR and ABCG2 in three MPM cell lines and normal mesothelial cells by quantitative RT-PCR. Furthermore, we evaluated the chemotherapeutic resistance associated with each CSC marker by determining the change in CSC marker-mRNA levels as an index of drug-resistance following treatment with either cisplatin or pemetrexed. We demonstrate the expression of CSC markers: CD133, Bmi-1, uPAR and ABCG2 in both normal and MPM cell lines. Bmi-1+, uPAR+ and ABCG2+ cells show a distinct role in conferring chemoresistance to cisplatin and pemetrexed in the malignant setting. By contrast, these markers have no apparent participation in chemoresistance to drug treatments in normal mesothelial cells. Intriguingly, CD133 revealed chemoresistant properties in both normal mesothelial and malignant pleural mesothelioma cells. This study provides evidence of putative CSCs conferring drug-resistance to cisplatin and pemetrexed in MPM cell lines. Specific targeting of these drug-resistant cells, while considering the functional heterogeneity of the MPM subtypes, may contribute to more focused and effective chemotherapeutic regimens for malignant pleural mesothelioma.

OBJECTIVES: The aim of this randomised clinical trial was to investigate if a laser fluorescence device is able to discriminate between sound and carious approximal sites and between enamel and dentinal lesions, as well as to find appropriate cut-off values. METHODS: One hundred and seventeen sound or uncavitated carious sites in permanent molars were visually and radiographically examined, then either opened or not, after which their laser fluorescence was measured. Forty-three lesions were opened, the caries removed and the clinically identified caries depths were registered in addition to the radiographical scoring. Seventy-four sites were radiographically deemed sound or had enamel caries and were not opened. Here, the radiographical scorings were registered. RESULTS: Taking the radiographic scoring as gold standard for all investigated approximal sites, sound sites (D(0), n=40) showed significantly lower laser fluorescence measurements than carious sites (D(1-4), n=77) (Mann-Whitney test, P<0.025) suggesting a cut-off at 7 (sensitivity=0.68, specificity=0.7). Comparing measurements of D(0-2) (n=74) and D(3,4) (n=43), the results were also different by a statistically significant amount (P<0.025) and the cut-off calculated to be 16 (sensitivity=0.6, specificity=0.84). A fair positive correlation between laser fluorescence values and radiographical scoring was found (rho=+0.47, P<0.01). Analysing the 43 opened lesions with their clinically found lesion depths as gold standard, there was a fair positive correlation to the laser fluorescence values (rho=+0.34, P=0.03) and a moderately strong correlation to the radiographic scoring (rho=+0.67, P<0.01). CONCLUSION: The device may be an adjunct tool in the approximal detection of caries along with established procedures.

Purpose To update American Society of Clinical Oncology/American Society of Hematology recommendations for use of erythropoiesis-stimulating agents (ESAs) in patients with cancer. Methods An Update Committee reviewed data published between January 2007 and January 2010. MEDLINE and the Cochrane Library were searched. Results The literature search yielded one new individual patient data analysis and four literature-based meta-analyses, two systematic reviews, and 13 publications reporting new results from randomized controlled trials not included in prior or new reviews. Recommendations For patients undergoing myelosuppressive chemotherapy who have a hemoglobin (Hb) level less than 10 g/dL, the Update Committee recommends that clinicians discuss potential harms (eg, thromboembolism, shorter survival) and benefits (eg, decreased transfusions) of ESAs and compare these with potential harms (eg, serious infections, immune-mediated adverse reactions) and benefits (eg, rapid Hb improvement) of RBC transfusions. Individual preferences for assumed risk should contribute to shared decisions on managing chemotherapy-induced anemia. The Committee cautions against ESA use under other circumstances. If used, ESAs should be administered at the lowest dose possible and should increase Hb to the lowest concentration possible to avoid transfusions. Available evidence does not identify Hb levels � 10 g/dL either as thresholds for initiating treatment or as targets for ESA therapy. Starting doses and dose modifications after response or nonresponse should follow US Food and Drug Administration–approved labeling. ESAs should be discontinued after 6 to 8 weeks in nonresponders. ESAs should be avoided in patients with cancer not receiving concurrent chemotherapy, except for those with lower risk myelodysplastic syndromes. Caution should be exercised when using ESAs with chemotherapeutic agents in diseases associated with increased risk of thromboembolic complications. Table 1 lists detailed recommendations. This guideline was developed through a collaboration between the American Society of Clinical Oncology and the American Society of Hematology and has been published jointly by invitation and consent in both Journal of Clinical Oncology and Blood.

We have previously shown that EphB4 and ephrin-B2 are differentially expressed in the mammary gland and that their deregulated expression in the mammary epithelium of transgenic mice leads to perturbations of the mammary parenchyma and vasculature. In addition, overexpression of EphB4 and expression of a truncated ephrin-B2 mutant, capable of receptor stimulation but incapable of reverse signalling, confers a metastasising phenotype on NeuT initiated mouse mammary tumours. We have taken advantage of this transgenic tumour model to compare stem cell characteristics between the non-metastasising and metastasising mammary tumours. We analysed the expression of the proliferation attenuating p21(waf) gene, which was significantly increased in the metastasising tumours. Moreover, we compared the expression of CK-19, Sca-1, CD24 and CD49f as markers for progenitor cells exhibiting a decreasing differentiation grade. Sca-1 expressing cells were the earliest progenitors detected in the non-metastasising NeuT induced tumours. The metastasising NeuT/EphB4 tumours were enriched in CD24 expressing cells, whereas the metastasising NeuT/truncated ephrin-B2 tumours contained in addition significant amounts of CD49f expressing cells. The same cell populations were also enriched in mammary glands of single transgenic MMTV-EphB4 and MMTV-truncated ephrin-B2 females indicating that deregulated EphB4-ephrin-B2 signalling interferes with the homeostasis of the stem/progenitor cell pool before tumour formation is initiated. Since the same cell populations are enriched in the normal tissue, primary mammary tumours and metastases we conclude that these progenitor cells were the origin of tumour formation and that this change in the tumour origin has led to the acquisition of the metastatic tumour phenotype.

Cancer most probably originates from stem/progenitor cells and exhibits a similar cell hierarchy as normal tissues. Moreover, there is growing evidence that only the stem cells are capable of metastasis formation. We have previously shown that overexpression of a dominant negative ephrin-B2 mutant interferes with mammary gland differentiation and confers a metastatic phenotype to NeuT-induced mammary tumors with an increase in cells with stem/progenitor characteristics. To investigate the role of ephrin-B2 in the control of the mammary stem cell niche, we analyzed the mammary stem and progenitor cell populations in transgenic mice overexpressing the mutant ephrin-B2. Quantification by FACS analysis revealed a significant increase of cells in the basal/alveolar cell-, the bi-potent progenitor- and the stem cell-enriched fractions. Moreover, the supposed precursors of estrogen receptor-positive cells were elevated in the stem cell-enriched fraction. In contrast, the epithelium from transgenic mice overexpressing the native ephrin-B2 gene showed an augmentation of the luminal cell- and the bi-potent progenitor-enriched fractions. Repopulation assays revealed that the epithelial cells of truncated ephrin-B2 transgenic epithelial cells have a higher regeneration capacity than those of controls and of native ephrin-B2 transgenic mice, confirming the augmentation of stem cells. Morphologically, these outgrowths exhibited impaired basal/luminal compartmentalization and epithelial polarization. These results demonstrate that deregulated ephrin-B2 expression interferes with the regulation of the stem cell niche and leads to a shift of the differentiation pathway and may thereby contribute to the acquisition of the metastatic phenotype long before carcinogenic growth becomes apparent.

The present research looked to explore the relationship between the emotional responses of college students to different hooking up behaviors. Seven hundred and nine undergraduates participated in a web-based survey that included a demographic questionnaire, SDS, PANAS, AUDIT, and a measure of hooking up. This measure examined the frequency with which they participated in eight different types of hooking up varying by degree of familiarity to their hook partner and whether or not the hook up was coital or non-coital, as well as their emotional responses to the behavior and their perception of the emotional responses of their partner. Results showed that both menand women experienced more positive emotional responses to hooking up behaviors than negative emotional responses. Men experienced significantly more positive emotional responses to hook up behaviors than did women. Women experienced significantly more negative emotional responses for hook ups that were coital with strangers than did men, while men experienced more positive emotional responses for hook ups that were coital with strangers, coital with acquaintances, and coital with partners that were previous romantic partners than did women. Men also experienced more positive emotional responses for hook ups that were non-coital with strangers and non-coital withacquaintances. Men tended to rate their partner’s positive emotional responses higher than what women reported experiencing, particularly for hook ups with less familiar partners. Women’s ratings of their partner’s negative emotional responses were lower than what men actually reported experiencing. The data collected provide several opportunities for future analyses to be conducted and this research will add to the relatively small body of literature on hooking up.
