This study develops a single-stream jet noise prediction model for a family of chevron nozzles. An original equation is proposed for the fourth-order space-time cross-correlations. They are expressed in flow parameters such as streamwise circulation and turbulent kinetic energy. The cross-correlations based on a Reynolds Averaged Navier-Stokes (RANS) flowfield showed a good agreement with those based on a Large Eddy Simulation (LES) flowfield. This proves that the proposed equation describes the cross-correlations accurately. With this novel source description, there is an excellent agreement between our model's far-field noise predictions and measurements1 for a wide range of frequencies and radiation angles. Our model captures the spectral shape, amplitude and peak frequency very well. This establishes that our model holds good for a family of chevron nozzles. As our model provides quick and accurate predictions, a parametric study was performed to understand the effects of a chevron nozzle geometry on jet noise and thrust loss. Chevron penetration is the underpinning factor for jet noise reduction. The reduction of jet noise per unit thrust loss decreases linearly with chevron penetration. The number of chevrons also has a considerable effect on jet noise.

Natural killer (NK) cell enhancing factor (NKEF) belongs to the newly defined peroxiredoxin (Prx) family. Its functions are to enhance NK cell cytotoxicity and to protect DNA and proteins from oxidative damage. In this study, a partial cDNA sequence of carp NKEF-B was isolated from thymus cDNA library. Subsequently, the full-length cDNA of carp NKEF-B was obtained by means of 3' and 5' RACE, respectively. The full-length cDNA of carp NKEF-B was 1022 bp, consisting of a 73 bp 5'-terminal untranslated region (UTR), a 355 bp T-terminal UTR, and a 594 bp open reading frame coding for a protein of 197 amino acids. Carp NKEF-B contained two consensus Val-Cys-Pro (VCP) motifs and three consensus cysteine (Cys-51, Cys-70 and Cys-172) residues. Sequence comparison showed that the deduced amino acid sequence of carp NKEF-B had an overall similarity of 74-96% to that of other species homologues. Phylogenetic analysis revealed that carp NKEF-B forms a cluster with other known teleost NKEF-Bs. Then, by PCR we obtained a 5.1 -k long genomic DNA of carp NKEF-B containing six exons and five introns. Realtime RT-PCR results showed that carp NKEF-B gene was predominantly detected in kidney and head kidney under un-infected conditions. Whereas under SVCV-infection condition, the expression of NKEF-B gene was significantly increased in blood cells, gill, intestine and spleen, but maintained in liver, and decreased significantly in kidney and head kidney. Finally, the rNKEF-B was constructed and expressed in Escherichia coli. By using an antibody against carp rNKEF-B, immunohistochemical study further indicated that NKEF-B positive cells are mainly some RBCs and a few epithelial cells in gill and intestine, and that under SVCV-infection condition, these positive cells or positive products in their cytoplasm were mainly increased in gill and spleen sections of carp. TRAIL (Apo2 ligand) described as a type II transmembrame protein belonging to the TNF superfamily can induce apoptotic cell death in a variety of cell types. In the present study, a putative cDNA sequence encoding the 299 amino acids of TRAIL (GC-TRAIL) and its genomic organization were identified in grass carp Ctenopharyngodon idella. The predicted GC-TRAIL sequence showed 44 and 41% identities to chicken and human TRAILs, respectively. In a domain search, a tumor necrosis factor homology domain (THD) was identified in the C-terminal portion of TRAILs. The GC-TRAIL gene consists of five exons, with four intervening introns, spaced over approximately 4 kb of genomic sequence. Analysis of GC-TRAlL promoter region revealed the presence of a number of putative transcription factor binding sites, such as Sp1, NF-kappaB, AP-1, GATA, NFAT, HNF, STAT, P53 and IRFI sequences which are important for the expression of other TNF family members. Phylogenetic analysis placed GC-TRAIL and the putative zebrafish (Danio rerio) TRAIL obtained from searching the zebrafish database into one separate cluster near mammalian TRAIL genes, but apart from the reported zebrafish TRAIL-like protein, indicating that the GC-TRAIL is an authentic fish TRAIL. Expression analysis revealed that GC-TRAIL is expressed in many tissues, such as in gills, liver, trunk kidney, head kidney, intestine and spleen. (c) 2005 Elsevier B.V. All rights reserved.

选取天然产物虎眼万年青作为研究对象,对一其化学成分及药理活性进行了系统的研究。针对不同成分极性的显著差异,分离、纯化出13种化合物,并鉴定了其中的弱极性的小分子物质。采用COZ超临界提取技术,对易挥发的19种成分分别作了鉴定。对其中具有药理活性的多糖、皂营及生物碱等成分,经进一步分离、纯化,并借助现代波谱技术,对其单一成分进行了跟踪、表征。研究了虎眼万年青不同组分的抗肿瘤、抗辐射作用。首次筛选出了有显著药理活性的虎眼万年青中性多糖53,并从非特异性免疫功能、体液免疫功能和细胞免疫功能等角度,即从细胞和分子水平较全面地研究了S3的免疫增强作用。结果表明,53能显著增强小鼠T淋巴细胞的转化、提高NK细胞细胞毒活性,不同程度的使CD3、CD4、CD4/CDS升高而使CDS下降,从分子水平证实了S3对辅助性T细胞(Th)的增强作用及对抑制性T细胞(Ts)的降低作用,推测Tl:可能是其主要的原发刺激细胞。此外,S3不仅能促进细胞因子IL-2的产生,还能使IL-1mRNA基因表达量显著增加,从基因水平进一步证实了53有提高小鼠免疫功能的作用。首次利用电喷雾质谱(ESI-MS),在较温和的条件下得到了具有较高稳定性的新型簇合物离子K+K(NO3)n:和NO3-(KNO3)m。并利用现代量子化学程序对低聚合度簇合物离子的可能组态进行了总成键能的计算,给出了相对稳定的组态。计算结果说明,n=2,3,6组态稳定性高的原因是总成键能高,而m≤4阴离子簇合物不能直接形成的原因是由于溶剂化作用。电喷雾质谱的进样过程与晶体的成核过程相类似,通过气相离子簇合物的组态信息和形成机理的研究,可以为晶体成核理论和化学键理论提供某些证据。

目前多数学者认为树鼩(Tree shrews)是灵长目攀居目树鼩科的一种小型哺乳动物。因其具有体型小、易饲养、价格低、新陈代谢和解剖结构与人类非常接近等优点,树鼩已经成为多种人类疾病研究的良好动物模型。树鼩作为乙型肝炎病毒(HBV)感染的动物模型已经被广泛应用。 自然杀伤性T细胞(Natrual killer T cells, NKT cells)可同时表达NK细胞和T细胞的表面标记,是CD1d限制性的特殊的淋巴细胞亚群,在机体的免疫反应中起重要作用。研究表明,在HBV感染及乙型肝炎的发病过程中,NKT细胞既有抑制和清除病毒的作用,也有加重肝炎及相关病症的负作用。因此,对NKT细胞的研究及恰当地调节将有助于理解和解决在研究乙肝防治中遇到的问题,但在树鼩中,至今尚无NKT细胞的相关报道。 目前公认的检测NKT细胞的手段是用加载了α-GalCer的CD1d四聚体检测。但至今尚无树鼩特异的相关抗体,给树鼩NKT细胞的研究造成很大障碍。因此,获得树鼩特异的CD1d四聚体是研究其NKT细胞特征和功能的前提。本论文作为其中一部分,目的是克隆树鼩CD1d编码区全长序列,为今后抗体的制备奠定基础。 本论文中,我们完成了相关工作并得到了以下结果:1、首次克隆得到了树鼩CD1d基因编码区的全长序列(1002bp),并预测了其氨基酸序列;2、通过比对分析得知其与灵长类的CD1d同源性较高,达到80%以上;同时,通过系统进化树分析,得知树鼩与灵长类亲缘关系较其它物种更近;3、首次分析了树鼩的CD1d mRNA在不同组织中的表达情况,发现树鼩的肝,肺中CD1d表达量较高,外周血和脾脏中也有少量表达。 总之,我们首次克隆得到了树鼩CD1d的编码区的全长序列,为今后制备CD1d四聚体进而研究树鼩的NKT细胞奠定了基础;同时首次分析了CD1d mRNA在树鼩的部分组织中的表达情况,结合以往其它物种中CD1d组织分布情况的检测结果,为树鼩中CD1d分子的研究提供了材料和数据基础。

川牛膝多糖(CP)是从传统中药川牛膝(Cyathula officinalis Kuan)中提取的一种活性多糖,现代药理研究表明川牛膝多糖是川牛膝许多生物活性的物质基础。本实验室前期进行了川牛膝多糖的提取、分离、结构鉴定及其部分活性研究,发现川牛膝中多糖含量非常高,在对川牛膝多糖活性的初步研究中也证实了其具有免疫调节作用。我们为了进一步了解其免疫调节活性,并为构效关系的研究奠定基础,对其进行了如下研究: 1. 通过体外毒性检测、淋巴细胞增殖实验、NK细胞杀伤活性和腹腔巨噬细胞吞噬中性红活性测定,发现川牛膝多糖在10~300μg/mL浓度范围内,对细胞无毒性作用;能够促进LPS诱导的B淋巴细胞增殖(P<0.01)、增强NK细胞杀伤活性(P<0.05)和PMΦ吞噬中性红活性(P<0.01),且随多糖浓度增高而增强;但其对ConA诱导的T淋巴细胞的增殖无促进作用(P>0.05)。 2. 通过正常小鼠体内淋巴细胞转化实验、迟发型变态反应分析、抗体生成细胞检测、碳粒廓清检测、腹腔巨噬细胞吞噬鸡红细胞活性和NK细胞活性测定,发现川牛膝多糖在适应性免疫方面能够促进SRBC免疫小鼠体内的抗体生成细胞的生成(P<0.01)和增强DNFB诱导的DTH(P<0.05),但对ConA诱导的脾淋巴细胞增殖无促进作用(P>0.05);在固有免疫方面能够提高小鼠碳粒廓清速率(P<0.05),PMΦ吞噬 CRBC 活性(P<0.01)和NK细胞杀伤活性(P<0.05)。同时还发现其对由环磷酰胺(Cy)引起的白细胞数下降具有很好的抑制作用(P<0.01)。 3. 为了获得结构明确、均一的保留活性的川牛膝多糖片段,为其作用机制、构效关系研究提供关键研究材料,我们开展了“保留免疫活性的最小片段”的分离制备的初步研究。建立并优化了川牛膝多糖的酸水解条件,发现在6%的样品浓度,0.025mol/L的硫酸浓度,65℃的水解温度,水解时间为8min的条件下可以得到一系列连续的多糖片段;采用Bio-Gel P2 分子筛柱层析分离得到5个级分,通过体外淋巴细胞增殖实验、NK细胞活性测定、腹腔巨噬细胞吞噬中性红实验发现其中的一个片段仍保留较强的免疫活性,并测得其分子量约为2057Da,为保留免疫活性的最小片段的进一步分离奠定了基础。 Cyathula officinalis Kuan is a commonly-used Traditional Chinese Medicine (TCM) with a wide range of pharmacological activities. Modern pharmacological researches showed the polysaccharide extracted from it (CP) is an important component for many bioactivities of this TCM. In the previous studies, we found CP showed significant immuno-regulative activities. In order to evaluate this activity systematically and lay foundations for revealling its immuno-regulative machanisms and the Structure -Function relationship, we carried out the following research works: 1. The in vitro immunoactivities of CP were evaluated by using normal mice immunocytes with respects to cytotoxicity, lymphocytes proliferation, NK activity and the ability of peritoneal macrophage phagocytizing neutral red. The polysaccharide showed no cytotoxicity below the concentration of 300 μg/mL, and could promote B lymphocytes proliferation (P<0.01), enhance NK activity (P<0.05) and the ability of peritoneal macrophage phagocytizing neutral red (P<0.01) at the concentration of 10-300 μg/mL. The above effects were positively correlated with the concentration of the polysaccharides. But it could not promote T lymphocytes proliferation (P>0.05). 2. The in vivo immunoactivities of CP were observed on normal mice through the following indices: splenic lymphocyte transformation efficiency, delayed-type allergy, antibody-forming cells activity (AFC), rate of carbon clearance, rate of peritoneal macrophage phagocytizing chicken red blood cell (CRBC) and NK activity, and its influence on the decline of the mouse leucocyte count induced by Cy. The polysaccharide at medium-dose enhanced delayed-type allergy (P<0.05)and NK activity(P<0.05) and increased the rate of carbon clearance(P<0.05), AFC activity(P<0.01) and the rate of peritoneal macrophage phagocytizing CRBC(P<0.01). The polysaccharides also effectively resisted the decline of the mouse leucocyte count induced by Cy(P<0.01). However, it couldn’t increase the splenic lymphocyte transformation efficiency(P>0.05). 3. Attempting to isolate and prepare the minimal fragments retaining activity with identical structure for further studying on immuno-regulative mechanism and Structure-Function relationship, we carried out the study on hydrolysis of CP, isolation of hydrolysed fragments, and the activity evaluation of the isolated fragments. CP with concentration of 6% was hydrolysed at 65℃ for 8 min with sulfuric acid of 0.025 mol/L,then the hydrolysate was separated using Bio-Gel P2 chromatography, 5 portions of fragments were obtained. The immunoactivities of these fragments were evaluated by using normal mice immunocytes with respect to lymphocytes proliferation, NK activity and ability of peritoneal macrophage phagocytizing neutral red. One fragment with relative molecular mass of 2057Da was found retaining immunoactivity.

采用ICR小鼠,转移受照射细胞的条件培养液来培养未受照射细胞,检测YAC?I(小鼠T淋巴瘤)细胞对小鼠自然杀伤(NK)细胞的敏感性变化,测定小鼠脾细胞增殖能力,观察自由基清除剂二甲基亚砜(Dimethylsulfoxide,DMSO)照射前处理细胞的影响。初步研究了电离辐射旁效应(Bystandereffect,BE)及其可能的机理。结果显示,条件培养液培养的Yac?I细胞对NK细胞的敏感性明显升高。用条件培养液培养的小鼠脾细胞的增殖能力也明显受到抑制,提示辐射旁效应的存在,观察到DMSO对旁效应有一定的抑制作用,说明活性氧(Reactiveoxygenspecies,ROS)等自由基对旁效应有一定贡献。然而在淋巴细胞增殖实验ICM+1%DMSO组的数据显示DMSO对旁效应的抑制作用非常有限,表明除自由基外必定存在其他因素介导旁效应的发生。

目的通过受照射的细胞培养液培养靶细胞,并用二甲基亚砜(DMSO)对靶细胞预处理,检测其对自然杀伤(NK)细胞活性的影响,研究旁效应是否存在及其产生机理。方法以健康雄性ICR小鼠脾NK细胞活性检测经过不同剂量照射的细胞培养液对小鼠淋巴瘤YacⅠ细胞的损伤程度,同时观察DMSO对该细胞的保护作用,即对旁效应的抑制作用。结果应用不同剂量60Coγ射线照射的细胞培养液来培养YacⅠ细胞,引发该细胞不同程度的损伤,表现在YacⅠ细胞对小鼠脾NK细胞的敏感性增强(P<0.01);DMSO预处理细胞对旁效应有一定的抑制作用,特别在0.25和0.5Gy组,对细胞的保护作用明显。结论60Coγ射线照射能诱发YacⅠ细胞的培养液介导的旁效应。活性氧(ROS)是诱发辐射旁效应的因素之一,清除ROS可部分抑制辐射旁效应。

目的 研究用自然杀伤 (NK)细胞活性的变化检测肿瘤细胞Yac Ⅰ在6 0 Coγ射线照射后是否存在旁观者效应 (旁效应 ) ,以及其旁效应剂量学和时间效应特征。方法 以健康ICR小鼠脾NK细胞活性为生物终点 ,检测NK细胞对不同条件培养肿瘤细胞Yac Ⅰ的活性 ,用培养基介导法观察旁效应 ,同时 ,培养不同时间观察其时间规律。结果 每个剂量组NK细胞活性的均值与对照细胞NK细胞活性均值差异有显著性 (P <0 0 5 )。照射后分别培养 0和 2 4h ,各剂量组间的NK细胞活性均值差异无显著性。结论 6 0 Coγ射线照射能诱发肿瘤细胞Yac Ⅰ的旁效应。旁效应在低剂量时较为明显。

Purpose: To estimate the biological risks to the immune system of the type of space radiation, 12C6+, encountered by cosmonauts during long-term travel in space. Materials and methods: The Kun-Ming strain mice were whole-body irradiated by 12C6+ ion with 0, 0.01, 0.05, 0.075, 0.2, 0.3, 0.5, 0.75, 1 or 2 Gy, at a dose rate of 1 Gy/min. At 35 days after irradiation, the thymus and spleen weights were measured, the natural killer (NK) cells activity of spleen was determined by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT), and the interferon-gamma (IFN-gamma) levels in serum and thymus were detected with enzyme-linked immunosorbent assays (ELISA). Results: The results showed that the thymus weight, IFN-gamma levels in serum and the activity of splenic NK-cells had significantly increased at a dose of 0.05 Gy. With further dose increase, the weight of spleen continued to increase but the weight of thymus, IFN-gamma level and NK-cells activity declined. Conclusions: These results suggest that the dose of 0.05 Gy irradiation has a stimulatory effect on mouse immunity; this effect declined with increasing dose.

We present results on the system size dependence of high transverse momentum di-hadron correlations at root s(NN) = 200 GeV as measured by STAR at RHIC. Measurements in d + Au, Cu + Cu and Au + Au collisions reveal similar jet-like near-side correlation yields (correlations at small angular separation Delta phi similar to 0, Delta eta similar to 0) for all systems and centralities. Previous measurements have shown Chat the away-side (Delta phi similar to pi) yield is suppressed in heavy-ion collisions. We present measurements of the away-side Suppression as a function of transverse momentum and centrality in Cu + Cu and Au + Au collisions. The suppression is found to be similar in Cu + Cu and An + An collisions at a similar number of participants. The results are compared to theoretical calculations based on the patron quenching model and the modified fragmentation model. The observed differences between data and theory indicate that the correlated yields presented here will further constrain dynamic energy loss models and provide information about the dynamic density profile in heavy-ion collisions. The contribution of B meson decays to nonphotonic electrons, which are mainly produced by the semileptonic decays of heavy-flavor mesons, in p + p collisions at root s = 200 GeV has been measured using azimuthal correlations between nonphotonic electrons and hadrons. The extracted B decay contribution is approximately 50% at a transverse momentum of p(T) >= 5 GeV/c. These measurements constrain the nuclear modification factor for electrons from B and D meson decays. The result indicates that B meson production in heavy ion collisions is also suppressed at high p(T).

200 GeV corresponding to baryon chemical potentials (mu(B)) between 200 and 20 MeV. Our measurements of the products kappa sigma(2) and S sigma, which can be related to theoretical calculations sensitive to baryon number susceptibilities and long-range correlations, are constant as functions of collision centrality. We compare these products with results from lattice QCD and various models without a critical point and study the root s(NN) dependence of kappa sigma(2). From the measurements at the three beam energies, we find no evidence for a critical point in the QCD phase diagram for mu(B) below 200 MeV.

We report the first three-particle coincidence measurement in pseudorapidity (Delta eta) between a high transverse momentum (p(perpendicular to)) trigger particle and two lower p(perpendicular to) associated particles within azimuth |Delta phi| < 0.7 in root s(NN) = 200 GeV d + Au and Au + Au collisions. Charge ordering properties are exploited to separate the jetlike component and the ridge (long range Delta eta correlation). The results indicate that the correlation of ridge particles are uniform not only with respect to the trigger particle but also between themselves event by event in our measured Delta eta. In addition, the production of the ridge appears to be uncorrelated to the presence of the narrow jetlike component.
