Mutations in Na+-glucose transporters (SGLT)-2 and hepatocyte nuclear factor (HNF)-1 alpha genes have been related to renal glycosuria and maturity-onset diabetes of the young 3, respectively. However, the expression of these genes have not been investigated in type 1 and type 2 diabetes. Here in kidney of diabetic rats, we tested the hypotheses that SGLT2 mRNA expression is altered; HNF-1 alpha is involved in this regulation; and glycemic homeostasis is a related mechanism. The in vivo binding of HNF-1 alpha into the SGLT2 promoter region in renal cortex was confirmed by chromatin immunoprecipitation assay. SGLT2 and HNF-1 alpha mRNA expression (by Northern and RT-PCR analysis) and HNF-1 binding activity of nuclear proteins (by EMSA) were investigated in diabetic rats and treated or not with insulin or phlorizin (an inhibitor of SGLT2). Results showed that diabetes increases SGLT2 and HNF-1 alpha mRNA expression (similar to 50%) and binding of nuclear proteins to a HNF-1 consensus motif (similar to 100%). Six days of insulin or phlorizin treatment restores these parameters to nondiabetic-rat levels. Moreover, both treatments similarly reduced glycemia, despite the differences in plasma insulin and urinary glucose concentrations, highlighting the plasma glucose levels as involved in the observed modulations. This study shows that SGLT2 mRNA expression and HNF-1 alpha expression and activity correlate positively in kidney of diabetic rats. It also shows that diabetes-induced changes are reversed by lowering glycemia, independently of insulinemia. Our demonstration that HNF-1 alpha binds DNA that encodes SGLT2 supports the hypothesis that HNF-1 alpha, as a modulator of SGLT2 expression, may be involved in diabetic kidney disease.

The effect of glucose on the intracellular pH (pH(i)) recovery rate (dpH(i)/dt) and Na(+)-glucose transporter (SGLT) localization was investigated in HEK-293 cells, a cell line that expresses endogenous NHE1, NHE3, SGLT1, and SGLT2 proteins. The activity of the Na(+)/H(+) exchangers (NHEs) was evaluated by using fluorescence microscopy. The total and membrane protein expression levels were analyzed by immunoblotting. In cells cultivated in 5 mM glucose, the pH(i) recovery rate was 0.169 +/- A 0.020 (n = 6). This value did not change in response to the acute presence of glucose at 2 or 10 mM, but decreased with 25 mM glucose, an effect that was not observed with 25 mM mannitol. Conversely, the chronic effect of high glucose (25 mM) increased the pH(i) recovery rate (similar to 40%, P < 0.05), without changes in the total levels of NHE1, NHE3, or SGLT1 expression, but increasing the total cellular (similar to 50%, P < 0.05) and the plasma membrane (similar to 100%, P < 0.01) content of SGLT2. Treatment with H-89 (10(-6) M) prevented the stimulatory effect of chronic glucose treatment on the pH(i) recovery rate and SGLT2 expression in the plasma membrane. Our results indicate that the effect of chronic treatment with a high glucose concentration is associated with increased NHEs activity and plasma membrane expression of SGLT2 in a protein kinase A-dependent way. The present results reveal mechanisms of glucotoxicity and may contribute to understanding the diabetes-induced damage of this renal epithelial cell.

Glutamine is the most important donor of NH(3) in kidney playing an important role in acid-base buffering system. Besides this effect, glutamine presents many other relevant functions in the whole body, such as a precursor of arginine in adult and neonates. In addition to these effects, some studies have shown that glutamine can potentiate renal disease. In the present study, the effect of short-term treatment (15 days) with glutamine on control and diabetic rats was investigated. Using biochemical, histological and molecular biology analysis from control and diabetic rats we verified that glutamine supplementation increase in pro-inflammatory interleukins (IL)-1 beta and IL-6 content in renal cortex and induce alteration in glomerular characteristics. This study showed that short-term treatment with glutamine in association with increased glucose levels could cause important alterations in glomerular morphology that may result in fast progression of kidney failure.

Introduction: Cytokines (IL-6, IL-10 and TNF-alpha) are increased after exhaustive exercise in the rat retroperitoneal (RPAT) and mesenteric adipose tissue (MEAT) pads. On the other hand, these cytokines show decreased expression in these depots in response to a chronic exercise protocol. However, the effect of exercise with overload combined with a short recovery period on pro-and anti-inflammatory cytokine expression is unknown. In the present study, we investigated the regulation of cytokine production in the adipose tissue of rats after an overtraining-inducing exercise protocol. Methods: Male Wistar rats were divided into four groups: Control (C), Trained (Tr), Overtrained (OT) and recovered overtrained (R). Cytokines (IL-6, TNF-alpha and IL-10) levels and Toll Like Receptor 4 (TLR4), Nuclear Factor kBBp65 (NF-kBp65), Hormone Sensitive Lipase (HSL) and, Perilipin protein expression were assessed in the adipose tissue. Furthermore, we analysed plasma lipid profile, insulin, testosterone, corticosterone and endotoxin levels, and liver triacylglycerol, cytokine content, as well as apolipoprotein B (apoB) and TLR4 expression in the liver. Results: OT and R groups exhibited reduced performance accompanied by lower testosterone and increased corticosterone and endotoxin levels when compared with the control and trained groups. IL-6 and IL-10 protein levels were increased in the adipose tissue of the group allowed to recover, in comparison with all the other studied groups. TLR-4 and NF-kBp65 were increased in this same group when compared with both control and trained groups. The protein expression of HSL was increased and that of Perilipin, decreased in the adipose in R in relation to the control. In addition, we found increased liver and serum TAG, along with reduced apoB protein expression and IL-6 and IL-10 levels in the of R in relation to the control and trained groups. Conclusion: In conclusion, we have shown that increases in pro-inflammatory cytokines in the adipose tissue after an overtraining protocol may be mediated via TLR-4 and NF-kBp65 signalling, leading to an inflammatory state in this tissue.

Several studies have implicated the renin angiotensin system in the cardiac hypertrophy induced by thyroid hormone. However, whether Angiotensin type 1 receptor (AT(1)R) is critically required to the development of T(3)-induced cardiomyocyte hypertrophy as well as whether the intracellular mechanisms that are triggered by AT(1)R are able to contribute to this hypertrophy model is unknown. To address these questions, we employed a selective small interfering RNA (siRNA, 50 nM) or an AT(1)R blocker (Losartan, 1 mu M) to evaluate the specific role of this receptor in primary cultures of neonatal cardiomyocytes submitted to T(3) (10 nM) treatment. The cardiomyocytes transfected with the AT(1)R siRNA presented reduced mRNA (90%, P < 0.001) and protein (70%, P < 0.001) expression of AT(1)R. The AT(1)R silencing and the AT(1)R blockade totally prevented the T(3)-induced cardiomyocyte hypertrophy, as evidenced by lower mRNA expression of atrial natriuretic factor (66%, P < 0.01) and skeletal alpha-actin (170%, P < 0.01) as well as by reduction in protein synthesis (85%, P < 0.001). The cardiomyocytes treated with T(3) demonstrated a rapid activation of Akt/GSK-3 beta/mTOR signaling pathway, which was completely inhibited by the use of PI3K inhibitors (LY294002, 10 mu M and Wortmannin, 200 nM). In addition, we demonstrated that the AT(1)R mediated the T(3)-induced activation of Akt/GSK-3 beta/mTOR signaling pathway, since the AT(1)R silencing and the AT(1)R blockade attenuated or totally prevented the activation of this signaling pathway. We also reported that local Angiotensin I/II (Ang I/II) levels (120%, P < 0.05) and the AT(1)R expression (180%, P < 0.05) were rapidly increased by T(3) treatment. These data demonstrate for the first time that the AT(1)R is a critical mediator to the T(3)-induced cardiomyocyte hypertrophy as well as to the activation of Akt/GSK-3 beta/mTOR signaling pathway. These results represent a new insight into the mechanism of T(3)-induced cardiomyocyte hypertrophy, indicating that the Ang I/II-AT(1)R-Akt/GSK-3 beta/mTOR pathway corresponds to a potential mediator of the trophic effect exerted by T(3) in cardiomyocytes.

Ischemia followed by reperfusion is known to negatively affect mitochondrial function by inducing a deleterious condition termed mitochondrial permeability transition. Mitochondrial permeability transition is triggered by oxidative stress, which occurs in mitochondria during ischemia-reperfusion as a result of lower antioxidant defenses and increased oxidant production. Permeability transition causes mitochondrial dysfunction and can ultimately lead to cell death. A drug able to minimize mitochondrial damage induced by ischemia-reperfusion may prove to be clinically effective. We aimed to analyze the effects of nicorandil, an ATP-sensitive potassium channel agonist and vasodilator, on mitochondrial function of rat hearts and cardiac HL-1 cells submitted to ischemia-reperfusion. Nicorandil decreased mitochondrial swelling and calcium uptake. It also decreased reactive oxygen species formation and thiobarbituric acid reactive substances levels, a lipid peroxidation biomarker. We thus confirm previous reports that nicorandil inhibits mitochondrial permeability transition and demonstrate that nicorandil inhibits this process by preventing oxidative damage and mitochondrial calcium overload induced by ischemia-reperfusion, resulting in improved cardiomyocyte viability. These results may explain the good clinical results obtained when using nicorandil in the treatment of ischemic heart disease.

Iron supplementation in hemodialysis patients is fundamental to erythropoiesis, but may cause harmful effects. We measured oxidative stress using labile plasma iron (LPI) after parenteral iron replacement in chronic hemodialysis patients. Intravenous iron saccharate (100 mg) was administered in patients undergoing chronic hemodialysis (N = 20). LPI was measured by an oxidant-sensitive fluorescent probe at the beginning of dialysis session (T0), at 10 min (T1), 20 min (T2), and 30 min (T3) after the infusion of iron and at the subsequent session; P < 0.05 was significant. The LPI values were significantly raised according to the time of administration and were transitory: -0.02 +/- 0.20 mu mol/L at the beginning of the first session, 0.01 +/- 0.26 mu mol/L at T0, 0.03 +/- 0.23 mu mol/L at T1, 0.09 +/- 0.28 mmol/L at T2, 0.18 +/- 0.52 mmol/L at T3, and -0.02 +/- 0.16 mmol/L (P = 0.001 to 0.041) at the beginning of the second session. The LPI level in patients without iron supplementation was -0.06 +/- 0.16 mmol/L. Correlations of LPI according to time were T1, T2, and T3 vs. serum iron (P = 0.01, P = 0.007, and P = 0.0025, respectively), and T2 and T3 vs. transferrin saturation (P = 0.001 and P = 0.0003, respectively). LPI generation after intravenous saccharate administration is time-dependent and transitorily detected during hemodialysis. The LPI increment had a positive correlation to iron and transferrin saturation.

The catalase mimetic complex Mn(III)-salen chloride (EUK8) was found to be pro-oxidant under low hydrogen peroxide concentrations. The increase in the fluorescence rate of the probe 1,2,3-dihydrorhodamine (DHR) in solution, as well as the carbonyl content of human serum albumin were found to be maximum at H(2)O(2):EUK8 molar ratios ranging from 0 to 2, supporting previous findings regarding the mechanism of EUK8 catalase activity and the formation of highly oxidative Mn(V)-O(2-) species. This pro-oxidant effect is precluded by the presence of glutathione. Cytotoxicity to HeLa cells, as probed by increased rate of oxidation of intracellular DHR, was not observed. Our findings suggest that the combination of H(2)O(2) and EUK8 at specific molar ratios, in the absence of reductants/antioxidants, induces the oxidation of organic molecules. It is shown that the fluorimetric determination of pro-oxidant activity of metal complexes is more sensitive than the colorimetric quantification of protein carbonyl content. The implications of our findings with respect to the somewhat confusing results arising from in vivo studies of EUK8 and other Mn(III) anti-oxidant metal complexes are discussed.

Problem: Arbetsmiljön är viktig för människors välbefinnande. Hälsofrämjande faktorer antas inte bara styrka individens fysiska och psykiska hälsa, utan även företagets konkurrenskraft och lönsamhet. I uppsatsen undersöker vi hur företaget SSAB i Borlänge arbetar med hälsa genom att tillämpa ett hälsofrämjande perspektiv och utgå från teorin om Känslan av sammanhang (KASAM). Ledarskapsvärderingar har betydelse för medarbetares hälsa (Hanson, 2004) och vi undersöker vilken uppfattning ett antal chefer har om sina roller i det hälsofrämjande arbetet och deras syn på medarbetarundersökningen HälsoSAM som företagshälsovården på SSAB bedriver. Arbetsbelastningen på medarbetare och chefer ökar till följd av sparkrav, samtidigt som resurserna minskar (Gatu, 2003). Följden blir en större risk för ohälsa och ett sätt att minska sjukfrånvaron är att genomföra hälsofrämjande insatser (Prevent, 2001). Syfte: Syftet med studien är att utifrån ett hälsofrämjande perspektiv förklara vad chefer har för möjligheter att skapa förutsättningar för att främja medarbetarnas hälsa på SSAB i Borlänge. Metod: Den metod som ligger till grund för uppsatsen baseras på ett kvalitativt angreppssätt där semistrukturerade intervjuer samt litteraturstudier genomförts för att samla information. Analys: Ledarens beteende påverkar medarbetarna. Hög arbetsbelastning på SSABs chefer leder till lägre närvaro bland medarbetarna, vilket i sin tur ger sämre förutsättningar för att främja hälsan. HälsoSAM kartlägger medarbetarnas hälsoläge, både välbefinnande och arbetskapacitet. Resultaten är vägledande i det hälsofrämjande arbetet men överbelastning, tidsbrist och kunskapsbrist hos cheferna bidrar till att uppföljningen inte blir systematisk. För ett väl fungerande hälsoarbete krävs systematik. Slutsats: Hälsofrämjande processer i arbetslivet skapas genom balans mellan krav och resurser. På SSAB i Borlänge hindras hälsoarbetet av tidsbrist och kunskapsbrist hos cheferna. Kostnadsbesparingar påverkar hälsoarbetet negativt och systematiken i uppföljningsarbetet blir lidande. Ansvaret för hälsoarbetet läggs på företagshälsovården, men utan stöd från chefer blir inte hälsoarbetet en naturlig del i verksamheten. Det är viktigt att i rådande situation se vad främjande av hälsa kan ge tillbaka till företaget i både ekonomiska och kvalitativa termer. Insikt om detta ökar chefernas incitament att prioritera hälsofrämjande processer och organisera arbetet så att människor har förutsättningar att hantera, kontrollera och klara av sina uppgifter.

Entrepreneurs are portrayed as salient drivers of regional development and for a number of years nascent entrepreneurs have been studied in a large number of countries as part of the Global Entrepreneurship Monitor project and the Panel Study of Entrepreneurial Dynamics. Scholars have devoted much effort to investigating factors that determine how individuals engage in entrepreneurial activities, with most of the discussion limited to business start-ups. However, since this type of project does not follow identical nascent entrepreneurs over time, limited knowledge exists about their development and whether they stay in this nascent phase for a long time. In practice, it is common for entrepreneurs to run a business and at the same time work in wage work, so-called combining entrepreneurs. In Sweden, almost half of all business owners combine wage work with a business. However, not all combining entrepreneurs will eventually decide to leave the wage work and invest fully in the business. Consequently, much research has focused on the first step of entering entrepreneurship full time, but less has focused on the second step, the transition from the combining phase to full-time self-employment. The aim of this thesis is therefore to contribute to the theory of entrepreneurship by gaining a deeper understanding of combining entrepreneurs and their motives and intentions. In the context of combining entrepreneurs, the theory of identity, resources and choice overload has been used to examine how entrepreneurs’ age (when starting the business), entrepreneurial tenure (the length of engagement in the side-business), hours spent (weekly involvement in the side-business), involvement in entrepreneurial teams (leading the business with one or more partners) and involvement in networks (business networks) influence their passion for engaging in entrepreneurship while sustaining wage work. Different categories of combining entrepreneurs and their intentions have also been examined. A survey was administered to 1457 entrepreneurs within the creative sector in two counties in Sweden (Gävleborgs County and Jämtlands County). Since there were no separate mailing lists to only combining entrepreneurs, the survey was sent to all entrepreneurs within the chosen industry and counties. The total response rate was 33.5 percent and of them 57.6 percent combined, yielding 262 combining entrepreneurs who answered the questionnaire. The survey was then followed up with eight focus group interviews and two single interviews to validate the answers from the questionnaire. The results indicate three types of combining entrepreneurs: nascent – with the intention to leave the combining phase for a transition into full-time self-employment, lifestyle – with the intention to stay in the combining phase, and occasional – with the intention to leave the combining phase for full-time wage work and close down the business. Transitioning fully to self-employment increases with the individual’s age. Also, a positive interactive effect exists with involvement in entrepreneurial networks. The results also indicate that the ability to work with something one is passionate about is the top motive for combining wage work with a side-business. Passion is also more likely to be the main motive behind the combining form among individuals who are older at business start-up, but passion is less likely to be the main motive behind the combining form among individuals who spend more time on the business. The longer the individual has had the side-business, the less likely passion is the main motive behind the combining form, and passion is less likely to be the main motive among those who are part of an entrepreneurial team.

Dentro da literatura de Comportamento do Consumidor e Teoria da Decisão existe considerável corpo teórico que analisa sentimentos negativos e reações adversas no processo decisório de compras de produtos de alto e baixo envolvimento. Vários fenômenos são identificados como negativos no processo, principalmente a Confusão do Consumidor, que compreende três dimensões: i) muitas informações similares sobre produtos, ii) muitas informações sobre diferentes produtos e iii) informações falsas e ambíguas. Tal fenômeno, no entanto, parece ser moderado por um conjunto de variáveis, como o Envolvimento, a Experiência e a Restrição de Tempo (moderadoras da relação entre Confusão do Consumidor e Intenção de Compra). Este fato foi identificado através de entrevistas em profundidade. Os resultados das entrevistas permitiram identificar as variáveis moderadoras, assim como a existência do fenômeno e sua relação com a decisão final de compra. Na segunda fase da pesquisa, supõe-se que indivíduos com baixo Envolvimento e Restrição de Tempo possuam uma propensão maior à confusão. No Estudo 2 foram utilizados como moderadores o Envolvimento e a Restrição de Tempo, ambos manipulados por instrução, sendo as variáveis dependentes a Intenção de Compra e a Confusão do Consumidor. Os resultados do Estudo 2 permitiram inferir que existem diferenças significativas entre os grupos, quando analisada a variável Confusão do Consumidor, mas, em alguns grupos, a Intenção de Compra não era significativamente diferente. No Estudo 3 foram manipuladas a Experiência (forte e fraca) e a Confusão do Consumidor, sendo a variável dependente a Intenção de Compra. Os resultados do Estudo 3 também permitiram inferir que existem diferenças significativas entre os grupos na Intenção de compra, quando consideradas baixa ou alta confusão, assim como Experiência forte ou fraca. Na última fase da pesquisa foram destacadas as estratégias dos consumidores para lidar com o fenômeno Confusão do Consumidor. Tais estratégias, muitas vezes, são mediadoras de comportamentos posteriores, como a compra do produto. No Estudo 4 manipulou-se a Confusão do Consumidor em duas de suas dimensões. Foi possível destacar a preponderância da estratégia por busca de informações e postergação da decisão, quando o consumidor se depara com situações confusas.

Através desta pesquisa, analisa-se a lei 9.957/2000, que instituiu o procedimento sumaríssimo no processo do trabalho. Procura-se identificar os modelos de procedimentos utilizados na tramitação do processo trabalhista, notadamente o procedimento sumaríssimo e sua eficácia, dentro da estrutura organizacional da Justiça do Trabalho. Busca-se demonstrar a necessidade do cumprimento da lei n. 9.957/2000, como instrumento viável para uma efetiva prestação jurisdicional da Justiça do Trabalho, nas ações de valor abaixo de quarenta salários mínimos. Para atingir tais propósitos, abordam-se, inicialmente, aspectos teóricos fundamentais relacionados ao acesso à justiça, aos princípios constitucionais da duração razoável do processo e da segurança jurídica e, ainda, a interpretação e integração da legislação trabalhista. Em um segundo momento, demonstra-se a necessidade de reforma na estrutura organizacional da Justiça do Trabalho para uma integral aplicabilidade do procedimento sumaríssimo, em face da sobrecarga de processos em tramitação nas varas do trabalho. Analisa-se, também, a visão dos operadores do Direito do Trabalho, jurisdicionados e serventuários da Justiça do Trabalho sobre o procedimento sumaríssimo, na qualidade de instrumento eficaz na busca de uma prestação jurisdicional célere e, por isso, efetiva. Por fim, expõe-se a proposta de criação de varas do trabalho específicas para julgamento de processos tramitados pelo procedimento sumaríssimo, como instrumento de celeridade da atividade jurisdicional e de redução do tempo de duração do processo trabalhista.

O trabalho ocupa um inegável espaço na vida dos indivíduos, sendo este rico de sentido individual e social, é um meio de produção da vida de cada um ao prover subsistência, criar sentidos existenciais ou contribuir na estruturação da identidade e da subjetividade. O cenário atual de adoção e uso das Tecnologias de Informação e Comunicação pelas organizações e pessoas, demonstra um crescimento da incorporação de dispositivos tecnológicos que têm o potencial de alterar as características espaciais e temporais do trabalho, aumentando a mobilidade do trabalhador. Considerando-se o fato de que o indivíduo ao deslocar-se, ao mover-se pelo espaço, anexa ou remodela sua identidade pessoal, profissional, cultural, social ou política a partir da mobilidade, o objetivo final desta pesquisa foi refletir sobre de que maneira os impactos positivos e negativos gerados pela intensa mobilidade geográfica demandados pelo exercício da profissão, são percebidos por trabalhadores docentes de curso de pós-graduação lato sensu em Administração que se deslocam de um território para outro(s), com a finalidade de exercerem suas atividades docentes. Realizou-se para tanto uma revisão bibliográfica acerca de sentido do trabalho, apresentando, a seguir, algumas reflexões sobre as alterações ocorridas nas relações de trabalho em decorrência de mutações no mundo do trabalho, abordando, por fim, o tema Mobilidade da força de trabalho. Após esta etapa, buscou-se compreender, na ótica desses trabalhadores docentes, por meio de entrevistas pessoais analisadas à luz do método de análise de conteúdo, os caminhos percorridos e significados atribuídos por eles a estas vivências, procurando perceber como os trabalhadores docentes em destaque representam o olhar sobre si e sobre os outros, visando demonstrar o significado (positivo e negativo) da mobilidade espacial, as práticas cotidianas e estratégias de sociabilidade e interação com e no local de destino. Pode-se concluir que, apesar de haver alguns fatores que podem tornar os constantes deslocamentos geográficos destes trabalhadores um elemento negativo para eles, uma vez que são fontes de alta demanda física e mental, geradoras de estresse, sobrecarga, tensão emocional, cansaço, ansiedade, desânimo, frustração e descontentamento em relação ao trabalho, levando-os ao afastamento constante do contexto social, podendo resultar em consequências graves para a saúde deste trabalhador. Estes constantes deslocamentos geográficos tornam-se, contudo, um elemento positivo, uma vez que possibilitam o incremento às suas práticas docentes, por meio das vivências enriquecidas pelos contactos estabelecidos, pelas aprendizagens realizadas e pelas capacidades de relacionamento em meios multiculturais adquiridas, tornando-se espaço de autorrealização, inserção e reconhecimento social, prestígio, status e de senso de utilidade.

Esta dissertação busca investigar a hipótese de sobrecarga de opções, denominada em inglês como choice overload, associada ao aumento de complexidade e da quantidade de informações disponíveis, ou information overload, abordada e discutida por pesquisadores como O’Donoghue e Rabin (1998), Iyengar e Lepper (2000), Schwartz (2002), Iyengar, Jiang e Huberman (2003) e Gourville e Soman (2005). Neste trabalho, a investigação será no contexto da indústria de produtos de investimento brasileira. Para isso, através de uma pesquisa empírica, simulamos a decisão de um investidor comum diante de diferentes cenários propostos, com menos ou mais opções e menos ou mais informações. Em linha com as pesquisas citadas, evidências mostraram a importância de se considerar e avaliar esses efeitos, ainda que hipóteses previamente estabelecidas não tenham sido confirmadas e experimentos mais amplos precisem ser realizados. Neste estudo, quando a oferta se resume a apenas um fundo de investimento, mesmo que claramente melhor do que o cenário inicial, um alto índice de 32% ainda decide procrastinar a decisão, ou seja, apresentar apenas uma opção, o extremo oposto de choice overload, não se apresentou como uma solução eficaz.
