Bioinspired Reductionistic Peptide Engineering for Exceptional Mechanical Properties

A simple solution-processing and self-assembly approach that exploits the synergistic interactions between multiple hydrogen bonded networks and aromatic interactions was utilized to synthesize molecular crystals of cyclic dipeptides (CDPs), whose molecular weights (similar to 0.2 kDa) are nearly three orders of magnitude smaller than that of natural structural proteins (50-300 kDa). Mechanical properties of these materials, measured using the nanoindentation technique, indicate that the stiffness and strength are comparable and sometimes better than those of natural fibres. The measured mechanical responses were rationalized by recourse to the crystallographic structural analysis and intermolecular interactions in the self-assembled single crystals. With this work we highlight the significance of developing small molecule based bioinspired design strategies to emulate biomechanical properties. A particular advantage of the successfully demonstrated reductionistic strategy of the present work is its amenability for realistic industrial scale manufacturing of designer biomaterials with desired mechanical properties.

Hetero-atom Doped Carbon Nanotubes for Dye Degradation and Oxygen Reduction Reaction

We report the synthesis of nitrogen doped vertically aligned multi-walled (MWNCNTs) carbon nanotubes by pyrolysis and its catalytic performance for degradation of methylene blue (MB) dye & oxygen reduction reaction (ORR). The degradation of MB was monitored spectrophotometrically with time. Kinetic studies show the degradation of MB follows a first order kinetic with rate constant k=0.0178 min(-1). The present rate constant is better than that reported for various supported/non-supported semiconducting nanomaterials. Further ORR performance in alkaline media makes MWNCNTs a promising cost-effective, fuel crossover tolerance, metal-free, eco-friendly cathode catalyst for direct alcohol fuel cell.

Enhanced Metastatic Potential in a 3D Tissue Scaffold toward a Comprehensive in Vitro Model for Breast Cancer Metastasis

Metastasis is clinically the most challenging and lethal aspect of breast cancer. While animal-based xenograft models are expensive and time-consuming, conventional two-dimensional (2D) cell culture systems fail to mimic in vivo signaling. In this study we have developed a three-dimensional (3D) scaffold system that better mimics the topography and mechanical properties of the breast tumor, thus recreating the tumor microenvironment in vitro to study breast cancer metastasis. Porous poly(e-caprolactone) (PCL) scaffolds of modulus 7.0 +/- 0.5 kPa, comparable to that of breast tumor tissue were fabricated, on which MDA-MB-231 cells proliferated forming tumoroids. A comparative gene expression analysis revealed that cells growing in the scaffolds expressed increased levels of genes implicated in the three major events of metastasis, viz., initiation, progression, and the site-specific colonization compared to cells grown in conventional 2D tissue culture polystyrene (TCPS) dishes. The cells cultured in scaffolds showed increased invasiveness and sphere efficiency in vitro and increased lung metastasis in vivo. A global gene expression analysis revealed a significant increase in the expression of genes involved in cell cell and cell matrix interactions and tissue remodeling, cancer inflammation, and the PI3K/Akt, Wnt, NF-kappaB, and HIFI signaling pathways all of which are implicated in metastasis. Thus, culturing breast cancer cells in 3D scaffolds that mimic the in vivo tumor-like microenvironment enhances their metastatic potential. This system could serve as a comprehensive in vitro model to investigate the manifold mechanisms of breast cancer metastasis.

Estudio del efecto de la regulación de la iluminación en pollos de engorde de la linea Hubbard Peterson en la granja avícola Belén, El Viejo, Chinandega

Con el fin de determinar el programa de iluminación más adecuado que permita maximizar los rendimientos productivos y disminuir los costos de producción en pollos de engorde, se realizó el presente ensayo en condiciones de producción comercial, bajo un sistema tradicional. El total de aves utilizadas fueron 196, pertenecientes a la línea Hubbard Peterson, de un día de nacidas, mixtas. Estas fueron alojadas en una galera (rancho de palma) con dimensiones de 29 m de largo * 1O m de ancho* 5 m de alto, y capacidad total de alojamiento de 2900 aves; por un período de 42 días. Para efectos experimentales las aves fueron agrupadas aleatoriamente en dos cubículos, a razón de 98/ grupo, utilizando una densidad de alojamiento de 9 8 aves 1m2. Los tratamientos evaluados fueron: T1= 23 hrs luz + 1 hr de oscuridad y T2 = 23 hrs luz + 1 hr de oscuridad hasta las dos semanas, en adelante hasta el sacrificio sólo luz natural. Las variables de respuesta fueron: Peso Final (PF) Consumo de Alimento (CAl) , Ganancia Media Diaria (GMD) e índice de Conversión Alimenticia (ICA), Margen Bruto (MB) y Relación Beneficio Costo (8\C). Para el análisis estadístico se utilizó un DCA con arreglo bifactoríal y prueba de separación de medías por Tukey, mediante el paquete estadístico SAS, 1991. Con base en los resultados de los análisis, no se encontraron diferencias significativas entre los tratamientos para las variables GMD e ICA; no así para CAL, y PF. Los valores promedios semanales a lo largo del período (6 semanas), pera las variables evaluadas, fueron los siguientes: Para T1, CAl= 87.37g, GM0=57.17g, ICA= 1.66 y PF= 1.895kg: para T2, CAL= 83.68g, GMD= 54.81g, ICA= 1.58 y PF= 1.833kg. En el análisis financiero, el tratamiento con un periodo de 12 horas de iluminación, se obtuvo una reducción del 66.66% en los costos de iluminación, y una superioridad del 19% en el margen bruto y 6.15% en la relación beneficio costo en relación al periodo de 23 horas luz.

Agua de mar como promotor de crecimiento en pollos de engorde arbor acres de cero a seis semanas, La Unión, Pasaquina, El Salvador

La industria avícola productora de carne de pollo, ha requerido la modernización tecnológica para mejorar los parámetros productivos recurriendo entre otras medidas al uso de estimulantes del crecimiento reforzados con antibióticos (Sánchez R.1996). Por esta razón, el productor avícola cada día se esfuerza por mejorar la producción a menores costos y evitar perdidas económicas ocasionadas por surgimiento de enfermedades en las aves, incluyendo por tanto en la ración alimenticia o agua de bebida, soluciones alternativas como el uso de Exclusión competitiva para establecer microflora intestinal mas resistente, como el uso de promotores de crecimientos químicos. (Nurmi y Rantala 1973). Es así que con este trabajo de investigación: “Uso del Agua de Mar como Promotor de crecimiento Natural (PN) en pollos de engorde en la granja avícola de La Unión – Pasaquina El Salvador”. Se establecieron tratamientos diferentes con promotores de crecimiento y uno de ellos fue el Agua de Mar implementado como Promotor Natural (PN), el cual fue aplicado en el agua de bebida. El estudio permitió determinar una dosis específica de 250 ml, lo que conllevó a obtener resultados como el consumo de alimento en los pollos de engorde con (PN)fue de 461.56kg, y de 460.95kg para (PQ), conversión alimenticia menor en el (PN) con un promedio de 2.51 kg mientras que en el PQ (súper promotor) demostró un promedio de 2.69kg, alcanzando un peso el (PN) de 2.07kg. Siendo menor el peso alcanzado (PQ) con 1.83kg. así como la condición organoléptica de la carne con PN resulto con categoría de muy buena (MB), demostrando con este estudio el efecto positivo del Agua de Mar como promotor de crecimiento, Además, por primera vez se utiliza el Agua de Mar como promotor de crecimiento en el desarrollo de pollos de engorde.
