O vírus da anemia infecciosa das galinhas (CAV, de “chicken anemia virus”), causa doença em aves jovens, caracterizada por anemia aplástica, atrofia linfóide generalizada e conseqüente imunodepressão. A doença clínica ocorre quando os pintos são infectados durante as duas primeiras semanas de vida e pode ser evitada se as reprodutoras passarem anticorpos em quantidade suficiente para sua progênie. Após as duas semanas de idade, as aves podem se infectar com o vírus, porém não desenvolverão a doença clínica. As matrizes infectadas durante o período de reprodução não demonstram sinais clínicos, nem alterações na postura, fertilidade dos ovos e viabilidade embrionária. Após algumas semanas, desenvolvem anticorpos que irão controlar a infecção e serão passados para o ovo, protegendo total ou parcialmente a progênie durante a fase de suscetibilidade a doença. O objetivo deste trabalho foi determinar o estado imune das matrizes, através da determinação da prevalência dos anticorpos em indivíduos e lotes de matrizes, determinar em que fase da criação as matrizes se infectaram e verificar se os títulos de anticorpos das matrizes eram suficientes para proteger a sua progênie. Fizeram parte deste estudo, 20 empresas da Região Sul do País (Paraná, Santa Catarina, Rio Grande do Sul), totalizando 64 lotes de matrizes não vacinadas e 12 lotes de matrizes vacinadas contra o CAV. Foram analisados 1709 soros através de um “kit” de ELISA indireto (KPL). A criação das matrizes foi dividida em quatro fases (Recria, 1ª, 2ª e 3ª fase de produção) para a coleta das amostras e constatou-se que todos os lotes de matrizes de corte não vacinados possuíam aves com anticorpos contra o CAV, 88,86% das matrizes foram soropositivas e 52,33% destas matrizes apresentaram títulos protetores para a progênie. Isto demonstrou que a maioria das aves se infectou durante a sua vida, entretanto, 47,67% das matrizes produziram uma progênie suscetível ou parcialmente protegida. A prevalência de matrizes não vacinadas soropositivas manteve-se constante a partir da fase de recria até o pico de produção, caindo no último terço da produção. Em todas as matrizes vacinadas foram encontrados anticorpos contra o CAV com títulos protetores para a progênie.

O vírus da anemia das galinhas – CAV (chicken anemia virus) está presente em praticamente todos os países com produção avícola investigados, inclusive no Brasil. O CAV causa a doença chamada de anemia infecciosa das galinhas em aves jovens, que se caracteriza por anemia, aplasia de medula óssea, retardo no crescimento, mortalidade variável (2 a 20%), atrofia de órgãos linfóides e imunodepressão. O controle da anemia infecciosa das galinhas é baseado na transferência de imunidade passiva das matrizes à progênie. A transferência de anticorpos maternos via ovo em nível suficiente é uma forma de prevenir a transmissão vertical do vírus, reduzindo assim a ocorrência de surtos da doença. Este trabalho teve como objetivo comparar o desempenho entre frangos nascidos positivos e negativos para a presença de anticorpos contra o CAV. Para isso, foram utilizados dois lotes de matrizes pesadas, sendo um vacinado e o outro não. Com a progênie obtida dessas matrizes de ambos os lotes, foram constituídos três tratamentos com 50 fêmeas e 50 machos cada: T1 - pintos oriundos de matrizes vacinadas com títulos protetores; T2 - pintos oriundos de matrizes não vacinadas com títulos médios e; T3 - pintos oriundos de matrizes não vacinadas e negativas ou com títulos baixos. Todos os tratamentos permaneceram em regime de criação intensiva usual por 47 dias em 5 propriedades diferentes, portanto o experimento teve 5 repetições. Os dados analisados foram o peso inicial e final, conversão alimentar e mortalidade. Como resultado, foi observado que não houve diferença significativa (p>0,05) no peso final entre os tratamentos com relação as fêmeas. Já, nos machos, os frangos negativos (T3) foram significativamente (p<0,05) mais pesados que os frangos positivos (T1 e T2). Não houve diferença significativa (p>0,05) entre os tratamentos em relação à mortalidade e a conversão alimentar. A análise do soro e histologia do timo determinaram a não ocorrência do desafio durante o período de criação dos frangos. A vacinação das matrizes contra o CAV não gerou títulos de anticorpos superiores aos obtidos nas matrizes não vacinadas e infectadas naturalmente. Este estudo indicou que a presença de anticorpos contra o CAV em frangos de corte, seja através da vacinação ou da infecção natural das matrizes, não gerou uma progênie com melhor desempenho nas condições de criação testadas.

Shrimp farming is one of the activities that contribute most to the growth of global aquaculture. However, this business has undergone significant economic losses due to the onset of viral diseases such as Infectious Myonecrosis (IMN). The IMN is already widespread throughout Northeastern Brazil and affects other countries such as Indonesia, Thailand and China. The main symptom of disease is myonecrosis, which consists of necrosis of striated muscles of the abdomen and cephalothorax of shrimp. The IMN is caused by infectious myonecrosis virus (IMNV), a non-enveloped virus which has protrusions along its capsid. The viral genome consists of a single molecule of double-stranded RNA and has two Open Reading Frames (ORFs). The ORF1 encodes the major capsid protein (MCP) and a potential RNA binding protein (RBP). ORF2 encodes a probable RNA-dependent RNA polymerase (RdRp) and classifies IMNV in Totiviridae family. Thus, the objective of this research was study the IMNV complete genome and encoded proteins in order to develop a system differentiate virus isolates based on polymorphisms presence. The phylogenetic relationship among some totivirus was investigated and showed a new group to IMNV within Totiviridae family. Two new genomes were sequenced, analyzed and compared to two other genomes already deposited in GenBank. The new genomes were more similar to each other than those already described. Conserved and variable regions of the genome were identified through similarity graphs and alignments using the four IMNV sequences. This analyze allowed mapping of polymorphic sites and revealed that the most variable region of the genome is in the first half of ORF1, which coincides with the regions that possibly encode the viral protrusion, while the most stable regions of the genome were found in conserved domains of proteins that interact with RNA. Moreover, secondary structures were predicted for all proteins using various softwares and protein structural models were calculated using threading and ab initio modeling approaches. From these analyses was possible to observe that the IMNV proteins have motifs and shapes similar to proteins of other totiviruses and new possible protein functions have been proposed. The genome and proteins study was essential for development of a PCR-based detection system able to discriminate the four IMNV isolates based on the presence of polymorphic sites

Visceral leishmaniasis hás adapted in the past 20 years to periurban and urban areas, and in Natal, Rio Grande do Norte, it became endemic. Thid study aimed to evaluate the environmental and social aspectsof Leishmania chagasi infection and its epidemiologic transmission chain in an urban, periurban and rural area of Parnamirim-RN. A study with three sections was conducted: Section 1: Sectional study of the human and canine infection by L. chagasi and its environmental and social determinants. Section 2: Observational longitudinal cohort to evaluate the dynamics of the canine infection. Section 3: Longitudinal study to evaluate the behavior of Lu. Longipalpis vector and the seasonal factors related to its dynamics. To include in the study the hauses were randomly selected and georreferenciated. Montenegro skin test was done in the human population and blood samples were collected for anti-Leishmania antibody detection. The canine population was examinated for L. chagasi infection by RIFI, ELISA and ELISA for rK39. An entomologic surveillance was monthly done with CDC light trapsin 10 houses of each locality. Quantitative and qualitative analyses was done using STATISC 6.0. Probality and prediction maps were done using ArcGis 9.0 model. In the human population L. chagasi infection was associated with the area of the hause, age, sex, population densyti, vegetation, kind of the floor of thr hause, water and resides destiny. In the canine population L. chagasi infection was associated with the breed, size, time of living in the hause, presence of dogs in the neighborhood, presence of horses and donkeys in the neighborhood, vegetation, kind of the floor and walls of the hause. The human infection was associated with canine infction only when analyzed taking into account the locality. In the prospective study, serum conversion and antibody lost observed in 30,8% and 22% of the animals examined, respectively. The human infection rate by L. chagasi was 24,6%, by the presence of anti-Leishmania antibody and 38,6% by the Montenegro skin test. The canine infection rate 32,5% by the presence of anti-Leishmania antibody. The vector Lu longipalpis showed an atypical behavior. These results indicate that environmental and social factors are important variables associated with L. chagasi infection in humans and canines, with punctual association of thr last two. Control measures of the infection on the studied points are necessary, in the aim to reduce the endemic focus of the disease in the study area. O objetivo deste trabalho foi avaliar a eficiência de transmissão do vírus do mosaico das nervuras, por ninfas e adultos de Aphis gossypii Glov., em duas cultivares de algodoeiro e registrar a evolução dos sintomas da doença. O trabalho foi realizado em casa de vegetação do Departamento de Fitossanidade da FCAV/UNESP, em Jaboticabal (SP). Ninfas e adultos ápteros criados em plantas infectadas da cultivar CNPTA ITA 90 foram transferidos para as plantas de algodoeiro das cultivares Coodetec 402 e CNPA ITA 90, com dois pares de folhas verdadeiras, onde permaneceram por 96 e 48 horas respectivamente. Para verificar a evolução da doença nas plantas, os sintomas foram avaliados até 60 dias após o confinamento dos afídeos, baseando-se numa escala de notas visuais. Os sintomas da doença foram inicialmente observados 25 dias após a inoculação pelos afídeos e se intensificaram com o desenvolvimento das plantas. A transmissão do vírus foi realizada mais eficientemente pelos adultos ápteros, do que pelas ninfas.

The demographic and epidemiological transition process caused by a declining in birth rates and in mortality, also changes occurred in morbidity and mortality is represented by the increasing of the aging population and the raising of chronic diseases. These diseases are characterized by multiple etiologies, risk factors, long latency period, a prolonged evolution, non-infectious origin and it has association with functional impairment and disability. Thus, elderly with chronic non-communicable disease has priority because they belong to a vulnerable group to get affection of comorbidities in aging, with increased demand and spending on health services. This study is aimed to analyse the understanding of elderly people with chronic non comunicable disease in the medium complexity service as a contribution to the improvement of health care in the city of Natal / RN. This is a descriptive and exploratory study with a quantitative approach, carried out at the Specialized Center for Elderly Health Care and at the Pescadores Hospital. The population was composed of 4,180 persons with a sample of 124 elderly aged above 60 years, attended in these medium complexity services. The instrument, a structured form, adapted from a questionnaire for monitoring risk and protective factors for chronic disease of the Ministry of Health. To collect data was was used the interview form containing demographic data, habits, health status and health care services. The results were processed using the Statistical Package for Social Science, version 18.0, analyzed by simple statistics. It was found that most seniors were female, predominantly between 70 and 74 years old, married, with a brown skin tone and Catholic religion, more than half had incomplete basic education, family income between one to two minimum wages and living with their families. Regarding the interviewers lifestyle, 94.4%, of them ate chicken and 97.6%, fruits, it was observed a reduction in smoking, alcoholism habits and physical activity according to the increasing age, 58.1 and 18.5% had insomnia18,5 % used sleeping pills. The elderly (51.6%) reported using services in times of sickness, seeking primary care at first (30.6%), 52% did not receive referral and was looking for free demand (38.7%). The most reported morbidity was hypertension, followed by musculoskeletal disorders. Regarding the difficulties in seeking health services, the delay in treatment and the waiting line were the most cited by the elderly. Almost all of them reported no activities to promote health in these services and those who received individual counseling on chronic diseases. Almost always, the health professionals who care of them, were mostly doctors followed by nurses. Based on the results presented, it is considered that the health services of medium complexity must undergone a more continuous dialogue with other attention level and focus on actions of health promotion and prevention. It is also recommended the necessity for qualified professionals to delivery health care to elderly and the implementation of protocols by a multidisciplinary health team, intending to provide better and continous care for the elderly with chronic diseases. The healthcare professionals who served them, were mostly physicians, followed by nurses. Through the results presented, it is considered that the medium complexity healthcare services need to perform a more continuous dialogue with the other levels of attention focusing attention to the health promotion and prevention actions. It is also recommended the necessity for qualified professionals to delivery healthcare for the elderly, in addition, a protocol implementation for the multidisciplinary health care team, to provide better care, and also the care continuity to elderly with chronic diseases

Periodontal disease is a complex inflammatory and infectious condition that immune host, front of the microbial aggressions, can lead to disease progression, resulting in tissue destruction. The tissue damage induces the release of regulatory molecules, which protective roles and / or destructive, including proteins VEGF (vascular endothelial growth factor vascular) and HIF-1 α (hypoxia-induced factor α -1). Thus, this study investigated, quantitatively and comparatively, the immunohistochemical expression of VEGF (vascular endothelial growth factor) and HIF-1 α (hypoxia induced factor 1-α), proteins involved in inflammation, angiogenesis and hypoxia, in human gingival tissues. Therefore, 75 samples of gingival tissues were examined. Thirty samples were chronic periodontitis, 30 with chronic gingivitis and 15 healthy gingival. After sections analysis, positives and negatives inflammatory and endothelial cells in the connective tissue were counted and converted into percentage. Data were statistically analyzed using Kruskal-Wallis test and Spearman correlation. The results showed that both proteins marked. It was observed higher immunoreactivity for HIF-1 α at chronic gingivitis and periodontitis specimens compared to healthy sites, however, no statistically significant differences were observed among them (p>0.05). The VEGF immunostaining showed similarity among the cases of periodontitis, gingivitis and healthy gingiva. Moderate and positive correlation statistically significant differences were verified for the expressions of VEGF and HIF-1α in gingival health (r = 0,529, p = 0.04). Thus, it can be conclude that possibly the route of HIF-1 α, is activated in periodontal disease may have involvement of the protein in pathogenesis and progression of disease, and that activation of VEGF, can be in addition to being triggered transcription by HIF-1 α may be also influenced by other additional factors such as the action of periodontal microorganisms endotoxins

Periodontal disease is an infectious disease resulting from the immunoinflammatory response of the host to microorganisms present in the dental biofilm which causes tissue destruction. The objective of this study was to evaluate the immunohistochemical expression of cyclophilin A (CYPA), extracellular matrix metalloproteinase inducer (EMMPRIN) and matrix metalloproteinase 7 (MMP-7) in human specimens of clinically healthy gingiva (n=32), biofilm-induced gingivitis (n=28), and chronic periodontitis (n=30). Immunopositivity for CYPA, EMMPRIN and MMP-7 differed significantly between the three groups, with higher percentages of staining in chronic periodontitis specimens, followed by chronic gingivitis and healthy gingiva specimens (p < 0.001). Immunoexpression of CYPA and MMP-7 was higher in the intense inflammatory infiltrate observed mainly in cases of periodontitis. Analysis of possible correlations between the immunoexpression of EMMPRIN, MMP-7 and CYPA and between the expression of these proteins and clinical parameters (probing depth and clinical attachment loss) showed a positive correlation of CYPA expression with MMP-7 (r = 0.831; p < 0.001) and EMMPRIN (r = 0.289; p = 0.006). In addition, there was a significant positive correlation between probing depth and expression of MMP-7 (r = 0.726; p < 0.001), EMMPRIN (r = 0.345; p = 0.001), and CYPA (r = 0.803; p < 0.001). These results suggest that CYPA, EMMPRIN and MMP-7 are associated with the pathogenesis and progression of periodontal disease

Este estudo avalia os fatores maternos e fetais envolvidos na transmissão vertical do HIV-1 em 47 pares de mãe e filho. As variáveis comportamentais, demográficas e obstétricas foram obtidas mediante entrevista; os dados referentes ao parto e ao recém-nascido, dos prontuários das maternidades. Durante o terceiro trimestre de gestação foi realizada a contagem da carga viral materna e dos linfócitos T CD4+. A média de idade foi de 25 anos e 23,4% das gestantes eram primigestas, e o fator comportamental mais prevalente foi não usar preservativos. Dentre as gestantes, 48,9% tinham células CD4+ superior a 500 células/mm³ e 93,6% se enquadravam na categoria clínica A; 95,7% submeteram-se à profilaxia com zidovudina durante a gestação ou no parto, a qual foi ministrada a todos os recém-nascidos; 50,0% delas foram submetidas à cesárea eletiva. Apesar de expostas a vários fatores de risco e protetores, nenhuma criança tornou-se infectada. A transmissão vertical resulta de um desequilíbrio entre os fatores, com predomínio dos de risco sobre os protetores.
