The rock pigeon (Columba livia) may serve as a reservoir for several pathogenic agents that can be transmitted to poultry, wildlife, domesticated pets, and/or humans via excreta, secretions, or dust from feathers. In addition, ingestion of infected pigeons by wild and domestic animals can also transmit these pathogenic agents. The health status of 126 free-living pigeons in an urban area was evaluated by microbiologic culture for Salmonella and serologic testing for the presence of antibodies for Toxoplasma gondii and for Newcastle disease virus (NDV) from 120 and 109 pigeons, respectively. After drawing blood, the birds were euthanized, and fragments of the liver, spleen, lungs, and gonads, and feces were cultured for Salmonella spp. Salmonella spp. was isolated from 10 birds (7.94%), of which 8 were Salmonella typhimurium, one was Salmonella enterica subsp. enterica serotype 4,12 and one was Salmonella enterica subsp. enterica serotype 4,12,i. Six of 109 pigeons (5.50%) were positive for NDV antibodies when using the hemagglutination inhibition test. Toxoplasma gondii antibodies were detected by immunofluorescence in one of 120 sera tested (0.83%). The results indicate that feral rock pigeons were exposed to NDV and T gondii, although the exposure was low. In addition, these birds had Salmonella spp. and could disseminate this pathogen in the environment.

Doença de Chagas é uma antropozoonose causada por Trypanosoma cruzi que tem os cães como importante reservatório da doença na América do Sul. Este trabalho teve como objetivo avaliar a ocorrência da infecção natural pelo T. cruzi em cães de uma área rural do estado de Mato Grosso do Sul, Brasil. Foram utilizados os testes de imunofluorescência indireta (IFI) e ensaio imunossorvente ligado a enzima (ELISA) em 75 cães residentes na área. Foram detectados anticorpos em 45,3% (n=34) e 24,0% (n=18) nos testes de IFI e ELISA, respectivamente. A real prevalência da infecção foi confirmada como 22,7% (n=17) pelo critério de positividade em ambos os testes. Os resultados obtidos confirmam a infecção chagásica nos cães dessa região.

Several diseases involve the nervous system of cattle, among which infections with Rabies Virus and Bovine Herpes Virus 5 (BoHV-5) are noteworthy. In order to detect seropositive animals to BoHV-5, 156 Brahman-Zebu bovines blood samples from Colombia's eastern plains were analyzed through seroneutralization assay; the area has a history of animals dying with nervous symptoms and which rule out the disease of rabies. All animals were over one year old and randomly selected from two different herds reporting no vaccination for infectious bovine rhinotracheitis in a period exceeding one year. Results indicated seropositivity for BoHV-5 in 91 cases (58.4%), of which 88 were also seropositive for bovine herpes virus 1 (BoHV-1), while 41 were seronegative for both agents. 22/64 seronegative cases for BoHV-5 were seropositive for BoHV-1 and 2/43 seronegative cases for BoHV-1 were seropositive for BoHV-5, and consequently, these animals could be only infected by encephalitis herpes virus. With these initial findings, emphasis its placed on the need establish the true impact of the disease in Colombia and proposes the epidemiological surveillance of cattle in the region studied in order to establish mechanisms for control of viral infection.

O objetivo deste estudo foi o de procurar evidências da transmissão vetorial da doença de Chagas, nos domicílios e peridomicílios de indivíduos residentes em municípios da região de Botucatu, que tiveram xenodiagnóstico positivos. Foram estudados 58 indivíduos e foi coletada amostra do sangue para a realização de exames laboratoriais. Os resultados deste estudo mostraram que os indivíduos, de ambos os grupos, tinham baixa escolaridade e exerciam profissões que não exigiam qualificações técnicas. Houve discreto predomínio de indivíduos do sexo feminino. Quando comparadas às condições anteriores, verificou-se que houve discreta melhora nas condições de habitação, por outro lado, houve aumento de moradores em zona rural. Os indivíduos nascidos antes de 1983, apresentaram conhecimento e contato com triatomídeo estaticamente mais elevado quando comparado com os nascidos a partir 1983. A análise e comparação dos resultados das sorologias, referentes aos hemaglutinação passiva indireta, imunofluorescência indireta e ensaio imunoenzimático, mostrou que o ELISA apresentou maior sensibilidade. Os resultados deste estudo mostram que a população nascida a partir de 1983 não conhecia o vetor transmissor da doença de Chagas.

We examined the types of Epstein-Barr virus-associated nuclear antigen-1 (EBNA-1) gene carboxy (C)-terminal mutations occurring in Hodgkin's disease (HD) and reactive tissues from two different geographic regions. Previously reported EBNA-1 C-terminal region amino acid sequence variants, based on the amino acid at codon 487, include Prototype (P)-ala, which is found in the B95.8-derived prototype virus, P-thr, Variant (V)-leu, V-val, and V-pro. Using polymerase chain reaction (PCR) to amplify portions of the EBNA-1 gene, followed by DNA sequencing, we found a single EBNA-1 gene sequence variant in each tissue, whether reactive or neoplastic and whether from Brazil or the United States. Variant EBNA-1 gene sequences were more common in both neoplastic and non-neoplastic tissues from different geographic areas than the so-called prototype sequence. In the 17 Brazilian HD cases, 4 cases had P-thr variants and 13 had V-leu variants. In the six reactive tissues from Brazil, one had a P-ala variant, two had P-thr variants, and three had V-leu variants. In the 12 American HD cases, 2 had P-ala variants, 6 had P-thr variants, and 4 had V-leu variants. The 11 American reactive tissues included 2 P ala variants, 5 P-thr variants, and 4 V-leu variants. In both countries, there were similar variant EBNA-1 sequences present in normal tissues and HD cases. Compared with the P ala and P-thr cases, the V-leu cases were more likely to have the 30-bp latent membrane protein 1 (LMP1) gene deletion (P = 0.0075). In addition, cases of HD with the V-leu were statistically associated with a substitution of asparagine for glutamine at codon 322 of the C-terminal portion of the LMP1 gene. Our results suggest that any variation in EBNA-1 gene sequence is caused by a polymorphism present in pre-existing viral strains in the underlying population, and not a mutation occurring during oncogenesis. The human buccal micronucleus cytome assay (BMCyt) is one of the most widely used techniques to measure genetic damage in human population studies. Reducing protocol variability, assessing the role of confounders, and estimating a range of reference values are research priorities that will be addressed by the HUMNXL, collaborative study. The HUMNXL, project evaluates the impact of host factors, occupation, life-style, disease status, and protocol features on the occurrence of MN in exfoliated buccal cells. In addition, the study will provide a range of reference values for all cytome endpoints. A database of 5424 subjects with buccal MN values obtained from 30 laboratories worldwide was compiled and analyzed to investigate the influence of several conditions affecting MN frequency. Random effects models were mostly used to investigate MN predictors. The estimated spontaneous MN frequency was 0.74 parts per thousand (95% CI 0.52-1.05). Only staining among technical features influenced MN frequency, with an abnormal increase for non-DNA-specific stains. No effect of gender was evident, while the trend for age was highly significant (p < 0.001). Most occupational exposures and a diagnosis of cancer significantly increased MN and other endpoints frequencies. MN frequency increased in heavy smoking (>= 40 cig/day. FR = 1.37:95% CI 1.03-.82) and decreased with daily fruit consumption (FR = 0.68; 95% CI 0.50-0.91). The results of the HUMNXL, project identified priorities for validation studies, increased the basic knowledge of the assay, and contributed to the creation of a laboratory network which in perspective may allow the evaluation of disease risk associated with MN frequency. Considering the high incidence of dogs with acute bacterial cystitis (BC) and the relationship among inflammation, genotoxicity, and carcinogenesis, we conducted a case-control study comparing the frequency of deoxyribonucleic acid (DNA) lesions assessed by the comet assay between disease-free animals (13 males and 13 females) and cytology-confirmed cases of acute BC (12 males and 12 females), which was mainly caused by Staphylococcus sp. (40%) and Escherichia coli (35%). The results show no increase in DNA damage in cells obtained by bladder washings and no influence of age, sex, and breed due to acute BC. In conclusion, DNA damage was seemingly not associated with the infection by specific bacteria.

This study was carried out to clarify the real role that was played by the budgerigars (Melopsittacus undulatus) in the epidemiological plan, under the perspective of its being an infection source of the Newcastle Disease Virus (NDV). For this, the study used Specific-Pathogen-Free chicks (SPF) that were housed with budgerigars that were inoculated with a pathogenic strain (velogenic viscerotropic) of NDV (EID5o =10815/0.1 mL) pathogenic to chickens, by the ocular-nasal via. Each group was composed by 10 SPF chicks and 5 budgerigars. After 5 days of the inoculation of the budgerigars with NDV, SPF chicks were put together with each group of budgerigars, so that there was a direct contact between both species. Cloaca) swabs and blood samples were collected in both species (budgerigars and SPF chicks) after 13 and 19 days post-challenge, respectively, for genome viral excretion by Reverse Transcription Polymerase Chain Reaction (RTPCR) and antibody's search by the inhibition of hemmaglutination test (HI). Budgerigars did not demonstrate any clinical signs of Newcastle Disease (ND). They were refractory to the clinical disease with the NDV. However, antibody titres from inhibition of Hemagglutination (HI) test were detected 9 and 21 days after challenge. Therefore, it was demonstrated the state of carrier of NDV in this species. In SPF chicks allocated with infected budgerigars, NDV genome was detected 13 and 19 days after challenge. Thus, the transmission of the pathogenic virus from the budgerigars to SPF chicks that were housed together was evident until 19 days of the experimental infection with this pathogen. This reveals the importance of the budgerigars from the epidemiological point of view as a potential source of infection of the NDV to commercial chickens that could be raised near this species.

The aim of this study was to evaluate the importance of vaccination against Newcastle Disease (ND) in lovebirds (Agapornis roseicollis) and to investigate the state of carrier of the virus (NDV) in this species. There were used 48 lovebirds, distributed at random into 4 experimental groups: GI (Ulster 2C strain), Gil (B1 strain), Gill (LaSota strain) and GIV (non-vaccinated group). At 12 months of age, all groups were challenged with a pathogenic virus (NDV) suspension (ElD50 = 1081510.1 mL) and a group of Specific-Pathogen-Free (SPF) chicks were used as control of the virus. Cloaca) swabs from each bird were collected after 9, 14 and 21 days post-challenge for detection of genome viral excretion by Reverse Transcription Polymerase Chain Reaction RT-PCR. Lovebirds of GI, Gil and Gill did not demonstrate any signs of ND. They were refractory to the clinical disease. In lovebirds from the control group, NDV genome was detected 9 and 21 days after challenge. Therefore it was demonstrated the state of carrier of NDV by lovebirds. In birds from the vaccinated groups, genome viral excretion was not detected by RT-PCR. It was also demonstrated the importance of the vaccination in the suppression of the state of virus carrier of ND in lovebirds.

Aiming to improve the diagnosis of canine leishmaniasis (CanL) in an endemic area of the Northwest region of São Paulo State, Brazil, the efficacy of parasitological, immunological and molecular diagnostic methods were studied. Dogs with and without clinical sips of the disease and positive for Leishmania, by direct parasite identification on lymph node smears and/or specific antibody detection by ELISA, were selected for the study. According to the clinical signs, 89 dogs attending the Veterinary Hospital of UNESP in Aracatuba (SP, Brazil) were divided into three groups: symptomatic (36%), oligosymptomatic (22%) and asymptomatic (22%). Twenty-six dogs from an area non-endemic for CanL were used as negative controls (20%). Fine-needle aspiration biopsies (FNA) of popliteal lymph nodes were collected and Diff-Quick (R)-stained for optical microscopy. Direct immumofluorescence, immunocytochemistry and parasite DNA amplification by PCR were also performed. After euthanasia, fragments of popliteal lymph nodes, spleen, bone marrow and liver were collected and processed for HE and immunohistochemistry. Parasite detection by both HE and immunohistochemistry was specifically more effective in lymph nodes, when compared with the other organs. Immunolabeling provided higher sensitivity for parasite detection in the tissues. In the symptomatic group, assay sensitivity was 75.61% for direct parasite search on Diff-Quick (R)-stained FNAs, 92.68% for direct immunofluorescence, 92.68% for immunocytochemistry and 100% for PCR; the corresponding values in the other clinical groups were: 32, 60, 76 and 96% (oligosymptomatic), and 39.13, 73.91, 100 and 95.65% (asymptomatic). Ten young partridges (Rhynchotus rufescens) were vaccinated with the lentogenic strain of Newcastle disease virus. Another eight unvaccinated birds were kept in close contact with the treated flock. Antibodies levels were measured over the course of 3 mo in all birds using the hemagglutination inhibition (HI) test and the liquid-phase blocking enzyme-linked immunosorbent assay (LPB-ELISA). The LPB-ELISA was standardized, and the results were compared with those obtained with the HI test. Antibodies increased after 23 days postvaccination in 16 birds with no side effects as determined by both the HI test and the LPB-ELISA.
