Transformation of small avascular masses of tumor cells into rapidly progressive cancers is triggered by the angiogenic switch, a process that involves vascular endothelial growth factor (VEGF) signaling. We have shown that VEGF enhances the survival and angiogenic potential of endothelial cells by activating the Bcl-2-CXCL8 signaling axis. The purpose of this study was to evaluate the effect of a small-molecule inhibitor of VEGF receptors (PTK/ZK) on the initial stages of head and neck tumor angiogenesis. In vitro, PTK/ZK blocked head and neck tumor cell (OSCC3 or UM-SCC-17B)-induced Bcl-2 and CXCL8 expression in endothelial cells. Oral administration of PTK/ZK decreased xenograft head and neck tumor microvessel density, and inhibited Bcl-2 and CXCL8 expression in tumor-associated endothelial cells. Analysis of these data demonstrates that PTK/ZK blocks downstream targets of VEGF signaling in endothelial cells, and suggests that PTK/ZK may inhibit the angiogenic switch in head and neck tumors. Reproductive experience (i.e., pregnancy and lactation) induces physiological changes in mammals. We recently showed that a previous reproductive experience can modulate the activity of dopaminergic hypothalamic systems while decreasing serum prolactin (PRL) levels and oxidative burst activity in peritoneal macrophages. Dopamine receptor antagonists increase serum PRL levels, and both PRL and dopamine receptors might be involved in the modulation of macrophage activity, providing a means of communication between the nervous and immune systems. The present study evaluated the in vitro effects of PRL and the dopamine receptor 02 antagonist domperidone (DOMP) on the peritoneal activity of macrophages from primiparous and multiparous female rats during lactation. Oxidative bursts and phagocytosis in peritoneal macrophages were evaluated by flow cytometry. Primiparous and multiparous Wistar rats, during the period of lactation (i.e., days 5-7 after parturition) were used. Samples of peritoneal fluid from these rats were first incubated with PRL (10 and 100 nM) for different periods of time. The same procedure was repeated to evaluate the effects of DOMP (10 and 100 nM). Our results showed that macrophages from multiparous rats respond more effectively to in vitro incubation with PRL, especially with regard to oxidative bursts and the percentage of phagocytosis. Additionally, these effects were more pronounced after 30 min of incubation. These data suggest that reproductive experience is associated with a reduction in serum PRL levels, and cells in experienced female animals, including their macrophages, become more sensitive to the effects of PRL (C) 2011 Elsevier Ireland Ltd. Using the classical twin design, this study investigates the influence of genetic factors on the large phenotypic variance in inspection time (IT), and whether the well established IT-IQ association can be explained by a common genetic factor. Three hundred ninety pairs of twins (184 monozygotic, MZ; 206 dizygotic, DZ) with a mean age of 16 years participated, and 49 pairs returned approximately 3 months, later for retesting. As in many IT studies, the pi figure stimulus was used and IT was estimated from the cumulative normal ogive. IT ranged from 39.4 to 774.1 ms (159 +/- 110.1 ms) with faster ITs (by an average of 26.9 ms) found in the retest session from which a reliability of .69 was estimated. Full-scale IQ (FIQ) was assessed by the Multidimensional Aptitude Battery (MAB) and ranged from 79 to 145 (111 +/- 13). The phenotypic association between IT and FIQ was confirmed (- .35) and bivariate results showed that a common genetic factor accounted for 36% of the variance in IT and 32% of the variance in FIQ. The maximum likelihood estimate of the genetic correlation was - .63. When performance and verbal IQ (PIQ & VIQ) were analysed with IT, a stronger phenotypic and genetic relationship was found between PIQ and IT than with VIQ. A large part of the IT variance (64%) was accounted for by a unique environmental factor. Further genetic factors were needed to explain the remaining variance in IQ with a small component of unique environmental variance present. The separability of a shared genetic factor influencing IT and IQ from the total genetic variance in IQ suggests that IT affects a specific subcomponent of intelligence rather than a generalised efficiency. The genetic relationship between lower (information processing speed), intermediate (working memory), and higher levels (complex cognitive processes as indexed by IQ) of mental ability was studied in a classical twin design comprising 166 monozygotic and 190 dizygotic twin pairs. Processing speed was measured by a choice reaction time (RT) task (2-, 4-, and 8-choice), working memory by a visual-spatial delayed response task, and IQ by the Multidimensional Aptitude Battery. Multivariate analysis, adjusted for test-retest reliability, showed the presence of a genetic factor influencing all variables and a genetic factor influencing 4- and 8-choice RTs, working memory, and IQ. There were also genetic factors specific to 8-choice RT, working memory, and IQ. The results confirmed a strong relationship between choice RT and IQ (phenotypic correlations: -0.31 to -0.53 in females, -0.32 to -0.56 in males; genotypic correlations: -0.45 to -0.70) and a weaker but significant association between working memory and IQ (phenotypic: 0.26 in females, 0.13 in males; genotypic: 0.34). A significant part of the genetic variance (43%) in IQ was not related to either choice RT or delayed response performance, and may represent higher order cognitive processes.

Amultidisciplinary collaborative study examining cognition in a large sample of twins is outlined. A common experimental protocol and design is used in The Netherlands, Australia and Japan to measure cognitive ability using traditional IQ measures (i.e., psychometric IQ), processing speed (e.g., reaction time [RT] and inspection time [IT]), and working memory (e.g., spatial span, delayed response [DR] performance). The main aim is to investigate the genetic covariation among these cognitive phenotypes in order to use the correlated biological markers in future linkage and association analyses to detect quantitativetrait loci (QTLs). We outline the study and methodology, and report results from our preliminary analyses that examines the heritability of processing speed and working memory indices, and their phenotypic correlation with IQ. Heritability of Full Scale IQ was 87% in the Netherlands, 83% in Australia, and 71% in Japan. Heritability estimates for processing speed and working memory indices ranged from 33–64%. Associations of IQ with RT and IT (−0.28 to −0.36) replicated previous findings with those of higher cognitive ability showing faster speed of processing. Similarly, significant correlations were indicated between IQ and the spatial span working memory task (storage [0.31], executive processing [0.37]) and the DR working memory task (0.25), with those of higher cognitive ability showing better memory performance. These analyses establish the heritability of the processing speed and working memory measures to be used in our collaborative twin study of cognition, and support the findings that individual differences in processing speed and working memory may underlie individual differences in psychometric IQ.

Genetic and environmental sources of covariation among the P3(00) and online performance elicited in a delayed-response working memory task, and psychometric IQ assessed by the multidimensional aptitude battery, were examined in an adolescent twin sample. An association between frontal P3 latency and task performance (phenotypic r = -0.33; genotypic r = -0.49) was indicated, with genes (i.e. twin status) accounting for a large part of the covariation ( > 70%). In contrast, genes influencing P3 amplitude mediated only a small part (2%) of the total genetic variation in task performance. While task performance mediated 15% of the total genetic variation in IQ (phenotypic r = 0.22; genotypic r = 0.39) there was no association between P3 latency and IQ or P3 amplitude with IQ. The findings provide some insight into the inter-relationships among psychophysiological, performance and psychometric measures of cognitive ability, and provide support for a levels-of-processing genetic model of cognition where genes act on specific sub-components of cognitive processes.

Body mass index (BMI), a simple anthropometric measure, is the most frequently used measure of adiposity and has been instrumental in documenting the worldwide increase in the prevalence of obesity witnessed during the last decades. Although this increase in overweight and obesity is thought to be mainly due to environmental changes, i.e., sedentary lifestyles and high caloric diets, consistent evidence from twin studies demonstrates high heritability and the importance of genetic differences for normal variation in BMI. We analysed self-reported data on BMI from approximately 37,000 complete twin pairs (including opposite sex pairs) aged 20-29 and 30-39 from eight different twin registries participating in the GenomEUtwin project. Quantitative genetic analyses were conducted and sex differences were explored. Variation in BMI was greater for women than for men, and in both sexes was primarily explained by additive genetic variance in all countries. Sex differences in the variance components were consistently significant. Results from analyses of opposite sex pairs also showed evidence of sex-specific genetic effects suggesting there may be some differences between men and women in the genetic factors that influence variation in BMI. These results encourage the continued search for genes of importance to the body composition and the development of obesity. Furthermore, they suggest that strategies to identify predisposing genes may benefit from taking into account potential sex specific effects.

A major component of variation in body height is due to genetic differences, but environmental factors have a substantial contributory effect. In this study we aimed to analyse whether the genetic architecture of body height varies between affluent western societies. We analysed twin data from eight countries comprising 30,111 complete twin pairs by using the univariate genetic model of the Mx statistical package. Body height and zygosity were self-reported in seven populations and measured directly in one population. We found that there was substantial variation in mean body height between countries; body height was least in Italy (177 cm in men and 163 cm in women) and greatest in the Netherlands (184 cm and 171 cm, respectively). In men there was no corresponding variation in heritability of body height, heritability estimates ranging from 0.87 to 0.93 in populations under an additive genes/unique environment (AE) model. Among women the heritability estimates were generally lower than among men with greater variation between countries, ranging from 0.68 to 0.84 when an additive genes/shared environment/unique environment (ACE) model was used. In four populations where an AE model fit equally well or better, heritability ranged from 0.89 to 0.93. This difference between the sexes was mainly due to the effect of the shared environmental component of variance, which appears to be more important among women than among men in our study populations. Our results indicate that, in general, there are only minor differences in the genetic architecture of height between affluent Caucasian populations, especially among men.

Analisa o curso de Pedagogia pela perspectiva das/os alunas/os concluintes, tomando a produção de subjetividades como eixo central. Potencializa os discursos pelas narrativas das/os alunas/os, a forma como se constituíram como professoras/es e pedagogas/os no decorrer do curso, fazendo as correlações com as normatizações curriculares e as dimensões vivenciadas por elas/es. Toma o currículo como um conjunto de elementos discursivos, normativos, de conversas, de compartilhamento de experiências, e potencializa-o através dos encontros, de uma formação para o outro, das lacunas prescritivas, formando assim uma complexa produção de subjetividades, sempre coletiva, que se esforça para abandonar os clichês dos significados e as molaridades da prescrição curricular. Tem como principais intercessores teóricos ao problematizar as produções subjetivas no que tange ao currículo, Gilles Deleuze, Félix Guattari e Janete Magalhães Carvalho. Utiliza a cartografia como acompanhamento de processos em que não há uma coleta de dados, mas uma produção desses, pautada no recurso metodológico das narrativas – formas de conversações em que há o principal recurso na produção dos dados. Assim, o processo tornou-se intenso e delicado, mas muito feliz, por ter priorizado as muitas formas de vivenciar esse currículo, sem uma individualização dos sujeitos, mas sempre a partir da perspectiva de uma subjetividade a-centrada, sem rostidade.

O presente trabalho versa sobre modos de vida no contemporâneo referente às práticas em torno de medicamentos ansiolíticos. Propomos um modo de pesquisa que não visa a descoberta de verdades, mas a escuta e a escrita das sensibilidades em processo na atualidade. Apropriamo-nos do cinema e da entrevista, intercessores deste trabalho, para dar vazão às vozes que se proferem em torno dessa temática. Os filmes: A pele que habito, Medianeras e O palhaço, somado a duas entrevistas com usuários de ansiolíticos, compõe o corpo desta pesquisa. As análises e a escuta deste campo nos levaram a uma discussão paradoxal entre modos de vida os quais, ao mesmo tempo em que nos lança em ritmos cada vez mais velozes e adoecedores - resultado das forças capitalísticas, tecnológica e midiática – também produzem desejo de anestesiamento do corpo, para suportar a saturação a qual nos encontramos. Corpos que não têm agüentado mais os novos ritmos de vida e, por isso, pedem uma nova ética, uma política de recusa aos ritmos estafantes. Uma discussão com base nos autores Deleuze, Guattari, Nietzsche, Spinoza e Foucault.

A pesquisa analisa as relações entre interculturalidade, práxis e educação escolar indígena Tupinikim e Guarani do município de Aracruz, Espírito Santo, Brasil. Investiga a práxis da educação intercultural no espaço da educação escolar indígena como meio de revitalização das culturas Tupinikim e Guarani. Objetiva problematizar a formação inicial e continuada dos professores indígenas; discutir a práxis da interculturalidade no contexto da educação escolar indígena; e, identificar outros espaços educativos da cultura e educação indígena. Analisa aspectos teóricos e práticos sobre cultura (WILLIAMS, 2008; BRANDÃO, 1989; FORQUIN, 1993; CANDAU, 2011; GEERTZ, 1989), interculturalidade (D‘AMBROSIO,1996; FLEURI, 2002; 2003; SCANDIUZZI, 2009;), identidade e alteridade (MELIÁ, 2000; FREIRE, 1981; 1987; LITAIFF, 2004) e práxis (FREIRE, 1989; VÁSQUEZ, 2011; SEMERARO, 2006) e educação (escolar) indígena de acordo com a legislação vigente. Realiza pesquisa interpretativa (GEERTZ, 1989) na educação escolar indígena junto aos professores indígenas Guarani das Aldeias de Boa Esperança e Três Palmeiras (2009-2010) e professores indígenas Tupinikim da Aldeia de Comboios (2011-2013) na perspectiva de um diálogo intercultural. Contribuem nos processos investigativos para produção, sistematização e análise de dados a realização de observações, entrevistas semiestruturadas, registros no caderno de campo, fotografias, gravações em áudio e em vídeo e análise documental sobre a educação escolar indígena de Aracruz. (ANDRÉ, 2007; GIL, 1999; 2004). Os resultados deste trabalho levantam questões relativas a duas realidades de educação escolar nas comunidades indígenas pesquisadas que se constituem em aspectos de sobrevivência e desencadeia formas para interagir e reagir em defesa de sua identidade e dignidade. Nesse sentido, a escola é um local de vivências e de encontro, vista e sentida pelas lideranças e pela comunidade como uma possibilidade real para desenvolver um elo entre as formas tradicionais de vida e as formas contemporâneas. O desafio de garantir uma escola nestes termos significa concretizar a proposta de um projeto de educação escolar para os povos indígenas, constituído por especificidades de como trabalhar a terra, pelo reconhecimento de suas tradições, das línguas e da memória coletiva. Distante de apresentar respostas conclusivas propõe uma educação escolar, coletiva e participativa, que critica e dialoga com todos os envolvidos no processo educativo.
