Pristimerin has been shown to be cytotoxic to several cancer cell lines. In the present work, the cytotoxicity of pristimerin was evaluated in human tumor cell lines and in human peripheral blood mononuclear cells (PBMC). This work also examined the effects of pristimerin (0.4; 0.8 and 1.7 mu M) in HL-60 cells, after 6, 12 and 24 h of exposure. Pristimerin reduced the number of viable cells and increased number of non-viable cells in a concentration-dependent manner by tripan blue test showing morphological changes consistent with apoptosis. Nevertheless, pristimerin was not selective to cancer cells, since it inhibited PBMC proliferation with an IC50 of 0.88 PM. DNA synthesis inhibition assessed by 5-bromo-2'-deoxyuridine (BrdU) incorporation in HL-60 cells was 70% and 83% for the concentrations of 0.4 and 0.8 mu M, respectively. Pristimerin (10 and 20 mu M) was not able to inhibit topoisomerase 1. In AO/EB (acridine orange/ethidium bromide) staining, all tested concentrations reduced the number of HL-60 viable cells, with the occurrence of necrosis and apoptosis in a concentration-dependent manner, results in agreement with trypan blue exclusion findings. The analysis of membrane integrity and internucleosomal DNA fragmentation by flow cytometry in the presence of pristimerin indicated that treated cells underwent apoptosis. The present data point to the importance of pristimerin as representative of an emerging class of potential anticancer chemicals, exhibiting an antiproliferative effect by inhibiting DNA synthesis and triggering apoptosis. Objetivou-se estudar a influência da artrite encefalite caprina no hemograma de cabras durante a lactação. Para tanto, 38 matrizes F1 Anglo-Nubiana x Saanen, com idade entre 14 e 38 meses e escore corporal entre dois e três, foram previamente testadas por IDGA e Western Blot para a divisão em grupo soropositivo (n = 19) e soronegativo (n = 19). Os grupos foram mantidos em piquetes distintos, sem contato físico, recebendo o mesmo manejo nutricional e sanitário. O sangue foi coletado por venipunção da jugular, mensalmente, desde o parto até o fim da lactação. Foram determinados os valores do eritrograma e do leucograma. em dados momentos, os soropositivos apresentaram alterações morfológicas nas hemácias, com diminuição significativa do VCM, aumento significativo do HCM e CHCM. Não houve diferença significativa entre grupos na contagem de hemácias e volume globular. Os valores absolutos de monócitos e eosinófilos dos soropositivos, em algumas ocasiões, encontraram-se significativamente superiores aos dos soronegativos e acima da faixa de normalidade para a espécie caprina.

Realizou-se pesquisa objetivando determinar o melhor entre quatro pontos de colheita da haste floral de crisântemos de maço do tipo pompom cv. White Polaris a concentração mais adequada de sacarose para tratamento de pulsing dessas hastes: estas foram colhidas em estufa de produção comercial, nos pontos preestabelecidos e transportadas para laboratório, onde foram totalmente imersas em água de torneira, à sombra, durante três horas. Selecionaram-se as hastes pela uniformidade do seu desenvolvimento e cortaram-nas sob água na base do caule, entre 50 e 60 cm, e identificadas, o que permitiu avaliar as mudanças morfológicas associadas às inflorescências individuais. As hastes foram distribuídas e mantidas nos diferentes tratamentos de pulsing durante 24 horas, à luz branca contínua de 1.500 lux, 60 a 90% de umidade relativa do ar e temperatura ambiente de 25 ± 2°C. Após o tratamento de pulsing, as hastes foram transferidas para água destilada, permanecendo por 10 horas sob luz branca contínua, nas mesmas condições de laboratório citadas. A vida floral em vaso começou a ser avaliada na instalação do experimento, após o tratamento de pulsing, e terminou quando as folhas e pétalas perderam a turgescência e o valor decorativo. As hastes colhidas em estádio de botão (25 e 50% de abertura das inflorescências apicais) não alcançaram o ponto de abertura adequado em nenhuma das seis concentrações de sacarose (0 a 146,07 mol/m³); as concentrações de 116,9 146,1 mol/m³, porém, estimularam, em geral, a abertura de botões. Os pontos de colheita 1 e 2 (100 e 75% de abertura das inflorescências apicais respectivamente) apresentaram bons resultados em todas as concentrações de sacarose.

0 objetivo do trabalho foi determinar o melhor tratamento pós-colheita para manutenção floral e aumento da longevidade de crisântemo de maço do tipo pompom (Dendranthema grandiflorum (Ramat.) S. Kitamura) cv. White Polaris. Estabeleceu-se como ponto de colheita o momento em que as hastes apresentavam três inflorescências apicais com as pétalas externas em ângulo de 45° em relação à horizontal. Durante o ensaio em laboratório, as hastes, colhidas em estufa de produção comercial, após totalmente imersas em água de torneira, à sombra, durante três horas, foram cortadas sob água na base do caule entre 50 e 60 cm. As hastes foram distribuídas nos diferentes tratamentos de pulsing durante 24 horas, com luz contínua de 1.500 lux, 60 a 90% de umidade relativa do ar e temperatura ambiente de 25 ± 2°C. No primeiro experimento, testou-se a eficiência de 8-hidroxiquinolina (8-HQ) e tiabendazole (TBZ) como germicidas de manutenção da qualidade na solução de pulsing; testaram-se, também, dois reguladores de crescimento, a saber: ácido giberélico (GA3), 6-benzilaminopurina (6-BA) ou a mistura dos dois, com o objetivo de preservar a cor e a turgidez da folhagem. Os melhores resultados foram com 8-HQ (0,69 mol/m³) e GA3 (0,058 mo1/m³). No segundo experimento, avaliaram-se os seguintes inibidores de etileno: tiossulfato de prata (STS), nitrato de prata (AgNO3) e cloreto de cobalto (COC1(2)). A melhor resposta foi obtida com AgNO3 (2,9 e 4,4 mo1/m³).

Este experimento foi realizado para avaliar os efeitos da suplementação de enzimas exógenas (pancreatina suína) em microdietas sobre o crescimento, a sobrevivência e as alterações morfológicas do trato digestório de larvas de pacu, Piaractus mesopotamicus. Foram testados oito programas alimentares: alimentação exclusiva com náuplios de Artemia (AV); alimentação exclusiva com dieta microparticulada com (DMP) ou sem (DM) suplementação enzimática; substituição, aos cinco dias, dos náuplios de Artemia por dietas inertes com (AV5DMP) ou sem (AV5DM) suplementação; e substituição dos náuplios aos dez dias por dietas com (AV10DMP) ou sem (AV10DM) suplementação. O experimento teve duração de 28 dias. Larvas que receberam o alimento vivo durante todo o período experimental apresentaram maiores médias de peso. O efeito negativo da supressão do alimento vivo sobre o crescimento das larvas foi verificado tanto na substituição aos cinco dias como aos dez dias. No entanto, nas avaliações biométricas subseqüentes, observaram-se efeitos positivos da suplementação enzimática; a partir do 20º dia de experimento, as larvas que receberam a dieta suplementada com enzima exógena apresentaram peso médio estatisticamente superior ao daquelas alimentadas com a dieta sem suplementação. As diferenças morfológicas mais evidentes proporcionadas pela suplementação enzimática foram observadas nas larvas que receberam substituição alimentar aos cinco dias. As diferenças foram relativas à quantidade de grânulos de zimogênio no pâncreas e às inclusões supranucleares no intestino. As larvas submetidas à transição alimentar aos dez dias de experimento já apresentavam diferenciação morfológica do sistema digestório mais avançada, assemelhando-se muito às larvas do tratamento com alimento vivo. Os resultados deste experimento indicam que a suplementação com pancreatina proporcionou efeitos positivos sobre o crescimento e a sobrevivência das larvas de pacu.

Objective: the purpose of this study was to evaluate, by scanning electron microscopy (SEM), the effects of Nd:YAG laser irradiation applied perpendicular or parallel to the root canal dentin wall. Methods: Thirty human teeth were divided into two groups: Group A (20 roots), laser application with circular movements, parallel to the dentin root surface; and Group B (10 roots), roots cut longitudinally and laser applied perpendicular to the root surface. Group A was subdivided into A1 (10 roots), laser application with 100 mJ, 15 Hz and 1.5 W; and A2 (10 roots) with 160 mJ, 15 Hz, and 2.4 W. Group B was subdivided into B1 (10 hemisections) and B2 (10 hemi-sections) with parameters similar to A I and A2. Four applications of 7-sec duration were performed, with a total exposure of 28 sec. SEM evaluations were made in the cervical, middle, and apical thirds, with 500X and 2000X magnifications. Morphological changes scores were attributed, and the results were submitted to Kruskal Wallis statistical test (5%). Results: Significant statistical differences were found between groups A and B (p = 0.001). In groups A1 and A2, few areas of dentin melting were observed. In groups B1 and B2, areas of melting dentin covering dentin surface were observed. Conclusions: It was concluded that intracanal laser application with circular movements (parallel to the surface) produces limited morphological changes in root canal dentin wall.

Diabetes mellitus can lead to reproductive disorders that in turn result in weakened fertility brought about by morphofunctional changes in the testes and accessory sex glands. However, doubts persist concerning the basic biology of the secretory epithelial cells and the stroma of the coagulating gland of diabetic mice. Thus, the objective of the present study was to analyze the histological and ultrastructural changes associated with stereology of the coagulating gland of mice with alloxan-induced diabetes, and of spontaneously diabetic mice. Sixteen mice of the C57BL/6J strain, and eight non-obese diabetic (NOD) mice were used. The animals were divided into three groups: 1) control (C), 2) alloxan diabetic (AD), and 3) NOD. Thirty days after the detection of diabetic status in group 2, all of the animals were killed and then perfused with Karnovsky's solution through the left cardiac ventricle. The coagulating gland was then removed and processed for morphometric study by light microscopy and electron microscopy. The results showed thickening of the stroma, atrophy of secretory epithelial cells, and disorganization of the organelles involved in the secretory process in both NOD and alloxan-induced mice. Thus, it may be concluded that the coagulating gland suffered drastic morphological changes, and consequently impaired glandular function, in the presence of diabetes mellitus type I in both NOD and AD mice. Purpose: To evaluate the effect of various bleaching agents on the cemento-enamel junction (CEJ) of human teeth by scanning electron microscopy (SEM) analysis. Methods: 30 intact teeth were selected and longitudinally sectioned, yielding 60 specimens. Thirty specimens served as controls; the other 30 were divided into six groups with five specimens each (n= 5) and bleached according to six protocols (Group 1: External bleaching with 10% carbamide peroxide; Group 2: External bleaching with 35% hydrogen peroxide; Group 3: External bleaching with 35% hydrogen peroxide; Group 4: Internal/external bleaching with 35% hydrogen peroxide; Group 5: Internal/external bleaching with 35% hydrogen peroxide; and Group 6: Intracoronal bleaching with a paste of sodium perborate mixed with 9% hydrogen peroxide). After treatment the specimens were prepared and examined in a scanning electron microscope. Results: the bleaching agents used in this study caused morphological changes in the CEJ and increased dentin exposure.

The biometric relationship between the weigth and the carapace width in the swimming crab A. cribrarius was compared with the results from other portunid crabs studied previously. During November/1988 to October/1989, a total of 403 specimens (189 males and 214 females) were collected with otter-trawl nets in north coast of the São Paulo State, Brazil. The animals were measured (carapace width excluding lateral spines = LC! and weighed (wet weight = PE). The empiric points of this relation were fit according to the power function (Y = a.X(b)) for each sex, maturation phases and total of individuals. The relation PE x LC indicates that the mole's growth changes during the ontogenesys from isometric (in juvenile phase) to allometric positive (in adult phase). For the females the growth is isometric in the two phases. The weight grows in a higher proportion than the carapace width variable (allometric positive growth). The data can be grouped in a single equation (PE = 7.85.10(-5).LC(3.14)) for the convertion between the variables there was a greater similarity between the equations obtained far each sex. In spite of this, the males present the fattening grade value (''a'') slightly higher than that of the females, possibly because of the greater size reached in its devellopment. The mean weight of the males is greater than the females one (p < 0.01). In the range 80 proves 90mm the males were more abundant, probably due to the females terminal ecdysis is near this size. The females only have the mean weight greater than the males in the 60 proves 70mm range (p < 0.01) when the puberty molt occurs and they present morphological changes in their reproductive system.

In the present study morphological changes occurring in the neuromuscular junctions (NMJ) of the extensor digitorum longus (EDL) and soleus muscles from albino rats (Rattus norvegicus) submitted to experimental chronic alcoholism were evaluated. Seventy two male animals aged 4 months and weighing on average 400 g were divided into three groups: control, alcoholic and isocaloric. Six rats from each group were anesthetized and sacrificed after 5, 10, 15 and 18 months. The NMJ did not show detectable morphological changes in either muscle after treatment when examined by light microscopy. With respect to the dimensions, statistical analysis demonstrated a tendency to a statistically significant treatment x time interaction for the length of soleus muscle NMJ. The ultrastructural study, however, revealed that the NMJ of the soleus muscle of animals submitted to 18 months of experimental alcoholism presented important morphological alterations. Characteristically, the NMJ of these muscles is located on an elevation on the surface of the muscle fiber, presenting a regular round, oval or elliptical shape and continuous and not very deep synaptic grooves. Approximately 30% of the NMJ of alcoholic rats are irregular in shape, with the sarcolemmal elevations typical of the synapse region being flattened on at least one side, with discontinuous synaptic grooves, and deep and punctiform contacts of the synaptic buds. These data suggest that, although skeletal muscle has a greater natural resistance against the direct or indirect effects of alcohol, some submicroscopic morphological alterations are detectable in the NMJ, especially in muscles with oxidative metabolism (soleus) following long periods of ingestion of alcohol. The addition of calcium chloride eletrolyte to sodium polyphosphate solutions lead to Calcium polyphosphate coacervates. The effects of a thermal treatment were investigated with the objective to increase the relative stability of the obtained material. Thermogravimetry analysis indicates that coacervates became less hydrophilic and more thermally stable after the thermal treatment. Crystallization was identified through differential scanning calorimetry and X-ray diffraction. Morphological changes were observed after the thermal treatment by scanning electron microscopy. N-2 adsorption-desorption isotherms suggest that both materials, thermally treated or not, display type IV isotherms, low superficial area and mesoporous structure. Stability experiments in solutions at different pH values show that the thermally treated calcium polyphosphate is relatively more stable than the non-treated coacervate.

Morphological changes of coagulating gland of Wistar rats submitted to the experimental chronic alcoholism were noted. Ultrastructurally, it was observed reduction of the granular endoplasmic reticulum cisternae and nuclei with basal position and irregular shape. The epithelial cells presented pycnotic nuclei and the mitochondrial cristae disappeared, characteristic of cellular degeneration.

The cytotoxicity of four calcium hydroxide-based root canal sealers (Sealapex, CRCS, Apexit, and Sealer 26) and one zinc oxide-eugenol-based sealer (Fill Canal) was evaluated microscopically for morphological changes in rat peritoneal macrophages. The least cytotoxic sealer was Fill Canal, followed in increasing order of cytotoxicity by CRCS, Sealer 26, Apexit, and Sealapex. Copyright © 2000 by The American Association of Endodontists.
