The objective of this update was to perform an evaluation of articles published in recent years about the Arg972 polymorphism of IRS-1 gene and its role in diabetes. It was used the database site of National Center of Biotechnology Information (NCBI - PUB- -MED). For search of articles the terms “polymorphisms on IRS-1 and diabetes” were used. Approximately 54% of the articles found were related to the polymorphism of interest and of these, approximately 68% the polymorphism was linked to various forms of diabetes. The Arg972 polymorphism is associated with decreased insulin synthesis and secretion with reduced stimulation of the activity of insulin pathway PI3K, MAPK and thymidine incorporation. It more frequently in patients with type 2 diabetes and gestational diabetes, showing a direct connection with the status of obesity and insulin resistance.

Although equines have participated in the forming and development of several civilizations around the world since their domestication 6,000 years ago in comparison to other species that have zootechnical interest, few researches have been done related to animal breeding area, especially in Brazil. Some reasons for that are difficulties associated with the species as well as operational aspects. However, developments in genetics in the last decades contributed to a better understanding of the traits related to reproduction, heath, behavior and performance of domestic animals, including equines. Recent technologies as next generation sequencing methods and the high density chips of SNPs for genotyping allowed some advances in the researches already done. These researches used basically the candidate gene strategy, and identified genomic regions related to diseases and syndromes and, more recently, the performance in sport competition and specific abilities. Using these genomic analysis tools, some regions related to race performance have been identified and based on this information; genetic tests to select superior animals for racing performance have started to be available in the market.

There is a high incidence of pituitary-dependent hyperadrenocorticism (PDH) in Poodle dogs, with family members being affected by the disease, suggesting a genetic involvement. Tpit is an obligate transcription factor for the expression of pro-opiomelanocortingene and for corticotroph terminal differentiation. The aim of the present study was to screen the Tpit gene for germline mutations in Poodles with PDH. Fifty Poodle dogs (33 female, 8.71 +/- 2.8 years) with PDH and 50 healthy Poodle dogs (32 females, 9.4241 2.8 years) were studied. Genomic DNA was isolated from peripheral blood, amplified by PCR and submitted to automatic sequence. No mutation in the coding region of Tpit was found, whereas the new single nucleotide polymorphism p.S343G, in heterozygous state, was found in the same frequency in both PDH and control groups. We concluded that Tpit gain-of-function mutations are not involved in the etiology of PDH in Poodle dogs.

O melhoramento genético de abelhas Apis mellifera é uma ferramenta essencial e de caráter obrigatório para o sucesso e desenvolvimento da indústria apícola. Práticas de manejo, troca das rainhas e inseminação instrumental constituem os pilares fundamentais de um programa que vise o aumento de determinada característica quantitativamente. A diversidade de climas no Brasil, junto com a possibilidade de direcionar a produção de diferentes produtos, fazem deste um país com oportunidades no campo internacional da apicultura. Programas de melhoramento genético, em grande escala, precisam ser feitos a partir de populações em massa, ou seja, um programa de melhoramento deverá iniciar-se a partir de um grande número de colmeias para serem selecionadas, com a finalidade de desenvolver e estruturar um programa de melhoramento genético que tenha como objetivo incrementar os níveis de produção e assim aumentar as médias de produção por colmeia em geral. Algumas características podem ser utilizadas para selecionar as colmeias, como: produção de própolis; a qualidade da própolis coletada; comportamento higiênico; taxa de infestação de varroa e incidência de Nosema. Isso garante a seleção das melhores colmeias como base do programa de melhoramento genético. Adicionalmente, padronização de práticas de manejo, troca constante de rainhas, produção de zangões, avaliação contínua da produção e a inseminação instrumental são práticas constantes nos apiários do programa de melhoramento em própolis.

[ES] El movimiento de genes entre individuos y poblaciones es esencial para la evolución de la vida. Dada la inmovilidad inherente a los organismos vegetales, el flujo genético entre individuos y poblaciones de una misma especie tendrá lugar mediante los eventos de dispersión del polen y las semillas, siendo la polinización la fase reproductora que más contribuye a la distribución de la diversidad genética y conectividad de las poblaciones. Con el uso de 8 loci microsatélites, se ha analizado la dispersión efectiva de los gametos masculinos de la palmera canaria (Phoenix canariensis), infiriendo en los patrones del transporte de polen de esta especie.

[ES] Dentro del contexto de la Biología de la Conservación de especies, los principios y la base de la conservación de la diversidad genética o sencillamente de la conservación genética está alcanzando de forma general, una importancia muy relevante en los últimos años. La genética de poblaciones y sus técnicas y algoritmos, están proporcionando a la ciencia de la conservación una precisión de análisis sin precedente, que se basa fundamentalmente en el mantenimiento del potencial adaptativo de las especies, especialmente en el contexto actual de cambio climático. También proporciona una base teórica que contribuye a entender los mecanismos que dan lugar a los cambios evolutivos y afectan el patrón genético observado en las poblaciones naturales y restituidas contribuyendo a proporcionar guías de manejo para aplicarse en los taxones amenazados. Pero además, la genética participa en un concepto integrado de conservación de la biodiversidad necesario para definir los métodos, concretar los objetivos e identificar las prioridades requeridas en cada programa de conservación y recuperación. No obstante, la importancia de los factores genéticos en la conservación de una especie, especialmente a gran escala, permanece hoy día como una cuestión abierta, existiendo un importante debate respecto a su importancia relativa y su auténtica contribución en la Biología de la Conservación y en la elaboración y ejecución de planes de conservación y recuperación de especies amenazadas.

Pharmacogenetic testing provides an outstanding opportunity to improve prescribing safety and efficacy. In Public health policy pharmacogenetics is relevant for personalized therapy and to maximize therapeutic benefit minimizing adverse events. CYP2D6 is known to be a key enzyme responsible for the biotransformation of about 25-30% of extensively used drugs and genetic variations in genes coding for drug-metabolizing enzymes might lead to adverse drug reactions, toxicity or therapeutic failure of pharmacotherapy. Significant interethnic differences in CYP2D6 allele distribution are well established, but immigration is reshaping the genetic background due to interethnic admixture which introduces variations in individual ancestry resulting in distinct level of population structure. The present thesis deals with the genetic determination of the CYP2D6 alleles actually present in the Emilia-Romagna resident population providing insights into the admixture process. A random sample of 122 natives and 175 immigrants from Africa, Asia and South America where characterized considering the present scenario of migration and back migration events. The results are consistent with the known interethnic genetic variation, but introduction of ethnic specific variants by immigrants predicts a heterogeneous admixed population scenario requiring, for drugs prescription and pharmacogenetics studies, an interdisciplinary approach applied in a properly biogeographical and anthropological frame. To translate pharmacogenetics knowledge into clinical practice requires appropriated public health policies, possibly guiding clinicians to evaluate prospectively which patients have the greatest probability of expressing a variant genotype.
