Transcriptional regulation is fundamental for the precise development of all organisms. Through tight regulation, necessary genes are activated at proper spatial and temporal patterns, while unnecessary genes are repressed. A large family of regulator proteins that have been demonstrated to be involved in various developmental processes by activation and repression of target genes is the homeodomain family of proteins. To date, the function of many of these homeoproteins has been elucidated in diverse species. However, the molecular mechanism underlying the function of these proteins has not been fully understood. In this study, the molecular mechanism of the function of a LIM-homeoprotein, Lim1, was examined. In addition to the homeodomain, Lim1 contains two LIM domains that are highly conserved among species. This high conservation along with data from in vitro studies on Xenopus Lim1 suggests that the LIM domains might be important for the function of Lim1 as a transcriptional regulator. Here, the functional importance of the LIM domains of Lim1 was determined by using a novel gene-targeting strategy in mouse embryonic stem (ES) cells. A cre-loxP system was used in conjunction with the unique genomic organization of Lim1 to obtain four types of mutant ES cell lines that would allow for the in vivo analysis of the function of both the LIM domains of Lim1 together and also singularly. These four mutant Lim1 alleles either contained base-pair changes at the LIM encoding exons that alters zinc-binding amino acids of the LIM domains or contained only exogenous loxP sequences in the first intron of Lim1, which serves as the control allele. These mutations in the LIM domains would presumably abolish the zinc-finger tertiary structure of the domain and thus render the domain non-functional. Mice carrying mutations at both the LIM domains of Lim1, L1L2, die around E10 without anterior head structures anterior to rhombomere 3, identical in phenotype to the Lim1 null mutants in spite of the presence of mutant Lim1 RNA. This result demonstrates that the integrity of both the LIM domains are essential for the function of Lim1. This is further supported by the phenotype of mice carrying mutation at only the second LIM domain of Lim1, L2. The L2 mice although still carrying one intact Lim1 LIM domain, also die in utero. The L2 mice die at varying times, from around E8 to E10 with anterior defects in addition to other axial defects which have yet to be fully characterized. The results of this study so far demonstrates that the integrity of both LIM domains are required for the function of Lim1. ^

The etiology of the vast majority of osteosarcoma deaths has not been explained. A possible explanation might be lifetime ingestion of radium from environmental sources which might give rise to differential risk. This study was an effort toward understanding the role of naturally occurring radium in the etiology of bone cancer. Furthermore, there was an interest in the interaction of between radionuclides and selenium; the latter believed to be a potential anticarcinogen.^ Two approaches were used to evaluate the association between environmental radium, selenium and osteogenic sarcoma: (1) spatial and temporal patterns of osteogenic sarcoma mortality in Texas were described for the period from 1969 to 1988; and (2) a case-control study was performed using 974 osteosarcoma deaths and category-matched controls selected from other deaths to evaluate the association between this disease and residency history as an indirect measure of radium and selenium exposures.^ Analyses and comparison of mortality in a population exposed to regions of elevated levels of radium 226,228 and elevated levels of selenium in drinking water with those in a matched control population have resulted in three observations: (1) there appeared to be a slight protective effect for residing in areas high in radium; (2) there were no significant differences between cases and controls when observed for length of residence or residence in urban/rural regions of high or low radium; and (3) although regions high in selenium appeared to have a decreased risk for bone cancer and urban areas in regions of elevated selenium showed an increased risk of bone cancer, these differences were not significant. ^

Wildfires are part of the Mediterranean ecosystem, however, in Israel all wildfires are human caused, either intentionally or un-intentionally. In this study we aimed to develop and test a new method for mapping fire scars from MODIS imagery, to examine the temporal and spatial patterns of wildfires in Israel in the 2000s and to examine the factors controlling Israel's wildfire regime. To map the fires we used two 'off-the-shelf' MODIS fire products as our basis-the 1 km MODIS Collection 5 fire hotspots, the 500 m MCD45A1 burnt areas-and we created a new set of fire scar maps from the 250 m MOD13Q1 product. We carried out a cross comparison of the three MODIS based wildfire scar maps and evaluated them independently against the wild fire scars mapped from 30 m Landsat TM imagery. To examine the factors controlling wildfires we used GIS layers of rainfall, land use, and a Landsat-based national vegetation map. Wildfires occurred in areas where annual rainfall was above 250 mm, mostly in areas with herbaceous vegetation. Wildfire frequency was especially high in the Golan Heights and in the foothills of the Judean mountains, and a high correspondence was found between military training zones and the spatial distribution of fire scars. The use of MODIS satellite images enabled us to map wildfires at a national scale due to the high temporal resolution of the sensor. Our MOD13Q1 based mapping of fire scars adequately mapped large (>1 km**2) fires with accuracies above 80%. Such large fires account for a large proportion of all fires, and pose the greatest threats. This database can aid managers in determining wildfire risks in space and in time.

Large-scale environmental patterns in the Humboldt Current System (HCS) show major changes during strong El Niño episodes, leading to the mass mortality of dominant species in coastal ecosystems. Here we explore how these changes affect the life-history traits of the surf clam Mesodesma donacium. Growth and mortality rates under normal temperature and salinity were compared to those under anomalous (El Niño) higher temperature and reduced salinity. Moreover, the reproductive spatial-temporal patterns along the distribution range were studied, and their relationship to large-scale environmental variability was assessed. M. donacium is highly sensitive to temperature changes, supporting the hypothesis of temperature as the key factor leading to mass mortality events of this clam in northern populations. In contrast, this species, particularly juveniles, was remarkably tolerant to low salinity, which may be related to submarine groundwater discharge in Hornitos, northern Chile. The enhanced osmotic tolerance by juveniles may represent an adaptation of early life stages allowing settlement in vacant areas at outlets of estuarine areas. The strong seasonality in freshwater input and in upwelling strength seems to be linked to the spatial and temporal patterns in the reproductive cycle. Owing to its origin and thermal sensitivity, the expansion and dominance of M. donacium from the Pliocene/Pleistocene transition until the present seem closely linked to the establishment and development of the cold HCS. Therefore, the recurrence of warming events (particularly El Niño since at least the Holocene) has submitted this cold-water species to a continuous local extinction-recolonization process.

The Yangtze River Basin downstream of China's Three Gorges Dam (TGD) (thereafter referred to as "downstream" basin) hosts the largest cluster of freshwater lakes in East Asia. These lakes are crucial water stocks to local biophysical environments and socioeconomic development. Existing studies document that individual lakes in this region have recently experienced dramatic changes under the context of enduring meteorological drought, continuous population growth, and extensive water regulation since TGD's initial impoundment (i.e., June, 2003). However, spatial and temporal patterns of lake dynamics across the complete downstream Yangtze basin remain poorly characterized. Using daily MODIS imagery and an advanced thematic mapping scheme, this study presents a comprehensive monitoring of area dynamics in the downstream lake system at a 10-day temporal resolution during 2000-2011. The studied lakes constitute ~76% (~11,400 km**2) of the total downstream lake area, including the entire +70 major lakes larger than 20 km**2. The results reveal a decadal net decline in lake inundation area across the downstream Yangtze Basin, with a cumulative decrease of 849 km**2 or 7.4% from 2000 to 2011. Despite an excessive precipitation anomaly in the year 2010, the decreasing trend was tested significant in all seasons. The most substantial decrease in the post-TGD period appears in fall (1.1%/yr), which intriguingly coincides with the TGD water storage season. Regional lake dynamics exhibit contrasting spatial patterns, manifested as evident decrease and increase of aggregated lake areas respectively within and beyond the Yangtze Plain. This contrast suggests a marked vulnerability of lakes in the Yangtze Plain, to not only local meteorological variability but also intensified human water regulations from both the upstream Yangtze main stem (e.g., the TGD) and tributaries (e.g., lakes/reservoirs beyond the Yangtze Plain). The produced lake mapping result and derived lake area dynamics across the downstream Yangtze Basin provides a crucial monitoring basis for continuous investigations of changing mechanisms in the Yangtze lake system.

The oocyte nuclear antigen of the monoclonal antibody 32-5B6 of Xenopus laevis is subject to regulated nuclear translocation during embryogenesis. It is distributed in the cytoplasm during oocyte maturation, where it remains during cleavage and blastula stages, before it gradually reaccumulates in the nuclei during gastrulation. We have now identified this antigen to be the enzyme S-adenosylhomocysteine hydrolase (SAHH). SAHH is the only enzyme that cleaves S-adenosylhomocysteine, a reaction product and an inhibitor of all S-adenosylmethionine-dependent methylation reactions. We have compared the spatial and temporal patterns of nuclear localization of SAHH and of nuclear methyltransferase activities during embryogenesis and in tissue culture cells. Nuclear localization of Xenopus SAHH did not temporally correlate with DNA methylation. However, we found that SAHH nuclear localization coincides with high rates of mRNA synthesis, a subpopulation colocalizes with RNA polymerase II, and inhibitors of SAHH reduce both methylation and synthesis of poly(A)+ RNA. We therefore propose that accumulation of SAHH in the nucleus may be required for efficient cap methylation in transcriptionally active cells. Mutation analysis revealed that the C terminus and the N terminus are both required for efficient nuclear translocation in tissue culture cells, indicating that more than one interacting domain contributes to nuclear accumulation of Xenopus SAHH.

Os reservatórios urbanos estão suscetíveis a uma variedade de interferências antropogênicas que acarretam grande variabilidade espacial e temporal. Contudo, possuem uma dinâmica própria na qual o hidroclima e micro e macro-eventos meteorológicos atuam sobre os processos físicos, químicos e biológicos resultando em respostas particulares de cada corpo de água. No presente estudo a existência de padrões espaciais e temporais na formação de florescimentos de algas, cianobactérias e macrófitas no reservatório Guarapiranga, São Paulo, SP, foi avaliada por meio de experimento de curta escala de tempo durante o evento da entrada de uma frente fria. Foram amostrados 64 pontos em todo o reservatório, e o estudo intensivo de florescimento algal e de cianobactérias em dois ciclos nictemerais, em um ponto selecionado no reservatório. Um modelo tridimensional de hidrodinâmica foi aplicado ao estudo compartimentalizado dos tempos de residência e imagens de satélite foram analisadas para determinação de padrões temporais e espaciais durante períodos de tempo mais amplos. Os resultados revelaram que os períodos mais favoráveis ao surgimento de florescimentos de cianobactérias são geralmente os meses mais quentes, de dezembro e janeiro, ou aqueles em que ocorrem estratificações mais fortes como no fim do inverno, em julho, e após as primeiras chuvas nos meses de setembro e outubro. Existem padrões espaciais recorrentes na formação dos florescimentos, controlados em grande parte pela ação do vento, que no reservatório Guarapiranga é predominantemente nas direções leste e sudeste empurrando os florescimentos na direção da foz dos tributários Embu Mirim e Embu Guaçu e ocasionalmente na direção da foz do rio Parelheiros. As simulações hidrodinâmicas evidenciam as forçantes que determinam os padrões observados e reforçam a importância de se discretizarem os tempos de residência de diferentes compartimentos do reservatório. As séries temporais amplas permitiram a determinação da qualidade da água em cada região e fornecem subsídios para o futuro manejo do reservatório. Como esse comportamento não se restringe ao reservatório Guarapiranga, o tipo de modelagem aqui utilizada pode ser útil para obter informações importantes no processo de planejamento e seleção de medidas para o gerenciamento de reservatórios urbanos tropicais polimíticos, em geral.

The Amon mud volcano (MV), located at 1250 m water depth on the Nile Deep Sea Fan, is known for its active emission of methane and non-methane hydrocarbons into the hydrosphere. Previous investigations showed a low efficiency of hydrocarbon-degrading anaerobic microbial communities inhabiting the Amon MV center in the presence of sulphate and hydrocarbons in the seeping subsurface fluids. By comparing spatial and temporal patterns of in situ biogeochemical fluxes, temperature gradients, pore water composition and microbial activities over three years, we investigated why the activity of anaerobic hydrocarbon degraders can be low despite high energy supplies. We found that the central dome of the Amon MV, as well as a lateral mud flow at its base, showed signs of recent exposure of hot subsurface muds lacking active hydrocarbon degrading communities. In these highly disturbed areas, anaerobic degradation of methane was less than 2% of the methane flux. Rather high oxygen consumption rates compared to low sulphide production suggest a faster development of more rapidly growing aerobic hydrocarbon degraders in highly disturbed areas. In contrast, the more stabilized muds surrounding the central gas and fluid conduits hosted active anaerobic hydrocarbon-degrading microbial communities. Furthermore, within three years, cell numbers and hydrocarbon degrading activity increased at the gas-seeping sites. The low microbial activity in the hydrocarbon-vented areas of Amon mud volcano is thus a consequence of kinetic limitations by heat and mud expulsion, whereas most of the outer mud volcano area is limited by hydrocarbon transport.

Increasingly, large areas of native tropical forests are being transformed into a mosaic of human dominated land uses with scattered mature remnants and secondary forests. In general, at the end of the land clearing process, the landscape will have two forest components: a stable component of surviving mature forests, and a dynamic component of secondary forests of different ages. As the proportion of mature forests continues to decline, secondary forests play an increasing role in the conservation and restoration of biodiversity. This paper aims to predict and explain spatial and temporal patterns in the age of remnant mature and secondary forests in lowland Colombian landscapes. We analyse the age distributions of forest fragments, using detailed temporal land cover data derived from aerial photographs. Ordinal logistic regression analysis was applied to model the spatial dynamics of mature and secondary forest patches. In particular, the effect of soil fertility, accessibility and auto-correlated neighbourhood terms on forest age and time of isolation of remnant patches was assessed. In heavily transformed landscapes, forests account for approximately 8% of the total landscape area, of which three quarters are comprised of secondary forests. Secondary forest growth adjacent to mature forest patches increases mean patch size and core area, and therefore plays an important ecological role in maintaining landscape structure. The regression models show that forest age is positively associated with the amount of neighbouring forest, and negatively associated with the amount of neighbouring secondary vegetation, so the older the forest is the less secondary vegetation there is adjacent to it. Accessibility and soil fertility also have a negative but variable influence on the age of forest remnants. The probability of future clearing if current conditions hold is higher for regenerated than mature forests. The challenge of biodiversity conservation and restoration in dynamic and spatially heterogeneous landscape mosaics composed of mature and secondary forests is discussed. (c) 2004 Elsevier B.V. Present paper is a review on the Ráckeve-Soroksár Danube in ecological standpoint. The goal of this study is to collect and evaluate all of available publications in that conception, concerning this Danube arm. Phytoplankton, zooplankton, macroinvertebrates, vertebrates, macrophytes and also water chemistry, water management, geographical description are presented. The review comprises the main studies beginning with the earliest faunistic publications up to the recent ecological, multidisciplinary investigations. Spatial and temporal patterns likewise water quality are considered as important. Additionally checklist of aquatic invertebrate and vertebrate fauna are given based on data from literature.

This study aimed to describe the spatial and temporal patterns of variation in trophic state and its relation with the structure and dynamics of planktonic community of a large reservoir located in semi-arid tropical region of Northeast Brazil. The reservoir Armando Ribeiro Gonçalves is the biggest reservoir of the Rio Grande do Norte State and is responsible for about 53% of all surface water accumulated in the State. The samples of water and plankton were taken monthly in 10 points distributed throughout the longitudinal axis of the reservoir and over a full hydrological cycle. The samples were collected to determine concentrations of total phosphorus, total nitrogen, chlorophyll a and suspended solids (fixed and volatile) and for determining the composition and abundance of phytoplanktonic and zooplanktonic species. During the study period, the reservoir was characterized as eutrophic and there was no trend of increasing the trophic state of the reservoir in the period of drought. The concentrations of total phosphorus and suspended fixed solids decreased towards the dam while the N:P ratios increased in the same direction due to the reduction in the phosphorus concentrations and relative constancy in the nitrogen concentrations. The N:P ratios observed were indicative of greater limitation by phosphorus than by nitrogen. However, as concentrations of both nutrients were high and the water transparency was very low, with secchi depth usually lower than one meter, it seems likely that the planktonic primary production of the reservoir is more limited by the availability of light than the availability of nutrients. High nutrient concentrations coupled with low availability of light may explain the continuing dominance of filamentous cyanobacteria such as Cylindrospermopsis raciborskii in the plankton of the reservoir.These cyanobacteria are potentially toxic and pose a serious environmental problem because it compromises the water quality for public supply, recreation and fishing when present in high densities as in this study. The mesozooplankton of the reservoir was dominated by the calanoid Notodiaptomus cearensis and the cladoceran Diaphanosoma spinulosum. In general, the structure of zooplankton community seems to be particularly influenced by the spatial variation of cyanobacteria. The results of the regression analyses show that both the chlorophyll a concentrations and the cyanobacteria biovolume were more strongly correlated with the nitrogen than with phosphorus and that the water transparency was more strongly correlated with algal biomass than with other sources of turbidity. The maximum load of phosphorus to attain the maximum permissible concentration of total P in the reservoir was estimated in 63.2 tonnes P/ year. The current external P load to the reservoir is estimated in 324 tonnes P / year and must be severely reduced to improve the water quality for water supply and allow the implementation of aquaculture projects that could contribute to the socio-economic development of the region
