Enzymatic cellulose degradation is a heterogeneous reaction requiring binding of soluble cellulase molecules to the solid substrate. Based on our studies of the cellulase complex of Clostridium thermocellum (the cellulosome), we have previously proposed that such binding can be brought about by a special "anchorage subunit." In this "anchor-enzyme" model, CipA (a major subunit of the cellulosome) enhances the activity of CelS (the most abundant catalytic subunit of the cellulosome) by anchoring it to the cellulose surface. We have subsequently reported that CelS contains a conserved duplicated sequence at its C terminus and that CipA contains nine repeated sequences with a cellulose binding domain (CBD) in between the second and third repeats. In this work, we reexamined the anchor-enzyme mechanism by using recombinant CelS (rCelS) and various CipA domains, CBD, R3 (the repeat next to CBD), and CBD/R3, expressed in Escherichia coli. As analyzed by non-denaturing gel electrophoresis, rCelS, through its conserved duplicated sequence, formed a stable complex with R3 or CBD/R3 but not with CBD. Although R3 or CBD alone did not affect the binding of rCelS to cellulose, such binding was dependent on CBD/R3, indicating the anchorage role of CBD/R3. Such anchorage apparently increased the rCelS activity toward crystalline cellulose. These results substantiate the proposed anchor-enzyme model and the expected roles of individual CipA domains and the conserved duplicated sequence of CelS.

The Huntington disease (HD) phenotype is associated with expansion of a trinucleotide repeat in the IT15 gene, which is predicted to encode a 348-kDa protein named huntington. We used polyclonal and monoclonal anti-fusion protein antibodies to identify native huntingtin in rat, monkey, and human. Western blots revealed a protein with the expected molecular weight which is present in the soluble fraction of rat and monkey brain tissues and lymphoblastoid cells from control cases. In lymphoblastoid cell lines from juvenile-onset heterozygote HD cases, both normal and mutant huntingtin are expressed, and increasing repeat expansion leads to lower levels of the mutant protein. Immunocytochemistry indicates that huntingtin is located in neurons throughout the brain, with the highest levels evident in larger neurons. In the human striatum, huntingtin is enriched in a patch-like distribution, potentially corresponding to the first areas affected in HD. Subcellular localization of huntingtin is consistent with a cytosolic protein primarily found in somatodendritic regions. Huntingtin appears to particularly associate with microtubules, although some is also associated with synaptic vesicles. On the basis of the localization of huntingtin in association with microtubules, we speculate that the mutation impairs the cytoskeletal anchoring or transport of mitochondria, vesicles, or other organelles or molecules.

In the glomeruli of the granule cell layer of mammalian cerebellum, neuronal extensions are interconnected by numerous small, nearly isodiametric (diameters up to 0.1 micron), junctions previously classified as puncta adherentia related to the vinculin-containing, actin microfilament-anchoring junctions of the zonula adherens of epithelial and certain other cells. Using immunofluorescence and immunoelectron microscopy, we have found, however, that these junctions are negative for E- and VE-cadherin, for desmosomal cadherins, and also for vinculin, alpha-actinin, and desmoplakin, but they do contain, in addition to the protein plakoglobin common to all forms of adhering junctions, the plaque proteins alpha- and beta-catenin and the transmembrane glycoprotein M-cadherin previously found as a spread--i.e., not junction bound--plasma membrane protein in certain fetal and regenerating muscle cells and in satellite cells of adult skeletal muscle. We conclude that these M-cadherin-containing junctions of the granule cell layer represent a special type of adhering junction, for which we propose the term contactus adherens (from the Latin contactus, for touch, site of bordering upon, also influence), and we discuss the differences between the various adhering junctions on the basis of their molecular constituents.

Compósitos de polímeros de polietileno linear de baixa densidade (LLDPE) possuem baixo desempenho mecânico devido principalmente à sua fraca interação, intermolecular, entre a cadeia polimérica e a carga. Uma maneira de minimizar esse baixo desempenho mecânico se faz com a mudança da estrutura química da poliolefina com a inserção de um grupo polar a sua cadeia, ou seja, faz-se a funcionalização das poliolefinas. O sistema de funcionalização adotado foi o processamento reativo, no qual foi utilizado para este sistema de processamento o misturador de dupla rosca acoplado a um reâmetro de torque. Neste trabalho, os grupos polares inseridos à cadeia dos polímeros de LLDPE\'s de copolímeros 1-buteno e 1-octeno (LLDPE-but e LLDPE-oct) foram o anidrido maléico (AM) e o anidrido tetrahidroftálico (ATF). Para a confecção dos compósitos foram utilizadas as cargas de microesferas de sílica modificada, no qual foi inserido compostos silanados em sua superfície (3-aminopropilsilano - APS - e trimetoxiclorosilano TMCISi) para estudo de interação com as poliolefinas funcionalizadas. Neste trabalho foram realizados ensaios de caracterização térmica, vibracional além de análises de torque do polímero fundido, análises do grau de reticulação e ensaios mecânicos de tração por elongação. Na caracterização térmica foram utilizadas as técnicas: termogravimetria (TG) e calorimetria exploratória diferencial (DSC). Na caracterização vibracional utilizou-se a espectroscopia fotoacústica no infravermelho (PAS-IR) e a espectroscopia de espalhamento Raman. Pela técnica PAS-IR foi possível comprovar a inserção dos anidridos à cadeia das poliolefinas assim como foi possível verificar a interação entre o polímero funcionalizado e a carga. Pelas técnicas térmicas de DSC e TG foi possível verificar mudanças das propriedades do compósito frente aos polímeros originais ou funcionalizados. Os ensaios mecânicos comprovaram que os compósitos de polímeros funcionalizados possuem maior elongação e tensão à ruptura comparada aos compósitos dos LLDPE\'s não funcionalizados

The mechanical behaviour of transventilated façades performed by natural stone is necessarily based on the correct execution of both anchoring elements on the stone cladding as in the ones corresponding to the enclosure support, either with brick masonry walls or reinforced concrete walls. In the case studied in the present work, the origin of the damages suffered on the façade of a building located in Alcoy has been analyzed, where the detachment of part of the outer enclosure occurred. This enclosure is a transventilated façade formed by Bateig Blue stone tiles. To this end, “in situ” tests of the anchoring systems employed have been performed, as well as laboratory tests of mechanical characterization of the material and of different types of anchor, comparing these results with those obtained in both the simplified analytical models of continuum mechanics as developed by the Finite Element Method (FEM).

Chiral rhodium hybrid nanocatalysts have been prepared by covalent anchorage of pyrrolidine-based diphosphine ligands onto functionalized CNTs. This work constitutes the first attempt at covalent anchoring of homogeneous chiral catalysts on CNTs. The catalysts, prepared with two different chiral phosphines, were characterized by ICP, XPS, N2 adsorption and TEM, and have been tested in the asymmetric hydrogenation of two different substrates: methyl 2-acetamidoacrylate and α-acetamidocinnamic acid. The hybrid nanocatalysts have shown to be active and enantioselective in the hydrogenation of α-acetamidocinnamic acid. A good recyclability of the catalysts with low leaching and without loss of activity and enantioselectivity was observed.

A synthetic procedure to prepare novel materials (surface-mediated fillings) based on robust hierarchical monoliths is reported. The methodology includes the deposition of a (micro- or mesoporous) silica thin film on the support followed by growth of a porous monolithic SiO2 structure. It has been demonstrated that this synthesis is viable for supports of different chemical nature with different inner diameters without shrinkage of the silica filling. The formation mechanism of the surface-mediated fillings is based on a solution/precipitation process and the anchoring of the silica filling to the deposited thin film. The interaction between the two SiO2 structures (monolith and thin film) depends on the porosity of the thin film and yields composite materials with different mechanical stability. By this procedure, capillary microreactors have been prepared and have been proved to be highly active and selective in the total and preferential oxidation of carbon monoxide (TOxCO and PrOxCO).

We describe a small case series that provides preliminary evidence of the usefulness of a new capsule-anchoring device for the management of subluxated cataracts. Three eyes of 3 patients with traumatic subluxated cataract causing a significant visual loss were enrolled. Phacoemulsification was performed in all cases with implantation of a capsule-anchoring device (AssiAnchor) because partial zonular dehiscence was present. A significant visual improvement was achieved in the 3 cases. The capsular bag was well centered and the anchors firmly attached to the capsulorhexis and sclera at 12 months postoperatively. The capsule-anchoring device was helpful in managing traumatic subluxated cataracts, enabling effective centration of the intraocular lens–capsular bag complex and, consequently, effective visual restoration.

tRNAs are charged with cognate amino acids by aminoacyl-tRNA synthetases (aaRSs) and subsequently delivered to the ribosome to be used as substrates for gene translation. Whether aminoacyl-tRNAs are channeled to the ribosome by transit within translational complexes that avoid their diffusion in the cytoplasm is a matter of intense investigation in organisms of the three domains of life. In the cyanobacterium Anabaena sp. PCC 7120, the valyl-tRNA synthetase (ValRS) is anchored to thylakoid membranes by means of the CAAD domain. We have investigated whether in this organism ValRS could act as a hub for the nucleation of a translational complex by attracting other aaRSs to the membranes. Out of the 20 aaRSs, only ValRS was found to localize in thylakoid membranes whereas the other enzymes occupied the soluble portion of the cytoplasm. To investigate the basis for this asymmetric distribution of aaRSs, a global search for proteins interacting with the 20 aaRSs was conducted. The interaction between ValRS and the FoF1 ATP synthase complex here reported is of utmost interest and suggests a functional link between elements of the gene translation and energy production machineries.

La propuesta de la ciudad de Sochi como sede de los Juegos Olímpicos de Invierno y de la 16° fecha del Gran Premio de Fórmula 1 de 2014, y como una de las sedes de la futura Copa Mundial de Futbol 2018, son una ratificación de la importancia estratégica que representa el Cáucaso para Rusia. Asimismo, el anclaje en el Cáucaso y en las costas del Mar Negro, con toda la inversión en infraestructura y en seguridad que representa la organización de eventos deportivos mundiales, implica la puesta en juego de una estrategia geopolítica que expresa la importancia de Rusia como potencia regional. En el calidoscopio de poder e intereses que se despliegan en la región caucásica, la sombra de Rusia se proyecta con fuerza. Abordar el rol que ejerce este país en el Cáucaso Sur, permitirá comprender las perspectivas actuales de la política exterior de Rusia en esta región, así como conocer de qué modo se despliega en el territorio el juego estratégico del poder a través de eventos deportivos que se observan desde todos los lugares del mundo.

Ce mémoire se penche sur les discours citoyens des Lavallois lorsqu’il est question de l'altérité ethnoculturelle dans leur quartier de banlieue. Cela implique de poser un regard sur le contexte spécifique de la ville de Laval pour y déchiffrer les arrimages entre rapport à l'espace et rapport à l'altérité ethnique dans l'expérience des répondants. Cette recherche qui se situe donc à la croisée des domaines des relations ethniques, de la géographie sociale et des études urbaines permet d’ancrer les représentations de l’altérité ethnoculturelle dans l’espace social et physique de la banlieue. Dans ce cadre, le discours de citoyens lavallois issus de deux quartiers ayant connu différents processus de croissance urbaine et de diversification ethnique sont analysés. Pour une majorité de répondants, les changements urbains récents témoignent d'une diversification de l'espace habité, ce qui est généralement mal perçu. Les manifestations d’altérité ethnoculturelle dans l’espace public et privé lavallois sont aussi associées à une détérioration du lien social dans la communauté du quartier. Au terme de cette étude, je soutiens que la présence d'immigrants est imbriquée dans les récents changements urbains dans la ville de Laval et que ces derniers sont directement associés à la perte du statut convoité de banlieue idéale.

Seriocarpa rhizoides Diehl 1969 was collected in abundance from the calcareous sand of the Josephine Bank (between Portugal and Madeira) during the "Meteor" seamount cruises in 1967. Attachment in this loose soft substratum is effected by fine anchoring strands of the tests. Two irregular series of small polycarp-like hermaphrodite bodies which are embedded in a connective tissue lie directly below the endostyle, forming a tubular compound gonad, but without common ducts. The intermediate nature of the reproductive system with respect to arrangement and structure increases our knowledge about the polygenetic relations of the stylid-genera. Some of the hitherto known ecological facts point to the presumed "seamounts effect" on this species.

In the past 10 years, the use of floating breakwaters as temporary coastal structures has become increasingly widespread in the United States as an inexpensive means for suppressing waves. However, as with any new technology, there have been many failures and a substantial number of imaginative, successful innovations. One of the chief problems contributing to the failure rate has been a lack of awareness by designers of reliable, up-to-date technical information. As part of a large research effort to remedy this problem, a survey was conducted on field experience with floating breakwaters in the Eastern United States. Results of the survey confirmed that state-of-the-art technical literature is not being properly disseminated. Structures built according to early design manuals were shown to have failed before the completion of their design life. Conversely, floating breakwaters built to the standards set by recent research have fared well and show promise of meeting their design golas. The weakest areas of the present technology are flotation and the anchoring systems. It is recommended that a concentrated research effort be directed toward these problem areas; it is also recommended that the monitoring of state-of-the-art projects continue. (Author).

Maria McCann paints a dark picture of masculinity and its effects in her novel As Meat Loves Salt (2001). The violent Jacob Cullen struggles with his masculinity as he faces the intricacies of religion, sexuality and politics in the midst of the English Civil War where he falls in love with fellow soldier Christopher Ferris. By using R.W. Connell and James Messerschmidt’s framework for the hierarchy of masculinities, I explore masculinities on local, regional and global levels and emphasized femininity in a close reading of McCann’s novel. My aim is not only to analyse the masculinities of the novel but also to use the framework to redefine toxic masculinity in order to make it a useable concept when analysing masculinities in literature. I redefine toxic masculinity because it lacks a clear definition anchored in an established framework used to study masculinity that does not see masculinity as inherently toxic. I believe that anchoring it to Connell and Messerschmidt’s framework will make it a useable concept. Due to the novel’s relationship to the Bible, I will use masculinity studies done on David and Jesus from the Bible to compare and reveal similarities with the masculinities in the novel, how they appear on the local, regional and global levels in the novel and its effects. I draw parallels between the love story in As Meat Loves Salt to the love story of David and Jonathan in the Bible by using queer readings of David and Jonathan in order to explore how masculinity affects the relationships and how the novel uses these two love stories as a study of toxic masculinity and how it relates it to hegemonic masculinity.
