Le virus de l’hépatite C (VHC) infecte ~185 millions d’individus dans le monde. Malgré le développement des nouvelles thérapies dirigées contre le VHC, on compte deux millions de nouvelles infections chaque année, en particulier dans les pays sous-développés et les populations marginalisées. Comme pour la plupart des virus à infection chronique, le développement d’un vaccin prophylactique efficace est limité par le manque de caractérisation des déterminants de la mémoire immunitaire protectrice lors des épisodes de réinfection naturelle chez les êtres humains. Le VHC représente un modèle unique au sein des virus à infection chronique pour étudier l’immunité protectrice. En effet ~30% des patients infectés par le VHC peuvent être guéris suite à un premier épisode d’infection spontanément. Dans cette thèse, nous avons étudié l’immunité protectrice contre le VHC dans une cohorte d’utilisateurs de drogues par injection qui sont à risque d’être infectés ou réinfectés. Notre hypothèse est que la majorité des patients qui ont résolu une première infection par le VHC sont protégés contre le développement d’une infection chronique s’ils sont réexposés. Cette immunité protectrice est associée à la présence des cellules T CD4 et CD8 polyfonctionnelles qui possèdent des fréquences, magnitudes et avidités élevées. La capacité protectrice des cellules T mémoire contre les séquences variables du VHC est dépendante de la diversité et flexibilité du répertoire de leurs récepteurs de cellules T (TCR), qui reconnaissent les séquences variables des épitopes ciblés. Notre premier objectif était de définir et détailler les déterminants de l’immunité protectrice conférée par les cellules T spécifiques du VHC. Nos résultats ont montré que la protection pendant l’épisode de réinfection était associée à une augmentation de la magnitude et du spectre des réponses spécifiques par les cellules T CD4 et CD8 polyfonctionnelles, ainsi que par l’apparition d’une population de cellules T tétramère+ CD8+ effectrices qui expriment faiblement le marqueur CD127 (CD127lo) lors du pic de la réponse. Chez les patients qui ont développé une infection chronique pendant l’épisode de réinfection, nous avons observé une expansion très limitée des cellules T CD4 et CD8. Le séquençage des épitopes ciblés par les cellules T CD8 chez ces patients qui sont non-protégés a montré que les séquences de ces épitopes sont différentes des séquences de référence qui étaient utilisées pour tous les essais immunologiques de cette étude. Le deuxième objectif était d’analyser la dynamique du répertoire des TCRs des cellules T CD8 spécifiques chez les patients protégés versus les patients non-protégés. Nos résultats ont montré que le répertoire des cellules T CD8 spécifiques est plus focalisé que chez les patients protégés. En plus, nous avons observé que les clonotypes qui forment le répertoire chez les patients protégés sont distincts de ceux chez les patients non-protégés. Ces clonotypes chez les patients protégés ont montré de plus grandes avidité et polyfonctionnalité que leurs homologues chez les patients non-protégés. En conclusion, nos résultats suggèrent que la protection contre le développement d’une infection chronique pendant l’épisode de réinfection par le VHC est associée à une augmentation de la magnitude, du spectre et de la fonctionnalité des réponses des cellules T spécifiques, ainsi qu’à un répertoire des TCRs plus focalisé composé des clonotypes distincts qui possèdent de plus grandes avidité et polyfonctionnalité que chez les patients non-protégés. L’homologie des séquences des souches virales entre les différents épisodes de l’infection est un déterminant majeur de l’établissement d’une protection efficace. Ces résultats ont donc des implications très importantes pour le développement d’un vaccin prophylactique contre le VHC et d’autres virus à infection chronique.

This thesis covers various aspects of viral diseases affecting shrimp aquaculture. The research component of this thesis can be divided into four areas. The areas covered are: I) A study to determine the prevalence of WSSV among the crustaceans in the Vembanad estuary, the shrimp aquaculture farms surrounding the estuary, and the sea off Cochin coast, India using two , sets of nested PCR primers. 2) An investigation to compare the sequence of six major structural proteins of WSSV; vp28, vp26, vp 19, vp68, vp281, vp466 from different geographical locations with that of an isolate from India. 3) Simultaneous occurrence of HPV, IHHNV, MBV and WSSV in postlarvae of P. monodon from hatcheries in India was monitored by Polymerase Chain Reaction. 4) A real time PCR procedure was developed for the quantitative analysis of WSSV infection. The viral load of postlarvae from hatcheries in Kerala meant for aquaculture was also determined using the quantitative PCR.

In general the behavioural sciences have contributed very little to filariasis research.Man's actions in creating vector breeding sites have been noted and discussed frequently inthe frlariasis literature. but virtually no systematic studies of these fonns of behavior have been undertaken (Dunn. 1979). Human behavioural observations and inquires into values and attitudes atfecting behavior that inhibits or promotes vector breeding are essential if any progress is to be made in developing self help programmes of vector control. Therefore, a systematic study on the socio-economic aspect of the community is warranted before undertaking any control programme against filariasis. In view of this the present study has been carried out which reveals the knowledge, attitude and practice concerning the causation, transmission, treatment and control of the disease. Socio economic factors that influence the creation and maintenance of vector breeding habitats were identified. characterization and ranking of these sociological factors will be helpful in identifying the determinants of human behavioural changes towards the containment of the disease- Information on the existing indigenous perception of the disease and the factors that hinder the control will be useful in developing a sound strategy from the human angle, which can be put to practical use.

Immortal cell lines have not yet been reported from Penaeus monodon, which delimits the prospects of investigating the associated viral pathogens especially white spot syndrome virus (WSSV). In this context, a method of developing primary hemocyte culture from this crustacean has been standardized by employing modified double strength Leibovitz-15 (L-15) growth medium supplemented with 2% glucose, MEM vitamins (1 ), tryptose phosphate broth (2.95 g l 1), 20% FBS, N-phenylthiourea (0.2 mM), 0.06 lgml 1 chloramphenicol, 100 lgml 1 streptomycin and 100 IU ml 1 penicillin and hemolymph drawn from shrimp grown under a bio-secured recirculating aquaculture system (RAS). In this medium the hemocytes remained viable up to 8 days. 5-Bromo-20-deoxyuridine (BrdU) labeling assay revealed its incorporation in 22 ± 7% of cells at 24 h. Susceptibility of the cells to WSSV was confirmed by immunofluoresence assay using a monoclonal antibody against 28 kDa envelope protein of WSSV. A convenient method for determining virus titer as MTT50/ml was standardized employing the primary hemocyte culture. Expression of viral genes and cellular immune genes were also investigated. The cell culture could be demonstrated for determining toxicity of a management chemical (benzalkonium chloride) by determining its IC50. The primary hemocyte culture could serve as a model for WSSV titration and viral and cellular immune related gene expression and also for investigations on cytotoxicity of aquaculture drugs and chemicals

White Spot Syndrome Virus (WSSV) is the most devastating disease affecting shrimp culture around the world. Though, considerable progress has been made in the detection and molecular characterization of WSSV in recent years, information pertaining to immune gene expression in shrimps with respect to WSSV infection remains limited. In this context, the present study was undertaken to understand the differential expression of antimicrobial peptide (AMP) genes in the haemocytes of Penaeus monodon in response to WSSV infection on a time-course basis employing semi-quantitative RT-PCR. The present work analyzes the expression profile of six AMP genes (ALF, crustin-1, crustin-2, crustin-3, penaeidin-3 and penaeidin-5), eight WSSV genes (DNA polymerase, endonuclease, immediate early gene, latency related gene, protein kinase, ribonucleotide reductase, thymidine kinase and VP28) and three control genes (18S rRNA, β-actin and ELF) in P. monodon in response to WSSV challenge. Penaeidins were found to be up-regulated during early hours of infection and crustin-3 during late period of infection. However, ALF was found to be up-regulated early to late period of WSSV infection. The present study suggests that AMPs viz. ALF and crustin-3 play an important role in antiviral defense in shrimps. WSSV gene transcripts were detected post-challenge day 1 itself and increased considerably day 5 onwards. Evaluation of the control genes confirmed ELF as the most reliable control gene followed by 18S rRNA and β-actin for gene expression studies in shrimps. This study indicated the role of AMPs in the protection of shrimps against viral infection and their possible control through the up-regulation of AMPs

Wenn man die Existenz von physikalischen Mechanismen ignoriert, die für die Struktur hydrologischer Zeitreihen verantwortlich sind, kann das zu falschen Schlussfolgerungen bzgl. des Vorhandenseins möglicher Gedächtnis (memory) -Effekte, d.h. von Persistenz, führen. Die hier vorgelegte Doktorarbeit spürt der niedrigfrequenten klimatischen Variabilität innerhalb den hydrologischen Zyklus nach und bietet auf dieser "Reise" neue Einsichten in die Transformation der charakteristischen Eigenschaften von Zeitreihen mit einem Langzeitgedächtnis. Diese Studie vereint statistische Methoden der Zeitreihenanalyse mit empirisch-basierten Modelltechniken, um operative Modelle zu entwickeln, die in der Lage sind (1) die Dynamik des Abflusses zu modellieren, (2) sein zukünftiges Verhalten zu prognostizieren und (3) die Abflusszeitreihen an unbeobachteten Stellen abzuschätzen. Als solches präsentiert die hier vorgelegte Dissertation eine ausführliche Untersuchung zu den Ursachen der niedrigfrequenten Variabilität von hydrologischen Zeitreihen im deutschen Teil des Elbe-Einzugsgebietes, den Folgen dieser Variabilität und den physikalisch basierten Reaktionen von Oberflächen- und Grundwassermodellen auf die niedrigfrequenten Niederschlags-Eingangsganglinien. Die Doktorarbeit gliedert sich wie folgt: In Kapitel 1 wird als Hintergrundinformation das Hurst Phänomen beschrieben und ein kurzer Rückblick auf diesbezügliche Studien gegeben. Das Kapitel 2 diskutiert den Einfluss der Präsenz von niedrigfrequenten periodischen Zeitreihen auf die Zuverlässigkeit verschiedener Hurst-Parameter-Schätztechniken. Kapitel 3 korreliert die niedrigfrequente Niederschlagsvariabilität mit dem Index der Nord-Atlantischen Ozillations (NAO). Kapitel 4-6 sind auf den deutschen Teil des Elbe-Einzugsgebietes fokussiert. So werden in Kapitel 4 die niedrigfrequenten Variabilitäten der unterschiedlichen hydro-meteorologischen Parameter untersucht und es werden Modelle beschrieben, die die Dynamik dieser Niedrigfrequenzen und deren zukünftiges Verhalten simulieren. Kapitel 5 diskutiert die mögliche Anwendung der Ergebnisse für die charakteristische Skalen und die Verfahren der Analyse der zeitlichen Variabilität auf praktische Fragestellungen im Wasserbau sowie auf die zeitliche Bestimmung des Gebiets-Abflusses an unbeobachteten Stellen. Kapitel 6 verfolgt die Spur der Niedrigfrequenzzyklen im Niederschlag durch die einzelnen Komponenten des hydrologischen Zyklus, nämlich dem Direktabfluss, dem Basisabfluss, der Grundwasserströmung und dem Gebiets-Abfluss durch empirische Modellierung. Die Schlussfolgerungen werden im Kapitel 7 präsentiert. In einem Anhang werden technische Einzelheiten zu den verwendeten statistischen Methoden und die entwickelten Software-Tools beschrieben.

The traditional task of a central bank is to preserve price stability and, in doing so, not to impair the real economy more than necessary. To meet this challenge, it is of great relevance whether inflation is only driven by inflation expectations and the current output gap or whether it is, in addition, influenced by past inflation. In the former case, as described by the New Keynesian Phillips curve, the central bank can immediately and simultaneously achieve price stability and equilibrium output, the so-called ‘divine coincidence’ (Blanchard and Galí 2007). In the latter case, the achievement of price stability is costly in terms of output and will be pursued over several periods. Similarly, it is important to distinguish this latter case, which describes ‘intrinsic’ inflation persistence, from that of ‘extrinsic’ inflation persistence, where the sluggishness of inflation is not a ‘structural’ feature of the economy but merely ‘inherited’ from the sluggishness of the other driving forces, inflation expectations and output. ‘Extrinsic’ inflation persistence is usually considered to be the less challenging case, as policy-makers are supposed to fight against the persistence in the driving forces, especially to reduce the stickiness of inflation expectations by a credible monetary policy, in order to reestablish the ‘divine coincidence’. The scope of this dissertation is to contribute to the vast literature and ongoing discussion on inflation persistence: Chapter 1 describes the policy consequences of inflation persistence and summarizes the empirical and theoretical literature. Chapter 2 compares two models of staggered price setting, one with a fixed two-period duration and the other with a stochastic duration of prices. I show that in an economy with a timeless optimizing central bank the model with the two-period alternating price-setting (for most parameter values) leads to more persistent inflation than the model with stochastic price duration. This result amends earlier work by Kiley (2002) who found that the model with stochastic price duration generates more persistent inflation in response to an exogenous monetary shock. Chapter 3 extends the two-period alternating price-setting model to the case of 3- and 4-period price durations. This results in a more complex Phillips curve with a negative impact of past inflation on current inflation. As simulations show, this multi-period Phillips curve generates a too low degree of autocorrelation and too early turnings points of inflation and is outperformed by a simple Hybrid Phillips curve. Chapter 4 starts from the critique of Driscoll and Holden (2003) on the relative real-wage model of Fuhrer and Moore (1995). While taking the critique seriously that Fuhrer and Moore’s model will collapse to a much simpler one without intrinsic inflation persistence if one takes their arguments literally, I extend the model by a term for inequality aversion. This model extension is not only in line with experimental evidence but results in a Hybrid Phillips curve with inflation persistence that is observably equivalent to that presented by Fuhrer and Moore (1995). In chapter 5, I present a model that especially allows to study the relationship between fairness attitudes and time preference (impatience). In the model, two individuals take decisions in two subsequent periods. In period 1, both individuals are endowed with resources and are able to donate a share of their resources to the other individual. In period 2, the two individuals might join in a common production after having bargained on the split of its output. The size of the production output depends on the relative share of resources at the end of period 1 as the human capital of the individuals, which is built by means of their resources, cannot fully be substituted one against each other. Therefore, it might be rational for a well-endowed individual in period 1 to act in a seemingly ‘fair’ manner and to donate own resources to its poorer counterpart. This decision also depends on the individuals’ impatience which is induced by the small but positive probability that production is not possible in period 2. As a general result, the individuals in the model economy are more likely to behave in a ‘fair’ manner, i.e., to donate resources to the other individual, the lower their own impatience and the higher the productivity of the other individual. As the (seemingly) ‘fair’ behavior is modelled as an endogenous outcome and as it is related to the aspect of time preference, the presented framework might help to further integrate behavioral economics and macroeconomics.

We extend a previous model of the Neolithic transition in Europe [J. Fort and V. Méndez, Phys. Rev. Lett. 82, 867 (1999)] by taking two effects into account: (i) we do not use the diffusion approximation (which corresponds to second-order Taylor expansions), and (ii) we take proper care of the fact that parents do not migrate away from their children (we refer to this as a time-order effect, in the sense that it implies that children grow up with their parents, before they become adults and can survive and migrate). We also derive a time-ordered, second-order equation, which we call the sequential reaction-diffusion equation, and use it to show that effect (ii) is the most important one, and that both of them should in general be taken into account to derive accurate results. As an example, we consider the Neolithic transition: the model predictions agree with the observed front speed, and the corrections relative to previous models are important (up to 70%)

Los tratamientos para aumentar los niveles de cúmulo de diferenciación 4 – CD4 en personas que padecen la enfermedad ocasionada por el Virus de la Inmunodeficiencia Humana (VIH), son importantes tanto para el mejoramiento del bienestar de los pacientes, como para el buen funcionamiento de las instituciones de salud. La presente investigación compara la intervención farmacológica de dos líneas de tratamiento, Lamivudina, Zidovudina, Efavirenz contra Efavirenz, Emtricitabina, Disoproxilo de Tenofovir que se encuentran en la recomendación de esquema de primera línea según la Guía Práctica Clínica (2014). Se evaluó el efecto costo-efectivo de estos dos tratamientos basado en el aumento de los niveles de CD4 a lo largo de tres tiempos diferentes (inicial, 6 y 12 meses) y los costos de los medicamentos de acuerdo a los precios en Colombia según el SISMED en el año 2014. Se realizó un análisis de varianza factorial con medidas repetidas, un árbol de decisiones y un análisis de costo-efectividad incremental (ACEI). Se obtuvo información de 546 pacientes, tanto hombres como mujeres, de la Institución Asistencia Científica de Alta Complejidad S.A.S de la ciudad de Bogotá. Se encontró que el esquema 1 (Lamivudina, Zidovudina, Efavirenz) fue considerado más efectivo y menos costoso que el tratamiento 2 (Efavirenz, Emtricitabina, Disoproxilo de Tenofovir), sin embargo no se evidenció una alta frecuencia de efectos adversos que pueda contribuir a la escogencia de un tratamiento u otro. De acuerdo a estos resultados la institución o los médicos tratantes tienen una alternativa farmacoeconómica para la toma de decisión del tratamiento a utilizar y así iniciar la terapia antirretroviral de pacientes que conviven con VHI con carga viral indetectable.

Previous studies have argued that the autocorrelation of the winter North Atlantic Oscillation (NAO) index provides evidence of unusually persistent intraseasonal dynamics. We demonstrate that the autocorrelation on intraseasonal time-scales of 10–30 days is sensitive to the presence of interannual variability, part of which arises from the sampling of intraseasonal variability and the remainder of which we consider to be “externally forced”. Modelling the intraseasonal variability of the NAO as a red noise process we estimate, for winter, ~70% of the interannual variability is externally forced, whereas for summer sampling accounts for almost all of the interannual variability. Correcting for the externally forced interannual variability has a major impact on the autocorrelation function for winter. When externally forced interannual variability is taken into account the intrinsic persistence of the NAO is very similar in summer and winter (~5 days). This finding has implications for understanding the dynamics of the NAO.
