A cultura do feijoeiro está sujeita à incidência de várias doenças que acarretam perdas significativas na produção, dentre as quais encontra-se a murcha-de-curtobacterium ou murcha bacteriana, causada por Curtobacterium flaccumfaciens pv. flaccumfaciens (Cff). Atualmente a murcha-de-curtobacterium tem se constituído em um novo problema para a cultura do feijoeiro em várias regiões brasileiras. A resistência genética tem sido o meio mais eficiente no controle da doença, porém a possibilidade da existência de variabilidade genética presente em isolados de Cff, pode ser uma conseqüência maléfica ao melhoramento visando a obtenção de cultivares de feijoeiro resistentes, especialmente na estabilidade e durabilidade da resistência. Neste sentido, o presente trabalho teve como objetivo o estudo da variabilidade genética de 26 isolados de Curtobacterium flaccumfaciens, 20 dos quais provenientes de feijoeiro (Cff), coletados em diferentes regiões do Brasil, quatro provenientes de coleções internacionais (Cff) e dois endofíticos de citros (C. flaccumfaciens). Foram utilizados dois pares de oligonucleotídeos, CffFOR2-CffREV4 e CF4-CF5, avaliando-se na especificidade em reação de PCR, para a caracterização dos 26 isolados. No estudo da variabilidade genética, utilizou-se da técnica rep-PCR e os iniciadores REP, ERIC e BOX. A partir do padrão eletroforético gerado pela amplificação dessas seqüências repetitivas no DNA genômico dos 26 isolados bacterianos, foram realizadas análises pertinentes (UPGMA SM) e obtenção de um dendograma. Considerando-se um índice de similaridade de 75%, os isolados foram distribuídos em quatro grupos distintos. Os isolados de Cff provenientes do Paraná e DF foram separados em grupos diferentes, enquanto que isolados endofíticos de citros não formaram um grupamento distinto dos de feijoeiro. Os isolados procedentes do Estados de São Paulo mostraram-se geneticamente heterogêneos, alguns se agruparam com o isolado de USA, Santa Catarina e de citros, enquanto outros agruparam-se com isolados provenientes da França e Paraná. A avaliação da especificidade dos dois pares de oligonucleotídeos, mostrou que os oligonucleotídeos CffFOR2-CffREV4 detectaram todos os 26 isolados. Os oligonucleotídeos CF4-CF5 apresentaram menor especificidade não detectando dois isolados de Cff de feijoeiro (2928) e (2936) e os isolados endofíticos de citros. Portanto como ferramenta para detecção de Cff em feijoeiro os oligonucleotídeos CffFOR2-CffREV4 revelaram ser os mais indicados.

O Pinus caribaea var. hondurensis Barret & Golfari tem elevada importância como espécie comercialmente plantada; aproximadamente 1,8 milhões de hectares estão ocupados por plantios desta espécie no Brasil. O trabalho teve como objetivo verificar por meio de marcadores microssatélites a variabilidade genética em Pinus caribaea var. hondurensis, bem como sua manutenção durante o processo de melhoramento genético, dentro de uma população- base de melhoramento, uma população de matrizes selecionadas e uma população melhorada F1. Para a realização das análises foi necessária a transferência de primers desenvolvidos para locos microssatélites de outras espécies do gênero. Dos 20 pares de primers testados, 8 foram transferidos para a espécie (RPS 25b, RPS 150, PSM 2, PR 4.6, PtTX 2037, PtTX 3029, RPTest 01 e RPTest 09). Verificou-se a existência de endogamia entre e dentro das populações estudadas, e o maior valor observado entre as populações foi F ST = 0,0213 (população base e F1). A heterozigosidade média observada e a heterozigosidade esperada na população-base foram, respectivamente, H0 = 0,2469 e He = 0,2489. A maior distância genética (D = 0,0119) foi observada entre as populações-base e a população melhorada F1. Através da distância genética entre as matrizes, foram indicados 10 cruzamentos potenciais entre as matrizes mais contrastantes, almejando a obtenção de vigor de híbrido nas progênies obtidas a partir destes cruzamentos.

A Egeria najas é uma espécie aquática submersa nativa da bacia hidrográfica do rio Paraná. Com o represamento das águas do rio para geração de energia elétrica a espécie tem mudado seu comportamento de colonização dos leitos dos rios e ocorrido em grandes maciços dentro da represa de Jupiá e rios afluentes. Essa planta tem causado problemas por obstruir a passagem de água para as turbinas de geração de energia elétrica. Coletas de material vegetativo foram realizadas na represa e afluentes do rio Paraná para análise da variabilidade isoenzimática e de DNA. A análise isoenzimática mostrou haver somente quatro classes de diferentes biotipos. No entanto, utilizando-se os procedimentos de RAPD, observou-se que a espécie possui grande variabilidade genética. O represamento das águas também está permitindo o acumulo de variabilidade no local e promovendo um aumento de variabilidade por meio de possíveis cruzamentos entre genótipos diferentes. Os resultados também possibilitaram inferir sobre as possíveis rotas de migração de material genético para colonização da represa e rios afluentes.

Curtobacterium flaccumfaciens pv. flaccumfaciens (Cff) agente causal da murcha-de-curtobacterium em feijoeiro (Phaseolus vulgaris), é um patógeno vascular de difícil controle. A doença foi detectada pela primeira vez no Brasil na safra das águas de 1995, no Estado de São Paulo. Por se tratar de uma doença de difícil controle, a resistência genética tem sido a melhor opção. O objetivo deste trabalho foi avaliar a reação de genótipos de feijoeiro à murcha-de-curtobacterium, frente a 333 acessos pertencentes ao banco de germoplasma de feijoeiro do Instituto Agronômico de Campinas (IAC). Oportunamente, foram selecionados genótipos de feijoeiro altamente resistentes e suscetíveis, com a finalidade de comparar a colonização de Cff no vaso do xilema a partir da visualização sob microscopia eletrônica de varredura. Os resultados da triagem da resistência genética em genótipos de feijoeiro indicaram a existência de variabilidade genética nas amostras dos 333 genótipos avaliados, ao isolado de Cff Feij 2634. Os materiais foram classificados em 4 grupos de resistência: 29 genótipos (8,7%) comportaram-se como altamente resistentes, 13 genótipos (3,9%) como resistentes, 18 genótipos (5%) como moderadamente resistentes e 273 genótipos (81%) suscetíveis. A partir dos resultados obtidos, cerca de 18% dos genótipos de feijoeiros, desde altamente resistentes à moderadamente resistentes, poderão ser úteis para o programa de melhoramento genético como fonte de genes para resistência a Cff. Através da microscopia eletrônica de varredura, foram observadas em genótipos altamente resistentes, várias aglutinações da bactéria envolvidas por filamentos e estruturas rendilhadas sob pontuações da parede do vaso do xilema, não verificados em genótipos suscetíveis, o que sugere a ativação de mecanismos de defesa estruturais e bioquímicos nas plantas resistentes.

No presente trabalho foram coletados acessos de Eichhornia crassipes (aguapé) nos reservatórios das hidrelétricas de Barra Bonita, Bariri, Três Irmãos, Ilha Solteira, Salto Grande, Promissão, Ibitinga, Nova Avanhandava, Mogi-Guaçu, Euclides da Cunha, Jaguari, Jurumirim, Jupiá, Paraibuna e Porto Primavera, do Estado de São Paulo. Estes acessos foram submetidos a um estudo de variabilidade genética por meio de RAPD. Os primers utilizados foram OP X02, OP X07, OP X11 e OP P10 (TTCCGCCACC, GAGCGAGGCT, GGAGCCTCAG, TCCCGCCTAG, respectivamente). Dentre os acessos coletados e analisados, 21 apresentaram índice de identidade genética acima de 0,90. O dendrograma gerado com dados entre populações revelou forte coerência com a distribuição geográfica dos reservatórios que continham as plantas de aguapé. A variabilidade genética encontrada entre os acessos coletados nos diferentes reservatórios estudados foi elevada, considerando que a principal via de reprodução dessa espécie é a vegetativa.

O manejo de plantas daninhas em ambientes aquáticos requer cuidado diferencial e específico, a fim de evitar a contaminação ou alteração nas funções dos corpos hídricos e otimização do custo-benefício das operações. O estudo das características genéticas de populações de plantas daninhas aquáticas fornece informações que podem auxiliar no seu controle e manejo. A alface-d'água é uma planta aquática flutuante livre amplamente distribuída em todo o Brasil, mas é em ambientes aquáticos eutrofizados que essa e outras espécies de rápido desenvolvimento causam problemas sociais e econômicos, devido à grande massa vegetal produzida. Este estudo caracterizou geneticamente populações de alface-d'água coletadas em 15 reservatórios de hidrelétricas (Barra Bonita-BAB, Bariri-BAR, Ibitinga-IBI, Chavantes-CHA, Salto Grande-SAG, Jurumirim-JUR, Promissão-PRO Jaguari-JAG, Nova Avanhandava-NAV, Mogi-Guaçu-MOG, Limoeiro-LIM, Três Irmãos-TRI, Ilha Solteira-ILS, Jupiá-JUP e Porto Primavera-PPR) do Estado de São Paulo. As análises foram realizadas no NUPAM (Núcleo de Pesquisas Avançadas em Matologia), ligado à FCA/UNESP, campus de Botucatu-SP. A técnica utilizada no estudo da diversidade genética foi o RAPD. Os materiais amostrados nos reservatórios do Estado foram muito similares em sua maioria. As populações de NAV, MOG, IBI, JUR, PRO e CHA foram idênticas geneticamente. BAB e SAG, LIM e TRI também foram muito parecidas, apresentando índice de distância genética de 0,0093 e 0,0178, respectivamente. A grande maioria dos reservatórios estudados (93%) apresentou distâncias inferiores a 0,30, formando um grupo definido. No entanto, a população de Jupiá, em média, foi a que apresentou maior diversidade genética (0,45).

No melhoramento genético de espécies florestais, uma população base ou indivíduos superiores pré-selecionados tem importância fundamental para a manutenção do programa. Indivíduos de melhores procedências e de ampla base genética propiciam a obtenção de ganhos de forma contínua. O objetivo deste trabalho foi avaliar a diversidade genética em duas populações-núcleo de Eucalyptus grandis. Foram avaliados 39 indivíduos, sendo 19 pertencentes à população 1 e 20, à população 2, utilizando-se 14 primers microssatélite. Os fragmentos foram identificados e analisados a partir dos programas GeneScan e Genotyper, utilizando-se um sequenciador automático ABI Prism 3100. O número de alelos encontrados para cada primer variou de cinco a 15 para a população 1 e, de 8 a 18 para a população 2. A heterozigosidade estimada foi maior na população 2, 0,869, contra 0,843 na população 1. A média da distância genética entre os indivíduos da população 1 foi 0,6220 e na população 2 foi 0,6112. Com a caracterização molecular dos indivíduos destas populações foi construído um banco de dados que permitirá, a partir dos parâmetros de genética de populações, monitorar esses programas de melhoramento em diferentes ciclos de seleção.

As a professor in Curso de Licenciatura em Letras, from Campus Avançado Profa Maria Eliza Albuquerque Maia (CAMEAM),do Estado do Estado do Rio Grande do Norte (UERN), in the town of Pau do Ferros, in the state of Rio Grande do Norte, we had the chance to carry out several writing activities , as well as guiding re-writing activities for the texts produced. From this experience, we started looking at the need of reflecting upon the writing process in higher education. Thus, we aim at analyzing, in this research, the methodology used in the moment of carrying out the writing practices activities in higher education, investigating, in particular, the rewriting practices, concerning the operations used for carrying out such activities, as well as the sense effects produced from the alterations which were made in the texts. Our theoretical foundation is grounded on a conception of text as a verbal action , what reveals a socio-interactional view of the language (MARCUSCHI, 2008; SAUTCHUK, 2003). As the production of written texts, our research focus, we assume that, for this activity, we deal with distinct figures (active writer and internal reader), so that we can, apart from writing, reflecting upon our writing and, this way, deciding about operations which are carried out to make the alterations which are necessary to the rewriting of our texts (SAUTCHUK, 2003). Still about the theoretical foundations used in this research, we made use of the theories from the Textual Analysis of Discourse (TAD) which discusses the belief on the evidence on the existence of the texts, which is opposite to the fixist view of textuality which believes that the texts exist by themselves. (ADAM, 2008; [2005]2010). Under this perspective, we have also adopted, the concepts which come from genetics criticism which is concerned about the relation between text and genesis, using as objects documents which bring traits of the text in progress, on the ground that the text is the result of work in progress, and the writing practice, on the other hand, as an activity in a continuous movement (HAY, [1975]2002; DE BIASI, [2000]2010; GRÉSILLON, 1989; [1990]2008; [1992]2002; SALLES, 2008a). The methodology in this research is an ethnography-based one, an approach which focuses on the process, as well as is meaning-based. To understand the objectives proposed in our research, we made use of different procedures of collecting data which include an ethnographic study, such as: observation, note-taking, document analysis. The data which were analyzed were collected during the semester of 2008.2, in a first term classroom of Curso de Letras from CAMEAM, when we were able to collect twenty-one written texts and all of them were rewritten based on rewriting activities, what provides a corpus of forty-two texts which will be analyzed based on the linguistics operations identified by Generative Grammar and adopted by Lebrave and Grésillon (2009). From these analyses, we were able to confirm that writing is a process, and rewriting has become an extremely important activity for this process. Still due to these data, we observed that substitution was the most used operation by text authors. We believe that this result is justified by the fact that the substitution, according to what proposes the Genetic Criticism, constitutes the source of all erasure, from which one can easily make a change in writing. Regarding the operations of addition and deletion, we found that they were used in quantitative terms, almost equivalently, which can be explained when we see that the two operations require, by the author of the text, different strategies from those used for the replacement, what includes , respectively, adding or removing a segment. Finally, we found out that the shift operation was the least used, since it works with a segment that will not be replaced, added or deleted, but transferred to another place of text, which requires a greater ability of the author to perform this operation and not compromising the meaning of his/her writing. As a result, we hope to contribute to the reflection on the teaching of writing, considering, in a particular way, those with a Bachelor in Arts. Our analysis will contribute to the teaching of Portuguese language, specifically for activities that guide the production of texts in order to explore with students the ability to rewrite their own text

The fauna of Brazilian reef fishes comprises approximately 320 species distributed along the coast of the mainland and islands ocean. Little is known about the levels of connectivity between their populations, but has been given the interest in the relations between the offshore and the islands of the Brazil, in a biogeographical perspective. The oceanic islands Brazilian hosting a considerable number of endemic species, which are locally abundant, and divide a substantial portion of its reef fish fauna with the Western Atlantic. Among the richest families of reef fish in species are Pomacentridae. This study analyzed through analysis of sequences of the mitochondrial DNA control region (D-loop), the standards-breeding population of C. Multilineata in different areas of the NE coast of Brazil, involving both oceanic islands (Fernando de Noronha Archipelago and of St. Peter and St. Paul) and continental shelf (RN and BA). To this aim, partial sequences were used in the region HVR1 of mtDNA (312pb). The population structure and parameters for the estimates of genetic variability, molecular variance (AMOVA), estimation of the index for fixing (FST) and number of migrants were determined. The phylogenetic relationships between the populations were estimated using neighbor-joining (NJ) method. A group of Bayesian analysis was used to verify population structure, according to haplotype frequency of each individual. The genetic variability of populations was extremely high. The populations sampled show moderate genetic structure, with a higher degree of genetic divergence being observed for the sample of the Archipelago of St. Peter and St. Paul. At smaller geographical scale, the sample of Rio Grande do Norte and the Archipelago of Fernando de Noronha do not have genetic differentiation. Three moderately differentiated population groups were identified: a population group (I), formed by the Rio Grande do Norte (I') and the archipelago of Fernando de Noronha (I''), and two other different groups formed by the island population of the archipelago of Saint Peter and St. Paul (II) and Bahia (III). The genetic patterns found suggest that the species has suffered a relatively recent radiation favoring the absence of shared haplotypes. C. multilineata seems to constitute a relatively homogenous population along the West Atlantic coast, with evidence of a moderate population genetic structure in relation to the Archipelago of St. Peter and St. Paul. These data supports the importance of the dispersal larvae by marine current and the interpopulation similarity this species.

Brazil is one of the major centers of diversity for polyploid cotton plants; these plants belong to the genus Gossypium, which has three known species: G. hirsutum, G. barbadense and G. mustelinum. The Northeast is the only region where the three species occur, the last group being endemic. Northeast s cotton plants can be important sources of variability for genetic breeding. It is believed that great part of local diversity is being lost, due to economic, political, cultural and agricultural problems. In an attempt to mitigate this loss and delineate conservation strategies it is necessary to know how the species are found where they occur. The objective was to characterize and determine how plants are maintained in situ in the states of Maranhão, Piauí, Ceará, Rio Grande do Norte and Paraíba at the beginning of the XXI century. The in situ characterization of G. hirsutum and G. barbadense was conducted through structured interviews with the cotton plants owners and through the analysis of the environment. The data were collected during expeditions undertaken between the years 2004 to 2005. Twenty-two plants were collected in the state of Paraíba, forty-four in the state of Rio Grande do Norte, one hundred and forty-six in the state of Ceará, forty in the state of Maranhão and ninety-one plants in the state of Piauí. All plants collected in the states of Paraíba and Rio Grande do Norte belonged to moco type. Moco cotton plants also predominated in the other states, representing 92%, 62% and 78% of plants collected in Ceará, Piauí and Maranhão, respectively. The other cotton plants collected belong to the species G. barbadense. The cotton plants were found in situ as dooryard plants, roads side, feral populations, cultivation or local varieties. Great part were dooryard plants (45.2%), being major in Piauí and Maranhão. Cultivation predominated in Ceará; in Rio Grande do Norte feral populations were the most frequent and, in Paraíba, local varieties. The maintenance of moco plants is related, mainly, to the phytotherapic domestic use (20.9%) and to confection of lamp wicks (29.7%). Few inhabitants in Paraíba, Rio Grande do Norte, Piauí and none in Maranhão used harvest the plants, storage the seeds or gin; however, in Ceará, 40.5% of owners affirmed that they harvested and commercialized the fiber. It was found that the maintenance of species is dependent of the fragile cultural habits of local inhabitants, therefore the maintenance in situ is not a suitable way to conservation of genetic resources. Gossypium mustelinum Miers ex Watt is the only cotton species native from Brazil. It is endemic of the semi-arid region from North-east of the country, where it occur near from resilient water sources. The threats to the in situ conservation of the populations are caused by human interference in its habitat, mainly by excessive cattle graze and deforestation. Establish efficient strategies of in situ conservation depend on the accomplishment of a diagnosis of how the specie is found in its natural environment, and the knowledge about the genetic structure of the populations. The objectives of this work were i) to determine the in situ conditions of two populations present in rivers from basin of Rio Paraguaçu at the Bahia State, ii) to evaluate the structure and genetic variability presented in both populations, iii) to establish in situ and ex situ conservation strategies. It were realized collection in november 2007, when was realized in situ characterization of G. mustelinum. SSR markers were used for analyze 218 genotypes deriving from two populations of the G. mustelinum, localized at Tocó river and the Capivara river. The allelic frequencies, the heterozigosity and the F statics were estimated. All the plants were classified as wild and natives, and there was no evidence of the use the plants or its parts. The populations showed different conservation conditions in situ. Few plantlets were found in sites with excessive cattle feed, an indication that the damages in young plants should be high enough to compromise the renovation of the populations. On the other hand, populations were well preserved when the anthropic damages was low or inexistent. The 14 SSR primer pairs amplified 17 loci with a medium number of 5 alleles per locus (a total of 85 alleles). The high level of endogamy estimated (FIS=0,808) and the low observed heterozygosity (H0=0,093) were indicatives that the populations reproduce mainly by selfing, geitonogamy and crosses between related individuals. The genetic diversity was high (HE=0,482) and the differentiation between the populations was very high (FST=0,328). At least two sites from both populations of G. mustelinum must be preserved to achieve suitable in situ conservation. Actions that preserve the gallery forest and keep the cattle away should implemented, and could be as simple as erecting a fence. It is not possible anticipated if the in situ preservation will be possible. Therefore collections and ex situ preservation of representative specimens are essential to conserve the genetic diversity of native G. mustelinum

Brazil has been considered one of the diversity centers of Gossypium barbadense species. It is believed that a relatively big erosion genetic process occurs with the species, due to economic, cultural and agricultural problems. A local diagnostic about species situation is the first step for reducing the diversity loss and establishing conservation strategies in situ. This research aimed the identification of the presence of Gossypium populations, characterization, determination of the main risks and collection of the accesses to store in germoplam banks, in Para and Amapa States. Expeditions were conducted in November 2004. An interview was carried out with the plant proprietor for characterizing in situ of G. barbadense species and of the environment where the plants were inserted. On hundred seventy nine plants in 22 municipal districts were collected in Para State and 117 plants in nine municipal districts in Amapa State. The majority of plants belong to G. barbadense species (98% in Amapa and 94% in Para). Plants occur in back yards, beside roads and spontaneously. That ones from back yards were more abundant (97% in Amapa and 95% in Para) and maintained as medicinal plants as the principal reason. Plants in natural environments in both states evaluated were not found, therefore, the creation of reserves and the application of others conventional methods of maintenance in situ are not applicable. The plant proprietors do not use to store or process seeds. Seed storage was reported as a practice by only 1% of the plant proprietors from Para and 11% from Amapa. The most plants collected were from two to three years of age (58% in Amapa and 93% in Para). As conclusions G. barbadense is the species most spread in the two studied states and are found in back yards. In Amapa State the botanical variety barbadense or Quebradinho is predominant, whereas in Para State the predominant variety is brasiliense or Rim-de-boi. The gray mold, causal organism Amphobotrys ricini, is one of the major diseases of castor bean. Difficulties in managing plant disease arises form the limited understanding of the genetic structure of A. ricini, their complexity and variability make it difficult to control. Genetic structure can be used to infer the relative impact of different forces that influence the evolution of pathogen populations, that allow to predict the potencial for pathogen populations to envolve in agricultural ecosystems. Growers protect their crop by applying fungicides, but there aren t fungicides to provide significant control of gray mold of castor bean. The objectives of this work were use RAPD to determine the genetic structure of A. ricini subpopulations in Paraíba and assay the sensitivity of A. ricini isolates to azoxystrobin and carbendazim. To determine the genetic structure of A. ricini subpopulations in Paraíba, 23 isolates were colleted from two different geographic location (subpopulation). These isolates were analysed by RAPD using 22 random decamer primers, purchased from OPERON, produced a total of 80 markers polimorphics. The resulting matrixes were analysed using PopGene version 1.32. Sensitivity to azoxystrobin and carbendazim of 30 isolates, colleted form Paraíba and Alagoas, was estimated based on spore germination and colony growth inhibition. The stock solutions were added toV8 medium after sterilization to produce final concentrations of 0, 0.01, 0.1, 1, 10, and 100 µg/ml of carbendazim and 0, 0.001, 0.01, 0.1, 1, and 10 µg/ml of azoxystrobin. All statistical analyses were performed using SAS to estimate the dose that inhibited fungal growth by 50% (ED50 values). The genetic diversity within subpopulations (Hs=0,271) accounted for 92% of the total genetic diversity (Ht=0,293), while genetic diversity between subpopulations (Gst = 0,075) represented only 7,5%. The estimated number of migrants per generation (NM ) was 6,15. Nei s average gene identity across 80 RAPD loci was 0,9468. Individual ED50 values, for the 30 isolates screened for their sensitivity to azoxystrobin, ranged From a maximum of 0,168 µg/ml to a minimum of 0,0036 µg/ml. The ED50 values for carbendazim varied within the range of 0,026 to 0,316 µg/ml
