INTRODUCTION: The aim of this study was to assess the epidemiological characteristics of Trypanosoma cruzi-infected mothers and the live birth conditions of neonates.METHODS:A serological survey with IgG-specific tests was conducted using dried blood samples from newborn infants in the State of Minas Gerais. T. cruzi infection was confirmed in mothers through positive serology in two different tests, and infected mothers were required to have their infants serologically tested after the age of 6 months. The birth conditions of the neonates were obtained from the System of Information on Live Births database.RESULTS:The study included 407 children born to T. cruzi-infected mothers and 407 children born to uninfected mothers. The average age of seropositive mothers was 32 years (CI95% 31.3-32.6), which was greater than the average age of seronegative mothers - 25 years (CI95% 24.8-25.2). The mothers' level of education was higher among uninfected mothers (41% had 8 or more years of education, versus 22% between the infected mothers). Vaginal delivery was more frequent among infected mothers. There was no evidence of inter-group differences with respect to the child's sex, gestational age, birth weight or Appearance, pulse, grimace, activity and respiration (APGAR) scores at 1 and 5 minutes.Conclusions:The level of education and the greater number of previous pregnancies and cases of vaginal delivery reflect the lower socioeconomical conditions of the infected mothers. In the absence of vertical transmission, neonates had similar health status irrespective of the infection status of their mothers.

An analysis on the bigger influencers in terms of design-thinking and new product development, as Tim Brown or Paul Trott, lead to a better understanding on how There App should evolve, keeping in mind the customer desires and technical features.

Abstract: INTRODUCTION: To characterize Trypanosoma cruzi (TcI) isolated from a Panstrongylus megistus specimen found in one of the biggest metropolitan areas of Latin America, the relationship between the TcI group of T. cruzi and the transmission cycle in the urban environment was studied. METHODS: The T. cruzi strain, Pm, was isolated in a culture medium from the evolutionary forms present in the hindgut of a live male specimen of P. megistus found in the Jabaquara subway in São Paulo City. The sample from the triatomine showed trypomastigote forms of Trypanosomatidae, which were inoculated in the peritoneum of Balb/c mice. The sample was then inoculated in Liver Infusion Tryptose medium and J774 cells for the molecular identification and characterization of the parasite. The Pm strain of T. cruzi was identified by isolation in axenic culture medium, and based on the morphology, cell infection, growth kinetics, and molecular characterization. RESULTS: After isolation, the protozoan was identified as T. cruzi. No parasites were detected in the peripheral blood of the animal, which can be a characteristic inherent to the strain of T. cruzi that was isolated. Cell invasion assays were performed in triplicate in the J774 cell line to confirm the invasive ability of the Pm strain and revealed amastigote forms of the parasite within macrophages. CONCLUSIONS: Our biological and molecular characterizations helped understand parasite-host interactions and their evolutionary history in context of the associations between vectors, ecotopes, hosts, and groups of the parasite.

PURPOSE: In previous papers, we described a new experimental model of congenital diaphragmatic hernia in rabbits, and we also reported noninvasive therapeutic strategies for prevention of the functional and structural immaturity of the lungs associated with this defect. In addition to lung hypoplasia, pulmonary hypertension, biochemical, and structural immaturity of the lungs, the hemodynamics of infants and animals with congenital diaphragmatic hernia are markedly altered. Hence, cardiac hypoplasia has been implicated as a possible cause of death in patients with congenital diaphragmatic hernia, and it is hypothesized to be a probable consequence of fetal mediastinal compression by the herniated viscera. Cardiac hypoplasia has also been reported in lamb and rat models of congenital diaphragmatic hernia. The purpose of the present experiment was to verify the occurrence of heart hypoplasia in our new model of surgically produced congenital diaphragmatic hernia in fetal rabbits. METHODS: Twelve pregnant New Zealand rabbits underwent surgery on gestational day 24 or 25 (normal full gestational time - 31 to 32 days) to create left-sided diaphragmatic hernias in 1 or 2 fetuses per each doe. On gestational day 30, all does again underwent surgery, and the delivered fetuses were weighed and divided into 2 groups: control (non-surgically treated fetuses) (n = 12) and congenital diaphragmatic hernia (n = 9). The hearts were collected, weighed, and submitted for histologic and histomorphometric studies. RESULTS: During necropsy, it was noted that in all congenital diaphragmatic hernia fetuses, the left lobe of the liver herniated throughout the surgically created defect and occupied the left side of the thorax, with the deviation of the heart to the right side, compressing the left lung; consequently, this lung was smaller than the right one. The body weights of the animals were not altered by congenital diaphragmatic hernia, but heart weights were decreased in comparison to control fetuses. The histomorphometric analysis demonstrated that congenital diaphragmatic hernia promoted a significant decrease in the ventricular wall thickness and an increase in the interventricular septum thickness. CONCLUSION: Heart hypoplasia occurs in a rabbit experimental model of congenital diaphragmatic hernia. This model may be utilized for investigations in therapeutic strategies that aim towards the prevention or the treatment of heart hypoplasia caused by congenital diaphragmatic hernia.

When sports fans attend live sports events, they usually engage in social experiences with friends, family members and other fans at the venue sharing the same affiliation. However, fans watching the same event through a live television broadcast end up not feeling so emotionally connected with the athletes and other fans as they would if they were watching it live, together with thousands of other fans. With this in mind, we seek to create mobile applications that deliver engaging social experiences involving remote fans watching live broadcasted sports events. Taking into account the growing use of mobile devices when watching TV broadcasts, these mobile applications explore the second screen concept, which allows users to interact with content that complements the TV broadcast. Within this context, we present a set of second screen application prototypes developed to test our concepts, the corresponding user studies and results, as well as suggestions on how to apply the prototypes’ concepts not only in different sports, but also during TV shows and electronic sports. Finally, we also present the challenges we faced and the guidelines we followed during the development and evaluation phases, which may give a considerable contribution to the development of future second screen applications for live broadcasted events.

Neurological disorders are a major concern in modern societies, with increasing prevalence mainly related with the higher life expectancy. Most of the current available therapeutic options can only control and ameliorate the patients’ symptoms, often be-coming refractory over time. Therapeutic breakthroughs and advances have been hampered by the lack of accurate central nervous system (CNS) models. The develop-ment of these models allows the study of the disease onset/progression mechanisms and the preclinical evaluation of novel therapeutics. This has traditionally relied on genetically engineered animal models that often diverge considerably from the human phenotype (developmentally, anatomically and physiologically) and 2D in vitro cell models, which fail to recapitulate the characteristics of the target tissue (cell-cell and cell-matrix interactions, cell polarity). The in vitro recapitulation of CNS phenotypic and functional features requires the implementation of advanced culture strategies that enable to mimic the in vivo struc-tural and molecular complexity. Models based on differentiation of human neural stem cells (hNSC) in 3D cultures have great potential as complementary tools in preclinical research, bridging the gap between human clinical studies and animal models. This thesis aimed at the development of novel human 3D in vitro CNS models by integrat-ing agitation-based culture systems and a wide array of characterization tools. Neural differentiation of hNSC as 3D neurospheres was explored in Chapter 2. Here, it was demonstrated that human midbrain-derived neural progenitor cells from fetal origin (hmNPC) can generate complex tissue-like structures containing functional dopaminergic neurons, as well as astrocytes and oligodendrocytes. Chapter 3 focused on the development of cellular characterization assays for cell aggregates based on light-sheet fluorescence imaging systems, which resulted in increased spatial resolu-tion both for fixed samples or live imaging. The applicability of the developed human 3D cell model for preclinical research was explored in Chapter 4, evaluating the poten-tial of a viral vector candidate for gene therapy. The efficacy and safety of helper-dependent CAV-2 (hd-CAV-2) for gene delivery in human neurons was evaluated, demonstrating increased neuronal tropism, efficient transgene expression and minimal toxicity. The potential of human 3D in vitro CNS models to mimic brain functions was further addressed in Chapter 5. Exploring the use of 13C-labeled substrates and Nucle-ar Magnetic Resonance (NMR) spectroscopy tools, neural metabolic signatures were evaluated showing lineage-specific metabolic specialization and establishment of neu-ron-astrocytic shuttles upon differentiation. Chapter 6 focused on transferring the knowledge and strategies described in the previous chapters for the implementation of a scalable and robust process for the 3D differentiation of hNSC derived from human induced pluripotent stem cells (hiPSC). Here, software-controlled perfusion stirred-tank bioreactors were used as technological system to sustain cell aggregation and dif-ferentiation. The work developed in this thesis provides practical and versatile new in vitro ap-proaches to model the human brain. Furthermore, the culture strategies described herein can be further extended to other sources of neural phenotypes, including pa-tient-derived hiPSC. The combination of this 3D culture strategy with the implemented characterization methods represents a powerful complementary tool applicable in the drug discovery, toxicology and disease modeling.

Durante 13 meses (de janeiro de 1986 a janeiro de 1987), quatro trilhas, de aproximadamente 2,5 km de comprimento cada uma, cortando diferentes ambientes de mata de terra firme, foram seguidas diariamente por um mínimo de dois pesquisadores para observar animais forrageando no chão o em diferentes estratos da floresta. O local de estudo foi em porção de floresta protegida em Tucuruí, estado do Pará. Foram investigadas por observação contínua no campo as tendências sazonais de utilização de ítens de alimentos preferidos demonstrados por quelôneos: os jabutis Geochelone carbonaria, G. denticulata e a aperema Platemys platicephala; por roedores: o quatipuru Sciurus gilvigularis e a cutia Dasyprocta aguti; por cervídeos: o veado-mateiro Mazama americana e o veado fuboca Mazama guazoubira; e por primatas: o macaco-prego Cebus apella, o guariba Alouatta belzebul, o cuxiú Chiropotes satanas , o macaco-mão-de-ouro Saimiri sciureus, e o sagui preto Saguinus midas. Todos esses animais foram estudados através de observação contínua nessas trilhas de transectos, estabelecidas na mata de terra firme. As observações foram feitas pela manhã entre 06:00 e 10:00 horas e à tarde, entre 15:00 e 19 horas. O estudo demonstrou que as dietas dessas espécies animais de habitat de terra firme depende da disponibilidade de alimento na floresta na estação do ano. A importância desses ítens de alimento é demonstrada pela mudança sazonal da atividade de forrageamento, dependendo do ítem alimentar em oferta. Os jabtutis (Geochelone carbonaria e G. denticulata) alimentam-se basicamente de frutos disponíveis entre setembro e janeiro, sendo esses mesmo frutos compatilhados por cutia (Dasyprocta aguti), enquanto o macaco-prego (Cebus apella) expande sua dieta baseada em frutas, em dois picos, um entre janeiro e março, com um máximo de atividade em fevereiro, e outro pico entre julho e dezembro. Essas mesmas frutas são também compartilhadas por Chiropotes satanas. Embora o guariba (Alouatta belzebul) seja observado comendo grande porcentagem de folhas, nas estações de produção de frutos, porém, durante os dois picos anuais, (fevereiro e dezembro) tornam-se oportunisticamente frugívoros.

Foi realizado um trabalho para determinar o aproveitamento alimentar da farinha de carne e ossos (FCO), farinha de vísceras de aves (FVA) e farinha de peixe (FP) em tartaruga-da-amazônia, por meio dos coeficientes de digestibilidade aparente (CDA) da matéria seca (MS), proteína bruta (PB), extrato etéreo (EE) e energia bruta (EB). Os animais experimentais foram 136 filhotes provenientes do Projeto Quelônios da Amazônia, no estado do Mato Grosso, mantidos em caixas com renovação de água e temperatura média de 29ºC. Os CDA foram determinados com dietas contendo 0,1% do marcador óxido de crômio III (Cr2O3). Os CDA da MS, PB, EE e EB foram, respectivamente, de 79,10; 87,61; 93,83 e 79,61% para FCO; 92,45; 94,89; 96,55 e 92,71% para FV e 93,53; 95,13; 94,05 e 93,18% para FP. Os melhores coeficientes foram obtidos com a farinha de peixe e a farinha de vísceras de aves.

The present work explores the best conditions for the enzymatic synthesis of poly (ethylene glutarate) for the first time. The start-up materials are the liquids; diethyl glutarate and ethylene glycol diacetate, without the need of addition of extra solvent. The reactions are catalyzed by lipase B from Candida antarctica immobilized on glycidyl methacrylate-ter-divinylbenzene-ter-ethylene glycol dimethacrylate at 40 °C during 18 h in water bath with mechanical stirring or 1 h in ultrasonic bath followed by 6 h in vacuum in both the cases for evaporation of ethyl acetate. The application of ultrasound significantly intensified the polyesterification reaction with reduction of the processing time from 24 to 7 h. The same degree of polymerization was obtained for the same enzyme loading in less time of reaction when using the ultrasound treatment. The degree of polymerization for long-term polyesterification was improved approximately 8-fold due to the presence of sonication during the reaction. The highest degree of polymerization achieved was 31, with a monomer conversion of 96.77%. The ultrasound treatment demonstrated to be an effective green approach to intensify the polyesterification reaction with enhanced initial kinetics and high degree of polymerization.

El presente proyecto, continuación de otro donde se desarrolló un Manual de Buenas Prácticas Pecuarias (BPP) relativas al bienestar animal, tiene como objetivo implementar estas BPP en establecimientos productores de carne vacuna, poniendo énfasis en la capacitación de los productores ganaderos para que logren adoptar la cultura del trabajo que proponen los sistemas de gestión de calidad. Para la realización del proyecto planteado se dispone de dos establecimientos productores de carne vacuna ubicados en la Provincia de Córdoba cuyo tamaño y sistema productivo responden a la media de productores ganaderos. También se propone la implementación de estas BPP en los campos de la Universidad Católica de Córdoba para que estos sirvan de modelo a otros establecimientos del medio, permitiendo no sólo la mejora en los resultados productivos de los mismos sino también la formación y capacitación de profesionales, alumnos y productores en temas relacionados al bienestar animal y las BPP. Las tareas a desarrollar serán: 1) Caracterización de los establecimientos en donde se implementarán las BPP. 2) Capacitación de los productores y técnicos, pertenecientes a los establecimientos seleccionados, en Sistemas de Gestión de Calidad (BPP) y en los contenidos del Manual desarrollado anteriormente. 3) Determinación de actividades en cada uno de los establecimientos (definir responsables, realizar un cronograma, realizar auditorías, priorizar “no conformidades”, proponer y poner en marcha acciones de mejora, monitorear la performance de la implementación del sistema y evaluar los indicadores). Una vez lograda la implementación de la primera etapa que recomienda el Manual desarrollado se agregarán las siguientes actividades para la capacitación y difusión a terceros: 4) Convocatoria de otros productores para su introducción en los Sistemas de Gestión de Calidad (BPP relativas al bienestar animal). 5) Realización de actividades grupales de capacitación (charlas, grupos de discusión y talleres) en los establecimientos usados como referentes. 6) Análisis de la performance del plan y de la satisfacción de las personas involucradas. 7) Propuesta de implementación de acciones correctivas a la capacitación de terceros. 8) Evaluación general del plan. Con la concreción del proyecto se espera: 1- Lograr que los establecimientos ganaderos implementen el sistema de calidad (BPP) propuesto para que puedan ser utilizados como modelo y brindar capacitación para éste y sucesivos trabajos de este tipo. 2- Lograr la incorporación de los conceptos que involucran los Sistemas de Gestión de Calidad y la adopción de la cultura de trabajo que dichos sistemas proponen, en los establecimientos que participen de la capacitación. 3- Constituir un grupo de trabajo interdisciplinario, capacitado y con experiencia en Sistemas de Gestión de Calidad para hacer un trabajo multiplicador. 4- Colaborar con la sustentabilidad de los campos ganaderos a través de los sistemas de gestión de calidad que tienen como primordiales objetivos la calidad e inocuidad de los alimentos, la salud, el bienestar de las personas, el cuidado del ambiente y el bienestar animal.

El objetivo del presente proyecto es evaluar las distintas causales de estrés en el bovino de abasto y, por ende, desarrollar un Manual ilustrado de Buenas Prácticas Pecuarias (BPP), referente al bienestar animal, para los establecimientos de producción bovina de carne de base pastoril que resulte de fácil transferencia y aplicación en nuestro sector productivo ganadero con la finalidad de minimizar, a través de la implementación del mismo, el impacto negativo que el inadecuado manejo de los animales en el campo, durante el transporte y en los corrales del frigorífico, tiene sobre los parámetros productivos y la calidad de la res y de la carne. Para ello se proponen realizar las siguientes actividades: 1. Determinar las principales causas de estrés en bovinos para faena que causan pérdidas cuantitativas y cualitativas en la res y la carne (carnes de corte oscuro o DFD, golpes o manchas verdes). 2. Visitar establecimientos productores de carne vacuna para la recopilación de información y de fotografías necesarias para la confección del Manual ilustrado de Buenas Prácticas Pecuarias (BPP) referido al bienestar del animal. 3. Diagramación y edición del Manual.

Este proyecto se ubica dentro del programa SUPPRAD que trabaja en red a lo largo del país en áreas desfavorecidas y con pequeños productores. En el contexto del uso sustentable de los recursos naturales y de promoción del desarrollo, el mejoramiento genético es una herramienta fundamental porque aporta beneficios a ambos componentes. Por eso se ha tomado para este proyecto como objetivos generales estudiar las estrategias de implementación de programas de mejoramiento genético de fibra en rumiantes menores y Camélidos en las áreas desfavorecidas donde actúa el programa. Se pretende aquí identificar genotipos de mayor eficiencia productiva en fibra en rumiantes menores y Camélidos y aplicar los conocimientos adquiridos en programas de mejoramiento productivo que tiendan a mejorar el ingreso del pequeño productor para finalmente contribuir al desarrollo de producciones sustentables en distintas regiones del país. Esto se va a desarrollar a través de estudios de estructura poblacional a nivel de pequeños productores de Llamas de la provincia de Jujuy, Córdoba, La Pampa y Río Negro; productores trashumantes de caprinos criollos con aptitud de fibra del área noroeste de Neuquén; pequeños productores de la pampa de Olaen (Córdoba). Por otra parte y a partir de los planteles experimentales establecidos en los campos de productores se organiza la toma de registros de las distintas variables a describir genéticamente: peso de vellón, peso de parche de vellón (predictor del peso de vellón), diámetro medio de las fibras, coeficientes de variación, frecuencia de rizos, rinde al descerdado (vellones mixtos), frecuencias y diámetro de tipos de fibra y tipos de folículos en cortes de piel. Por otra parte se diseñarán objetivos de mejoramiento para las distintas especies a partir de la información genética generada en este proyecto y la información económica recopilada a partir de informaciones de mercado y de los resultados de ensayos de textiles específicos. Como resultado final se pretende elaborar estrategias de abordajes de los mejoramientos productivos de las distintas áreas en estudio y de los distintos tipos de productores involucrados.

La explotación comercial de la llama facilita la aparición de patologías bacterianas y parasitarias que producen importantes pérdidas económicas. Su prevención y el tratamiento exigen establecer regímenes terapéuticos eficaces y seguros que eviten la aparición de resistencias. La ausencia de especialidades farmacéuticas propias para esta especie y la falta de información científica sobre el comportamiento de los fármacos de uso habitual en llamas, obligan al veterinario a establecer pautas terapéuticas “fuera de prospecto”, a partir de su experiencia en otras especies. Esto conlleva el riesgo de alcanzar o bien niveles tóxicos o bien sub-terapéuticos, estos últimos facilitan la aparición de resistencias bacterianas transmisibles a otras poblaciones. Pretendemos describir el comportamiento cinético de dos antimicrobianos de amplio espectro y uso exclusivo en veterinaria (enrofloxacina y marbofloxacina). Los datos obtenidos nos permitirán evaluar la seguridad y eficacia de diferentes regímenes terapéuticos que puedan dar respuesta a las necesidades clínicas en esta especie. El proyecto que abordamos se encuentra en una línea de aprovechamiento de los recursos naturales dentro de un espíritu de compromiso social, desarrollo sostenible, bienestar animal y salud pública.
