GABAB receptors associate with Gi/o-proteins that regulate voltage-gated Ca(2+) channels and thus the intracellular Ca(2+) concentration ([Ca(2+)]i), there is also reported cross-regulation of phospholipase C. These associations have been studied extensively in the brain and also shown to occur in non-neural cells (e.g. human airway smooth muscle). More recently GABAB receptors have been observed in chick retinal pigment epithelium (RPE). The aims were to investigate whether the GABAB receptor subunits, GABAB1 and GABAB2, are co-expressed in cultured human RPE cells, and then determine if the GABAB receptor similarly regulates the [Ca(2+)]i of RPE cells and if phospholipase C is involved. Human RPE cells were cultured from 5 donor eye cups. Evidence for GABAB1 and GABAB2 mRNAs and proteins in the RPE cell cultures were investigated using real time PCR, western blots and immunofluorescence. The effects of the GABAB receptor agonist baclofen, antagonist CGP46381, a Gi/o-protein inhibitor pertussis toxin, and the phospholipase C inhibitor U73122 on [Ca(2+)]i in cultured human RPE were demonstrated using Fluo-3. Both GABAB1 and GABAB2 mRNA and protein were identified in cell cultures of human RPE; antibody staining was co-localized to the cell membrane and cytoplasm. One-hundred μM baclofen caused a transient increase in the [Ca(2+)]i of RPE cells regardless of whether Ca(2+) was added to the buffer. Baclofen induced increases in the [Ca(2+)]i were attenuated by pre-treatment with CGP46381, pertussis toxin, and U73122. GABAB1 and GABAB2 are co-expressed in cell cultures of human RPE. GABAB receptors in RPE regulate the [Ca(2+)]i via a Gi/o-protein and phospholipase C pathway.

O presente trabalho tem por objetivo o estudo crítico-teórico da comediografialatina pré-clássica e portuguesa humanista nas figuras de Plauto e Gil Vicente,considerando os aspectos relativos ao conceito de família, de casamento e de vidaprivada. Investigando as obras "" e "Comédia do Viúvo", realizamosAululariainicialmente um minucioso estudo da função e importância social destes conceitosnos contextos específicos da Roma Antiga, no período republicano, e de Portugal natransição da Idade Média para o Renascimento. Procedendo então a análises dasobras, à luz dos temas propostos, identificaremos como se expressam nodesenvolvimento temático no interior da encenação. A partir do conceito decomicidade e possíveis estratégias para sua obtenção fundamentadas em teóricoscomo Henri Bergson e Vladimir Propp, procuramos encontrar pontos deconvergência e divergência entre os tratamentos temáticos dos autores de modo aelucidar também a influência do modo de pensar social como elemento decisivo daexpressão literária. Tal estudo do elemento cômico será aprofundado tendo comoponto de partida trabalhos filosóficos e teóricos de Gilles Deleuze, Mikhail Bakhtin eFriedrich Nietzsche

Introducción: La mastitis granulomatosa idiopática es una enfermedad crónica benigna, rara y de etiología desconocida; tiende a confundirse con otras enfermedades debido a síntomas similares. Este estudio pretende identificar y cuantificar las características demográficas, los antecedentes ginecoobstétricos relevantes y las manifestaciones clínicas prediagnósticas de esta enfermedad Metodología: Se realizó una revisión sistemática con análisis agrupado de datos tipo meta análisis. Se utilizó una estrategia de búsqueda en PubMed. Todos los estudios relacionados con la definición, manifestaciones clínicas, diagnóstico, tratamiento y pronóstico de la mastitis granulomatosa idiopática fueron elegibles. Las variables de interés fueron edad, país, antecedente de contracepción hormonal, tiempo de evolución, tiempo desde el último embarazo, diagnóstico inicial, y manifestaciones clínicas previas a la consulta. No hubo restricción en fechas de publicación. Resultados: Fueron incluidas 641 mujeres con diagnóstico de MGI reportadas en 68 publicaciones que cumplieron los criterios de selección. La edad media fue 35.9 años, 14.1% de ellas estaba embarazada o lactando, el antecedente de consumo de anticonceptivos hormonales fue 21% y el tiempo promedio desde el último parto fue de 3.9 años. La afectación ocurre principalmente en mama izquierda y en cuadrante superoexterno. El cáncer de mama y el absceso mamario son diagnósticos diferenciales en la consulta. Discusión: El diagnóstico de MGI es un reto para el ginecólogo desde la consulta inicial. Debido a que sus manifestaciones clínicas no son específicas, su diagnóstico parece apuntar a la necesidad de un proceso de descarte de otras patologías más frecuentes e incluso de peor pronóstico. Plasmodium falciparum, the most lethal malarial parasite, expresses an ortholog for the protein kinase C (PKC) activator RACK1. However, PKC has not been identified in this parasite, and the mammalian RACK1 can interact with the inositol 1,4,5-trisphosphate receptor (InsP3R). Therefore we investigated whether the Plasmodium ortholog PfRACK also can affect InsP3R-mediated Ca(2+) signaling in mammalian cells. GFP-tagged PfRACK and endogenous RACK1 were expressed in a similar distribution within cells. PfRACK inhibited agonist-induced Ca(2+) signals in cells expressing each isoform of the InsP3R, and this effect persisted when expression of endogenous RACK1 was reduced by siRNA. PfRACK also inhibited Ca(2+) signals induced by photorelease of caged InsP3. These findings provide evidence that PfRACK directly inhibits InsP3-mediated Ca(2+) signaling in mammalian cells. Interference with host cell signaling pathways to subvert the host intracellular milieu may be an important mechanism for parasite survival. (C) 2009 Elsevier Inc. All rights reserved.

Study of K isomerism in the transfermium region around the deformed shells at N=152, Z=102, and N=162, Z=108 provides important information on the structure of heavy nuclei. Recent calculations suggest that the K-isomerism can enhance the stability of such nuclei against alpha emission and spontaneous fission. Nuclei showing K isomerism have neutron and proton orbitals with large spin projections on the symmetry axis which is due to multi quasiparticle states with aligned spins K. Quasi-particle states are formed by breaking pairs of nucleons and raising one or two nucleons in orbitals near the Fermi surface above the gap, forming high K (multi)quasi-particle states mainly at low excitation energies. Experimental examples are the recently studied two quasi-particle K isomers in 250,256-Fm, 254-No, and 270-Ds. Nuclei in this region, are produced with cross sections ranging from several nb up to µb, which are high enough for a detailed decay study. In this work, K isomerism in Sg and No isotopes was studied at the velocity filter SHIP of GSI, Darmstadt. The data were obtained by using a new data acquisition system which was developed and installed during this work. 252,254-No and 260-Sg were produced in fusion evaporation reactions of 48-Ca and 54-Cr projectiles with 206,208-Pb targets at beam energies close to the Coulomb barrier. A new K isomer was discovered in 252-No at excitation energy of 1.25 MeV, which decays to the ground state rotational band via gamma emission. It has a half-life of about 100 ms. The population of the isomeric state was about 20% of the ground state population. Detailed investigations were performed on 254-No in which two isomeric states (275 ms and 198 µs) were already discovered by R.-D. Herzberg, but due to the higher number of observed gamma decays more detailed information about the decay path of the isomers was obtained in the present work. In 260-Sg, we observed no statistically significant component with a half life different from that of the ground state. A comparison between experimental results and theoretical calculations of the single particle energies shows a fair agreement. The structure of the here studied nuclei is in particular important as single particle levels are involved which are relevant for the next shell closure expected to form the region of the shell stabilized superheavy elements at proton numbers 114, 120, or 126 and neutron number 184. K isomers, in particular, could be an ideal tool for the synthesis and study of these isotopes due to enhanced spontaneous fission life times which could result in higher alpha to spontaneous fission branching ratios and longer half lifes.

A number of mathematical models for predicting growth and final height outcome have been proposed to enable the clinician to 'individualize' growth-promoting treatment. However, despite optimizing these models, many patients with isolated growth hormone deficiency (IGHD) do not reach their target height. The aim of this study was to analyse the impact of polymorphic genotypes [CA repeat promoter polymorphism of insulin-like growth factor-I (IGF-I) and the -202 A/C promoter polymorphism of IGF-Binding Protein-3 (IGFBP-3)] on variable growth factors as well as final height in severe IGHD following GH treatment. DESIGN, PATIENTS AND CONTROLS: One hundred seventy eight (IGF-I) and 167 (IGFBP-3) subjects with severe growth retardation because of IGHD were studied. In addition, the various genotypes were also studied in a healthy control group of 211 subjects.

C57BL/6, BALB/c, and CBA/Ca mouse strains with different MHC-I haplotypes were compared with respect to susceptibility to Neospora caninum infection. Groups of 5 mice received , , or tachyzoites of the NC-Liverpool isolate by intraperitoneal injection and were observed for disease symptoms. Humoral responses, splenocyte interferon-γ (IFN-γ) production, cerebral parasite loads, and histopathology were evaluated at human end points or the latest at 34 days postinfection (PI). The mortality rates in C57BL/6 mice were the highest, and relatively high levels of IgG1 antibodies were detected in those mice surviving till 34 days PI. In lymphocyte proliferation assays, spleen cells from C57BL6 mice stimulated with N. caninum antigen extract exhibited large variations in IFN-γ production. In BALB/c mice mortality was 0% at the lowest and 100% at the highest infection dose. Serologically they responded with high levels of both IgG2a and IgG1 subclasses, and lymphocyte proliferation assays of surviving mice yielded lower IFN-γ levels. CBA/Ca mice were the most resistant, with no animal succumbing to infection at a dose of and tachyzoites, but 100% mortality at tachyzoites. High IgG2a levels as well as increased IFN-γ in lymphocyte proliferation assays were measured in CBA/Ca mice infected with tachyzoites.
