The potato spindle tuber disease was first observed early in the 20th century in the northeastern United States and shown, in 1971, to be incited by a viroid, potato spindle tuber viroid (PSTVd). No wild-plant PSTVd reservoirs have been identified; thus, the initial source of PSTVd infecting potatoes has remained a mystery. Several variants of a novel viroid, designated Mexican papita viroid (MPVd), have now been isolated from Solanum cardiophyllum Lindl. (papita güera, cimantli) plants growing wild in the Mexican state of Aguascalientes. MPVd's nucleotide sequence is most closely related to those of the tomato planta macho viroid (TPMVd) and PSTVd. From TPMVd, MPVd may be distinguished on the basis of biological properties, such as replication and symptom formation in certain differential hosts. Phylogenetic and ecological data indicate that MPVd and certain viroids now affecting crop plants, such as TPMVd, PSTVd, and possibly others, have a common ancestor. We hypothesize that commercial potatoes grown in the United States have become viroid-infected by chance transfer of MPVd or a similar viroid from endemically infected wild solanaceous plants imported from Mexico as germplasm, conceivably from plants known to have been introduced from Mexico to the United States late in the 19th century in efforts to identify genetic resistance to the potato late blight fungus, Phytophthora infestans.

ADPglucose pyrophosphorylase (glucose-1-phosphate adenylyltransferase; ADP:alpha-D-glucose-1-phosphate adenylyltransferase, EC 2.7.7.27) catalyzes a key regulatory step in alpha-glucan synthesis in bacteria and higher plants. We have previously shown that the expression of the cDNA sequences of the potato tuber large (LS) and small (SS) subunits yielded a functional heterotetrameric enzyme capable of complementing a mutation in the single AGP (glgC) structural gene of Escherichia coli. This heterologous complementation provides a powerful genetic approach to obtain biochemical information on the specific roles of LS and SS in enzyme function. By mutagenizing the LS cDNA with hydroxylamine and then coexpressing with wild-type SS in an E. coli glgC- strain, >350 mutant colonies were identified that were impaired in glycogen production. One mutant exhibited enzymatic and antigen levels comparable to the wild-type recombinant enzyme but required 45-fold greater levels of the activator 3-phosphoglycerate for maximum activity. Sequence analysis identified a single nucleotide change that resulted in the change of Pro-52 to Leu. This heterologous genetic system provides an efficient means to identify residues important for catalysis and allosteric functioning and should lead to novel approaches to increase plant productivity.

Figs are rainforest keystone species. Non-strangler figs establish on the forest floor; strangler figs establish epiphytically, followed by a dramatic transition from epiphyte to free-standing tree that kills its hosts. Free-standing figs display vigorous growth and resource demand suggesting that epiphytic strangler figs require special adaptations to deal with resource limitations imposed by the epiphytic environment. We studied epiphytic and free-standing strangler figs, and non-strangler figs in tropical rainforest and in cultivation, as well as strangler figs in controlled conditions. We investigated whether the transition from epiphyte to free-standing tree is characterised by morphological and physiological plasticity. Epiphyte substrate had higher levels of plant-available ammonium and phosphate, and similar levels of nitrate compared with rainforest soil, suggesting that N and P are initially not limiting resources. A relationship was found between taxonomic groups and plant N physiology; strangler figs, all members of subgenus Urostigma, had mostly low foliar nitrate assimilation rates whereas non-strangler figs, in subgenera Pharmacocycea, Sycidium, Sycomorus or Synoecia, had moderate to high rates. Nitrate is an energetically expensive N source, and low nitrate use may be an adaptation of strangler figs for conserving energy during epiphytic growth. Interestingly, significant amounts of nitrate were stored in fleshy taproot tubers of epiphytic stranglers. Supporting the concept of plasticity, leaves of epiphytic Ficus benjamina L. had lower N and C content per unit leaf area, lower stomatal density and 80% greater specific leaf area than leaves of conspecific free-standing trees. Similarly, glasshouse-grown stranglers strongly increased biomass allocation to roots under water limitation. Epiphytic and free-standing F. benjamina had similar average foliar delta C-13, but epiphytes had more extreme values; this indicates that both groups of plants use the C-3 pathway of CO2 fixation but that water availability is highly variable for epiphytes. We hypothesise that epiphytic figs use fleshy stem tubers to avoid water stress, and that nitrate acts as an osmotic compound in tubers. We conclude that strangler figs are a unique experimental system for studying the transition from rainforest epiphyte to tree, and the genetic and environmental triggers involved.

This paper describes the first systematic study of nutritional deficiencies of greater yam (Dioscorea alata). Yam plants (cv. 'Mahoa'a') were propagated from tuber discs and grown in nutrient solution, with nutrients supplied following a modified programmed nutrient-addition method. After an establishment period of four weeks, deficiencies of nitrogen (N), phosphorus (P), potassium (K), calcium (Ca), magnesium (Mg), sulfur (S), iron (Fe), boron (B), manganese (Mn), copper (Cu), zinc (Zn), and molybdenum (Mo) were induced by omitting the relevant nutrient from the solution. Foliar symptoms were recorded photographically. Notably, deficiencies of the mobile macronutrients failed to induce senescence of oldest leaves, while vine growth and younger leaves were affected. Leaf blades of the main stem were sampled in sequence and analyzed chemically, providing the distribution of each nutrient from youngest to oldest leaves in both adequately supplied and deficient plants. The nutrient-concentration profiles, together with the visible symptoms, indicated that little remobilization of mobile macronutrients had occurred. For both macro- and micronutrients, young leaves gave the best separation of nutrient concentrations between well-nourished and deficient plants.

Root and shoot attributes of 12 indigenous perennial accessions of the wild mungbean (Vigna radiata ssp. sublobata) were evaluated in early and late summer sowings in the field in SE Queensland. All but one of the accessions were obtained from the Townsville-Charters Towers region of NE Queensland. In both sowings, the accessions developed thickened tap and lateral roots, the taproot thickening extending to a depth of 0.20-0.30m below the soil surface, depending on accession. The thickened lateral roots emerged from the taproot within 0.10m of the soil surface, and extended laterally up to 1.10 m, remaining close to the soil surface. Differences among the accessions in gross root morphology and phenology were relatively small. There were differences among the accessions in the production of seed, tuberised root, and recovered total plant biomass. Depending on accession and sowing date, the tuberised roots accounted for up to 31% of recovered plant biomass and among accessions, the root biomass was positively correlated with total plant biomass. In contrast, seed biomass represented only a small proportion of recovered plant biomass, up to a maximum of 14%, depending on accession and sowing date. Among accessions, the proportion of seed biomass tended to be negatively correlated with that of tuber biomass. The perennial trait appears to be unique to Australian accessions of wild mungbean obtained from coastal-subcoastal, speargrass-dominant woodlands of NE Queensland. Although the ecological significance of the trait remains conjectural, field observation indicates that it facilitates rapid plant re-growth following early summer rainfall, especially where dry-season. re has removed previous-season above-ground growth.

O potássio é um dos macronutrientes que as plantas absorvem em maior quantidade, influenciando na batateira a produtividade e a qualidade dos tubérculos. A introdução de novas variedades de batata para indústria implica a avaliação prévia da sua adaptação às condições de solo, clima e tecnologia cultural. Neste contexto, instalou-se um ensaio em Salvaterra-de-Magos, em pleno campo de cultivo, com o objetivo de avaliar o efeito de diferentes doses e modo de fraccionamento (à plantação e em cobertura, após a emergência) de potássio (K2O) na batata de indústria “VR0808”. Foram avaliados 5 tratamentos: (K0-0kg/ha, K1-100kg/ha, K2-200kg/ha, K3-100+50+50kg/ha e K4-200+50+50kg/ha). O dispositivo experimental foi o de parcelas totalmente aleatórias, com 4 repetições. Foi possível concluir que, em relação ao rendimento, com excepção do K4-200+50+50kg/ha, não existiram diferenças significativas entre os tratamentos com 0, 100, 200 e 100+50+50kg/ha de K2O. A mobilização do K2O de formas fixas e/ou de origem orgânica poderá explicar este resultado. Observou-se um decréscimo dos teores de Ca ao nível da folha e do teor de matéria seca dos tubérculos com o incremento do potássio.

O azoto é determinante na produtividade e na qualidade da batata para indústria. Com este trabalho pretendeu-se avaliar o efeito de diferentes doses de azoto na variedade de batata “Hermes”, em plena condições de cultivo do Vale do Tejo. Os tratamentos consistiram na aplicação de 0 (N0), 80 (N80), 160 (N160), 240 (N240) e 320 (N320) kg de azoto/ha, na forma de ENTECÒ 26. O ensaio foi instalado em parcelas totalmente aleatórias, com 4 repetições. Foi registado semanal a taxa de cobertura de solo pelo método da grelha até à sua cobertura total. Bissemanalmente, foram recolhidas plantas nos tratamentos N0 (0kgN/ha) e N160 (160kgN/ha) para avaliação do seu desenvolvimento e crescimento. No final do ciclo cultural avaliou-se a produção total, comercial, em peso e número de tubérculos. Avaliou-se ainda o peso específico, os teores em matéria seca dos tubérculos e a percentagem do azoto total na matéria seca das folhas e tubérculos. A adubação azotada influenciou a dimensão do aparelho fotossintético, através do estímulo de ramificações. Verificou-se uma maior produtividade de tubérculos nos tratamentos com maior aplicação de azoto, embora os acréscimos tenham sido menores a partir dos 160kg/ha. O azoto influenciou a percentagem de tubérculos de maior calibre.

Potato crop cycle is relatively short and presents high yield per area; therefore, it is a very demanding culture for available nutrients in the soil solution. Despite its importance and the large number of studies about the crop, there is little research on plant nutrition regarding the use of organomineral fertilizer. This study evaluated potato, cv. Cupid, development and productivity as a function of fertilization with pelletized organomineral fertilizer. The experiment was done in Perdizes, Minas Gerais, in the rainy season of 2014/2015. The experimental design was a randomized blocks, with factorial arrangement of 4 x 2 (doses x management) and a control with mineral fertilizer, with 3 repetitions. Organomineral fertilizer doses were 25, 50, 75 and 100% of the conventional mineral dose, which was 600 kg ha-1 K2SO4, 850 kg ha-1 NH4H2PO4, and 300 kg ha-1 (NH4)2SO4 of topdressing 19 days after planting (DAP). Fertilization managements were with or without topdressing at 19 DAP, when the potato was hilled. Two plants per plot were sampled at 36, 50, 64 and 81 DAP and analyzed for leaf, stem and dry matter contents. DRIS - Diagnosis and Recommendation Integrated System was applied at 36 DAP and the potatoes were harvested 112 DAP and subjected to tuber classification. Throughout the cycle, stem, leaf and tuber dry mass showed no significant differences between the fertilization managements. The doses of organomineral fertilizer and topdressing management does not affect productivity, and the lower doses (25%) were similar the greater ones and the control, with an average of 16.8 t ha-1, demonstrating that it is viable to make a single application of organomineral fertilizer at planting due to operational efficiency. The low yields observed were due to high rainfall and temperature, creating favorable conditions for the incidence of pests and diseases. According to DRIS, the organomineral dose 75% for topdressing, presented the best nutritional balance.

Cat’s claw creeper vine, Dolichandra unguis-cati (L.) Lohmann (syn. Macfadyena unguis-cati (L.) Gentry) (Bignoniaceae), is a major environmental weed in Australia. Two distinct forms of this weed (‘long’ and ‘short’ pod), with differences in leaf morphology and fruit size, occur in Australia. The long pod form has only been reported in less than fifteen localities in the whole of south-east Queensland, while the short pod form is widely distributed in Queensland and New South Wales. This study sought to compare growth traits such as specific leaf area, relative growth rate, stem length, shoot/root ratio, tuber biomass and branching architecture between these forms. These traits were monitored under glasshouse conditions over a period of 18 months. Short pod exhibited higher values of relative growth rates, stem length, number of tubers and specific leaf area than long pod, but only after 10 months of plant growth. Prior to this, long and short pod did not differ significantly. Higher values for these traits have been described as characteristics of successful colonizers. Results from this study could partly explain why the short pod form is more widely distributed in Australia while long pod is confined to a few localities.

Apart from morphology and genetic characteristics, species status of Pythium zingiberis and P. myriotylum may also be confirmed based on their pathogenicity and host range. An Australian putative P. zingiberis isolate and imported type isolates of P. myriotylum and P. zingiberis were subject to both in vitro and in vivo pathogenicity tests. In vitro tests were carried out on excised carrot, ginger, potato, radish, and sweet potato tuber/root sections, and on seeds and seedlings of cucumber, cauliflower, millet, rye, sweet corn, tomato, and wheat. In all assays conducted, the Australian isolate was found to be the most pathogenic, followed by type specimen of P. zingiberis (UOP 275), and then the type specimen P. myriotylum (CBS 254.70). An in vivo experiment on ginger plants at 35°C (with 10 h day light) in quarantine conditions showed that the ginger plants inoculated with the Australian isolate and also the type specimen of P. zingiberis died at 21 days after inoculation, whereas those inoculated with P. myriotylum CBS 254.70 were still green and healthy. Along with cardinal growth rate, the Australian isolate was confirmed to be closely related to P. zingiberis. This is also the first direct comparison in pathogenicity of P. zingiberis and P. myriotylum.

Helcystogramma sp., es una nueva especie de microlepidóptero que afecta desde la siembra hasta la cosecha del boniato o camote (Ipomoea batatas L. Lam.) en Cuba. Debido a la falta de conocimientos y a la escasa bibliografía relacionada con las especies del género Helcystogramma en nuestro país, nos propusimos como objetivo conocer aspectos bio-ecológicos de este microlepidóptero, con la finalidad de proporcionar nuevos conocimientos para un futuro manejo de esta plaga. El trabajo se realizó desde septiembre 2013 a septiembre de 2014, en el Instituto de Investigaciones de Viandas Tropicales (INIVIT), Santo Domingo, Villa Clara, Cuba. Los insectos se estudiaron por observación directa en campos de I. batatas. La larva de este microlepidóptero es muy ágil, dobla la hoja, la cual pega con hilos de seda, creando una protección para su desarrollo hasta el estado de pupa, se alimenta dentro de esta cavidad a partir de la misma hoja, sin afectar la epidermis inferior. Sus poblaciones han aumentado considerablemente, llegando a 20 larvas/m2 en verano. La especies Helcystogramma sp., se cita por primera vez para Cuba y además se registra como una nueva especie de insecto que se alimenta del boniato. ABSTRACT. Primer registro de Helcystogramma sp. (Lepidoptera: Gelechiidae) en Cuba. Helcystogramma sp. is a new species of microlepidoptera affecting from planting to sweet potato (I. batatas L. Lam.) harvest crop in Cuba. Due to the lack of knowledgze and limited literature relate with the Helcystogramma genus in our country, the goal of this study was to know the bioecological aspects of such microlepidoptera, in order to provide new insights for future management of this pest. The study was conducted from September 2013 to September 2014 at the Research Institute of Tropical Root and Tuber Crops, Bananas and Plantains (INIVIT), Santo Domingo, Villa Clara, Cuba. Insects were studied by direct observation in sweet potato (I. batatas) fields. The microlepidoptera larvae is very agile, folds the sheet which sticks with silk threads, creating a protection for its development to the pupal stage. The larvae will feed in the cavity from the same leaf without affecting the lower epidermis. Their populations have increased significantly, reaching 20 larvae / m2 in summer. The species Helcystogramma sp., is cited for the first time in Cuba and it is reported as a new species of insect that feeds on sweet potato leaves.
