Development of dairy organic probiotic fermented products is of great interest as they associate ecological practices and benefits of probiotic bacteria. As organic management practices of cow milk production allow modification of the fatty acid composition of milk (as compared to conventional milk), we studied the influence of the type of milk on some characteristics of fermented milks, such as acidification kinetics. bacterial counts and fatty acid content. Conventional and organic probiotic fermented milks were produced using Bifidobacterium animalis subsp. lactis HN019 in co-culture with Streptococcus thermophilus TA040 and Lactobacillus delbrueckii subsp. bulgaricus LB340. The use of organic milk led to a higher acidification rate and cultivability of Lactobacillus bulgaricus. Fatty acids profile of organic fermented milks showed higher amounts of trans-octadecenoic acid (C18:1, 1.6 times) and polyunsaturated fatty acids, including cis-9 trans-11. C18:2 conjugated linoleic (CLA-1.4 times), and alpha-linolenic acids (ALA-1.6 times), as compared to conventional fermented milks. These higher levels were the result of both initial percentage in the milk and increase during acidification, with no further modification during storage. Finally, use of bifidobacteria slightly increased CLA relative content in the conventional fermented milks, after 7 days of storage at 4 degrees C, whereas no difference was seen in organic fermented milks. Daily intake of conjugated linoleic acid (CLA) has been shown to reduce body fat accumulation and to increase body metabolism; this latter effect has been often associated with the up-regulation of uncoupling proteins (UCPs). Here we addressed the effects of a CLA-supplemented murine diet (similar to 2 % CLA mixture, cis-9, trans-10 and trans-10, cis-12 isomers; 45 % of each isomer on alternating days) on mitochondrial energetics, UCP2 expression/activity in the liver and other associated morphological and functional parameters, in C57BL/6 mice. Diet supplementation with CLA reduced both lipid accumulation in adipose tissues and triacylglycerol plasma levels, but did not augment hepatic lipid storage. Livers of mice fed a diet supplemented with CLA showed high UCP2 mRNA levels and the isolated hepatic mitochondria showed indications of UCP activity: in the presence of guanosine diphosphate, the higher stimulation of respiration promoted by linoleic acid in mitochondria from the CLA mice was almost completely reduced to the level of the stimulation from the control mice. Despite the increased generation of reactive oxygen species through oxi-reduction reactions involving NAD(+)/NADH in the Krebs cycle, no oxidative stress was observed in the liver. In addition, in the absence of free fatty acids, basal respiration rates and the phosphorylating efficiency of mitochondria were preserved. These results indicate a beneficial and secure dose of CLA for diet supplementation in mice, which induces UCP2 overexpression and UCP activity in mitochondria while preserving the lipid composition and redox state of the liver.

In functional dairy products, polyunsaturated fatty acids such as, conjugated linoleic acid (CLA) and alpha-linolenic acid (ALA) have been highlighted for their benefits related to prevention of some chronic diseases. In order to study the effect of type of milk (conventional vs. organic, characterized by a specific fatty acid composition), Bifidobacterium animalis subsp. lactis (BB12, B94, BL04 and HN019) counts, acidification activity and chemical composition (pH, lactose, lactic acid contents and fatty acids profile) were investigated before fermentation and after 24 h of products stored at 4 degrees C. Organic and conventional milk influenced acidification performance and bacteria counts, which was strain-dependent. Higher counts of BB12 were observed in organic milk, whereas superior counts of BL04 were found in conventional milk. Organic fermented milk showed lower levels in saturated fatty acids (FA) and higher in monounsaturated FA contents. Similarly, among bioactive FA, organic fermented milks have higher amounts of trans vaccenic acid (TVA-C18:1t), conjugated linoleic acid (CLA) and slightly higher contents of alpha-linoleic acid (ALA). The aim of this study was to determine and compare the fatty acid (FA) composition of colostrum and mature milk produced by nursing mothers of preterm and at-term newborns, in Florianopolis, SC, Brazil. Low contents of Eicosapentaenoic acid (EPA) (0.02%/colostrum and 0.01%/mature milk for preterm and term milk) and Docosahexaenoic acid (DHA) (colostrum group: 0.10%/preterm and 0.09%/term; mature milk: 0.05%/preterm and 0.03%/term) were determined. The comparison among the groups showed that the elaidic acid content was significantly higher (1.67%) in mature term milk. The content of rumenic acid (conjugated linoleic acid) was significantly higher in at-term colostrum compared with preterm colostrum. When considering the maturity of the milk, there was a significant increase in the percentage of this FA in the preterm group. The results show that, overall, the greatest differences observed were between the colostrums and mature milks for both groups and not between preterm and at-term mothers.

The objective in this study was to determine growth, carcass characteristics, chemical composition and fatty acid profile of the longissimus dorsi of crossbred Boer x Saanen kids fed castor oil. Twenty-four kids (12 males and 12 females) were assigned in a randomized complete block design with two treatments and twelve replications. Blocks were defined according to weight, gender and initial age of animals for the evaluation of performance. The experimental treatments consisted of two diets containing 900 g concentrate/kg: a control diet (without addition of oil) and another containing castor oil at 30 g/kg (on a dry matter basis). After they reached an average body weight of 25 kg, males were slaughtered for the evaluation of carcass characteristics, chemical composition and fatty acid profile of the longissimus dorsi muscle. The addition of castor oil in the diet did not affect the intake of dry matter, crude protein and neutral detergent fiber; the average daily gain; and feed conversion, but increased the ether extract intake. No difference was observed for the carcass characteristics, chemical composition of the meat, concentration of C18:2 cis-9, trans-11 (CLA) and total concentration of saturated, monounsaturated and polyunsaturated fatty acids and their relations; however, there was increase in the concentrations of C18:2 trans-10, cis-12 (CLA) and C20:4 omega-6. The addition of castor oil to the diet of crossbred Boer x Saanen kids containing a high content of concentrate did not promote benefit to the characteristics evaluated.

Conjugated linoleic acid (CLA) is a class of 28 positional and geometric isomers of linoleic acid octadecadienoic.Currently, it has been described many benefits related to the supplementation of CLA in animals and humans, as in the treatment of cancer, oxidative stress, in atherosclerosis, in bone formation and composition in obesity, in diabetes and the immune system. However, our results show that, CLA appears to be not a good supplement in patients with cachexia.

OBJETIVO: Avaliar os fatores de risco associados à ausência de aleitamento materno exclusivo (AME) em crianças <6 meses de vida na cidade de São Paulo, em 2008. MÉTODOS: Aplicou-se o questionário do Projeto Amamentação e Municípios-1998 (AMAMUNIC) a pais/responsáveis de crianças <6 meses de idade durante a Campanha Nacional de Vacinação contra Poliomielite. Cálculo da amostra por conglomerados com sorteio em dois estágios. Os fatores analisados foram idade e educação materna, peso de nascimento, sexo, tipo de parto, nascer em Hospital Amigo da Criança, presença de aleitamento materno precoce, uso de chupeta nas últimas 24 horas e mãe trabalhando fora de casa. Análise estatística por regressão logística binária com SPSS, versão 15.0, sendo significante p<0,05. RESULTADOS: Foram realizadas 724 entrevistas, das quais 275 referiram (39,1%) aleitamento materno exclusivo (Grupo I - GI) e 429 (60,9%) sem aleitamento materno exclusivo (Grupo II - GII). Houve diferenças entre os grupos quanto ao uso da chupeta nas últimas 24 horas (GI 32,3 versus GII 59,8%; p<0.001), mães trabalhando fora (GI 12,4 versus GII 24,8%; p<0.001) e idade da criança (GI 74,1±45,3 versus GII 105,8±49,5 dias; p<0,0001).Na análise multivariada, houve associação entre ausência de aleitamento materno exclusivo e uso de chupeta (OR 3,02; IC95% 2,10-4,36), mãe trabalhando fora (OR 2,11; IC95% 1,24-3,57) e idade da criança (OR 1,01; IC95% 1,01-1,02). CONCLUSÕES: O uso da chupeta nas últimas 24 horas associou-se à ausência de AME em crianças menores do que seis meses, seguido pelo trabalho materno fora de casa e pela idade da criança, que são importantes fatores a serem controlados em programas de promoção do aleitamento materno.

The aim of the first part of this thesis was to evaluate the effect of trans fatty acid- (TFA), contaminant, polycyclic aromatic hydrocarbon (PAH)- and oxidation productenriched diets on the content of TFA and conjugated linoleic acid (CLA) isomers in meat and liver of both poultry and rabbit. The enriched feedings were prepared with preselected fatty co-and by-products that contained low and high levels of TFA (low, palm fatty acid distillate; high, hydrogenated palm fatty acid distillate), environmental contaminants (dioxins and PCBs) (two different fish oils), PAH (olive oil acid oils and pomace olive oil from chemical refining, for low and high levels) and oxidation products (sunflower-olive oil blend before and after frying), so as to obtain single feedings with three enrichment degrees (high, medium and low) of the compound of interest. This experimental set-up is a part of a large, collaborative European project (http://www.ub.edu/feedfat/), where other chemical and health parameters are assessed. Lipids were extracted, methylated with diazomethane, then transmethylated with 2N KOH/methanol and analyzed by GC and silver-ion TLC-GC. TFA and CLA were determined in the fats, the feedings, meat and liver of both poultry and rabbit. In general, the level of TFA and CLA in meat and liver mainly varied according to those originally found in the feeding fats. It must be pointed out, though, that TFA and CLA accumulation was different for the two animal species, as well as for the two types of tissues. The TFA composition of meat and liver changes according to the composition of the oils added to the feeds with some differences between species. Chicken meat with skin shows higher TFA content (2.6–5.4 fold) than rabbit meat, except for the “PAH” trial. Chicken liver shows higher TFA content (1.2–2.1 fold) than rabbit liver, except for the “TRANS” and “PAH” trials. In both chicken and rabbit meats, the TFA content was higher for the “TRANS” trial, followed by the “DIOXIN” trial. Slight differences were found on the “OXIDATION” and “PAH” trends in both types of meats. In both chicken and rabbit livers, the TFA content was higher for the “TRANS” trial, followed by those of the “PAH”, “DIOXIN” and “OXIDATION” trials. This trend, however, was not identical to that of feeds, where the TFA content varied as follows: “TRANS” > “DIOXIN” >“PAH” > “OXIDATION”. In chicken and rabbit meat samples, C18:1 TFA were the most abundant, followed by C18:2 TFA and C18:3 TFA, except for the “DIOXIN” trial where C18:3 TFA > C18:2 TFA. In chicken and rabbit liver samples of the “TRANS” and “OXIDATION” trials, C18:1 TFA were the most abundant, followed by C18:2 TFA and C18:3 TFA, whereas C18:3 TFA > C18:2 in the “DIOXIN” trial. Slight differences were found on the “PAH” trend in livers from both species. The second part of the thesis dealt with the study of lipid oxidation in washed turkey muscle added with different antioxidants. The evaluation on the oxidative stability of muscle foods found that oxidation could be measured by headspace solid phase microestraction (SPME) of hexanal and propanal. To make this method effective, an antioxidant system was added to stored muscle to stop the oxidative processes. An increase in ionic strength of the sample was also implemented to increase the concentration of aldehydes in the headspace. This method was found to be more sensitive than the commonly used thiobarbituric acid reactive substances (TBARs) method. However, after antioxidants were added and oxidation was stopped, the concentration of aldehydes decreased. It was found that the decrease in aldehyde concentration was due to the binding of the aldehydes to muscle proteins, thus decreasing the volatility and making them less detectable.

This PhD thesis describes the application of some instrumental analytical techniques suitable to the study of fundamental food products for the human diet, such as: extra virgin olive oil and dairy products. These products, widely spread in the market and with high nutritional values, are increasingly recognized healthy properties although their lipid fraction might contain some unfavorable components to the human health. Zusammenfassung In der vorliegenden Arbeit wurden unterschiedliche Eigenschaften von tandem repetitiver DNA (trDNA) analysiert. Drei der untersuchten trDNA-Familien (Cla-Elemente, Alu-Elemente, 1,688-Satelliten-DNA) stammen aus der genomischen DNA von Insekten, während ein trDNA-Cluster artifiziell aus Hsp70 Promotoren hergestellt wurde. Untersucht wurde die Stabilität dieser trDNA-Sequenzfamilien innerhalb eines Plasmidvektors in E. coli bzw. nach Integration der trDNA auch im Genom von D. melanogaster. Der Schwerpunkt der Arbeit liegt jedoch in der Analyse des Einflusses von trDNA auf die Expression eines benachbarten Reportergens in D. melanogaster.Ziel der Untersuchungen zur Stabilität war es, Eigenschaften von trDNA-Clustern und Mechanismen aufzuzeigen, die die Stabilität derselben in E. coli und im Genom von D. melanogaster beeinflussen. Mit Ausnahme der Alu-Elemente zeigen alle trDNA-Familien eine deutliche Instabilität in E. coli. Am Beispiel der Cla-Elemente wurde gezeigt, daß spezifische Eigenschaften der trDNA-Familie, wie etwa die sequenzbedingte Krümmung der Helixachse, keinen Einfluß auf die Stabilität des trDNA-Clusters haben, sondern die Orientierung des trDNA-Clusters innerhalb des Vektors ausschlaggebend ist. Ein entscheidender Faktor könnte die Orientierung des trDNA-Clusters relativ zur Wanderungsrichtung der Replikationsgabel in E. coli sein. Am Beispiel des trDNA-Clusters aus artifiziellen Hsp70 Promotoren konnte gezeigt werden, daß verschiedene Rekombinationssysteme an der Instabilität in E. coli beteiligt sind. Die meisten beobachteten Deletionen von trDNA sind RecA-abhängig. Zusätzlich findet jedoch in einem kleinen Teil der Plasmide auch eine RecA-unabhängige Rekombination statt. Sowohl in E. coli als auch in D. melanogaster wurde als vorherrschender Mechanismus der trDNA-Instabilität die homologe Rekombination identifiziert. TrDNA-Cluster, die in E. coli deutlich instabil sind, können jedoch im Genom von D. melanogaster weitgehend stabil sein. Auch Faktoren, die in E. coli die Stabilität eines trDNA-Clusters beeinflussen, wie etwa die Orientierung, zeigen in D. melanogaster keinen Einfluß auf die Stabilität der trDNA-Cluster. Ergebnisse aus Stabilitätsuntersuchungen in E. coli können damit nicht ohne Überprüfung auf andere Organismen übertragen werden. Im Hauptteil der Arbeit sollte geklärt werden, ob trDNA generell die Expression benachbarter Gene beeinflußt und welche Eigenschaften der trDNA für diesen inhibitorischen oder stimulierenden Effekt verantwortlich sind. Keine der untersuchten trDNA-Familien zeigt einen inhibitorischen Effekt auf ein benachbartes Reportergen. Entgegen dem Modell von Dorer und Henikoff (1994) führen trDNA-Cluster nicht per se zu der Entstehung von Heterochromatin. Die Ergebnisse der vorliegenden Arbeit zeigen, daß sowohl ein trDNA-Cluster aus Cla-Elementen als auch aus Hsp70 Promotor-Elementen eine deutliche Steigerung der Expression des miniwhite-Reportergens bewirken. Diese Steigerung ist für beide trDNA-Familien unabhängig von der chromosomalen Lage des Transgens im Euchromatin. In beiden Fällen ist der Effekt von der Orientierung des trDNA-Clusters abhängig und verstärkt sich mit wachsender Kopienzahl der trDNA-Einheiten. Während eines der trDNA-Cluster aus trDNA-Einheiten besteht, die bekanntermaßen eine Promotoraktivität aufweisen (Hsp70 Promotoren), war für die Cla-Elemente kein Einfluß auf die Expression eines benachbarten Gens bekannt. Damit wurde für eine trDNA-Familie aus Chironomus nachgewiesen, daß sie auf ein benachbartes Gen ähnlich wirkt wie zusätzliche Promotoren. Die experimentellen Befunde unterstützen ein Modell, demzufolge die Cla-Elemente in gleicher Weise wie tandem repetitive Promotoren auf ein benachbartes Gen expressionssteigernd wirken. Die TATA-Box ist für das Modell ein wichtiges Strukturelement, da diese in beiden expressionssteigernden DNA-Sequenzen der trDNA-Cluster vorkommt und ausschließlich in einer Orientierung wirkt. Das Modell besagt, daß durch die Verbindung einer offenen Chromatinstruktur mit korrekt orientierten Bindungsstellen für TBP der Aufbau von vollständigen Transkriptionskomplexen an den tandem repetitiven Promotoren initiiert wird. Einer Perlenschnur ähnlich wären die Transkriptionskomplexe direkt verfügbar, nachdem ein Transkriptionskomplex den Promotor zur Transkription verlassen hat (Abb. 32). Dies würde zu der beobachteten Steigerung der Expression des Reportergens sowohl durch die tandem repetitiven Hsp70 Promotoren als auch durch die Cla-Elemente führen.

Die durch eine männchenspezifisch auftretende heterochromatische Bande und ein hemizygotes Cla-Element-Cluster gekennzeichnete geschlechtsbestimmende Region („sex determining region“: SDR) auf Chromosom III von C. riparius stellt ein frühes Stadium in der Evolution von Geschlechtschromosomen dar. Diese eindeutig lokalisierte chromosomale Region, die den molekular noch unbekannten männchen¬bestimmenden Faktor M enthalten muss, ist im Vergleich zu den Y-Chromosomen anderer Dipterenarten wie unter anderem M. domestica, die ebenfalls einen dominanten Männchenbestimmer besitzen, relativ klein. Aus diesem Grund bietet die SDR von C. riparius eine Möglichkeit, den männchenbestimmenden Faktor einzugrenzen und zu identifizieren. In der vorliegenden Arbeit konnte ein Bereich einer Größe von ca. 200 kb aus der SDR von C. riparius charakterisiert und analysiert werden. Durch bioinformatische Sequenzanalysen konnten an 20 Stellen der SDR mögliche Genstrukturen nachgewiesen werden. Von den gefundenen möglichen Genen ist bisher die Funktion in C. riparius unbekannt. Bei den Genen mit vermuteter Funktion deutet nichts eindeutig auf eine Beteiligung an der Geschlechtsbestimmung von C. riparius hin. Da allerdings davon auszugehen ist, dass für die Funktion des Männchenbestimmers M ein Gen rekrutiert wurde, welches zur Interaktion mit dem nachgeschalteten Gen der Geschlechts¬bestimmungskaskade fähig ist, muss die geschlechtsbestimmende Funktion des Gens M nicht unbedingt offensichtlich sein. Aus geschlechtsbestimmenden Genkaskaden anderer Dipteren bekannte Gene wie transformer und doublesex konnten im analysierten Bereich nicht nachgewiesen werden, obwohl zumindest zu doublesex homologe Gene im Genom von C. riparius vorkommen. Um möglicherweise proto-X- und proto-Y-Chromosom miteinander vergleichen zu können und einen Hinweis auf die chromosomale Herkunft der analysierten Sequenzen aus der SDR zu erlangen, wurden Sequenzen von 31 teilweise parallel liegenden BAC-Klonen aus der untersuchten Region verglichen. Dabei zeigte sich, dass die Klone zwei Gruppen bilden, deren Sequenzen sich durch 500 SNPs und 110 Indels unterschiedlicher Größe (1-800 Bp) unterscheiden, was für eine Herkunft von zwei sich erst seit kurzer Zeit unterscheidenden Geschlechtschromosomen spricht. Die zwölf größten dieser Indels wurden auf geschlechtsspezifische Unterschiede hin untersucht. Dabei zeigte sich, dass die Unterschiede zwischen den beiden Klongruppen zwar im 30 Jahre alten Laborstamm, der auch für die Konstruktion der durchsuchten BAC-Bibliotheken verwendet wurde, tatsächlich geschlechtsspezifisch sind, in zwei Wildfangpopulationen jedoch keine derartige Geschlechtsspezifität aufweisen. Somit kann keine Aussage zur Herkunft der untersuchten Klone aus der SDR von C. riparius getroffen werden, und es bleibt unklar, ob die analysierten Sequenzen vom proto-X oder vom proto-Y-Chromosom stammen.

Trans-10,cis-12 conjugated linoleic acid (CLA) supplementation causes milk fat depression in dairy cows, but CLA effects on glucose metabolism are not clear. The objective of the study was to investigate glucose metabolism, especially endogenous glucose production (eGP) and glucose oxidation (GOx), as well as hepatic genes involved in endogenous glucose production in Holstein cows supplemented either with 50 g of rumen-protected CLA (9% trans-10,cis-12 and 10% cis-9,trans-11; CLA; n=10) or 50 g of control fat (24% C18:2; Ctrl; n=10) from wk 2 before parturition to wk 9 of lactation. Animal performance data were recorded and blood metabolites and hormones were taken weekly from 2 wk before to 12 wk after parturition. During wk 3 and 9 after parturition, glucose tolerance tests were performed and eGP and GOx were measured by [U-(13)C] glucose infusion. Liver biopsies were taken at the same time to measure total fat and glycogen concentrations and gene expression of pyruvate carboxylase, cytosolic phosphoenolpyruvate carboxykinase, glucose-6-phosphatase, and carnitine palmitoyl-transferase 1. Conjugated linoleic acid feeding reduced milk fat, but increased milk lactose output; milk yield was higher starting 5 wk after parturition in CLA-fed cows than in Ctrl-fed cows. Energy balance was more negative during CLA supplementation, and plasma concentrations of glucose were higher immediately after calving in CLA-fed cows. Conjugated linoleic acid supplementation did not affect insulin release during glucose tolerance tests, but reduced eGP in wk 3, and eGP and GOx increased with time after parturition. Hepatic gene expression of cytosolic phosphoenolpyruvate carboxykinase tended to be lower in CLA-fed cows than in Ctrl-fed cows. In spite of lower eGP in CLA-fed cows, lactose output and plasma glucose concentrations were greater in CLA-fed cows than in Ctrl-fed cows. This suggests a CLA-related glucose sparing effect most likely due to lower glucose utilization for milk fat synthesis and probably because of a more efficient whole-body energy utilization in CLA-fed cows.
