O objetivo do presente trabalho é a investigação e o desenvolvimento de estratégias de otimização contínua e discreta para problemas de Fluxo de Potência Ótimo (FPO), onde existe a necessidade de se considerar as variáveis de controle associadas aos taps de transformadores em-fase e chaveamentos de bancos de capacitores e reatores shunt como variáveis discretas e existe a necessidade da limitação, e/ou até mesmo a minimização do número de ações de controle. Neste trabalho, o problema de FPO será abordado por meio de três estratégias. Na primeira proposta, o problema de FPO é modelado como um problema de Programação Não Linear com Variáveis Contínuas e Discretas (PNLCD) para a minimização de perdas ativas na transmissão; são propostas três abordagens utilizando funções de discretização para o tratamento das variáveis discretas. Na segunda proposta, considera-se que o problema de FPO, com os taps de transformadores discretos e bancos de capacitores e reatores shunts fixos, possui uma limitação no número de ações de controles; variáveis binárias associadas ao número de ações de controles são tratadas por uma função quadrática. Na terceira proposta, o problema de FPO é modelado como um problema de Otimização Multiobjetivo. O método da soma ponderada e o método ε-restrito são utilizados para modificar os problemas multiobjetivos propostos em problemas mono-objetivos. As variáveis binárias associadas às ações de controles são tratadas por duas funções, uma sigmoidal e uma polinomial. Para verificar a eficácia e a robustez dos modelos e algoritmos desenvolvidos serão realizados testes com os sistemas elétricos IEEE de 14, 30, 57, 118 e 300 barras. Todos os algoritmos e modelos foram implementados em General Algebraic Modeling System (GAMS) e os solvers CONOPT, IPOPT, KNITRO e DICOPT foram utilizados na resolução dos problemas. Os resultados obtidos confirmam que as estratégias de discretização são eficientes e as propostas de modelagem para variáveis binárias permitem encontrar soluções factíveis para os problemas envolvendo as ações de controles enquanto os solvers DICOPT e KNITRO utilizados para modelar variáveis binárias não encontram soluções.

Com o objetivo de aumentar o lucro de plantas químicas, a Otimização em Tempo Real (RTO) é uma ferramenta que busca determinar as condições ótimas operacionais do processo em estado estacionário, respeitando as restrições operacionais estabelecidas. Neste trabalho foi realizada a implementação prática de um ciclo RTO em um processo de destilação por recompressão de vapor (VRD), propileno-propano, da Refinaria de Paulínia (Petrobras S.A.), a partir de dados históricos da planta. Foram consideradas as principais etapas de um ciclo clássico de RTO: identificação de estado estacionário, reconciliação de dados, estimação de parâmetros e otimização econômica. Essa unidade foi modelada, simulada e otimizada em EMSO (Environment for Modeling, Simulation and Optimization), um simulador de processos orientado a equações desenvolvido no Brasil. Foram analisados e comparados dois métodos de identificação de estado estacionário, um baseado no teste estatístico F e outro baseado em wavelets. Ambos os métodos tiveram resultados semelhantes e mostraram-se capazes de identificar os estados estacionários de forma satisfatória, embora seja necessário o ajuste de parâmetros na sua implementação. Foram identificados alguns pontos estacionários para serem submetidos ao ciclo RTO e foi possível verificar a importância de partir de um estado estacionário para a continuidade do ciclo, já que essa é uma premissa do método. A partir dos pontos analisados, os resultados deste estudo mostram que o RTO é capaz de aumentar o ganho econômico entre 2,5-24%, dependendo das condições iniciais consideradas, o que pode representar ganhos de até 18 milhões de dólares por ano. Além disso, para essa unidade, verificou-se que o compressor é um equipamento limitante no aumento de ganho econômico do processo.

The use of electric pulses to deliver therapeutic molecules to tissues and organs in vivo is a rapidly growing field of research. Electrotransfer can be used to deliver a wide range of potentially therapeutic agents, including drugs, proteins, oligonucleotides, RNA and DNA. Optimization of this approach depends upon a number of parameters such as target organ accessibility, cell turnover, microelectrode design, electric pulsing protocols and the physiological response to the therapeutic agent. Many organs have been successfully transfected by electroporation, including skin, liver, skeletal and cardiac muscle, male and female germ cells, artery, gut, kidney, retinal ganglion cells, cornea, spinal cord, joint synovium and brain. Electrotransfer technology is relevant in a variety of research and clinical settings including cancer therapy, modulation of pathogenic immune reactions, delivery of therapeutic proteins and drugs, and the identification of drug targets by the modulation of normal gene expression. This, together with the capacity to deliver very large DNA constructs, greatly expands the research and clinical applications of in vivo DNA electrotransfer.

Diante da necessidade de inserção de políticas de desenvolvimento sustentável dentro do contexto das organizações, as empresas investem em recursos esperando que a adoção dessas práticas possa trazer benefícios econômicos e estratégicos, refletidos na imagem da organização e na sua valorização no mercado. Neste cenário, Índices de Sustentabilidade foram criados em escala global, e estes índices em geral, avaliam várias dimensões das relações da organização com a sociedade, meio ambiente e com os provedores de capital para a empresa. No Brasil, especificamente no ano de 2005, foi criado o Índice de Sustentabilidade Empresarial (ISE) para reunir as ações de empresas que incorporam em suas diretrizes de negócio práticas de responsabilidade social e sustentabilidade empresarial. Com isso, a presente dissertação tem por objetivo verificar qual o impacto no valor das ações de uma amostra de 43 empresas perante a entrada ou saída das mesmas das carteiras do ISE. A metodologia de estudo de evento buscou identificar os retornos anormais após as divulgações das carteiras do ISE, de 2005 a 2009, através do modelo de retorno ajustado ao risco e ao mercado. Os dados financeiros, antes e depois do evento, foram extraídos da base de dados da Bolsa de Valores de São Paulo. Os resultados alcançados indicaram relação positiva entre a participação de algumas empresas nas carteiras do ISE e o desempenho financeiro das suas ações, além de mostrar que os investimentos necessários em desenvolvimento sustentável não acarretam perdas, visto a predominância de retornos anormais positivos em todas as situações analisadas neste estudo.

Background The production of high yields of recombinant proteins is an enduring bottleneck in the post-genomic sciences that has yet to be addressed in a truly rational manner. Typically eukaryotic protein production experiments have relied on varying expression construct cassettes such as promoters and tags, or culture process parameters such as pH, temperature and aeration to enhance yields. These approaches require repeated rounds of trial-and-error optimization and cannot provide a mechanistic insight into the biology of recombinant protein production. We published an early transcriptome analysis that identified genes implicated in successful membrane protein production experiments in yeast. While there has been a subsequent explosion in such analyses in a range of production organisms, no one has yet exploited the genes identified. The aim of this study was to use the results of our previous comparative transcriptome analysis to engineer improved yeast strains and thereby gain an understanding of the mechanisms involved in high-yielding protein production hosts. Results We show that tuning BMS1 transcript levels in a doxycycline-dependent manner resulted in optimized yields of functional membrane and soluble protein targets. Online flow microcalorimetry demonstrated that there had been a substantial metabolic change to cells cultured under high-yielding conditions, and in particular that high yielding cells were more metabolically efficient. Polysome profiling showed that the key molecular event contributing to this metabolically efficient, high-yielding phenotype is a perturbation of the ratio of 60S to 40S ribosomal subunits from approximately 1:1 to 2:1, and correspondingly of 25S:18S ratios from 2:1 to 3:1. This result is consistent with the role of the gene product of BMS1 in ribosome biogenesis. Conclusion This work demonstrates the power of a rational approach to recombinant protein production by using the results of transcriptome analysis to engineer improved strains, thereby revealing the underlying biological events involved.

Understanding the structures and functions of membrane proteins is an active area of research within bioscience. Membrane proteins are key players in essential cellular processes such as the uptake of nutrients, the export of waste products, and the way in which cells communicate with their environment. It is therefore not surprising that membrane proteins are targeted by over half of all prescription drugs. Since most membrane proteins are not abundant in their native membranes, it is necessary to produce them in recombinant host cells to enable further structural and functional studies. Unfortunately, achieving the required yields of functional recombinant membrane proteins is still a bottleneck in contemporary bioscience. This has highlighted the need for defined and rational optimization strategies based upon experimental observation rather than relying on trial and error. We have published a transcriptome and subsequent genetic analysis that has identified genes implicated in high-yielding yeast cells. These results have highlighted a role for alterations to a cell's protein synthetic capacity in the production of high yields of recombinant membrane protein: paradoxically, reduced protein synthesis favors higher yields. These results highlight a potential bottleneck at the protein folding or translocation stage of protein production.

The slow down in the drug discovery pipeline is, in part, owing to a lack of structural and functional information available for new drug targets. Membrane proteins, the targets of well over 50% of marketed pharmaceuticals, present a particular challenge. As they are not naturally abundant, they must be produced recombinantly for the structural biology that is a prerequisite to structure-based drug design. Unfortunately, however, obtaining high yields of functional, recombinant membrane proteins remains a major bottleneck in contemporary bioscience. While repeated rounds of trial-and-error optimization have not (and cannot) reveal mechanistic details of the biology of recombinant protein production, examination of the host response has provided new insights. To this end, we published an early transcriptome analysis that identified genes implicated in high-yielding yeast cell factories, which has enabled the engineering of improved production strains. These advances offer hope that the bottleneck of membrane protein production can be relieved rationally.

Projects exposed to an uncertain environment must be adapted to deal with the effective integration of various planning elements and the optimization of project parameters. Time, cost, and quality are the prime objectives of a project that need to be optimized to fulfill the owner's goal. In an uncertain environment, there exist many other conflicting objectives that may also need to be optimized. These objectives are characterized by varying degrees of conflict. Moreover, an uncertain environment also causes several changes in the project plan throughout its life, demanding that the project plan be totally flexible. Goal programming (GP), a multiple criteria decision making technique, offers a good solution for this project planning problem. There the planning problem is considered from the owner's perspective, which leads to classifying the project up to the activity level. GP is applied separately at each level, and the formulated models are integrated through information flow. The flexibility and adaptability of the models lies in the ease of updating the model parameters at the required level through changing priorities and/or constraints and transmitting the information to other levels. The hierarchical model automatically provides integration among various element of planning. The proposed methodology is applied in this paper to plan a petroleum pipeline construction project, and its effectiveness is demonstrated.

Energy consumption has been a key concern of data gathering in wireless sensor networks. Previous research works show that modulation scaling is an efficient technique to reduce energy consumption. However, such technique will also impact on both packet delivery latency and packet loss, therefore, may result in adverse effects on the qualities of applications. In this paper, we study the problem of modulation scaling and energy-optimization. A mathematical model is proposed to analyze the impact of modulation scaling on the overall energy consumption, end-to-end mean delivery latency and mean packet loss rate. A centralized optimal management mechanism is developed based on the model, which adaptively adjusts the modulation levels to minimize energy consumption while ensuring the QoS for data gathering. Experimental results show that the management mechanism saves significant energy in all the investigated scenarios. Some valuable results are also observed in the experiments. © 2004 IEEE.

Silvia Baeva - In the Ministry of Education and Science’s system it has been talked about optimization of the school network; this optimization can be carried out in different directions and be supported by laws. An important aspect in the optimization of the school network is to reduce costs and increase overall efficiency in each school. We will formulate this aspect as a problem of the multicriteria decisions making and by appropriate numbers of methods and criteria it can be transformed to the problem of unicriterion optimization. This problem is separated into two stages: 1-th stage – determining the minimum number of classes in a school under certain statutory provisions for the size of each of them; 2-th stage – the appointment of a minimum number of teachers and achievement of maximal effectiveness of teaching and acquiring of knowledge and skills by students according to certain statutory provisions for teachers’ annual norms of subjects and number of hours in a particular subject area and the salaries of teachers. The achievement of maximum overall efficiency is a priority in all schools.

It is a crucial task to evaluate the reliability of manufacturing process in product development process. Process reliability is a measurement of production ability of reconfigurable manufacturing system (RMS), which serves as an integrated performance indicator of the production process under specified technical constraints, including time, cost and quality. An integration framework of manufacturing process reliability evaluation is presented together with product development process. A mathematical model and algorithm based on universal generating function (UGF) is developed for calculating the reliability of manufacturing process with respect to task intensity and process capacity, which are both independent random variables. The rework strategies of RMS are analyzed under different task intensity based on process reliability is presented, and the optimization of rework strategies based on process reliability is discussed afterwards.
